Development and Validation of a Simultaneous Analytical Method for 5 Residual Pesticides in Agricultural Products using GC-MS/MS (GC-MS/MS를 이용한 농산물 중 잔류농약 5종 동시시험법 개발 및 검증)

  • The aim of this research was to develop a rapid and easy multi-residue method for determining dimethipin, omethoate, dimethipin, chlorfenvinphos and azinphos-methyl in agricultural products (hulled rice, potato, soybean, mandarin and green pepper). Samples were prepared using QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) and analyzed using gas chromatography-tandem mass spectrometry (GC-MS/MS). Residual pesticides were extracted with 1% acetic acid in acetonitrile followed by addition of anhydrous magnesium sulfate (MgSO4) and anhydrous sodium acetate. The extracts were cleaned up using MgSO4, primary secondary amine (PSA) and octadecyl (C18). The linearity of the calibration curves, which waas excellent by matrix-matched standards, ranged from 0.005 mg/kg to 0.3 mg/kg and yielded the coefficients of determination (R2) ≥ 0.9934 for all analytes. Average recoveries spiked at three levels (0.01, 0.1, 0.5 mg/kg) and were in the range of 74.2-119.3%, while standard deviation values were less than 14.6%, which is below the Codex guideline (CODEX CAC/GL 40).

A Study on the Development of Microorganism Test Strips (대장균군 검사용 간이 시험지 개발)

  • The objective of this study was to develop a paper strip which could determine E. coli qualitatively and quantitatively in water, wastewater, drinks, or food. This paper strip method was a simple and rapid test method that determine E. coli by visual identification. In this study, nutrient culture media were formulated and characterized for optimum conditions. Paper strips were then prepared by impregnating into the media and dried at $60^\circ$C. The test procedure is quite simple to use. The paper strip was dipped into a sample, and excess sample was removed. The strip was then incubated at $37^\circ$C for 16 to 20 hours and the number of colonies on the strip was counted. The color of the colony spots produced by microorganisms varied depending on the media formulation. Violet-red spots were produced by E. coli. The test method was simple, rapid and no special laboratory equipment was necessary for visual identification. Therefore, this test method is applicable to on-site tests such as field tests or home tests. The paper strip method was compared with the standard agar plate method and Japanese commercial product. The method of the economical preparation of test strips was studied for production on industrial scale.

Specimens and method for evaluating the moisturizing ability of lip makeup products (립메이크업 제품의 보습능 평가용 시편과 측정법)

  • In this study, the method of evaluation of moisturizing ability of cosmetics using specimens was studied as an alternative method of clinical tests. Cosmetic spreadability, surface drying, and moisture loss rate of four specimens (Japanese cake (Maru mochi), Garaetteok, wheat flour, and agar) were evaluated. Also, the water loss rate of the specimens and the transepidermal waterloss were analyzed for 10 kinds of lip make-up products (5 kinds of lipstick, 3 kinds of lip balm, 2 kinds of lip gloss). The moisture loss rate (moisture evaporation sensitivity) of the agar specimen was highest in the order of agar> Garaetteok> Japanese cake> wheat flour. Agar specimens are the most suitable in terms of spreadability, surface crack, and raw material supply. Containers for the preparation of agar specimens are suitable for plastic materials with low heat transfer, which can produce convex, smooth surface specimens. In the evaluation of moisturizing ability of lip makeup products, there was a strong correlation between the water loss rate and the transepidermal waterloss rate measured with agar specimens. These results show that the proposed method can be used as one of the useful alternative test methods.

The Study on the Analysis Method of Tetrodotoxin in Puffer Fish (복어 중 테트로도톡신 분석법에 관한 연구)

  • The current standard for testing tetrodotoxin (TTX) in foodstuffs is the mouse bioassay (MBA) in Korea as in many other countries. However, this test suffers from potential ethical concerns over the use of live animals. In addition, the mouse bioassay does not test for a specific toxin thus a sample resulting in mouse incapacitation would need further confirmatory testing to determine the exact source toxin (e.g., TTX, STX, brevotoxin, etc.). Furthermore, though the time of death is proportional to toxicity in this assay, the dynamic range for this proportional relationship is small thus many samples must be diluted and new mice be injected to yield a result that falls within the quantitative dynamic range. Therefore, in recent years, there have been many efforts in this field to develop alternative assays. High performance liquid chromatography (HPLC) coupled with mass spectrometry (MS) has been emerged as one of the most promising options. A LC-MS-MS method involves solid-phase extraction (SPE) and followed by analysis using an electrospray in the positive ionization mode and multiple reactions monitoring (MRM). To adopt LC-MS-MS method as alternative standard for testing TTX, we performed a validation study for the quantification of TTX in puffer fish. This LC-MS-MS method showed good sensitivity as limits of detection (LOD) of $0.03{\sim}0.08{\mu}g/g$ and limits of quantification (LOQ) of $0.10{\sim}0.25{\mu}g/g$. The linearity ($r^2$) of tetrodotoxin were 0.9986~0.9997, the recovery were 80.9~103.0% and the relative standard deviations (RSD) were 4.3~13.0%. The correlation coefficient between the mouse bioassay and LC/MS/MS method was higher than 0.95.

  • A cross-over design is frequently used in clinical trials (especially in bioequivalence tests with a parametric method) for the comparison of two treatments. Missing values frequently take place in cross-over designs in the second period. Usually, subjects that have missing values are removed and analyzed. However, it can be unsuitable in clinical trials with a small sample size. In this paper, we compare single imputation methods in a $2{\times}2$ cross-over design when missing values exist in the second period. Additionally, parametric and nonparametric methods are compared after applying single imputation methods. A Monte-Carlo simulation study compares type I error and the power of methods.

Fundamentals of Ecotoxicity Evaluation Methods using Domestic Aquatic Organisms in Korea : (I) Fish (국내 생물종을 이용한 생태독성평가 기반연구 : (I) 어류)

  • The Ministry of Environment plans to introduce the Whole Effluent Toxicity (WET) system in Korea. The WET test is well established in developed countries with recognizing of the limitations of physicochemical analysis method and potential risk of chemicals in water medium. Therefore, it is essential to build the ecotoxicity infrastructure for the induction of WET test. In this study, we extensively collected the domestic and foreign toxicity test methods which employ native test species to Korea. And we suggested that the domestic ecotoxicity test methods with domestic test species in Korea through extracting the range of test conditions. Five domestic fish species selected were Carassiu auratus (Crucian carp), Cyprinus carpio(Common carp), Gasterosteus aculeatus (Three spine stickleback), Misgurnus anguillicaudatus (Oriental weather fish) and Oryzias latipes (Japanese medaka), The toxicity test methods with native test species to Korea were collected from the standard methods (OECD, U.S. EPA, ASTM), government reports, SCI papers and domestic papers. We collected the 32 test methods, and suggested the suitable aquatic toxicity test methods for fish. It is expected that this study could prove a useful information to establish the ecotoxicity test methods with domestic aquatic organisms in Korea henceforth.

Evaluation of Dry Rehydratable Film Method for Enumeration of Microorganisms in Meat, Dairy and Fishery Products (축.수산식품 중 미생물 분석을 위한 건조필름법 평가)

  • Contents of total aerobic bacteria, coliform, Escherichia coli, yeast, mold, and Staphylococcus aureus in meat, dairy, and fishery products were analyzed by dry rehydratable film method using 3M $Petrifilm^{TM}$ and compared against those obtained through conventional method. Two methods showed high correlations of 0.990-0.999, 0.975-0.999, 0.979-0.987, 0.978-0.984, and 0.999 for total aerobic bacteria, yeast and mold, coliform, E. coli, and S. aureus, respectively; therefore, dry rehydratable film method using 3M $Petrifilm^{TM}$ offers acceptable alternative to conventional method for enumeration of microorganisms in meat, dairy, and fishery products.

Replacement of the in vivo Bioassay for Erythropoietin with the in vitro Bioassay (Erythropoietin in vivo 시험법의 in vitro 대체 시험법 확립)

  • In vivo bioassays for biological medicines have been considered final resort to unequivocally assess the biological activities for them because there are some cases in which the biological activities obtained from in vivo bioassay and in vitro bioassay quite differ each other. The in vivo biological activity of EPO depends on its sialic acid contents which confer microheterogeneity-isoforms to this protein. We have devise a method which consists of a in vitro bioassay using BaF3 cell line and a capillary zone electrophoresis (CZE) for the measurement of the EPO isoform distribution. The biological activity of EPO obtained using in vitro bioassay with BaF3 cell line showed good correlation (C.V.(%) 7.34, 5.85, 8,16, 8.08, 8.8) to EPO content measured either spectrophotometric assay (A280 0.1 % =0.743) or radio immunoassay. The assay validation results of in vitro bioassay with 3 lot of in house EPO showed good results to EPO content measured either in vivo assay or radio immunoassay. and also showed good results the robustness of our method in terms of precision, accuracy, repeatability. The isoform distribution for EPO-BRP (1 : 1 mixture of epoetin-${\alpha}$ and epoetin-${\beta}$, European Pharmacopoeia) by CZE method resulted in isoform 2 through isoform 8. The major peaks in electrophoregram were composed of isoform 3 through 7. Our recombinant EPO (epoetin-${\alpha}$) having equivalent in vivo biological activity showed the isoform distribution of isoform 3 through 9. The major peaks consisted of isoform 4 through 8. The peak area of isoform 4 was always smaller than that of isoform 5. The preparations of recombinant epoetin-${\alpha}$ with lower in vivo biological activity than EPO-BRP showed the isoform 2 through 8 in their electrophoregrams whose major peaks consisted of the isoform 3 through 7. The peak area of isoform 4 was larger than that of isoform 5.

  • Water fleas are very important organisms in aquatic ecosystem because they are key constituents of food webs as either glazers or foods for predators. Daphnia magna is a representative test species and it has been extensively used in ecotoxicity evaluation. However, Daphnia magna has not been found yet in Korean water environment. There are limitations of using the foreign species such as Daphnia magna to reflect domestic situations in Korea. Consequently it is mandatory to find domestic species suitable for bioassay, and to develop corresponding toxicity test methods using the domestic species. In this study, we widely collected the domestic and foreign toxicity test methods performed by using domestic water fleas in Korea. The 58 test methods were collected from the standard methods (OECD, US EPA, ASTM), government reports, SCI papers and domestic papers. Ten domestic water fleas selected were Bosmina longirostris, Ceriodaphnia dubia, Ceriodaphnia reticulata, Daphnia obtusa, Daphnia puex, Moina macrocopa, Moina micrura, Simocephalus mixtus, Simocephalus serrulatus, and Simocephalus vetulus. We suggested the domestic ecotoxicity test methods using domestic water fleas in Korea through providing a range of test conditions, and future directions for toxicity test using water flea. This study could be a useful basis for establishing the aquatic toxicity test methods with domestic organisms in Korea.

  • Green algae are important primary producers in aquatic ecosystem, and they are sensitive test species in bioassay. Green algae are broadly used to assess the adverse effects of various chemicals by measuring the inhibition of metabolism, reproduction and survival. In this study, we extensively gathered domestic and foreign toxicity test methods conducted using green algae, which are distributed in Korean water environment. Selected eight domestic green algae were Chlamydomonas reinhardtii, Desmodesmus subspicatus (=Scenedesmus subspicatus), Scenedesmus abundans, Scenedesmus acutus, Scenedesmus quadricauda, Podohedriella falcata (=Ankistrodesmus falcatus), Pseudokirchneriella subcapitata (=Selenastrum capricornutum), and Chlorella vulgaris. Forty four test methods were collected from the standard test ones, government reports, SCI papers and Korean research papers. P. subcapitata and D. subspicatus are the most common test species recommended by the standard test methods. Initial cell density and dilution water were the main differences among the test methods we collected. We proposed the suitable ecotoxicity test methods based on domestic green algae in Korea. This study could be a fundamental basis to establish the ecotoxicity test methods by green algae distributed in Korea.
