Root bark extract of Cudrania tricuspidata reduces LPS-induced inflammation in macrophages of atherogenic mice

  • In this paper, we propose to evaluate the potential anti-inflammatory properties of root bark extract of Cudrania (C.) tricuspidata on lipopolysaccharide (LPS)-induced inflammation in macrophages of apolipoprotein E (ApoE) knockout (ApoE-/-) mice, murine model of atherosclerosis. Atherosclerosis is a chronic vascular inflammatory disease. C. tricuspidata is a small tree of the Moraceae family and its extract has anti-inflammatory activities. However, its role in the progress of atherosclerosis is not yet clear. To determine anti-inflammatory effects of C. tricuspidata in atherogenesis, we applied LPS in peritoneal macrophages of ApoE-/- mice and measured cell viability by CCK-8 and expression of pro-inflammatory cytokines by qRT-PCR following treatment with root bark extract of C. tricuspidata. Research data was expressed as differences between the cells treated with LPS and root bark extract and the cells treated with LPS alone (control) by a two-tailed non-parametric Mann-Whitney U-test using GraphPad Instat program. No cytotoxic effect was observed when the cells were treated with the extract at concentrations ≤ 100 ㎍/mL. The expression of inflammatory cytokines, including MCP-1, IL-1β, IFN-γ, TNF-α, and IL-6 were inhibited by the extract. These results indicated that the extract has an anti-inflammatory effect and therefore a possible role in the treatment of atherosclerosis.

  • This study investigated the anti-inflammatory effects of mixed extracts of Achyranthes japonica Nakai (Aj) and Aralia continentalis Kitagawa (Ac) (ratios of 1 : 2, 1 : 3, 1 : 5, 2 : 1, 3 : 1 and 5 : 1) on RAW264.7 macrophages. Cell toxicity was determined using a cell counting kit (CCK) assay. We evaluated anti-inflammatory effects of the mixed extracts of Aj and Ac by measuring interleukin $(IL)-1{\beta}$, IL-6, and tumor necrosis factor $(TNF){\alpha}$ using an enzyme-linked immunosorbent assay (ELISA) kit assay. The mixed extracts of Aj and Ac inhibited lipopolysaccharide (LPS)-induced $IL-1{\beta}$ and $TNF{\alpha}$ in LPS-stimulated macrophages. Comparing different ratios of the mixed extracts, the 2 : 1 ratio of Aj and Ac has much more potency and inhibited the production of $TNF{\alpha}$ in LPS-induced RAW264.7 cells. The results of the present study showed that the mixed extracts of Aj and Ac have potential anti-inflammatory effects on RAW264.7 macrophages. Therefore, these extracts may be used as a good source of functional foods for the protection against inflammatory diseases.

Non-saponin fraction of red ginseng inhibits monocyte-to-macrophage differentiation and inflammatory responses in vitro (홍삼 비사포닌 분획의 단핵세포 분화와 염증반응에 대한 억제효과)

  • The aim of this study was to investigate the effects of red ginseng-derived non-saponin fraction (NSF) on inflammatory responses and monocyte-to-macrophage differentiation in RAW264.7 and THP-1. NSF effectively inhibited inflammatory responses by downregulating nitric oxide (NO) production and protein levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). NSF ($2000{\mu}g/mL$) decreased the levels of NO, iNOS, and COX-2 by 33, 83, and 64%, respectively. NSF inhibited the differentiation of monocyte-to-macrophage by decreasing cell adherence along with downregulation of the cluster of differentiation molecule $11{\beta}$ ($CD11{\beta}$) and CD36. In addition, pro-inflammatory cytokines, such as tumor necrosis factor-alpha, interleukin 6, and monocyte chemoattractant protein 1 (MCP-1), were significantly reduced with NSF treatment. The NSF-mediated inhibition of inflammatory responses was due to the regulation of nuclear factor kappa-light-chain-enhancer of activated B cells ($NF-{\kappa}B$) and nuclear factor (erythroid-derived 2)-like 2 (Nrf2). NSF effectively suppressed the translocation of $NF-{\kappa}B$ into the nucleus, while nuclear Nrf2 and its target protein, heme oxygenase-1, levels were significantly increased.

  • Effects of Citrus sunki peel and its fermented product extracts on physiological and functional activities of cellular systems were investigated. Ethanol extract of Citrus sunki peel showed potent ROS-scavenging activity using 2',7'-Dichlorofluorescin diacetate as a fluorescent ROS probe in HepG2 cells. Fermented product of C. sunki peel extract markedly suppressed nitric oxide production in lipopolysaccharide (LPS)-activated RAW264.7 murine macrophage cells. Treatment with fermented product of C. sunki peel extract decreased intracellular protein levels of inducible nitric oxide synthase and cyclooxygenase II stimulated by LPS. High doses of fermented product lend to apoptotic cell death in CHO-IR cells.

Study on Immuno-stimulating Activity of ${\beta}$-Glucan Isolated from the Cell Wall of Yeast Mutant Saccharomyces cerevisiae IS2 (효모변이주 Saccharomyces cerevisiae IS2 세포벽 유래의 베타글루칸 면역활성능에 관한 연구)

  • Yeast cell wall mutant, Saccharomyces cerevisiae IS2 was screened by the NTG treatment of Saccharomyces cerevisiae KCTC 7911. The mutant was highly resistant to zymolase, which specifically degrades ${\beta}$-1,3-D-glucose chain of ${\beta}$-glucan and mechanical disruption by glass beads. These phenomena demonstrate that the yeast mutant has cell wall structure different from the wild-type. The ${\beta}$-glucan of yeast mutant and wild-type strains was recovered by sequential extraction with NaOH. The injection of ${\beta}$-glucan into the abdominal cavity of mouse resulted in an increase in the number of peritoneal immune cells, NO (nitric oxide) production, and phagocytic activity of macrophage. The number of immune cells was found to be $3.90{\times}10^6\;cells/10\;mL$ and $5.48{\times}10^6\;cells/10\;mL$ with the wild-type and mutant ${\beta}$-glucan, respectively. The effect on the NO production and phagocytic activity of mutant ${\beta}$-glucan were 1.69 and 1.43-fold higher than those of wild-type. These results indicate that the immuno-stimulating activity of alternated ${\beta}$-glucan from mutant yeast is higher than that of wild-type.

In vitro and Ex vivo Supplementation of Houttuynia cordata Extract and Immunomodulating Effect in Mice (어성초 추출물 첨가가 마우스 면역능 증진에 미치는 영향)

  • Houttuynia cordata THNUB (He; Uh-Sung-Cho) is a medicinal plant which has been widely used as a component of blood-building decoctions. This study was performed to investigate the immunomodulative effect of He in mice. In vitro experiment, the mice splenocytes proliferation and three kinds of cytokines (IL-1$\beta$, IL-6, TNF- $\alpha$) production by mice peritoneal macrophages cultured with six (methanol, hexane, chloroform, ethylacetate, butanol and water) fractions of He were used to indicate the immunomodulative effect. Ex vivo experiment, the different concentrations of He water extract was orally administrated every other day for two weeks. The production of cytokines IL-1$\beta$, IL-6, TNF- $\alpha$) secreted by activated macrophages and the mice splenocytes proliferation were used as an index for the immunocompetence. The supplementation of all six fractions of He enhanced the splenocytes proliferation at the level of 6.58$\pm$1.23∼47.82$\pm$5.48 compared to that of control in the range of 1∼50 $\mu\textrm{g}$/mL. IL-1$\beta$ production was significantly increased with the supplementation of chloroform and water extract of He. Higher level of IL-6 production was detected by the supplementation of ethylacetate, butanol and water extract. TNF - $\alpha$ production was enhanced by the supplementation of all six fractions of He. From the ex vivo study, the highest proliferation of splenocytes was seen from the mice orally administrated with the He water extract at the concentration of 500 mg/kg bw In case of cytokines production, IL-1$\beta$, IL-6, and TNF- $\alpha$ release by activated peritoneal macrophages were augmented by the oral administration of He water extract. These results indicated that He may enhance the immune function by regulating the splenocytes proliferation and cytokines production capacity in mice.

  • 석곡은 난초과의 여러해살이풀 Dendrobium moniliforme의 지상부를 건조한 것으로 예로부터 양위생진(養胃生津), 자음제열(滋陰除熱) 등의 효능이 있어 해열, 진통의 작용과 위액분비 촉진, 혈압강하의 작용이 있는 것으로 알려져 있다. 본 연구에서는 석곡의 항염증 작용 기전을 알아보기 위하여 석곡 열수추출물을 대식세포주에 처리하여 NO 및 IL-$1{\beta}$의 생성에 미치는 영향을 조사하였다. LPS로 자극된 대식세포주 RAW264.7 세포에서 석곡은 NO 및 IL-$1{\beta}$ 생성과 iNOS 단백질 발현을 저해하였으며, LPS에 의해서 활성화되는 ERK, p38, JNK 효소의 활성을 현저히 억제하였다. 이 결과들로 보아 석곡의 항염증 작용이 MAPK 활성 저해로 인한 NO 및 IL-$1{\beta}$ 생성의 억제 때문인 것으로 사료된다.

Combined Eeffect of Exercise and L-arginine Supplementation on Cardiovascular and Immune Responses in SHR (L-arginine 투여와 훈련이 SHR의 심혈관 반응과 면역력에 미치는 영향)

  • The purpose of this study is to investigate the combined effect of L-arginine supplementation and regular physical exercise on HR, BP, eNOS and Macrophage activation using SHR. To examine the differences among HR, BP, eNOS, and Macrophage activity levels, normotensive Wistar-Kyoto rats were used as a control. Thirty two male rats (six weeks old) were divided into four groups; eight WKY control (WKYC), eight SHR control (SHRC), eight SHR supplemented with L-arginine (SHRA), and eight SHR trained and supplemented with L-arginine (SHRTA). Obtained results were as follows : In the heart and blood pressure, there was significant differences anong the four group (p<.05) compare to SHRC. In the eNOS levels, there was significant differences among the four groups (p<.05) compare to SHRC. In the macrophage activity, there was significant differences among the four groups (p<.05) compare to SHRC. In conclusion, For the SHRC group, the level of eNOS is higher than that of WKYC, and we can expect tissue damage caused by toxic free radical. However, this can be stabilized by the L-arginine supplementation and regular physical training. we can also conclude regular aerobic training decrease cardiovascular stress caused by stabled macrophage activity. Therefore, we can trace it is the effect of training in SHR.

Inhibition of Tumor Growth through Macrophage Activation by Polysaccharide Fraction from Peonia japonica (PJ-P) (백작약 조다당분획에 의한 대식세포 활성화를 통한 암세포 증식 억제)

  • The immunomodulatory activity of PJ-P, a polysaccharide fraction extracted from Paeonia japonica, were reported in our previous paper. In the present study, we investigated that PJ-P inhibited cancer growth through activation of macrophages. The activities of peritoneal macrophage to induce tumor necrosis factor (TNF)-$\alpha$, interleukin-1 (IL-1)$\beta$, interleukin-6 (IL-6) and interleukin-12 (IL-12) as well as to ingest fluorescence-latex microbeads were enhanced by treatment of PJ-P. Direct cytocidal activity of PJ-P against cancer cells was not shown. However, in vitro, peritoneal macrophages treated with PJ-P had an activity to kill cancer cells. Furthermore, PJ-P significantly prolonged the survival of mice implanted intraperitoneally with B16F0 mel-anoma cells. These results suggest that PJ-P could be a useful immunomodulator and assistant of anti-tumor agent.

Optimun Conditions for Production of Mycelia and Extracellular Polysaccharide from Inonotus obliquus and Their Immunomodulating Acitivities (차가버섯의 균사체 및 세포외다당체의 생산조건과 면역활성)

  • Optium conditions for the production of mycelia and extracellular polysaccharide (EXPS) from submerged mycelial culture of Inonotus obliquus and their immunomodulating activities were investigated. The optmium production of mycelia and EXPS from I. obliquus was observed in mushroom complete medium (MCM). The optimum pH, temperature, and agitation speed for the production of mycelia and EXPS were 5.5, $25^{\circ}C$, and 150 rpm, respectively. The culture period for maximum production of mycelia (10.89 g/l) and EXPS (1.25 g/l) in shake flask cultivation was 11 days. The anticomplementary activity of intracellular polysaccharide (INPS) and EXPS form I. obliquus increased in a dose-dependent manner. Lysosomal enzyme activity of EXPS and INPS increased by 2.0- and 2.2-fold at $100{\mu}g/ml$ concentration, respectively, compared to the control group.
