  • Mode of safening action of N-(4-chlorophenyl)maleimide (CPMI) on metolachlor [2-chloro-N-(2-ethyl-6-methylphenyl)-N-(2-methoxy-l-methylethyl) acetamide] was investigated in sorghum(Sorghum bicolor L.). CPMI was synthesized by dehydration of N-(4-chlorophenyl)maleamic acid (CPMA) which was obtained from amination with maleic anhydride and 4-chloroaniline. Melting points of CPMA and CPMI (>95% purity) were $200-202^{\circ}C$ and $116-118^{\circ}C$, respectively. Growth response study indicated that seed treatment of CPMI increased tolerance of sorghum shoot to metolachlor approximately threefold. Sorghum shoot was more sensitive to injury caused by metolachlor and CPMI activity than the root. Metolachlor was initially absorbed by sorghum shoot and metabolized to the metolachlor-glutathione conjugate in CPMI-untreated and treated shoots. However, CPMI treatment significantly accelerated metabolism of $[^{14}C]$metolachlor in sorghum shoot, resulting in decrease in metolachlor content and increase in formation of the glutathione conjugate. It was concluded that the protection against metolachlor injury conferred by CPMI appeared to be correlated to detoxification of metolachlor in sorghum shoot tissue.

  • This experiment was conducted to determine the selective mechanism of cyhalofop-butyl ester on uptake, traslocation, and metabolism of the herbicide in both rice and Echinochloa crus-galli. Uptake and translocation of $^{14}C$-cyhalofop-butyl ester was higher in E. crus-galli than rice when treated to shoot. $^{14}C$-uptake by root of E. crus-galli increased rapidly at 30 minute after treatment and reached the maximum at 12 hoots after treatment. After that, uptake was leveled off. Uptake pattern in rice root was not significantly affected by the duration of herbicide treatment. In E. crus-galli, the absorbed $^{14}C$-cyhalofop-butyl ester seemed to be rapidly metabolized into free acid and the content of changed free acid was higher than rice.

  • O-Ethyl S-methyl ethylphosphonothioate was studied for chemical and metabolic oxidation using $^{31}P-NMR$ analyses. The chemical shifts of O-ethyl ethylphosphonic acid (2) which is one of major metabolites were changed with the variation of oxidation systems. $^{31}P-NMR$ chemical shifts of 2 were observed at 40.15ppm from oxidaton by MCPBA, 30.98 ppm by MMPP, 29.31 ppm from in vitro rat liver microsomal oxidation, and 29.10 ppm from in vivo metabolism in houseflies. $^{31}P-NMR$ chemical shift of 2 in two different solvents such as deutero-chloroform and deuterium oxide were observed at 30.70 ppm and 40.15 ppm, respectively. And those of the metabolites were also observed at around 30 ppm under the conditions of pH 3, 5.6 and 14 and 47.91 ppm under pH 1 which is a strong acidic condition. It could be explained that the ionized form of 2 should have greater shielding effect on phosphorus atom and hence shows upfield chemical shift in polar solvents and alkaline conditions. On the other hand, a protonated form under organic solvents and the strong acidic condition should have less shielding effect than its ionized form, shifting the peak downfield.

Effect of Organophosphorus Insecticides on the Inhibition of the Acetylcholinesterase Activities (유기인계(有機燐系) 살충제(殺?劑)가 Acetylcholinesterabe 활성저해(活性沮害)에 미치는 영향(影響))

  • The responses of brain acetylcholinesterase and plasma cholinesterase activities were examined in chicken given oral doses of an organophosphorus insecticides. Acute oral $LD_{50}$ of terbufos was 1.82mg/kg, and terbufos sulfone was 2.85mg/kg, terbufos sulfoxide, terbufoso xon, terbufosoxon sulfone and terbufosoxon sulfoxide were about $0.30{\sim}0.79mg/kg$, respectively. Acute oral $LD_{50}$ of phorate was 1.02mg/kg, and phorate sulfone was 1.73mg/kg, phorate sulfoxide, phoratoxon, phoratoxon sulfoxide and phoratoxon sulfone were about $0.36{\sim}0.63mg/kg$, respectively. Dosages of 50% inhibition for brain AChE and plasma ChE activities in chicken at 60 mins after oral administration of organop hosphorus insecticides were $35.3{\pm}6.8%\;and\;17.4{\pm}3.6%$ of acute oral $LD_{50}$, respectively. Activities of brain AChE and plasma ChE in chicken at 60 mins after oral administratration of acute oral $LD_{50}$ of organophosphorus insecticides were inhibited by $83.7{\pm}3.9%\;and\;93.3{\pm}2.2%$, respectively.

  • Microbial secondary metabolites of marine organisms are regarded as major sources of structurally and biologically novel compounds with numerous potential uses. Sponge-microbe associations are among the most interesting sources for exploring bioactive compounds. In this study, the bacterial strain Microbulbifer sp. (127CP7-12) was isolated from the Asian marine sponge Hymeniacidon sinapium collected at an intertidal zone on the west coast of Korea. Cultured bacteria produced a violet pigment, and optimal culture conditions for violet pigment production were investigated. Maximum production of the violet pigment from the strain culture was observed under the conditions of $25^{\circ}C$, pH 6.0, and 3% NaCl. Acetone provided better extraction of the pigment from fermented broth compared with ethanol and methanol. The proposed structure of the major component in the extracted crude pigment was determined via high-performance liquid chromatography, nuclear magnetic resonance, mass spectrometry, and UV spectra analyses, which showed that the metabolite was the promising bioactive compound violacein. This study describes the examination of marine bioactive materials from microbe-engaged metabolites and the ecological implications of the sponge-microbe association in a changing ocean.

Microbial Production of Carotenoids: Biological Functions and Commercial Applications (미생물에 의한 카로티노이드 생산; 생물학적 기능성 및 상업적 적용)

  • Carotenoids are isoprenoids with a long polyene chain containing 3 to 15 conjugated double bonds, which determines their absorption spectrum. They typically consist of a $C_{40}$ hydrocarbon backbone often modified by different oxygen-containing functional groups, to yield cyclic or acyclic xanthophylls. Much work has also been focused on the identification, production, and utilization of natural sources of carotenoid (plants, microorganisms and crustacean by-products) as an alternative to the synthetic pigment which currently covers most of the world markets. Nevertheless, only a few carotenoids (${\beta}-carotene$, lycopene, astaxanthin, canthaxanthin, and lutein) can be produced commercially by fermentation or isolation from the small number of abundant natural sources. The market and demand for carotenoids is anticipated to increase dramatically with the discovery that carotenoids exhibit significant anti-carcinogenic activities and play an important role in the prevention of chronic diseases. The increasing importance of carotenoids in the feed, nutraceutical food and pharmaceutical markets has renewed by efforts to find ways of producing additional carotenoid structures in useful quantities. Because microorganisms and plants synthesize hundreds of different complex chemical carotenoid structures and a number of carotenoid biosynthetic pathways have been elucidated on a molecular level, metabolic and genetic engineering of microorganisms can provide a means towards economic production of carotenoid structures that are otherwise inaccessible. The aim of this article is to review our current understanding of carotenoid formation, to explain the perceived benefits of carotenoid in the diet and review the efforts that have been made to increase carotenoid in certain microorganisms.

Studies on the Chromatin Isolated from the Organs of Animals Received Whole-body X-ray Irradiation (백서장기(白鼠臟器)에서의 Chromatin의 분리(分離)와 그 RNA 합성능(合成能)에 미치는 X-선전신조사(線全身照射)의 영향(影響)에 관(關)한 연구(硏究))

  • 1. Within experimental chromatin, the total protein: DNA ratio did not vary in the same organs of control and irradiated rats. However, the amount of RNA and total protein associated with the DNA varied considerably among the different types of chromatin. In particular, the content of chromatin was the highest in the irradiated tissue, and the lowest in the chromatin control tissue. RNA and total protein ratio of chromatins from brain, liver, testis and spleen declined with experimental organs. 2. There was the same quantitative relationship between the amount of RNA and the amount histone-protein associated with DNA in each chromatin. 3. RNA:DNA ratio of chromatin showed a $1.5{\sim}2$ times increase in the irradiated organs except brain. However, RNA:DNA ratio was decreased in chromatin by irradiation. 4. Histone-protein:Residual protein ratio was greatly varied among the organs. However, the effect was not found by irradiation. 5. Priming activity of chromatins showed a higher value in testis and the activity was greater in organs with higher metabolic activity. 6. Inhibition of Actinomycin D observable in chromatin for testis, liver, spleen and brain declined without relationship between irradiated and non-irradiated conditions. Ammonium sulfate in DNA of chromatin from histone showed increased priming activity with dissociation by Electrostatics. It may give different effect of ammonium sulfate on stimulation by property of chromatins. 7. It is suggested that the results support a proposal that the higher sensitivity of radioactive in testis, spleen by irradiated showed a increase and decrease lower-sensitivity of radioactive from brain, liver than did priming activity under the radioactive conditions.

Increasement of Secondary Metabolites and Antioxidative Activity in Panax ginseng Adventitious Root by Methyl Jasmonate (Methyl jasmonate 처리에 의한 인삼 (Panax ginseng C.A. Meyer) 부정근의 이차대사산물 및 항산화활성 증가)

  • This study was initiated to investigate the impacts of methyl jasmonate (MeJA) on adventitious root growth of Panax ginseng, the production of secondary metabolites, such as ginsenosides and phenolic compounds, and antioxidative activity. Among various concentrations of MeJA, $100\;{\mu}M$ MeJA increased the ginsenosides accumulation to 26.6 mg/g dry wt, about 8 times higher than the control in ginseng adventitious roots (GAR). In addition, $50\;{\mu}M$ MeJA increased the accumulation of phenolic compounds to 0.38 mg/g dry wt, about 3 times higher than control in GAR. This MeJA treatment was more effective in conditioned medium (CM) which obtained in bioreactor after 40 days of culture than in fresh medium (FM). Treatment of $100\;{\mu}M$ MeJA in CM increased the accumulation of ginsenosides (1.7 times) and phenolic compounds (1.2 times) more than in FM, respectively. Consequently, these high accumulation of ginsenosides and phenolic compounds by MeJA led to increase the antioxidative activities expressed to the DPPH scavenging activity (over $78.3\%$). The DPPH scavenging activity in control was $45.5\%$.

  • Positron Emission Tomography(PET) is nuclear medical tests which is a combination of several compounds with a radioactive isotope that can be injected into body to quantitatively measure the metabolic rate (in the body). Especially, Phenomena that increase (sing) glucose metabolism in cancer tissue using the $^{18}F$-FDG (Fluorodeoxyglucose) is utilized widely in cancer diagnosis. And then, Numerous studies have been reported that incidence seems high availability even in the modern diagnosis of dementia and Parkinson's (disease) in brain disease. When using a dynamic PET iamge including the time information in the static information that is provided for the diagnosis many can increase the accuracy of diagnosis. For this reason, clinical researchers getting great attention but, it is the lack of tools to conduct research. And, it interfered complex mathematical algorithm and programming skills for activation of research. In this study, in order to easy to use and enable research dPET, we developed the software based graphic user interface(GUI). In the future, by many clinical researcher using DIA-Tool is expected to be of great help to dPET research.

Changes in the Contents of Some Metabolites and Ions and in Some Enzyme Levels in Rice Plants Grown under Water-and Salt-stressed Condition (수분장애(水分障碍) 및 염장애(鹽障碍)하에서 수도체(水稻體)중 효소수준(酵素水準) 및 유기대사산물(有機代謝産物)과 무기(無機)이온 함량의 변화)

  • Two rice cultivars, Jinju and Iri 348, were used to compare the changes in the contents of some organic metabolites and ions and in some enzyme levels under water-and salt-stressed conditions. The water loss and proline accumulation under water and salt stresses were accelerated more in the salt-sensitive cultivar Iri 348 than in the salt-tolerant Jinju. The contents of crude protein, total free amino acids, proline and polyphenols increased under water-or salt-stressed rice, but that of reducing sugar increased under water stress only. The water-and salt-stresses induced the high ratio of low molecular organic solutes to crude protein in Jinju but not in Iri 348. The ratio of total free amino acids to crude protein increased under the stressed conditions was likely due to high protease activity. The contents of $Na^+$ and $Cl^-$ were higher in Iri 348 than in Jinju. Iri 348 had higher values of $Na^+/Ca^{2+}$ and monovalent/divalent of cations, but lower of $K^+/Na^+$ than Jinju Rice. The further studies should emphasize to set the correlations between these ratios and tolerance to water and salt stresses among rice cultivars.
