Cluster Analysis and Growth Characteristics of Hemp (Cannabis sativa L.) Germplasm (저 마약형 대마 유전자원의 생육특성 및 품종군 분류)

  • In order to breed hemp variety with nondrug type adapted to Korea, a total of 44 accessions were introduced from Center for plant breeding and reproduction research (CPRO) In Netherland and evaluated for plant growth characteristics and contents of cannabinoids such as tetrahydrocannbinol (THC), cannabidiol (CBD), Cannabinol (CBN) in Korean climatical and geographical condition. 1. In Korean geographical position which latitude is $34^{\circ}N$, days to flowering and stem length were shorter than in Netherlands which latitude is $52^{\circ}N$. But THC content was not shown significant difference between two areas. 2. Corrected accessions of hemp were classified into 3 variety groups by cluster analysis and 4 accessions including Korean local variety were not classified into any variety group. 3. Most hemp accessions have low THC contents but were not suitable for fiber production in Korea due to short days to flowering and stem length. 4. Among the introduced accessions, IH3 was suitable for breeding material of fiber hemp with non-drug type because of low THC content and similar days to flowering with Korean local variety.

A Study of the Predictive Effectiveness of Stem and Root Extracts of Cannabis sativa L. Through Network Pharmacological Analysis (네트워크 분석기반을 통한 대마 줄기 및 뿌리 추출물의 약리효능 예측연구)

  • Cannabis sativa is a plant widely cultivated worldwide and has been used as a material for food, medicine, building materials and cosmetics. In this study, we assessed the functional effects of C. sativa stem and root extracts using network pharmacology and confirmed their novel functions. The components in stem and root ethanol extracts were identified by gas chromatography-mass spectrometry analysis, and networks between the components and proteins were constructed using the STICHI database. Functional annotation of the proteins was performed using the KEGG pathway. The effects of the extracts were confirmed in lysophosphatidylcholine-induced THP-1 cells using real-time PCR. A total of 21 and 32 components were identified in stem and root extracts, respectively, and 147 and 184 proteins were linked to stem and root components, respectively. KEGG pathway analysis showed that 69 pathways, including the MAPK signaling pathway, were commonly affected by the extracts. Further investigation using pathway networks revealed that terpenoid backbone biosynthesis was likely affected by the extracts, and the expression of the MVK and MVD genes, key proteins in terpenoid backbone biosynthesis, was decreased in LPC-induced THP-1 cells. Therefore, this study determined the diverse function of C. sativa extracts, providing information for predicting and researching the effects of C. sativa.

Comparison of color and major components of hempseed oils extracted with pressuring and extruding methods (압착식, 압출식 착유 대마 종실유의 색깔과 주요성분 비교)

  • Hemp [Cannabis sativa L.] has been cultivated as a fiber crop for long history, but it was a good oil crop because its seed contain plenty of lipid which is high ratio of unsaturated fatty acid. Hemp seed oil was extracted with a extruding method in many countries including Europe. The color of oil extracted with extruding method is dark green which could be difficult to attract consumer's interest in Korea because of insufficient information about hemp seed oil. This study was conducted to know correct information about hemp seed oils which were extracted with pressuring and extruding methods. In extruding method, seeds were crushed during the extracting process and discharged oil cake in shape of thin ribbon, but maintained seed shape although the seed were slightly flatted in pressuring method. Oil yield were higher in the extruding method in comparison with pressuring method. The oil have lower degree of lightness but higher degree of greenness and yellowness in the extruding method in comparison with pressuring method because of higher content of chlorophyll A, B and carotenoid. In fatty acid composition, the ratio of palmitic acid, stearic acid, oleic acid and ${\gamma}$-linolenic acid were higher but linoleic acid and ${\alpha}$-linolenic acid were lower in the extruding method in comparison with pressuring method. The content of total tocopherol and ${\gamma}$-tocopherol were lower in the extruding method in comparison with pressuring method.

Optimum Sowing Date for Seed Production in Hemp (대마의 채종재배를 위한 파종적기)

  • This study was carried out to investigate the effects of sowing dates, Apr. 25, May 15, June 5 and June 25, on sex ratio, growth and seed yield of "Chonnam species", local variety of hemp plant. The results were following as. : In sex ratio according to the different seeding dates, the revealating rate of female and male plants were highest on June 25, and Apr. 25, respectively. The length of stem, the number of branches, nodes, and seed yield were increased on May 15, but these charateristics were rapidly reduced on June 25 because of short growing period. Yield of seeds was increased by 24% on May 15 comparing with 1.30ton /ha on Apr. 25.

Evaluation of Antimicrobial, Antioxidant, and Antithrombin Activities of the Rhizome of Various Dioscorea Species (마 품종별 메탄올 추출물의 항균, 항산화 및 항혈전 활성 평가)

  • Yams (Dioscorea spp.) have been used as medicinal and edible resources worldwide. In the present study, methanol extracts were prepared from various kinds of yam, including D. batatas (Gyeongbuk No. 1 and No. 4), D. alata L. (Gyeongbuk No. 5 and No. 6), and D. bulbifera and D. nipponica, and the antioxidant, antithrombin, and antimicrobial activities of these extracts were evaluated. The water content of the various yams was 54.7~83.2% (D. batatas> D. alata L.>D. nipponica>D. bulbifera) and the methanol extracts contained 0.59-1.59% (w/v) solid matter (D. alata L.>D. batatas >D. nipponica>D. bulbifera). The mean concentrations of total polyphenols, total flavonoids, and total sugars in the different types of yam were 49.82 mg/g, 9.79 mg/g, and 173.81 mg/g, respectively. Analysis of DPPH radical-scavenging activity showed that D. alata L. Gyeongbuk No. 6 had a strong antioxidant capacity (IC50=$142.30{\mu}g/mL$), whereas the other yams showed reduced levels of activity (IC50=371-$486{\mu}g/mL$). D. alata L. Gyeongbuk No. 6 showed a strong antithrombin activity against human thrombin. Upon treatment with a D. alata methanol extract (1.5 mg/mL), thrombin time was increased 7.35-fold compared with that when methanol alone was used. In an antimicrobial activity assay, the D. nipponica extract showed moderate antibacterial activity against Bacillus subtilis, Staphylococcus aureus, Proteus vulgaris, and Salmonella typhimurium. Our results indicate that different varieties of Dioscorea spp., including D. batatas, have useful biochemical attributes, including antioxidant, antithrombin, and antibacterial activities.

The Production-And-Marketing System and the Regional Division in a Traditional Industrial District: Hemp Fabric Handicrafts in Andong (안동 삼베 수공업산지의 생산유통체제와 지역분화)

  • The purpose of this study is to clarify the reason of the change of production-and-marketing system and of the regional division. The data for this study were collected by administering interviews with people engaging in hemp fabric industry: namely, craftsmen and managers in the production and marketing of "hemp fabric" handcraft. The summary of this study is as follows: First, the hemp fabric industry is a main subsidiary business of agriculture in Andong. The process of making hemp fabric heavily depends on manual labor. The process is divided into five stages: namely, cultivating hemp, pre-reeling, reeling, weaving, and colouring. Second, Andong "hemp fabric" handicrafts industry had been continuously growth until the late 1960s. During the period of Japan colonization, it was under boom condition: while the other traditional fabric industries were declined. In the 1970s, the decrease of the demand of hemp fabric was the result of mass production of substitute goods on factory system: while, in the 1980s, the growth of per capital income play an important role in bringing about the increased demand of hemp fabric. Third, in the 1980s. production-and-marketing system was changed as the result of the effort to improve the quality and the process, the advanced age of craftsmen, and the weak function of existing marketing systems. The social division of labor within the district is well developed between cultivating hemp, reeling and weaving. The social division of labor is also found in the partly process of handcratfs, and between the production and marketing. The social division of labor between production and marketing is not strongly developed to establish a reliable enterprise that develops a new product and opens a new market. Fourth, the spatial boundary of the production of hemp fabric handcrafts becomes limited into a special region based on the regional specialization of hemp cultivating and the differentiation of utilization of labor.

The characteristics and changes of traditional hemp textile production of Gurye at Jeollanam-do in modern times (1920s~1980s) (근대 시기(1920~1980) 전라남도 구례군의 전통 삼베 수공업 생산 방식의 특징과 변화)

  • This study investigated the characteristics of and changes in hemp textile production practices at Gurye in Jeollanam-do in modern times. To do this, in this study, both a literature review and fieldwork research were utilized as research methods. The literature review focused on Gurye's hemp data, and the fieldwork research involved two separate trials. Male and Female residents of Gurye who had experience in the area of hemp production from the 1920s to the 1980s participated in this study. The results were as follows. First, regarding hemp fiber cultivation in Gurye, hemp cultivation continued to the 1970s and there were no changes in hemp cultivation practices during the Japanese colonial-era. Second, there have been very important changes in hemp kilns over time. In the 1950s, there were both single-body kilns and separate-body kilns in Gurye. Later, specifically in the 1950s, a new type of kiln using an iron pot appeared, and the most modern kilns were concrete structures. Third, in Gurye, women cooperatively removed hemp husks immediately after stemming, subsequently bleaching the hemp by soaking it in lye or caustic soda. Over time, there have been changes in ash types and in soaking periods. Fourth, loom types changed from the traditional Korean back-strap loom to the treadle loom in the 1930s- to 1940s. Fifth, since the 1970s, the hemp textile output levels of Gurye have been reduced due to the westernization of clothing styles, the inflow of Chinese hemp fiber and government regulations pertaining to hemp cultivation.

Planting Conditions of Korean Cannabis Derived from Stable Isotope Ratio & Tetrahydrocannabinol Contents (안정동위원소 비율 및 테트라하이드로칸나비놀 함량 분석을 통한 한국산 대마의 재배 환경 추론)

  • Stable isotope ratio of carbon and nitrogen ($\delta^{13}C$ & $\delta^{15}N$), and $\Delta^{9}$-tetrahydrocannabinol (THC) contents were measured on 37 Korean cannabis and 10 commercial grade marijuana seized in Korea. Factors influencing on the measured values and their variations were investigated. $\delta^{13}C$ value of cannabis is specified mainly by water availability. Korean cannabis showed relatively low $\delta^{13}C$ values ranging -33.29$\sim$-27.01% (mean=-31.01%), which reflect geographic conditions of Korea where is rainy, especially during summer. $\delta^{15}N$ values, which reflect individual planting conditions, were relatively high up to -0.5$\sim$18.0% (mean=6.44%). It reflects characteristics of Korean cannabis growing wild in forest or cultivated in fertile soil. Tetrahydrocannabinol is the major hallucinogenic compound of cannabis. Ethanol extracts of cannabis leaves were derivatized by N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA), and the derivatives were analyzed by GC-MS in selected ion monitoring (SIM) mode. THC contents of Korean cannabis ranged 0.11$\sim$4.34% (mean=1.47%), which were relatively low compared with commercial grade marijuana.

Seed Production by Induction of Male Flowers on Female Plants of Hemp(Cannabis saliva L.) (대마 자주의 웅화 유도에 의한 자성종자 생산에 관한 연구)

  • This study was conducted to overcome the deliciousness by sex reversal in hemp plants. When female plants grown under long day length condition were transferred to short day length condition in 3th and 6th leaf stage, a few male flowers were formed and the gap of flowering days between sex reversed male and normal female flowers were around 10 days and 3 days, respectively. Although the flowering amounts were small, this short gaps of flowering days were enough for artificial pollination. When female plants grown under long day length condition were transferred to short day length condition just after treatment of silver nitrate in the vegetative stage, a great amount of male flower was formed and the gap of flowering days between sex reversed male and normal female flowers were only 1 to 3 days. The increased sex reversed male flowers and induced short gap of flowering days were adequate for natural wind pollination. When female seeds were cultivated, seed yield was 141 kg/10a, while it was 96 kg/10a when normal seeds cultivated.

Studies on the Phytoextraction of Cadmium and Lead Contaminated Soils by Plants Cultivation (토양중 카드뮴과 납의 Phytoextraction을 위한 식물재배 연구)

  • In order to select more proper plants for phytoextraction at the heavy metal polluted areas, 11 species of non-edible plants were cultivated at the cadmium(Cd) and Lead(Pb) treated soils and analyzed the content of the absorbed Cd and Pb in each part of plants. Plants include three fibers(Linum usitatissimum, Cannabis sativa, Gossypium spp.), three flowers(Calendula officinalis, Rhododendron lateritium, Portulaca grandiflora), and five trees(Pinus thunbergii, Magnolia kobus, $Populus\;nigra\;{\times}\;P.$ maximowiczii, Euonymus japonica, Fraxinus rhynchophylla). Yield of tree species were higher than that of fiber and flower species. Cd and Pb were highly accumulated in root rather than leaves and stems. The Cd content of plants was in the order Portulaca grandiflora > Calendula officinalis > Gossypium spp. > Linum usitatissimum, Pb was Cannabis sativa > Linum usitatissimum > Fraxinus rhynchophylla. Total absorbed Cd by each plant was in the order $Populus\;nigra\;{\times}\;P.$ maximowiczii > Euonymus japonica > Rhododendron lateritium, but Pb was $Populus\;nigra\;{\times}\;P.$ maximowiczii > Rhododendron lateritium > Euonymus japonica. Total absorbed Cd and Pb contents in plants were negatively correlated with the residual Cd and Pb in the treated soils. It was estimated that $Populus\;nigra\;{\times}\;P.$ maximowiczii, Euonymus japonica, Fraxinus rhynchophylla, and Rhododendron lateritium were the most effective species for phytoextraction in the polluted area considering yield and heavy metal uptake.
