Lactobacillus acidophilus KFCC12731와 Kluyveromyces fragilis KFCC35457의 대두유 발효중 당대사의 상호작용

  • 대두유중에서 L, acidophilus와 K. fragilis를 단독발효시키는 것 보다 혼합배양 함으로써 젖산의 생성이 촉진되었는데 균간의 어떠한 상호작용에 의하여 산의 생산이 증가되었는가를 연구하였다. 대두유와 Soywhey를 사용하였을 때의 젖산 생성속도는 L. acidophilus 단독보다는 K. fragilis 와 혼합배양하는 것이 빨랐다. 이들 균을 단독 또는 혼합배양하여 경시적으로 발효 대두유중의 당 성분을 HPLC로 분석하였다. 그 결과 L. acidophilus가 발효하기 어려운 sucrosed raffinose, stachyose를 K. fragilis가 생산하는 효소에 의하여 젖산균이 발효할 수 있는 glucose, fructose로 분해되기 때문에 혼합배양 함으로써 젖산 발효가 촉진된다는 것을 알았다.

  • Lactobccillus acidophilus와 Saccharomyces cerevisiae를 대두유에서 혼합 배양시켜 최적 젖산발효조건을 검토하였다. 대두유에서의 혼합배양시 20시간 이상 배양했을 때 산생성이 촉진되었고 배양온도는 34$^{\circ}C$ 부근으로 했을 때 가장 좋았다. 대두유에 sucrose와 skim milk가 젖산발효에 미치는 영향을 조사한 결과 sucrose 는 1.5% 첨가시 skim milk는 2.0% 첨가시 첨가하지 않고 배양할 때 보다 2배 정도의 산생성을 보였다.

  • 젖산균 Lactobacillus casei와 Saccharomyces uvarum을 사용하여 대두유중에서의 최적 젖산발효조건을 검토하였다. 젖산발효 중의 온도의 영향은 Saccharomyces uvarum과의 혼합배양에서 37-4$0^{\circ}C$가 가장 좋았다. 배양시간은 24시간일때, Lactobacillus casei 와 Saccharomyces uvarum 의 접종비는 2:1일때 산생성이 가장 좋았다. Sucrose와 skim milk의 혼합배양시 대두유의 젖산발효에 미치는 영향을 검토한 결과 sucrose의 경우 1.0% 첨가하였을 때 산생성은 높게 나타났으며 skim milk를 첨가했을 때는 첨가하지 않고 배양했을 때와 비교하여 별다른 차이를 보여주지 않았다.

Production of a Fermented Soymilk using a New Strain Leuconostoc mesenteroides KC51 Isolated from Kimchi (김치에서 분리한 Leuconostoc mesenteroides KC51을 이용한 대두 발효유의 제조)

  • 숙성 중인 김치에서 생육과 산 생성이 우수한 유산균을 분리하여 API 50 CHL kit로 확인하고 16S rDNA의 염기 서열을 분석한 결과 Leuconostoc mesenteroides로 동정되어 Leuconostoc mesenteroides KC51로 명명하였다. 대두에 10배의 증류수를 가하여 제조한 두유에 KC51 균주를 접종하고 $30^{\circ}C$에서 정치 배양한 결과, 생균수는 접종 후 9시간까지 급격히 증가하였으며 그 이후 완만하게 증가하여 배양 12시간에는 $2.70{\times}10^9$ CFU/g까지 증가하였다. 적정산도는 배양 12시간에 0.35%까지 증가하였으며,pH 는 pH 4.52까지 감소하였다. 이때 유기산 농도는 젖산과 초산의 함량이 각각 0.56%와 0.10%로 분석 되었다. 두유 발효액에서 Leuconostoc mesenteroides의 증식에 의한 점도의 증가는 관찰되지 않았으며, 관능적으로도 신맛을 제외한 항목에서 두유와 유사하였다. 또한 $4^{\circ}C$에서 두유 발효액은 2주일의 저장기간 동안 pH, 적정산도와 생균수의 변화는 미미하였다.

  • It has been investigated that preparation of cheese-like product from defatted soybean milk and changes during 6 weeks' ripening. Obtained results were as follows: 1. Defatted soybean milk was fermented at $37^{\circ}C$ for 4 hours with Streptococcus lactis YUFE L-4 without addition of any other carbon sources. 2. Optimum concentration of coagulant such as $MgCl_2{\cdot}6H_2O$ and $CaCl_2$ were 0.0125M and 0.0375M, respectively. 3. Coagulating time for 30 minutes was sufficient to make curd. 4. Optimal reaction temperature for coagulating were $50^{\circ}C$. 5. After 6 weeks' ripening, moisture content was approximately $62{\sim}63%$. Water and 10%-TCA soluble nitrogen contents of soybean cheese with Asp. saitoi protease were higher than those of Mucor rennet. Water soluble nitrogen content of Asp. saitoi protease cheese was 50.5% and 10%-TCA soluble nitrogen was 27.5%.

  • L. acidophilus를 대두유에 젖산발효 시킬 때 단독배양과 효모인 K. fragilis와의 혼합배양에 대하여 검토하였다. 젖산의 생성량은 젖산균 단독배양보다는 효모와 혼합배양 하는 것이 2.6배나 많았으며 L. acidophilus를 단독배양 하였을 경우 균의 생육속도와 PH 강하속도는 K. fragilis를 단독배양한 것에 비교하여 늦었다. 그러나 혼합배양 함으로써 생육속도와 PH 강하속도는 K, fragilis를 단독배양한 것보다 빨랐다. 단독 또는 혼합젖산발효에 미치는 온도의 영향을 조사한 결과는 L. acidophilus.는 37$^{\circ}C$, K. frahilis는 3$0^{\circ}C$, 두 균의 혼합배양시는 35-37$^{\circ}C$ 범위에서 산생성이 가장 좋았다. 혼합배양 시 검토한 젖산발효 최적조건은 L. acidophilus와 K. fragilis의 접종비율이 1:5, sucrose 0.5-1.0% SKIMMILK 1.5%를 각각 첨 가한 것이 좋았다.

Pretreatment of Soybean and Development of Fermentation Conditions of Chungkukjang for High Contents Isoflavone Production (고함량 이소플라본 생산을 위한 대두의 전처리 및 청국장 발효조건 확립)

  • The separation conditions for the enhancement of isoflavone contents in soybean by soaking and germination were developed, and the fermentation conditions of Chungkukjang were optimized by using response surface methodology analysis. The high-yield of isoflavone production was obtained when the conditions of soaking and gemination were at 25.6$^{\circ}C$, 7 hr and 29.1$^{\circ}C$, 42.4 hr, respectively. Total isoflavone contents in soybean (Daepung) by treatment were increased from 1,012 ppm to 1,912 ppm. Also, the optimum fermentation conditions of Chungkukjang were determined 39.96$^{\circ}C$, 1.32% (inoulum size) and 42.4 hr, respectively. Isoflavone contents in Chungkugjang under the optimum fermentation conditions were 1.3 times higher than the control (1,960 ppm).

A New Evaluation of Browning Reactions of Korean Traditional Soy Sauce Mash During Fermentation (대두발효식품의 새로운 갈변기작에 관한 연구)

  • To re-evaluate the browning reactions of fermented soybean products, soy sauce mash with added glucose and/or tyrosine was fermented for 152 days in the presence or absence of oxygen. Glucose negatively affected brown pigmentation either singly or with tyrosine. Tyrosine-added soy sauce mash initially browned at the same rate as the control mash until 127th day and then the former continued to brown at the same steady rate while the control mash stopped further browning. Aerobically incubated mash browned much more than anaerobically incubated one when the browning was compared on the 152nd day of fermentation. More than half of the mash browning was found to be due to the oxygen-related browning during the limited 152 days of fermentation time. Both oxygen-related and oxygen-unrelated browning reactions were found to contribute to the browning of soy sauce mash. Oxygen-related browning, however, was found to be more important than the Maillard browning reaction.

  • This study was carried out to investigate the oligosaccharide and amino acid utilization by mixed cultures during soy yogurt fermentation. Three types soy yogurt were prepared by fermenting with Lactobacillus acidophilus and Streptococcus thermophilus, Streptococcus thermophilus and Saccharomyces uvarum, Lactobacillus acidophilus and Saccharomyces uvarum. The utilized amount of oligosaccharide and amino acid was determined by HPLC during the fermentation period. The oligosaccharide and amino acid utilization efficiency of S. thermophilus and Sac. uvarum was greater than the other mixed cultures. It was found that Sac. uvarum produced enzymes which can convert oligosaccharide and common sugars in soy milk into glucose, galactose and fructose which can be fermented by L. acidophilus and S. thermophilus, and in turn stimulated acid production and amino acid utilization of the latter.

  • The effect of yogurt containing Hamcho (Salicornia herbacea) extract (Hamcho yogurt: HY) on the lipid concentrations in male Sprague-Dawley rats fed a cholesterol diet were studied. Rats were assigned three groups in the experiment; cholesterol diet (the CHOL group), cholesterol diet plus yogurt by lactic acid bacteria (the Y group) or cholesterol diet plus yogurt supplemented Hamcho extract by lactic acid bacteria (the HY group). Fermented milk with or without Hamcho extract in the diet supplemented at the levels 5.0% for 3 weeks. The concentrations of total cholesterol and bile acid in serum was significantly lower in the HY group than the CHOL group. This effect was also observed in rats fed a diet supplemented fermented milk. This cholesterol-lowering effect was more pronounced in the HY group compared to the Y group. The atherogenic index was significantly lower in the HY or Y groups than the CHOL group. The concentration of non esterified fatty acid in serum was significantly lower in the HY group than the CHOL and the Y groups. These results suggested that Hamcho yogurt exert the hypocholesterolemic effect in cholesterol fed rats.
