  • The investigations of specific growth rate, specific alcohol production rate, cell yield, alcohol yield of K. marxianus LG were performed according to the sugar concentrations, 50, 80, 110, 190, and 250g/l of extracted solution of Jerusalem Artichoke. The functipnal relationship between specific growth rate, specific alcohol production rate, and alcohol concentrations were devoted study to. In case of low concentration of alcohol, the fuctions were linear relationships. But in the region of high concentration of alcohol, they expressed the exponential relationships. The growth rate of K. marxianus was prohibited at higher than 50g/l of alcohol concentrations regardless of concentration of residual sugar. Cell and alcohol yield showed the maximum values around 25g/l of alcohol concentraton without being related to initial sugar concentrations.

  • Functional relationship between the Fermentation characteristics of Functional relationship between the Fermentation characteristics of S. cerevisiae SIV-89 and the fermentation time was investigated. According to the increase in the initial sugar cone. from 50g/L to 80 g/L, maximum specific growth rate and maximum specific alcohol production rate were increased until 0.35 and 1.98. But the two values were decreased with increase of initial sugar cone. in the region of more that 80 g/L. Maximum alcohol yield and biomass yield were 0.45 and 0.15 respectively. However those vlaue were found to be reduced with the argument of initial sugar concentration. Sugar conversion was decreased with sugar concentration. When the sugar concentration was more than 190 g/L, the conversion was dropped below 70%. The increase of alcohol concentration in the fermentation broth induced the phenomenon of decline of metabolism. In case of more than 80 g/L of alcohol conc., biomass growth and alcohol production were completely stopped regardless of remaining sugar concentration.

  • It was investigated that the influent sugar concentration and the dilution rate have an influence on the ethanol fermentation characteristics at steady state in a chemostat culture of K. fragilis using Jerusalem Artichoke juice as substrate. And also the optimum condition of high ethanol productivity and low residual sugar output in the ethanol production by K. fragilis, was clarified to be given at a dilution rate of 0.22/hr and at an influent sugar concentration of 85g/l.

  • Fermentation characteristics of Kluyveromyces fragilis CBS 1555 with Jerusalem artichoke juice, in extractive ethanol fermentation in aqueous two phase systems composed of polyethylene glycol 20000 (PEG) and crude dextran(Dx), were investigated as a function of initial sugar concentrations, concentrations of ethanol formed, or fermentation time. Both specific ethanol production rate increased with decrease in concentrations of PEG and Dx in two-phase systems. Without being related to the compositions of aqueous two-phase system, maximum specific cell growth rate and maximum specific ethanol production rate were showed in the initial sugar concentration fo $80g/\ell$ and $120g/\ell$, respectively. The inhibition effects of ethanol on specific cell growth rate and specific ethanol production rate decreased with decrease in PEG concentration and in the range of 2.5 to 5% Dx. Specific cell growth rate and specific ethanol production rate was fitted as an exponential function and a hyperbolic function, respectively, of the concentrations of ethanol formed. Overall ethanol productivity increased with increase in initial sugar concentrations, and also the required time for the maximum productivity was so. Ethanol production rate by the elapsed fermentation time showed the maximum value in the initial sugar concentration of $160g/\ell$.

  • 굴절율 측정에 의하여 당 성분을 정량하는 방법을 이용한 장치로, 글루코오스의 환원성에 기초하여 산화ㆍ환원 반응을 이용한 적정법이나 흡광도 측정법과 비교하여 시약이 필요없고 시간이 단축되는 장점이 있다. 그리고, 보통의 굴절율 측정기(예로 Abbe refractometer)가 당 농도 0.5%일 때의 굴절율 값 단위로 측정되는 한계를 극복하여 본 당 모니터링 장치는 바탕 물질과의 굴절율 차이를 증폭한 값으로 data(differential refractive index)를 얻을수 있으므로 글루코오스 농도 0.01% 단위까지 정량할 수 있어서 0.1% 이하의 저농도도 측정 가능하며 그 이상의 농도 범위에서도 측정 오차를 줄일수 있는 장점이 있다.   • The human granulocyte-macrophage colony-stimulating factor (hGM-CSF) was produced from cell suspension culture of transgenic tobacco which was transformed by using Agrobacterium harboring the hGM-CSF gene. To improve the production of hGM-CSF in batch culture system, the effects of initial sucrose concentration and inoculum size were investigated. The results show that the hGM-CSF production was not affected by small inoculum size in medium containing either low or high concentration of sucrose. However, the production of hGM-CSF was increased under increasing of the inoculum sizes and sucrose concentration. Under the combination of inoculum and sucrose concentration, the maximum hGM-CSF production of 720 $\mu$g/L was obtained at 90 g/L of initial sucrose concentration and 110 g/L of inoculum size.

  • The effect of concentration of carbon and nitrogen sources on the sporulation and the life cycles of three strains of Zygosaccharomyces was investigated. The results are as follows: 1) The good sporulation of Zygosaccharomyces bisporus, delbruekii, and Z. steineri was obtained on solid medium containing 0 to 0.001% of nitrogen and 10 to 20% of glucose. The high content of nitrogen was detrimental to sporulation and asci were formed under 0.01% of nitrogen. 2) It is widely accepted that the life cycle of Zygosaccharomyces proceeds in the following way: Ascospore...Vegetative cells...Conjugation of vegetative cells...Sporulation...Ascopores But zygotes of Z. bisporus proceeded to vegetative cells when transfered to the suitable medium for vegetative reproduction, and then formed asci.

  • To increase the production of ethanol by using sugar from lignocellulosic biomass, pentose and hexose have to be fermented simultaneously by yeast. The effects of mixed sugar and nitrogen on ethanol production by immobilized Pichia stipitis KCCM 12009 were investigated. When optimal mixed sugar and nitrogen concentration were 5% (Glucose/Xylose = 3:1) and 1%, respectively, ethanol concentration produced by immobilized P. stipitis was 19-20 g/L. In repeated fed-batch by immobilized P. stipitis, all glucose was consumed very quickly at 1-3% mixed sugar concentration. But, xylose consumption was decreased as the mixed sugar concentration increased. Also, ethanol (5.6 g/L) was stably produced and ethanol production rate was 0.13 g/$L{\cdot}h$ in immobilized cell reactor (ICR) with 1% mixed sugar (Glucose/Xylose = 3:1) as feeding media.

  • Fermentation characteristics of Jerusalem Artichoke of yeast K.fragilis CBS 1555 were investigated experimentally and quantitatively according to the change of initial sugar concentrations and initial PHs of fermentation broth. Initial sugar concentrations employed were 26, 45, 65, 105, 180, and 215g/1. And initial PHs of fermentation broth were 3, 5.5, 7 and 9. The maximum specific growth rate was observed as 0.4hr-1 at 65g/1 of initial sugar concentration. The maximum specific alcohol production rate was 1.68g/ghr at 105g/1 of initial sugar concentration Cell yield and ethanol yield represent the maximum values such as 0.14 and 0.49 respectively when the initial sugar concentration was 25g/1. The maximum of ethano1 fermentability, 97% was obtained at the initial concentrations, 26 and 45g/1. However, the maximum of total ethanol yield productivity was 2.78g/1hr when the initial concentration was 215g/1. And also the optimum PH was found 5.5 for both specific growth rate and specific alcohol production rate.

Effects of Concentrations of Glucose and Maltose on the Growth of Recombinant E. coli (재조합 대장균의 세포성장에 대한 포도당과 맥아당 농도의 영향)

  • The growth of recombinant E. coli and formation of the by-products were investigated. Glucose was consumed in approximately 8hours of cultivation when initial glucose concentration was 5g/1. When higher initial glucose concentrations were employed, it took approximately 16 to 20hours for the glucose to be consumed. When maltose or glucose was used as a carbon source, lactic acid and acetic acid were main by-products, while propionic acid and methanol production was not significant. Much by- products were formed at the higher initial carbon source concentrations. Higher concentration of lactic acid was observed when glucose was used, while higher concentration of acetic acid was observed when maltose was used. Lactic acid production affected the cell yield while acetic acid production affected the growth rate.
