Serum Fat Soluble Vitamins in Bile Duct Ligated Rats (담도 결찰한 백서에 있어서 담즙산 및 UDCA 투여에 따른 혈중 지용성 비타민의 농도 변화에 관한 연구)

  • Purpose: The aims of this study are to measure the serum levels of fat soluble vitamins (vitamin A and D) from bile duct ligated rats, and to evaluate the effect of oral bile acids administration to facilitate absorption of fat soluble vitamins. Methods: We measured serum ALT, total bilirubin, vitamin A, and vitamin D of Sprague-Dawley rats 1 week before and 4 weeks after experimental bile duct ligation. Rats were consisted with 3 groups. Group 2 had been fed bile acids and group 3 ursodeoxycholic acid after operation for 4 weeks. Multi-vitamin was given to all groups. Results: 1) Base line (mean value before duct ligation): ALT 74.2 IU, total bilirubin 0.26 mg/dL; vitamin D 13.01 ng/mL vitamin A $0.87\;{\mu}g/mL$, total bile acids $25.16\;{\mu}mol/L$. 2) Four weeks after ligation: ALT 100.7 IU, total bilirubin 2.58 mg/dL; vitamin D 7.89 ng/mL vitamin A $1.37{\mu}g/mL$, total bile acids $278.22\;{\mu}mol/L$. 3) 4 weeks after ligation, each group (group 1, group 2 and group 3) showed vitamin D (7.62, 8.10 and 7.99) ng/mL, vitamin A (1.68, 1.06 and 1.33) ${\mu}g/mL$, total bile acids (233.17, 345.80 and 268.57) ${\mu}mol/L$, which were statistically not significant. Conclusion: Serum level of vitamin A is increased after bile duct ligation although vitamin D is decreased. Oral administration of bile acids does not affect the serum levels of vitamin A and D in bile duct ligated rats.

  • Chitosan has been known to have hypocholesterolemic and hypolipidemic effects in animal studies. Chitosan also absorbs bile acids in vitro and in vivo, which might result in the hypocholesterolemic action. Trialkyl chitosan derivatives were prepared and tested for bile acid absorption activity in vitro. The derivatives showed enhanced absorption capacities which were comparable to cholestyramine.

폐염균 다당류 정제 방법의 개발 및 한국형 폐염균의 수집

  • 한국에서 유행하는 페염균 type을 결정하기 위한 기초 실험을 수행하여 서울대병원과 서울중앙병원의 환자로부터 분리한 88개의 폐염균주를 담즙산에 의한 용해도 및 Optochin에 대한 민감성 등을 측정하고 폐염균의 type을 결정하기 위해서 Double diffusion, Countercurrent immunoelectrophoresis, Rocket immunoelectrophoresis 방법 등으로 폐염균 항혈청과 반응하였다. 또한 ATCC로부터 구입한 폐염균주를 이용하여 배지조성의 변화에 따른 폐염균 생장을 측정하기 위해 CAT broth, Brain Heart Infusion broth, Defined Media에서 3가지 type의 균주(Type I, III, IV)를 접종하고 37$^{\circ}C$에서 배양하면서 1시간마다 550nm에서 흡광도를 측정하였다. 3가지 type (Type I, III, IV)의 폐염균주로부터 협막다당류를 phenol Iysis 추출법으로 폐염균 Capsule 다당류의 분리하여 Total Sugar 함량, Uronic acid 함량, 0-Acetyl 함량, 단백질 함량, 핵산 함량, 분자량분포 Countr-current immunoelectrophoresis 등을 실시하여 폐염균 백신 규격에 부합되는지 확인하였다. 수집한 88개의 폐염균주가 모두 담즙산에 의해 용해되고 6mm Optochin disc를 사용하였을때 성장 저지원이 모두 14mm 이상으로 나타나 감수성이 있어서 폐염균임을 확인하였으며 폐염균의 type 결정에 필요한 방법을 설정하였다. 배지조성에 따른 폐염균의 생장을 측정한 결과 CAT broth 에서는 Type I만이 잘 배양되었고 Type III, IV는 늦게 배양되었으나, Brain Heart Infusion broth에서는 3가지 Type이 모두 잘 배양되었고, Defined Media 에서는 Type I만이 성장되었을 뿐 나머지 두 Type은 배양되지 않았다. 본 실험방법으로 분리된 폐염균 다당류는 폐염균 백신 규격중의 6가지 항목에서 적합하였으며 Statens(Denmark)로부터 구입한 항체와 뚜렷한 침강선을 나타내었다.

  • Fucoidan from sea tangle (Laminaria japonica) was isolated by hot water extraction, and partially purified. The in-vitro glucose and bile acid retarding effects of the partially purified fucoidan were investigated. Fucoidan exhibited 27.06$\sim$21.42% of retarding index for glucose and 33.50$\sim$27.02% of retarding index for bile acid during in vitro dialysis experiment for 2 hours. These retarding effects on glucose and bile acid diffusion was considered as a relatively good or very good, suggesting the prevention from diabetes and arteriosclerosis of some extent.

  • Bile salt hydrolase (BSH, EC 3.5.1.24) activity, which cleaves amide bond between carboxyl group (bile acid) and amino group (glycine or taurine), is commonly detected in gut-associated species of human and animal. During the screening of BSH active strains from the fecal samples of elderly human volunteers, strain CU30-2 was isolated on the basis of the highly active BSH producing activity. A bsh gene of the isolate was cloned into the pET22b expression vector and overexpressed in Escherichia coli BL21 (DE3) Gold by induction with 1mM IPTG. The overexpressed BSH enzyme with 6x His-tag was purified with apparent homogeneity using a $Ni^+$-NTA agarose column and characterized. The BSH enzyme of E. faecalis CU30-2 exhibited approximately 50 times higher activity against glycol-conjugated bile salts than tauro-conjugated bile salts having the highest activity against glycocholic acid. Considering the prevalence of E. faecalis strains in the human GI tract and glycol-conjugates dominated bile acid composition of human bile, further study is needed to investigate the impact of the BSH activity exerted by E. faecalis strains to the host as well as to the BSH producing strains.

Hypocholesterolemic Effect of Yogurt Supplemented Salicornia herbacea Extract in Cholesterol-Fed Rats. (콜레스테롤 급여 흰쥐에서 함초 요구르트의 콜레스테롤 저하효과)

  • The effect of yogurt containing Hamcho (Salicornia herbacea) extract (Hamcho yogurt: HY) on the lipid concentrations in male Sprague-Dawley rats fed a cholesterol diet were studied. Rats were assigned three groups in the experiment; cholesterol diet (the CHOL group), cholesterol diet plus yogurt by lactic acid bacteria (the Y group) or cholesterol diet plus yogurt supplemented Hamcho extract by lactic acid bacteria (the HY group). Fermented milk with or without Hamcho extract in the diet supplemented at the levels 5.0% for 3 weeks. The concentrations of total cholesterol and bile acid in serum was significantly lower in the HY group than the CHOL group. This effect was also observed in rats fed a diet supplemented fermented milk. This cholesterol-lowering effect was more pronounced in the HY group compared to the Y group. The atherogenic index was significantly lower in the HY or Y groups than the CHOL group. The concentration of non esterified fatty acid in serum was significantly lower in the HY group than the CHOL and the Y groups. These results suggested that Hamcho yogurt exert the hypocholesterolemic effect in cholesterol fed rats.

Effects of Dietary Supplementation of Sea Tangle Extracts on the Excretion of Neutral Steroids and Bile Acid in Diabetic Rats (다시마 추출물의 급여가 당뇨쥐의 중성스테로이드와 담즙산 배설에 미치는 영향)

  • The present study was conducted to investigate the effect of dietary supplementation of sea tangle extracts on the excretion of neutral steroids and bile acid in diabetic rats. Sprague-Dawley rats were fed on AIN-76 based experimental diets containing methanol extracts (2%, w/w) or water extracts (4%, w/w) of sea tangle for 4 weeks. And then they were induced to be diabetic by receiving streptozotocin (45 mg/kg BW) intramuscularly. The dried fecal weight was increased significantly in water extracts group compared with control group. The fecal content of cholesterol was higher in extracts groups of sea tangle than in control group. But there was no significant difference in fecal excretion of cholesterol between methanol and water extracts groups. The fecal excretion of coprostanol was increased significantly in water extracts group compared with the control group. The fecal excretion of bile acid was increased significantly in sea tangle extracts groups compared to the control group. These data suggest that dietary supplementation of sea tangle extracts might reduce the incidence of atherosclerosis through increasing the excretion of neutral steroids.

Hypocholesterol Effect of Opuntia humifusa Extract on High Cholesterol Diet-induced Hypercholesterolemic Rats (고콜레스테롤혈증을 유도한 쥐를 대상으로 천년초 추출물의 콜레스테롤 저하 효과)

  • The aim of this study was to investigate the hypocholesterol effects of Opuntia humifusa extract in hypercholesterolemic rats. Rats (8-week-old, male) were randomly divided into four groups (n=4) as follows: N-control, normal diet; H-control, hypercholesterolemic diet; OH-1, 2% O. humifusa extract-supplemented hypercholesterolemic diet; OH-2, 4% O. humifusa extract-supplemented hypercholesterolemic diet. After 4 weeks, we observed that hypercholesterolemia induced significant increases in serum lipids (total cholesterol and LDL-cholesterol), hepatic lipids (total cholesterol and triglyceride), and hepatic function parameters (ALT, alanine aminotransferase; AST, aspartate aminotransferase) (P<0.05). Treatment with O. humifusa extract for 2 weeks normalized these indexes up to the levels of normal rats. O. humifusa extract tended to increase fecal lipid (H-control: 142.74 mg/day vs. OH-1: 214.05 mg/day; OH-2: 200.95 mg/day, P<0.05) and bile acid (H-control: 37.07 ${\mu}M/day$ vs. OH-1: 47.23 ${\mu}M/day$, OH-2: 47.93 ${\mu}M/day$, P<0.05) contents of hypercholesterolemic rats. We concluded that oral administration of O. humifusa extract effectively improved cholesterol metabolism in a hypercholesterolemic animal model induced by hypercholesterol diet.
