The composition and content of amino acid and free amino acid were separately compared in cap and stipe of fruitbody obtained from different color strains in winter mushroom and oyster mushroom strains. The result showed different kinds and amount of amino acids according to the mushroom parts, strains and kinds. Tryptophane was not detected in any mushrooms. Alanine was also not detected in all oyster mushroom strains. The content of glutamic acid was the highest and that of methionine, phenylalanine and cysteine were very low in all mushroom strains. When compared by parts, the ratio of amono acids showed similar tendency in composition. Generally the content of amono acids showed higher in caps than in stipes. The profile and content of free amino acid showed a lot of variation between cap and strip in the intra and inter mushrooms.
The present study aimed to investigate the biological activities and nutritional contents of extracts from three types of edible mushrooms (Pleurotus ostreatus, P. eryngii, and Hericium erinaceus) following different roasting treatments. Roasting by a household multi-oven (HR60) decreased the total polyphenol contents, as well as DPPH radical scavenging and nitrite scavenging activities compared with the fresh samples, and no significant difference was observed between the different roasting times. As a result of rotary drum roasting (CR) treatment, we could observe a decrease in the DPPH radical scavenging activity in all roasted mushroom samples compared with the fresh samples, whereas nitrite scavenging activity increased in roasted P. eryngii and H. erinaceus. Additionally, total polyphenol contents increased significantly in roasted H. erinaceus. In each roasted sample dried by HR60 and CR, the amount of palatable-taste amino acids (e.g., glutamic acid) and sweet-taste amino acids, including glycine, serine, alanine, and threonine, increased in P. ostreatus, whereas the amount of bitter amino acids, including leucine, isoleucine, histidine, valine, and methionine, decreased compared with the fresh samples. In P. eryngii and H. erinaceus samples, we also observed an increase in the amount of palatable and sweet-taste amino acids upon the roasting treatments; however, the amount of bitter amino acids increased simultaneously following the roasting treatments. These results suggest that CR treatment is more effective in increasing the biological activities and nutritional contents, such as palatable and sweet-taste amino acids, compared with HR60 treatment.
To develop a new cultivar of King oyster mushroom white variety (Pleurotus ferulae), GW10-68 as parental strain was selected by the method of Di-mon crossing between monokaryotic strain ASI 2798-24 derived from ASI 2798 and dikaryotic strain ASI 2803. The GW10-95(ASI 2803 ${\times}$ ASI 2798-24) was shown the best cultural characteristics, selected to be a new cultivar and designed as 'Beesan No.2'. The 'Beesan No.2' was formed incompatibility line distinctly in the confrontation growth of parental strains ASI 2803, ASI 2798 and Beesan No.1. Analysis of the genetic characteristics of the new cultivar 'Beesan No.2' showed a different DNA profile as that of the control strains, ASI 2803, ASI 2798 and 'Beesan No.1', when RAPD(Random Amplified Polymorphic DNA) primer URP4 was used. The optimum temperature and pH arrange for mycelial growth were $25^{\circ}C$ and pH5~8, respectively. Fruiting body production per bottle was about 123.3 g. And also the stipe was long. This new cultivar 'Beesan No.2' of Pleurotus ferulae was characterized white-like variety of King oyster mushroom in the color of stipe, that was long and low yield compared to that of other cultuvar 'Beesan No.1'. We therefore expect that this new strain will increase of export and substitute cultivar of King oyster mushroom.

This study was conducted to prepare healthy vinegars using the mycelia of various mushrooms (Cordyceps militaris, Phellinus baumi, Pleurotus cornucopiae, and Ganoderma lucidum). Analyses of the bioactive components of these vinegars revealed that those prepared using mushroom mycelial cultures on fermented brown rice had increased bioactive component levels compared to those cultured on unfermented brown rice. The highest beta-glucan contents (78.7 mg/g) were found in P. baumi cultures and 0.34mg/g of cordycepin was detected in C. militaris cultures. Regarding the chemical properties of these mushroom mycelia vinegars (MMVs), those prepared using C. militaris and P. cornucopiae cultures showed higher acidity and reduced sugar content. Sensory assessments of C. militaris and P. cornucopiae MMVs yielded scores of 7.3 and 6.6, respectively. Analyses of bioactive components revealed that C. militaris MMV contained high levels of polyphenols and 786 mg/L of cordycepin. P. cornucopiae MMV contained the highest amount of beta-glucan (20.9 mg/g).
For development of a method for differentiation of Pleurotus eryngii cultivars, simple sequence repeats (SSR) from whole genomic DNA sequence analysis was used for genotyping and two multiplex-SSR primer sets were developed. These SSR primer sets were employed to distinguish 12 cultivars and strains. Five polymorphic markers were selected based on the genotyping results. PCR using each primer produced one to four distinct bands ranging in size from 200 to 300 bp. Polymorphism information content (PIC) values of the five markers were in the range of 0.6627 to 0.6848 with an average of 0.6775. Unweighted pairgroup method with arithmetic mean clustering analysis based on genetic distances using five SSR markers classified 12 cultivars into two clusters. Cluster I and II were comprised of four and eight cultivars, respectively. Two multiplex sets, Multi-1 (SSR312 and SSR366) and Multi-2 (SSR178 and SSR277) completely discriminated 12 cultivars and strains with 21 alleles and a PIC value of 0.9090. These results might be useful in providing an efficient method for the identification of P. eryngii cultivars with separate PCR reactions.
This research aimed at developing the efficient method and device applicable to the inoculation of mushroom using (spawn was intended to utilize) liquid spawn for the stable production of mushroom. For the mass production of liquid culture, optimal inoculum volume and cultural period were $5{\sim}6%$ and 4 days for Flammulina velutipes and 4% and 5 days for Pleurotus ostreatus. Fruit body weight in 850 ml polythene bottle was highest at 6% or 15 ml liquid inoculum for P. ostreatus and $4{\sim}6%$ or $10{\sim}15ml$ for F. velutipes. Weight of fruit body by the application of liquid spawn inoculation system increased up to 33.7% for P. ostreatus and 32.8% for F. velutipes, respetively, compared to conventional spawn making system. The system of liquid spawn inoculation was successfully operated without malfunction in opening or closing the lid, and it took 26 min to inoculate 1200 bottles.
This study investigated the possibility of using pine tree sawdust in thinning-out tree as a substitute for poplar and douglas-fir tree sawdust in oyster mushroom cultivation. Mycelial growth was 10.8 cm in medium supplemented with 70% pine sawdust after 18 days of culture and 10.2 cm in control medium. Mycelial density showed high density with no significant difference between treatments. Fresh weight and diameter of fruiting bodies were slightly lower in medium supplemented with pine tree sawdust. The hardness of pine tree sawdust was slightly higher in the medium supplemented with 20% and 30% of pine tree sawdust. The L value showed a tendency to be higher in the pine added medium, but the a and b values d id not show any significant difference between the treatments. The yields of fruiting body were 157 g / 850 ml for the control and 170 g / 850 ml for the 40% added pine tree sawdust.

느타리버섯은 전남 장흥군에서 병재배로 재배된 '곤지 7호' 느타리버섯 품종을 $3^{\circ}C$에서 1일간 냉각시킨 후 실험에 사용하였다. 이산화탄소 처리는 10mm 두께의 아크릴로 제작한 밀폐 챔버($40{\times}70{\times}40cm$)를 이용하여 CO2가스를 주입하여 내부의 CO2 농도를 $30{\pm}1%$로 조정하여 3시간 동안 밀폐시켰으며 $3^{\circ}C$ 저온 저장고에서 수행하였다. 처리 후 포장필름은 3처리로 하여 PVC랩, 재배농가에서 이용하는 필름인 $20{\mu}m$ OPP필름, $30{\mu}m$ OPP필름을 재료로 하여 산소투과율을 $5,000cc/m2{\cdot}day{\cdot}atm$로 조정한 필름(OPP5)을 사용하였다. PVC랩 처리는 스티로폼 트레이에 옮긴 후 균일하게 한 겹으로 둘렀고, OPP와 OPP5는 봉지에 옮겨 봉지 입구 4cm 안쪽을 비닐접착기로 열접착하여 밀봉하였으며 $3^{\circ}C$에 저장하면서 필름내부의 기체조성, 경도, 색도, 이취, 종합선도 변화를 조사하였다. 포장된 느타리버섯의 포장재 내 이산화탄소 농도는 저장초기 0.03%에서 점차 증가하여 저장 3일 후 PVC랩은 3.5~3.9%, 나머지 처리는 18%이상으로 증가하였으며 OPP필름보다 OPP5필름의 경우 포장재 내 이산화탄소 농도가 더 낮게 유지되었다. 생체중 감소는 PVC랩에서 저장 26일 후 9.9% 이하로 육안으로 보이는 시들음이 관찰되었으며 나머지 처리들은 저장 26일 후까지 1.6% 이하의 중량감소율을 보였다. 느타리버섯 갓 경도는 유지되었으며 줄기표면의 황색도 $b^*$값은 PVC랩에서는 증가 경향을, OPP나 OPP5에서는 유지 경향을 보였다. 느타리버섯의 이취 및 전체적인 품질은 무처리 후 PVC랩 포장은 저장 6일 후에 이취가 상품성 한계로 발생하였고 OPP5 포장은 포장재 중 가장 이취발생이 늦게 발생하였으며 CO2 처리에 의해 지연되는 것을 알 수 있었다. 이런 결과를 종합하여 느타리버섯의 $3^{\circ}C$ 저장 중 품질유지기간은 각 PVC랩(6일) ${\rightarrow}$ 30% CO2+PVC랩(7일) ${\rightarrow}$ OPP(10일) ${\rightarrow}$ 30% CO2+OPP(17일) ${\rightarrow}$ OPP5(20일) ${\rightarrow}$ 30% CO2+OPP5(22일)로 조사되었다. CO2처리 후 산소투과율을 $5,000cc/m2{\cdot}day{\cdot}atm$으로 조정한 미세천공(OPP5)필름이 가장 높은 전체적 품질점수를 나타내었으며 상대적으로 낮은 이취발생, 갈변과 갓무름이 적어 높은 점수를 얻는데 영향을 준 것으로 보인다.
Pleurotus eryngii, a white rot fungus, produces two extracellular lignin-degrading enzymes, laccase and manganese peroxidase (MnP). Owing to these enzymes, P. eryngii efficiently degrades synthetic chemicals such as azo, phthalocyanine, and triphenyl methane dyes. In this study, we investigated the degradation processes of four aromatic dyes, congo red (CR), methylene blue (MB), crystal violet (CV), and malachite green (MG), by P. eryngii under solid and liquid culture conditions. CR and MG were the most quickly degraded under solid and liquid culture conditions, respectively. However, compared to CR, CV, and MG, MB was not degraded well under both culture conditions. The activities of ligninolytic enzymes (laccase and MnP) were also investigated. Laccase was identified to be the major enzyme for dye degradation. A positive relationship between decolorization and enzyme activity was observed for CR, MB, and CV degradation. In contrast, decolorization of MG ensued after high enzyme activity. These results indicate that the degradation process differs between MG and the other aromatic dyes. Therefore, P. eryngii could be a potential tool for the bioremediation of synthetic aromatic dye effluent.
We conducted an on-site application study at the livestock cooperative fertilizer plant to compare the composting period, temperature change, moisture content, and chemical properties between livestock manure compost using sawdust as a moisture regulator with those using spent oyster mushroom substrate. The composting period, moisture content, and fertilizer composition of compost containing spent oyster mushroom substrate did not differ from that of conventional compost mixed with sawdust after the first and second fermentation and post-maturation stages, it was suitable as a material for manufacturing livestock manure compost. The spent oyster mushroom substrate also lower the production cost of livestock manure compost by replacing the more expensive sawdust. The developed technology is expected to contribute towards the utilization of by-products of the oyster mushroom harvest while simultaneously producing high quality livestock manure compost.
