Production of Lignocellulytic Enzymes from Spent Mushroom Compost of Pleurotus eryngii (큰느타리버섯 수확 후 배지로부터 리그닌섬유소분해효소 생산)

  • The lignocellulytic enzymes including a-amylase (EC 3.2.1.1), lignin peroxidase (EC 1.11.1.14), laccase (EC 1.10.3.2), xylanase (EC 3.2.1.8), ${\beta}$-xylosidase (EC 3.2.1.37), ${\beta}$-glucosidase (EC 3.2.1.21) and cellulase (EC 3.2.1.4) were extracted from spent mushroom compost (SMC) of Pleurotus eryngii. Different extraction buffers and conditions were tested for optimal recovery of the enzymes. The optimum extraction was shaking incubation (200 rpm) for 2 h at $4^{\circ}C$. ${\alpha}$-Amylase was extracted with the productivity range from 1.20 to 1.6 Unit/SMC g. Cellulase was recovered with the productivity range from 2.10 to 2.80 U/gf. ${\beta}$-glucosidase and ${\beta}$-xylosidase productivities showed lowest recovery producing 0.1 U/g and 0.02 U/g, respectively. The P. eryngii SMCs collected from three different mushroom farms showed different recovery on laccase and xylanse, cellulase. Furthermore, the water extracted SMC was compared to commercial enzymes for its industrial application in decolorization and cellulase activity.

Influence of Soaking Time of Paddy Straw on the Mycelial Growth of Oyster Mushroom, Pleurotus spp. (배지의 침수조건이 느타리버섯의 균사생장에 미치는 영향)

  • The purpose of this study was to investigate the change of water content in mushroom substrate, paddy straw, and the mycelial growth of oyster mushroom by various soaking time. The water content of paddy straw was gradually increased as the soaking time was getting longer. These experiments also showed that the water content in the upper part of paddy straw was less than that of the low part. The mycelial growth was the good between 4 and 12 hours soaking treatment, showing faster growth and higher density in the lower part than the upper part. However we could not found any significant variation among 5 strains of oyster mushroom in the effects of the various soaking times and soaking part. The pH of the lower part substrates had a low pH compared with that of the upper part. When pathogens and mushroom mycelia were co-inoculated on paddy straw under various soaking conditions, the mushroom mycelial growth showed fast and high density according to the increase of the soaking time. The experiment related to water transport reveals that it is not easy to move into upper part, but only soaked part by adding water is available to absorb enough water. Old straw was much better than the straw new this year one for water absorption, mycelial growth and mycelial density.

Isolation Method of Mushroom Infesting Pests from Mushroom-Growing Compost (버섯배지에서 버섯해충류 분리방법)

  • We investigated the isolation method of mushroom infesting pests, Lycoriella mali, Coboldia fuscipes, Histiostoma sp. from mushroom-growing compost. Sugar solution of different densities (0, 10, 20, 30, 40, 50%) was tested to provide a means of seperating mushroom pests from the compost media. Thus, 40% sugar solution was suitable for isolation. The sieve size to entrap the pests was $30{\sim}140$ mesh; Lycoriella mali was trappped mainly $30{\sim}65$ mesh sieve, Coboldia fuscipes was caught mainly $30{\sim}100$ mesh sieve, Histiostoma sp. was trapped mainly $65{\sim}140$ mesh sieve. An isolation procedure was as follows; The infested compost was disintegrated in water and poured onto a set of 16, 30, 80, 140-mesh sieve. The material caught in the sieve is added in 40% sugar solution and then most compost particle were massed at the bottom while the supernatant contains mushroom pests. The upperlayer material was poured into a Seperatory funnel and the sediment at the bottom is drained off. The remaining material are washed off examination dish for study.

Genetic Variability of Pleurotus ostreatus Monospore Isolates by Random Amplified Polymorphic DNA Analysis (DNA 다형성 분석에 의한 느타리버섯 단포자 분리주의 유전적 변이성)

  • This study was carried out to obtain data concerning the genetic variability of Pleurotus ostreatus. Monospores of P. ostreatus were isolated and cultured to estimate differences in the rate of mycelial growth and genetic similarity among the isolates. Although the overall growth rates were slow compared to their parental dikaryon except 2-MI 4, significant differences in the rate of mycelial growth were observed among isolates. Random amplified polymorphic DNA (RAPD) analysis using twenty six random primers showed 345 polymorphic DNA bands from 35 monospore isolates and their parental dikaryon. DNA bands ranged from 0.1 to 4.0 Kb in size. Most various polymorphic DNA bands within monospore isolates were obtained when we used J (OPA-01) or W (OPB-04). The 36-MI 103 showed totally different band patterns from those of the others. RAPD analysis revealed that there is approximatly 75% genetic similarity between monospore isolates and their parental dikaryon except 36-MI 103, which showed only 49% genetic similarity. In addition, genetic similarity degrees were classified into four groups: I (parental dikaryon), II (fast growing type), III (moderate growing type), and IV (slow growing type). However, there is no correlation between mating type and mycelial growth rates.

Studies on the Volatile Aroma Components of Pleurotus ostreatus (느타리버섯의 향기(香氣) 성분(成分)에 관한 연구(硏究))

  • The volatile aroma concentrates were isolated from Pleurotus ostreatus by simultaneous distillation-extraction and analyzed by gas chromatography and combined gas chromatography-mass spectrometry. The aroma concentrates obtained had a typical raw or slightly cooked mushroom-like odor, and the yield was 123 ppm. Of 27 components identified, the major components were 3-octanone, 3-octanol, 2-octenal, n-octanol and 2, 4-decadienal, including 1-octen-3-ol contributing 67.06% of the total volatiles, and the $C_8$ compounds comprised about 80% of the total volatiles. On the other hand, the $C_6$ and $C_8$ alcohols in mushroom increased gradually with increase of standing time after homogenation, while aldehydes, ketones, and linoleic acid decreased, and also $C_6$ and $C_8$ compounds were significantly increased when linoleic and linolenic acid were added in homogenates. It appears that most of volatile aroma components in Pleurotus ostreatus were probably produced enzymatically from unsaturated fatty acids containing cis, cis-1, 4-pentadiene moiety.

Enzyme-Linked Immunosorbent Assays of Pseudomonas tolaasii, a Bacterial Brown Blotch Pathogen of Oyster Mushroom. (느타리버섯 세균성갈반병균 Pseudomonas tolaasii의 효소면역검출법)

  • For simple and rapid detection of Pseudomonas tolaasii (PT), a bacterial brown blotch pathogen of oyster mushroom, enzyme-linked immunosorbent assays (ELISA) were developed. To produce specific antibody, PT ($5{\times}10^7$ cfu) and Freund's adjuvant were subcutaneously immunized into rabbits several times. By using the antiserum showing the highest titer, we established noncompetitive and competitive ELISA's. Standard curves of the ELISA's showed that the detection limits were $2{\times}10^2$cfu/ml and $3{\times}10^2$cfu/ml, respectively When investigated by noncompetitive ELISA, cross reactivities of the anti-PT antibodies against P. agarici, P. reactans, and other fluorescent Pseudomonas spp. were very low (<1/10$^3$), but those against P. solanacearum, Erwinia chrysanthemi, Streptococcus mutans, Xanthomonas citri, and a fungus Fusarium oxysporum were almost none. However, when investigated by competitive ELISA, the reactivities against any other strains except PT were almost none. When the ELISA's were applied to 18 strains derived from mushrooms in order to identify PT, only 11 strains showing both pathogenicity and white line reactivity were obviously positive. These results showed that the ELISA's could be convenient tools to detect PT in accordance with existing methods.

Anti-cancer Activity of Lentinus edoeds and Pleurotus astreatus (표고버섯과 느타리 버섯의 항암효과)

  • The effects of mushrooms such as Lentinus edodes and Pleurotus ostreatus on anti-cancer activity through in vivo and in vitro experiments were powders of protein-bound polysaccharides in mushrooms were solubilized in 0, 5, 25 mg/kg saline, respectively and were used in vitro experiments. The in vivo experiments were carried out as followed: i) anti-cancer activities on Leukemia $(L_{1210})$, Hepatlicus cancer $(H_{22})$ and Sarcoma180/(S180), and ii) the effect on immune system through changes in intestine weight and the number of hemolytic plague forming cells. Protein-bound polysaccharides of all showed anti-cancer activity on $L_{1210}$ and fruit body of Lentinus edodes 25 mg/kg treatment group showed the highest inhibition rate (86%). Pleurotus ostreatus mycelial in medium of cultivate 25 mg/kg treatment. Fruid body of Lentinus edodes 25 mg/kg treatment group showed the highest inhibition rate (86% and 71%, respectively) on $H_{22}$ among them. The inhibition rates of fruit body and mycelial of Lentinus edodes 25 mg/kg treatment groups on S180 were 33.9% and 30.9%, respectively. Each samples of 50, 100, 200, $400\;{\mu}g/{\mu}L$ on in vitro cell toxicity test did not show significantly different cell death rates at P<0.05. In immune test, weights of liver and spleen were increased according to increase in conc. but were not significantly different at P<0.05. The weights of thymus were heavy in fruit body and mycelial of Lentinus edodes treatment group but were not significantly different at P<0.05. Hemolytic plague forming cells with antibody formation capability were significantly high in fruit body and mycelial of Lentinus edodes treatment samples.

Enhanced Antitumorigenicity and Antimutagenicity of Doenjang Prepared from Mushroom Mycelia-cultured Traditional Mejus (재래식 버섯균사체 된장의 항종양성과 항돌연변이성)

  • Antitumorigenic and antimutagenic activities of the doenjangs prepared from mushroom mycelia-cultured traditional mejus (designated to MTDJ) were investigated using the model of Sarcoma-180-induced mouse ascites cancer, and 2-amino-3-methylimidazo [4,5-f] quinoline (IQ) and aflatoxin B$_1$ (AFB$_1$) -mediated S. typhimurium mutagenicity, respectively. Antioxidative activity of MTDJ was also investigated using the mouse liver microsome system. Mushroom stains used for the preparation of the mushroom mycelia-cultured traditional mejus were Synryeong (Agaricus blazei), Yeonggi (Canoderma Iucidum), Sanghwang (Phellinus linteus), and Neutari (Pleurotus ostreatus). All MTDJS showed the enhanced antitumorigenicities (12% by Synryeong, 13% by Sanghwang, 16% by Yeonggi, and 19% by Neutari), antimutagenicity (6.1~20.8% for IQ and 3.1~10.2% for AFB$_1$), and antioxidative activity (6.6~46.5%), relative to the control doenjang. The $\beta$-D-glucan content (0.75~1.71 mg/g) of MTDJs was 3~8 times higher than that (0.22 mg/g) of the control doenjang. Genistein content (769~932 Ug/g) of MTDJS was also higher than that (728 Ug/g) of control doenjang The content of $\beta$-D-glucan and genistein was not exactly correlated to the antitumorigenicity and antimutagenicity of MTDJs. These results indicate that anti-tumorigenicity and antimutagenicity of MTDJS were elevated in comparison with the control doenjang, and the observed functions were, in part, derived from $\beta$-glucan and/or genistein in the MTDJS.

Effect of highly activated calcium on the growth of mushrooms (고활성칼슘의 버섯 생육에 대한 효과)

  • Highly Activated Calcium(below HAC) is the oxidized calcium made by dissolving shell materials with high voltage about 15,000V and high temperature ($1,500{\sim}5,000^{\circ}C$). This HAC is a material with a very high degree of purity without toxicant. This HAC decreases chemical reaction so the degree of being active and dissolving living material is outstanding. The effects of HAC on the propagation of mushrooms are following. In the case of the Pleurotus ostreatus, when controlling media moisture by mixing with the HAC, mycelium cultivating days were shortened by 2days. The day required for primordial formation after inoculation(DPI) were one day faster. The number of stem was 15 and individual weight was 248g/850cc, a 6.5% increase. In the case of Pleurotus eryngii, when controlling media moisture by mixing with the highly activated calcium, mycelium cultivating days were shortened by 3days. DPI were l day faster. The day required for colonization after inoculation was shortened by 6days and individual weight was 108.8g/850cc, a 9.7% increase. In the case of Flammulina velutipes, the highly activated calcium was the best for scraping up mycelium. Mycelium incubating days were shortened by 2days. DPI were shortened by 3days. The day required for colonization after inoculation was 1day faster and the period of cultivation was shortened by 3days. Individual weight was 165g/850cc, a 6.7% increase. In the case of Lentinula edodes, when mixing media with the highly cultivated calcium, cultivating days were shortened by 3 days. The days for becoming brown in color were 2 days faster and the days of the first harvest were shortened by 4 days. The weight of mushrooms was 169g/2kg, a 9.7% increase.

Effect of functional material addition on the growth and physiological activity materials in Pleurotus ostreatus (기능성물질 첨가시 느타리버섯 생육 및 생리활성물질의 변화)

  • This expriment was carried out on culture media which affect growth, development and physiological effect to Pleurotus ostreatus, when aqueous ferrum, red pepper powder, leaves of ginkgo biloba were added to culture media. The yield of fresh mushroom per 850ml PP bottle in chunchu-neutari was highest by 144g in 34.9g aqueous ferrum added, ferrous content don't affected to yield of mushroom, but variety of Pleurotus ostreatus affect to yield. Mineral content of fruit body and culture media in Suhan-neutari was different to mineral kind, ferrous content of furit body was similar to 0.31~0.43mg/kg when ferrous content of media was increased in 0.89mg/kg to 10.8mg/kg. The more content of red pepper powder in culture media was high, the more capsaicin content in fruit body was high. transferred capsaicin was highest by 6 mg/100g in Suhan-neutari. Total phenolics content were higher in culture media which was added aqueous ferrum and red pepper powder, but correlation don't showed total phenolics content among antioxydant activity, reducing power, chelating capacity. Macrophage activation was higher in Suhan-neutari than other Pleurotus spp.
