Effect of Sodium Hexametaphosphate on the Extractability of Sesame Meal Protein and Amino Acid Composition and Color of Its Protein Concentrate (Sodium Hexametaphosphate처리가 참깨박 단백질의 추출성과 농축단백질의 색도 및 아미노산 조성에 미치는 영향)

  • The effect of sodium hexametaphosphate(SHMP) on the extraction of defatted sesame meal protein and the color and amino acid composition of protein concentrate have been studied. The highest amount of protein could be extracted with 1.5% SHMP and the extraction was effective at pH 12.0. The extraction rate tended to increase with increasing the flour to solvent ratio and about 60% of protein was obtained when adjusted the ratio to 1 : 40. Color of sesame protein concentrate was slightly improved by SHMP treatment. Lysine and methionine content were decreased in SHMP-treated protein concentrate but valine and leucine content were increased.

  • 원자력발전소에서 생성되는 폐기물들의 양을 줄이는 것은 주요한 관심사이다. 방사성폐기물의 감용은 여러 방법으로 성취될 수 있는데, 본고에서는 액체폐기물 처리계통의 증발기를 효율적으로 운전하여 폐기물의 양을 감소시키는데 초점을 맞추었다. 이를 위해, 온도와 pH를 변화시키면서 농도가 다른 붕산수들의 침전상태가 조사되었는데, 배관의 보온을 고려하지 않았을 때는 붕산농도 $17wt.\%$까지, 고려했을 때는 $25wt.\%$까지 농축할 수 있음이 밝혀졌다. 또한, 붕산농도의 증가에 따른 시멘트고화체의 압축강도변화를 조사하였고 증발기 농축운전으로 얻어질 경제적 이들을 평가하였다.

  • Quality stability and utilization of sardine protein concentrates were investigated. pH, water activity and amino-nitrogen contents of autoclaved and boiled products were little changed during the storage of 60 days. Available lysine contents of the both products at the initial stage of storage were 5.58g/16g-N and 5.69g/16g-N, respectively. But the available lysine contents and digestibility of the both products decreased slightly with increasing of storage time. Lipophilic and hydrophilic brown pigment formation of the both products increased during storage of 60 days, but peroxide value(POV) and thiobarbituric acid(TBA) value decreased. Total amino acid contents of the both products were in the range of $88.99{\~}89.90g/16g-N$, and the predominant ones were glutamic acid, aspartic acid, leucine and lysine. From the sensory scores of model snack, it is concluded that the sardine protein concentrate can be used as a source material for snack.

  • There are various ions in seawater. In order to use seawater as the drinking water, some elements are to be concentrated and other elements are to be removed. To obtain these characteristics using seawater, it is necessary to adjust seawater quality. Because calcium and magnesium are especially healthful to human bodies, it is required to concentrate these elements. In this study, the technology to obtain the hard water from seawater by electerodialysis was investigated. After concentrated water was produced using nanofiltration membranes, sodium chloride was eliminated from the concentrated water by electrodialysis. The hard water production from seawater was successfully achieved using electrodialysis in this study.

  • A method to store concentrated milk in the liquid state at $-15^{\circ}C$ was developed, and quality changes during storage of milk were evaluated. Combined cryoprotectants (CCP) suitable for storing concentrated milk in the liquid state at $-15^{\circ}C$ were consisted of 17.74% sucrose, 8.87% glucose, 8.87% fructose, 2% glycerol, 0.25% sodium hexametaphosphate, 0.25% NaCl and 0.02% ascorbic acid. The amount of CCP to be added to concentrated milk to depress freezing point to $-15^{\circ}C$ was 38% by weight. Gelation due to protein denaturation was the most serious quality change during storage, which adversely affected appearance and utilization of the stored product. Gelation was observed after 3 weeks storage in the control, but it was not in milk with CCP throughout 18 weeks storage. Amount of protein precipitated increased in the control during storage, whereas there was no protein precipitated in milk with CCP. Surface color and peroxide value of the control and treatment did not change significantly during storage, and there were no marked differences between the control and treatment. These results indicated that quality of concentrated milk could be preserved, without gelation, by storing milk with CCP in the -liquid state at the frozen storage temperature. Besides, energy required for freezing preservation of milk could be significantly reduced by elimination of phase changes for freezing and thawing, and the stored product could be continuously processed for the final products without long waiting time for thawing.

Preparation of Ginseng Concentrate with High Content of Acidic Polysaccharide from White Tail Ginseng Marc (백삼 알코올 추출박을 이용한 산성다당체 다량 함유 백삼 농축액 제조)

  • Preparation of ginseng concentrate with high content of acidic polysaccharide from white tail ginseng marc that was obtained after preparation of white tail ginseng extract. As a result of extraction of white tail ginseng under various concentrations of ethanol (0∼90%), both amount of acidic polysaccharide and extraction yield decreased by increasing the ethanol concentration. However, acidic polysaccharide extracted by water from white tail ginseng marc was increased in accordance with the increase of ethanol concentration. The optimal condition for the extraction of acidic polysaccharide from the marc was treatment of $\alpha$-amylase in 390∼650 unit/g residue/15 mL of distilled water for 5 min at 4$0^{\circ}C$. The amount of acidic polysaccharide in water extract of the marc was increased from 8.3% to 10.5% by the treatment of $\alpha$-amylase. A new ginseng extract mixture was manufactured by mixing 50% ethanol extract of white tail ginseng and water extract of alcoholic residue in the ratio of 8:2 (w/w). Crude saponin content and acidic polysaccharide content were 10.5% and 17%, respectively. The mixture had a same crude saponin content and twice acidic polysaccharide content comparing to 50% ethanol extract of white tail ginseng. It suggests that preparation of new ginseng concentrate with high content of acidic polysaccharide from white tail ginseng marc has high potencies in the utilization of waste material.

  • Trace amount of cobalt in water samples was preconcentrated continuously with an organic precipitant and determined by flame atomic absorption spectrometry. The flow injection technique was used to preconcentrate cobalt by on-line direct precipitation with 1-nitroso-2-naphthol. The precipitation was dissolved with methyl isobutyl ketone (MlBK) and was sent to the flame. The optimum conditions for cobalt determination were determined and used to analyze Co samples. For 1.0 mL of sample, the enrichment factor was 13 and the sample throughput was about lO per hour for 0.5 ppm Co solution. The enrichment factor was increased to 68 fold for 10.0 mL. A semi-reference biologicaI sample was prepared and analyzed. The result was in good agreement with the expected value with RSD of 4%.

  • 전통 한약재의 일환인 황기, 당귀로부터 추출물의 획득은 일반적으로 열수추출법에 의하여 행해져 왔으나 추출물의 획득수율이 낮고 추출, 농축과정에서의 심란 열처리로 인하여 고유의 향이 소실되는 등 많은 문제점을 안고 있다. 이에 본 연구에서는 주정추출법을 시도하여 황기, 당귀 추출물의 획득수율을 열수추출법의 50, 39%(w/w) 대비 각각 15. 36% 증가한 65. 75%로 향상시킬 수 있었으며, 강한 고유의 향을 포집하고 있는 전혀 새로운 타입의 추출물을 얻을 수 있었다. 추출과정on서 열수추출법과의 차이는 추출용매를 물에서 주정으로 변경한 것과 1차 추출 후 얻어진 박을 이용하여 2차 추출을 행하였으며, 농축온도를 열수추출의 104$^{\circ}C$에 비하여 상대적으로 낮은 9$0^{\circ}C$에서 실시하였다. 이와 같이 처리온도와 거의 무관하게 주정은 고유의 향을 포집하는 뛰어난 효과를 나타내었다. 이 추출물을 물엿, 음료 등의 제품에 적용할 경우 별도의 인공향료 처리 없이 천연물 그대로의 가공으로도 충분한 고유의 향미를 부여할 수 있을 것으로 기대된다.

  • 1. 서론 : Nanofiltration(NF)은 분자량이 200-1000인 유기물과 음이온 및 다가 이온등을 일가 이온으로부터 선택적으로 분리함으로써 유기물의 농축과 이온의 선택적인 분리를 동시에 행할 수 있는 분리 공정이며, 순수의 제조, 과일과 야채주스의 농축, 저알콜 맥주와 와인의 제조, 낙농 산업에서 유제품의 농축, 섬유공업에서 염료와 이온의 분리 및 제지공업의 폐수 처리 등에 응용되고 있다. NF용 막재료로는 셀룰로오스 계열의 고분자나 폴리아미드, 폴리술폰, PAN등이 연구되어 왔으며 최근에는 친수성이 좋고 가교를 하지 않고도 이온 결합에 의하여 강도를 유지할 수 있다는 아이오노머의 성질을 이용하여 아이오너마를 Nanofiltration용 막재료로 이용하려는 연구가 이루어지고 있다.[1-3] 그러나 아이오노머의 미세구조와 유기물의 분리특성간의 관계에 관한 연구는 미흡한 실정이다. 따라서 본 연구에서는 분리막의 성형성을 향상시키고자 알킬메타크릴레이트가 도입된 아이오너머를 합성하고 알킬기의 길이나 이온의 함량 등 고분자의 구조에 따른 아이오노머막의 분리특성을 연구하여 아이오노머막의 NF용막으로서의 응용가능성을 살펴보았다.

Highly Sensitive Detection of Pathogenic Bacteria Using PDMS Micro Chip Containing Glass Bead (유리비드를 포함한 PDMS 마이크로칩을 이용한 고감도 감염성 병원균 측정에 관한 연구)

  • Here, we demonstrated simple nucleic acid, RNA, concentration method using polymer micro chip containing glass bead ($100\;{\mu}m$). Polymer micro chip was fabricated by PDMS ($1.5\;cm\;{\times}\;1.5\;cm$, $100\;{\mu}m$ in the height) including pillar structure ($160\;{\mu}m\;(I)\;{\times}\;80\;{\mu}m\;(w)\;{\times}\;100\;{\mu}m\;(h)$, gap size $50\;{\mu}m$) for blocking micro bead. RNA could be adsorbed on micro glass bead at low pH by hydrogen bonding whereas RNA was released at high pH by electrostatic force between silica surface and RNA. Amount of glass beads and flow rate were optimized in aspects of adsorption and desorption of RNA. Adsorption and desorption rate was measured with real time PCR. This concentrated RNA was applied to amplification micro chip in which NASBA (Nucleic Acid Sequence Based Amplification) was performed. As a result, E.coli O157 : H7 in the concentration of 10 c.f.u./10 mL was successfully detected by these serial processes (concentration and amplification) with polymer micro chips. It implies this simple concentration method using polymer micro chip can be directly applied to ultra sensitive method to measure viable bacteria and virus in clinical samples as well as environmental samples.
