Development of an Analytical Method for the Determination of Pyriofenone residue in Agricultural Products using HPLC-UVD (HPLC-UVD를 이용한 농산물 중 살균제 pyriofenone 분석법 확립)

  • Pyriofenone is an aryl phenyl ketone fungicide that is newly registered in Korea in 2013 to control powdery mildew on food. The objective of this study was to develop reliable and sensitive analytical method for determination of pyriofenone residue in agricultural products for ensuring the food safety. The pyriofenone residues in all samples(Korean melon, pepper, potato, mandarin, soybean, and hulled rice) were extracted with acetonitrile, partitioned with dichloromethane, and then purified with a silica cartridge. The purified samples were analyzed by HPLC-UVD and confirmed with LC-MS. The linear range of pyriofenone was 0.05~5 mg/kg with the correlation coefficient ($r^2$) > 0.999. Average recoveries of pyriofenone ranged from 72.8% to 99.5% at the spiked level of 0.05 and 0.5 mg/kg, while the relative standard deviation was 2.3%~6.4%. In addition, the limit of detection and limit of quantification were 0.01 and 0.05 mg/kg, respectively. The results revealed that the developed and validated analytical method was suitable for pyriofenone determination in agricultural products.

Herbicidal activity of a new benzenesulfonylurea compound, KSC-13906 (신규 benzenesulfonylurea 계 화합물 KSC-13906의 제초활성)

  • Herbicidal activities of newly synthesized experimental compound, KSC-13906 [Erythro N-{(4,6-dimethoxy pyrimidin-2-yl)aminocarbgnyl}-2-(2-fluoro-1- hydroxy-n-propyl)benzenesulfonamide, US Patent 5,461,025], were investigated in greenhouse and field for development as a paddy herbicide. KSC-13906 exihibited excellent weeding effects and selectivity to rice under submerged paddy condition in geenhouse. At the application rate of $2.5{\sim}5$ g ai/ha treated on surface of water, KSC-13906 effectively controlled three annual weeds such as Echinochloa crus-galli var. oryzicola. Scirpus juncoides, and Monochoria vaginalis as well as five perennial weeds such as Cyperus serotinus, Sagittaria pygmaea, Eleocharis kuroguwai, Potamogeton distinctus, and S. trifolia. However, there was no effect on Aneilema keisak and Lindernia procumbens. Especially, KSC-13906 effectively controlled Echinochloa crus-galli var. oryzicola at growth stages ranging from pre-emergence to 3-leaf stage. Antagonistic interaction was found between pyrazosulfuron-ethyl and mefenacet, but not between KSC-13906 ana mefenacet. KSC-13906 had high selectivity to transplanted rice, but caused phytotoxic effect on direct-seeded rice when treated before 3-leaf stage. Herbicidal activity of KSC-13906 on the Echinochloa crus-galli var. oryzicola was 7-folds greater than that of pyrazosulfuron-ethyl in terms of relative dose ratio of the herbicides. These results suggest that KSC-13906 possesses selective herbicidal potency in rice.

Persistence and Dislodgeable Residues of Chlorpyrifos and Procymidone in Lettuce Leaves under Greenhouse Condition (상추의 생산단계별 Chlorpyrifos 및 Procymidone의 잔류허용기준 설정)

  • Two pesticides commonly used for lettuce cultivation including chlorpyrifos and procymidone were subjected to a field residue trial to ensure safety of terminal residues in the harvest. After pesticides were applied at standard and double rates in a foliar spray, leaf persistence of their residues was investigated far 10 days prior to harvest. Even though far exceeded the tolerances, initial leaf residues were rapidly dissipated with time and remained only 0.4$\sim$7.2% of the residues in the harvest. As well fitted by the first-order kinetics, biological half-lives of the pesticide residues in lettuce leaves ranged 1.2$\sim$2.6 days. Slow dissipation of the residues in the harvest was observed during storage at room temperature and 4$^{\circ}C$ for 7 days. Portions of dislodgeable residues which resided in detergent washings decreased as time elapsed. Patterns in dissipation and distribution of dislodgeable residues were not largely affected by the application rate of pesticides. It is concluded that timing of pesticide application, that is, pre-harvest interval would be the first factor to determine the terminal residue level in edible portions of lettuce.

Studies on Cropping System of Upland Crop for the Sustainable Agriculture. (환경 농업 실천을 위한 밭작물 작부체계의 적용 조사)

  • This study was carried out to find out basic data of the various cropping systems controlling the soil environment by analysing the historical literature, the changes of cropping system and the expected income of cropping system of the upland crop. The results were summerized as follows ; 1. According to the literature of 'Gumyangjoprok' and 'Sanlimgyungjae', published on 15 to 16 century in Korea, the cropping system of barley, soybean, red bean, and millet, etc. was done together both the 'kunkyong' and 'kanjong'. 2. Since 1970s the staple food grains had been self-sufficient and the setting up of the rice seedbed became faster. However, the cropping system of the rice after barley was rapidly fallen from 83.7% to 4.0% in 1990s. Furthermore, the food production and the rate of arable land utilization were also rapidly fallen. 3. The most prospective cropping system is considered the soybean after barley, and root & tuber crops considering with the soil environment. 4. The expecting income of cropping system ranged from 940,000won to l,970,000won per 10a but that of barley after soybean cropping for 610,000won, and that of barley-after mung bean cropping for 613,000won. 5. The maintenance and the preservation of agricultural environment shall become positive by practicing the Integrated Nutrients Management(INM), and the Integrated Pest Management(IPM) with control using of fertilizer and pesticide presenting the excessive loads to the soil environment.

Control of Phythophthora capsici and residual characteristics by drenching of pesticides on tomato in hydroponic culture system (약제 관주처리에 의한 양액재배 토마토의 역병 방제 및 농약잔류 특성)

  • To establish effective and safe control method against Phytophthora root rot caused by Phytophthora capsici on tomato in hydroponic culture, three pesticides, oxadixyl copper hydroxide 8% WP, metalaxyl copper oxychloride 15% WP, and dimethomorph. dithianon 38% WP at 4 concentration levels were tested on potato dextrose agar medium inoculated with Phytophthora capsici. All pesticides inhibited mycelial growth, but two pesticides of them, metalaxyl copper oxychloride WP and dimethomorph. dithianon WP, were selected as effective pesticides for the efficacy test in a hydroponic culture. Forty days after transplanting of tomato seedlings, 4 ml of sporangia of P. capsici (about 25 sporangi/ml) per plot was inoculated around tomato plant root, and then 5 days after inoculation, the pesticides diluted at 5,000 times were drenched 1, 2 or 3 times per plot on the culture cube at 15 days interval. Fifteen days after drenching, tomato fruits and hydroponic culture solution were sampled for the analysis of pesticide residues. Dimethomorph was detected 0.001 and 0.003 mg/kg in tomato of the plots sprayed 2 and 3 times with dimethomorph dithianon WP of which detection levels were far below compared with 1.0 mg/kg of the Korean MRL of dimethomorph on tomato. Incidences of Phytophthora root rot were $30.5{\sim}50%$ in the plots drenched at 1 or 2 times with metalaxyl.copper oxychloride WP, and $16.7{\sim}25%$ in the plots treated with dimethomorph dithianon WP. However, there was no incidence of Phytophthora root rot in the plots treated at 3 times with both of pesticides, showing no phytotoxic effect. Based on the results, the drenching of these pesticides on the culture cube could be recommended as a very safe and effective control method for Phytophthora root rot in tomato.

Suggestion for Establishment of Temporary MRLs and Safe use Guideline of the Organophosphorus Insecticides in Jinpi (한약재 진피에 대한 유기인계 살충제의 잠정 MRL 설정 및 안전사용기준 제안)

  • Jinpi(Aurantii nobilis Pericarpium) is one of the most important material of oriental herbal medicine which is made from the peel of mandarin by washing with hot water and drying. Pesticides have necessarily used for mandarin cultivation according to their preharvest intervals (PHIs) but their maximum residue limits (MRLs) and PHIs for Jinpi are not established yet. This study is to know residue amount of organophosphorus insecticides in Jinpi and to establish the MRLs and PHIs for fenitrothion and phenthoate in Jinpi. Fenitrothion was sprayed once, twice and three times with 7 days interval before harvest. Its residue amount ranged from 0.14 to 1.17 mg/kg in mandarin, 0.59 to 4.02 mg/kg in its peel and 1.66 to 22.38 mg/kg in Jinpi. In case of phenthoate, it was sprayed with 10 days interval for 10 days before harvest. Its residue amounts in mandarin, its peel, and Jinpi ranged from 0.16 to 0.65, 0.69 to 2.41 and 1.69 to 11.3 mg/kg, repectively. Proposed MRLs of fenitrothion and phenthoate for Jinpi are suggested to 22.39 and 11.30 mg/kg, respectively. So we recommend PHIs of the pesticides that fenitrothion can be sprayed 3 times 7 days before harvest and in case of phenthoate, sprayed 3 times 10 days before harvest.

Residue safety on ethephon in soybean leaf by drenching and foliar application (에테폰의 관주처리와 엽면살포에 의한 콩 잎 중 잔류 안전성)

  • Ethephon is useful pesticide as ethylene precursor, which is an efficient plant hormone to produce functional secondary metabolites. However, the residual safety of ethephon was not studied on various crops. In here, the dissipation pattern of ethephon residue in soybean leaf was investigated both on the foliar and drenching applications. The biological half-lives of ethephon residues were 26.6, and 21.1 h on the once, and double foliar applications, respectively. Although the residue after three days from the final application was up to $60.6mg\;kg^{-1}$, the residue was below the limit of quantitation on the dried soybean leaf. In addition, drenching application of ethephon could increase the residue up to $36.3mg\;kg^{-1}$ after 20.1 h from the application, however, the treatment would not affect to the total phenol content significantly (p >0.01).

Residual Characteristics of Insecticide Dinotefuran in Asparagus under Greenhouse Condition (시설재배 아스파라거스 중 살충제 dinotefuran의 잔류특성)

  • This study was performed to investigate the residual characteristics of the insecticide dinotefuran in asparagus under greenhouse conditions from July to August and consequentially to obtain basic data for guidelines on the safe use of this pesticide in asparagus. Residues of dinotefuran in young stem of asparagus before and after growing mother stem were analyzed from samples collected at 0, 1, 3, 5, and 7 days after single application of a commercial formulation of dinotefuran (wettable powder, 10%) at the recommended dose (2,000 times dilution). The residue of dinotefuran in young stem of asparagus was analyzed by HPLC-UVD, and recovery of dinotefuran in young stem was tested at 0.5 and 1.0 mg/kg concentrations. As a result, the limit of quantitation (LOQ) of dinotefuran was 0.01 mg/kg, and the recovery of dinotefuran was in the range of 83.3-94.0% with a coefficient of variation less than 10%. Residues of dinotefuran in young stem of asparagus before and after growing the mother stem were lower than the tentative limit (0.05 mg/kg) from 5 and 3 days after single application, respectively. Based on these results, single application of dinotefuran (wettable powder, 10%) at the recommended dose at 7 days before harvest would have no deleterious effects on safety issues concerning pesticide residue. This result might provide basic information to construct guidelines for the safe use of dinotefuran in asparagus.

Improvement of an Analytical Method for Methoprene in Livestock Products using LC-MS/MS (LC-MS/MS를 이용한 축산물 중 살충제 메토프렌의 잔류분석법 개선)

  • The research aims to develop a rapid and easy analytical method for methoprene using liquid chromatography-tandem mass spectrometry (LC-MS/MS). A simple, highly sensitive, and specific analytical method for the determination of methoprene in livestock products (beef, pork, chicken, milk, eggs, and fat) was developed. Methoprene was effectively extracted with 1% acetic acid in acetonitrile and acetone (1:1), followed by the addition of anhydrous magnesium sulfate (MgSO4) and anhydrous sodium acetate. Subsequently, the lipids in the livestock sample were extracted by freezing them at -20℃. The extracts were cleaned using MgSO4, primary secondary amine (PSA), and octadecyl (C18), which were then centrifuged to separate the supernatant. Nitrogen gas was used to evaporate the supernatant, which was then dissolved in methanol. The matrix-matched calibration curves were constructed using 8 levels (1, 2.5, 5, 10, 25, 50, 100, 150 ng/mL) and the coefficient of determination (R2) was above 0.9964. Average recoveries spiked at three levels (0.01, 0.1, and 0.5 mg/kg), and ranged from 79.5-105.1%, with relative standard deviations (RSDs) smaller than 14.2%, as required by the Codex guideline (CODEX CAC/GL 40). This study could be useful for residue safety management in livestock products.

Residue Analysis to Establish an Index for the Safety Use of Propineb (Propineb의 안전사용(安全使用) 기준(基準) 설정(設定)을 위한 잔류분석(殘留分析))

  • Residue of Propineb in Sesame(Sesamum indicum L.) seed was determined to establish an index for the safety use of Propineb to Sesame. Evaluation was made on residual concentration of Propineb in Sesame seed as a function of application frequency and date when the mixed formulations of Propineb(56%), protectant fungicide, and Oxadixyl(8%), contact fungicide, were sprayed into Sesame leaves. Level of Propineb treatment was $0.028g/m^2$ with various combinations of application time from three to sixty days before harvest. Results are summarized as followings. 1. Recovery percentages of Propineb from Sesame seed were ranged from 84 to 96, and the minimum detectable limit of Propineb with the method employed in this experiment was 0.03mg/kg. 2. Residues of Propineb in Sesame seed were in the ranges of 0.14 to 1.38mg/kg, varying with frequency and date of Propineb application. 3. Residues of Propineb increased as increasing application frequency of Propineb or as being application date closer to harvest time. 4. Residue of Propineb in Sesame seed was decreased with time, showing to be fitted to the first-order kinetics. 5. Residues of Propineb in Sesame seen were, irresepective of treatments, lower than 2mg/kg, the Maximum Residue Limit(MRL) established by FAO/WHO. 6. Half-life of Propineb determined in this experiment was ranged from 12 to 16 days.
