Effect of Oligosaccharide Addition on Gelatinization and Retrogradation of Backsulgies (올리고당 첨가가 백설기의 호화와 노화에 미치는 영향)

  • Four different oligosaccharides used to determine their effects on gelatinization and retrogradation of Backsulgies(BSG), Korean traditional rice cake, were cyclodextrin, isomaltooligosaccharide, fructooligosaccharide and maltotetrose, with 3 or 6%(w/w), based on the rice flour. From the Amylograph and blue value data, adding 6% cyclodextrin into the rice flour fastened the gelatinzation process, and delayed the retrogradation in stored BSG at 4$\^{C}$ for up to 3 days, probably due to its cyclic structure. Using rheometer, the hardness of freshly made BSG added with cyclodextrin was significantly lower than that of control BSG, and increased with storage time in all BSGs at refrigerated temperature. When using maltotetrose in rice flour, the hardness of BSG was also significantly low, but slightly higher than that of cyclodextrin. Hunter “L” value in BSG decreased with the addition of oligosaccharides, but the higher the level of oligosaccharide in BSG, the insignificant the Hunter “a”,“b” values of BSG. From the X-ray diffraction studies, the rice flour showed typical A pattern, and the crystallinities of all BSG gave amorphous V type. The highest peaks of X-ray patterns in BSG added with 6% cyclodextrin or maltotetrose were, however, lower than that of the control, meaning the less crystalline, retrograded starch in the former BSGs. Based on sensory evaluation, BSG added with all oligosacchrides were not significantly different in taste, flavor, chewiness, moistness and overall preferences, with slightly darker color with longer storage time, compared to the control BSG.

  • 새로운 노화방지 물질로 개발한 3-APPA가 노화에 의해 야기되는 여러 변화들, 특히 세포 증식, 유전자 수준 및 단백질 수준에서의 collagen의 생합성 변화, 면역조직화학염색을 이용한 collagen 생합성의 변화등을 세포배양 및 동물실험을 통하여 측정하였다. MTT assay를 이용한 인체 피부 섬유아세포의 증식 실험에서 3-APPA는 무처치군에 비교해서 최고 2배의 섬유아세포 증식 효능을 나타내었으며, $^3$[H]-proline incorporation 방법을 이용한 단층세포 배양 및 3차원 dermal equivalent 섬유아세포 배양에서 무처치군 및 vitamin C 처리군에 비해 최고 1.5배의 collagen 생합성 증가를 나타내었다. 그러나 type I alpha-procollagen mRNA expression에는 영향을 미치지 않는 것으로 나타났다. H&E 염색을 이용한 hairless mice의 피부에 대한 형태학적 변화 및 type I pM procollagen antibody를 이용한 면역조직화학염색에서, 3-APPA는 collagen 생합성을 증가시키는 것으로 나타났다. 이상의 결과에서 3-APPA는 섬유아세포 배양 및 hairless mouse를 이용한 실험에서 피부 섬유아세포 증식을 촉진시키며 collagen 생합성을 증가시켜 피부노화를 억제 할 수 있는 물질임을 밝혔다.

  • 활성 산소종은 노화, 특히 피부노화의 원인 물질로 작용하고 있다. 피부는 자외선에 노출되어 있어 활성 산소종을 만드는 광화학적 반응들이 계속해서 일어나고 있다. 이들 활성 산소종들은 피부 세포 및 조직 손상을 주도한다. 이들은 항산화 효소와 비효소적 항산화제들로 구성된 항산화 방어망을 파괴함으로써 산화제/항산화제 균형을 산화 상태 쪽으로 기울게 한다. 결과적으로 계속된 산화적 스트레스는 지질 과산화, 단백질 산화, 간질 성분을 파괴시키는 단백질분해효소의 활성화, 탄력 섬유인 콜라겐과 엘라스틴의 사슬절단 및 비정상적인 교차결합, 히아루론산 사슬의 절단, 멜라닌 생성반웅 촉진, DNA 산화와 같은 생체 구성 성분들의 손상을 야기시킨다. 결국에는 탄력감수 주름살 및 기미.주근깨 둥으로 특징 지워지는 피부노화가 가속화된다. 따라서 피부노화 방지를 위해서는, 과잉의 활성 산소종 생성을 억제하고 또한 생성된 활성산소를 효율적으로 제거할 수 있는 시스템이 화장품의 처방에 반드시 포함될 필요가 있다. 즉, 산화제/항산화제 밸런스가 유지되는 피부의 항산화 방어 시스템 구축이 필요하다. 피부노화 방지에 있어서 천연물의 역할로 (1) 자외선 흡수제로서의 역할, (2) 항산화제로서의 역할, (3) 주름 개선제로서의 역할, (4) 미백제로서의 역할, (5) 항균\ulcorner항염작용 및면역 조절제로서의 역할에 대하여 살펴 보았다. 21세기는 본격적으로 기능성 화장품 시대가 개막될 것으로 예측하고 있다. 이에 맞춰 천연물들은 피부노화를 방지하는데 주도적인 역할을 할 것으로 기대된다.

Photo-aging regulation effects of newly bred Green ball apple (신품종 그린볼 사과의 광노화인자 조절효과)

  • In this study, extracts from the Green ball apple peel (GBE) and the newly bred green ball apple from Korea showed inhibition effects on photo-aging factor regulation associated with skin aging. To investigate the inhibition effect on photo-aging factor regulation in skin, GBE was treated with UVB to induce photo-aging related factors in CCD986sk fibroblast cells. Photo-aging factor regulation effects showed that GBE inhibited UVB-stimulated matrix metalloproteinase (MMP)-1 and MMP-9 protein synthesis in collagen type I alpha 2 chain (COL1A2), MMP-1, MMP-9, and tissue inhibitors of metalloproteinase (TIMP)-1 protein expression. The expression of COL1A2 and TIMP-1 protein was significantly increased. The mRNA expression levels of COL1A2, MMP-1, MMP-9, hyaluronan synthase (HAS)2, transforming growth factor (TGF)-β, and TIMP-1 were decreased by GBE. The expression of TIMP-1 and TGF-β, which are regulators involved in matrix metalloproteinase and type I procollagen expression, was found to increase with increasing expression of COL1A2. The expression of HAS2, which is involved in the production of hyaluronic acid, one of the structural proteins constituting the skin, was also confirmed. Therefore, GBE showed excellent efficacy against photo-aging factor regulation and could be used as functional material to prevent and treat skin aging.

The effects of antioxidant and anti-aging treatment of UVB-irradiated human HaCaT keratinocytes with ethanol extracts of colored rice varieties (유색미 에탄올 추출물의 산화방지 효과와 UVB에 유도된 인간유래 피부각질형성세포에서의 항노화 효과)

  • In this study, we investigated the antioxidant activities and anti-aging efficacy in terms of suppression of matrix metalloproteinases (MMPs) in UVB-irradiated HaCaT cells by adding the ethanol extracts of Josaengheogchal (JE) and Shintoheug rice (SRE). In the electron-donating ability and ABTS radical-scavenging assays, we observed that both JE and SRE had scavenging activities and in a collagenase inhibition assay, both extracts showed inhibition effects of over 73% at $1,000{\mu}g/mL$ concentration. The expression of MMP-1 and -3, when the extracts were treated with UVB $50mJ/cm^2$, irradiated human HaCaT keratinocytes, was analyzed by western blotting and reverse-transcription polymerase chain reaction (RT-PCR). The results showed that MMP-1 and -3 proteins and mRNAs were downregulated in a concentration-dependent manner in response to both extracts. Therefore, we expect that these compounds have a potential for the use as functional ingredients with anti-aging effects in the cosmetic and food industries.

  • 나이, 자외선 등 내외적인 여러 가지 스트레스에 의해 일어나는 피부노화 현상은 피부두께의 변화오 함께 세포수 감소 및 피부 신진대사 저하, 생체결합수 손실을 비롯하여 콜라겐, 엘라스틴 등을 세포간물질 감소로 인해 피부속이 바람 빠진 공처럼 함몰되고 골이 생겨 결국 주름지고 거칠은 피부표면의 형태로 나타난다. 피부 노화 현상에 대한 여러 가지 기능적, 구조적 변화를 살피고 피부노화를 억제하기 위한 화장품의 역할과 연구 동향을 살펴 본다.

The Effect of Kimchi Intake on Production of Free Radicals and Anti-oxidative Enzyme Activities in the Liver of SAM (김치가 노화촉진 쥐 간의 유리기 생성 및 항산화효소 활성에 미치는 영향)

  • This study was performed to investigate the effect of kimchi intake on antiaging characteristics in liver of senescence-accelerated mouse (SAM) in terms of free radical production and anti-oxidative enzyme activities. Two hundred twenty SAM were divided into four groups and fed kimchi diet for 12 months. Experimental groups were kimchi free AIN-76 diet (control) group, Korean cabbage kimchi diet (KCK) group, mustard leaf added (30%) Korean cabbage kimchi diet (MKCK) group, and mustard leaf kimchi diet (MLK) group. Amount of freez-dried kimchi added to the diet was 5% that is equivalent to 50 g of fresh kimchi. Concentrations of total free radical, OH radical, $H_2O$$_2$in the liver significantly increased as aged (p<0.05). But those free radical concentrations from kimchi diet groups were lower than those of control (p<0.05). Among kimchi groups, MKCK and MLK groues showed greater inhibiting effect than KCK. Antioxidant enzyme activities of Cu, Zn-SOD, Mn-SOD, GSH-px, catalase and GSH/GSSG in kimchi groups were significantly increased (P
