Purpose: The current study was conducted to evaluate the compatibility of UV-A blocking contact lens on eye protection with regular contact lens. Methods: The protective activity of regular contact lens (UV-A blocking: 20%) and UV-A blocking contact lens (UV-A blocking: 85%) on the denaturation of RNase A, catalase, and superoxide dismutase (SOD) induced UV-A irradiation were compared by acrylamide gel electrophoresis. The enzyme solutions were irradiated with UV-A for 1, 3, 6, 24 and 96 hours at the wavelength of 365 nm. Covering area with contact lenses were varied as 50%, 70% and 100% according to the calculation of blocking areas of anterior eye that could be covered with RGP lens, soft contact lens, and eye glasses, respectively. Results: Denaturations of RNase, catalase and SOD were exaggerated when they were exposed to UV-A for a longer period. The denaturation was effectively prevented by UV-A blocking contact lens compared to regular contact lens. The capability of UV-A blocking contact lens was considerably reduced when the covering area with contact lens decreased and exposure time to UV-A extended. Conclusion: Therefore, it would be suggested that wearing contact lens for a long time under sunlight is carefully considered since the activity of UV-A blocking contact lens against UV-A irradiation may not be enough to protect enzymes presented in eyes when exposure time to UV-A increased.
Experiments were performed to investigate changes in metabolic rate (MO2), critical oxygen saturation (Scrit) and hematological parameters of black rockfish, Sebastes schlegeli exposed to hypoxia at 15, 20 and 25℃. The MO2 was measured at an interval of 10 min using intermittent-flow respirometry. The normoxic standard metabolic rate (SMR) was 116.5±5.5, 188.6±4.2 and 237.4±6.8 mg O2/kg/hr, and Scrit was 22.1±1.2, 30.6±1.5 and 41.9±1.4% air saturation at 15, 20 and 25℃, respectively. Q10 values were 2.62 between 15 and 20℃, 1.58 between 20 and 25℃, and 2.04 over the full temperature range. In the investigation of blood (hematocrit and hemoglobin) and biochemical parameters (plasma cortisol, glucose, electrolyte and osmolality), the rockfish were subjected to Scrit for each temperature during 4 hr. All of hematological parameters of the rockfish exposed to hypoxic water were significantly higher than those of normoxic control. Moreover, blood and biochemical parameters of the rockfish maintained to normoxic water showed the tendency of increase with temperature, and were significantly higher at 25C. As a result of this experiment, it was found that physiological stress due to hypoxia increased at high temperature.
피조개, Scapharca broughtonii의 개체크기(작은 개체: 1.0-1.3cm, 큰 개체:4-5 cm), 수온(10, 15및 2$0^{\circ}C$), 염분(7,13,19,26 및 32 $\textperthousand$)에 따른 일반적인 대사경향과, 수온 1$0^{\circ}C$와 2$0^{\circ}C$ 그리고 부니 농도 25, 50, 100, 200, 400 600, 800및 1000ppm에서 15일간 노출시키면서 사망률, 산소소비율, 여수율 및 배설률을 측정한 결과는 다음과 같다.1. 일반적으로 피조개의개체 크기는 작은 개체일수록, 그리고 수온이 높을수록 비체중 산소소비율, 비체중 여수율 및 비체중 배설률은 높게 나타났다. 그리고 염분을 제외하고는 산소 소비율과 여수율은 염분농도의 증가에 따라 다소 증가하는 유사한 경향을 보인 반면, 비체중 배설률은 염분농도가 낮아질수록 낮은 양상을 나타내었다. 2. 1$0^{\circ}C$의 경우 작은 개체에서 LC$_{50}$은 557.11 ppm, 큰 개체에서 856.03 ppm이었고, 20%C의 경우, 작은 개체에서의 LC$_{50}$은 274.55 ppm, 큰 개체에서 346.26 ppm이었으며 1$0^{\circ}C$에 비해 2$0^{\circ}C$에서 특히 1000 ppm이상의 농도에서 사망률이 현저히 증가함으로써 부니에 의한 영향은 크게 나타났다.3.에 비해 2$0^{\circ}C$의 고수온구에서 부니의 농도가 높을수록 폐사율은 높게 나타났으며, 1$0^{\circ}C$와 2$0^{\circ}C$의 두 수온구에서 산소소비율과 여수율에 대한 각 농도별 부니의 영향은 유사한 경향을 나타내었다. 특히, 2$0^{\circ}C$에서 사망률이 현저히 증가하기 시작하는 부니농도 100ppm이상에서 산소소비율의 감소가 크게 나타났다. 그리고 암모니아태질소 배설률은 부니농도의 증가에 따라 차츰 증가를 보인후 600ppm이후부터는 다시 감소하는 경향을 보였다.

We examined the impacts of anthyopogenic pollutant (benzo〔a〕pyrene) on the growth and photosynthesis of five marine phytoplankton species (Skeletonema costatum, Heterosigma akashiwo, Prorocentrum dentatum, P. minimum, Aknshiwo sanguinea), which are dominant in Korean coastal water. After the 72 h exposure to benzo〔a〕pyrene, the dramatic decrease in cell numbers was observed in the range of 1 to 10 $\mu\textrm{g}$ L$^{-1}$ for S. costatum, P. minimum, P. dentatum, whereas for A. sanguinea and H. akashiwo at the low concentrations 0.1 to 1 $\mu\textrm{g}$ L$^{-1}$ . Among the 5 phytoplankton species, the highest growth inhibition concentration ($IC_{50}$/) was 6.20 $\mu\textrm{g}$ L$^{-1}$ for P. minimum, followed by 2.14 $\mu\textrm{g}$ L$^{-1}$ for P. dentatum, 1.68 $\mu\textrm{g}$ L$^{-1}$ for S. costatum, 0.74 $\mu\textrm{g}$ L$^{-1}$ for H. akashiwo, 0.10 $\mu\textrm{g}$ L$^{-1}$ for A. sanguinea. The five species exposed to the low concentration of 1 $\mu\textrm{g}$ L$^{-1}$ were recovered after transferring to new media, but the species exposed to the high concentrations of 10 and 100 $\mu\textrm{g}$ L$^{-1}$ were not recovered, with the exception of P. minimum. Those results indicate that the thecate dinoflagellate P. minimum is most tolerant to the chemical and the athecate dinoflagellate A. sanguinea is not. Geneyally, the cell-specific photosynthetic capacity of H. akashiwo exposed to the low concentrations of 0.1 and 1 $\mu\textrm{g}$ L$^{-1}$ was higher than that of the cells in the control, whereas the cells exposed to the high concentrations of 5 and 10 $\mu\textrm{g}$ L$^{-1}$ showed the negligible photosynthetic level by the first few days of the experiment. In the case of the cells exposed to the concentration of 5 $\mu\textrm{g}$ L$^{-1}$ , after 12 days of the experiment the photosynthetic capacity was increased toward the end of the experiment. This indicates that H. akashiwo may utilize the benzo〔a〕pyrene as a carton source for its growth when exposed to low concentrations. Results suggest that anthropogenic pollutants such as benzo〔a〕pyrene may have significant influence on the succession of phytoplankton species composition and the primary production in coastal marine environments.

Structural light weight aggregate concrete are made with both coarse and fine light weight aggregates, but it is common with the high strength concrete to replace all or part with normal weight sand be called class 1 structural light weight aggregate concrete. Fire resistance of structural light weight aggregate concrete are determined by properties of high water content ratio and explosive spalling. Especially, structural light weight aggregate concrete is occurred serious fire performance deterioration by explosive spalling stem from thermal stress and water vapor pressure. This study is concerned with experimentally investigating fire resistance of class 1 structural light weight concrete. From the test result, class 1 structural light weight concrete is happened explosive spalling. The decrease of cross section caused by explosive spalling made sharp increasing gradient of inner temperature.
This study was carried out to determine the optimal condition for successful and efficient c cryopreservation of zygotes, 1-cell embryos, using EFS40 which was 40% (v/v) ethylene glycol diluted in DPBS medium containing 30% Fic-oll (w/v) and 0.3 M sucrose. After mouse zygote produced by IVF was vitrified by two freezing methods, the post-warming survival rates of 1-cell zygotes were assessed as cleavage to the 2-cell stage and development into the hatching blastocysts at 5 day. In the one-step method, when embryos were directly exposed to the vitrification solution at 25$^{\circ}C$ for 1 min., survival and development rates of zygotes were 85.5% and 31.9% In the two-step method, embryos were equilibrated with a dilute 20% EG for 1, 3, 5 min. before 1 min. exposure to EFS40, re-spectively. However, the rates of development (17.7, 3.3, 0%) were lower than that of one-step method. The highest survival rate (95.9%) was obtained by one-step method which exposes embryos in EFS40 for 30 sec. In this condition, 63. 8% of cleaved 2-cell developed into hatching blastocysts. In the cell number of Total and ICM using differential labelling technique, there are no significant differences in the cell number of Total and ICM between blastocysts devel oped in vitrified-thawed embryos (63.2${\pm}$16.9, 1 13.5${\pm}$4.0) and control balstocysts (54.0${\pm}$15.2, 1 12.3${\pm}$4.6). Therefore, these results show that mouse zygotes can be successfully cryopreserved by a simple vitrification method although developmental rates of vitrified embryos were reduced. In conclusion, this proposed vitrifi cation procedures can be useful in the cryopreservation of mouse IVF zygotes.

This study was carried out to investigate in vitro/in vivo development of vitrified mouse oocytes. Mouse oocytes were vitrified using EFS30, 35 and 40 (30, 35 and 40% ethylene glycol, 18% ficoll and 0.5 M sucrose in M2 medium). After being exposed or vitrified-thawed, oocytes of normal morphology were inseminated in vitro by $1-2\times10^6/ml$ of epididymal sperm. The rates of fertilization, in vitro/in vivo development and cell number (inner cell mass and tropectoderm cell) of blastocysts in each treatment group were examined. The results obtained in these experiments were summarized as follows: The cleavage rates were obtained in EFS35 containing 35% ethylene glycol higher than in EFS30 and EFS40. The development rate of vitrified-thawed oocytes to two-cell stage after in vitro fertilization (51.1%) was significantly different compared to that of exposed to vitrification solution without cooling (60.0%) and control (68.2%) (p<0.05). However, there were no differences in the blastocyst formation from the cleaved embryos among groups (75.0, 73.3 and 80.0%). Also, the mean number of cells per blastocysts of vitrified group $(92.5{\pm}2.9)$ was similar to that of the exposed $(98.5{\pm}5.3)$ and control $(100.9{\pm}4.8)$. In vivo development of the blastocysts derived from vitrified-thawed oocytes resulted in fetal development (50.7%) and implantation rates (80.0%) which are very similar to those of control (58.2, 78.2%). These results suggest that mouse oocytes could be cryopreserved using vitrification solution (EFS35) based on ethylene glycol.
Physical sterilization methods using ultraviolet radiation and ionizing radiation such as gamma ray and electron beam are applied in various industry fields due to disinfection effects and economic efficiency but may also cause microbial mutation. In this research, Salmonella enterica and Escherichia coli strains were treated with ionizing and ultraviolet radiation and their survival rate, mutation rate, and DNA damage were studied to evaluate the genetic safety. The survival rate of the strains decreased drastically as the irradiation dose of ultraviolet ray, gamma ray, and electron beam increased, and over 90% of the strain was exterminated at a dosage of $0.40{\sim}25.06mJ/cm^3$, 0.11~0.22 kGy, 0.14~0.53 kGy respectively. In SOS / umu-test, genotoxicity causing DNA damage was identified in all samples. In Ames test, back-mutation rate increased to $3.82{\times}10^{-4}$ and $9.84{\times}10^{-6}$ respectively when exposed to ultraviolet ray and gamma ray. At exposure to ultraviolet ray, gamma ray, and electron beam with dosage of over 99.99% extinction rate of S. enterica TA100, back-mutation rate increased 347 times, 220 times, 0.6 times respectively to the spontaneous back-mutation rate. Rifampicin resistance mutation rate of E. coli CSH100 exposed to ultraviolet ray, gamma ray, and electron beam was $2.46{\times}10^{-6}$, $1.66{\times}10^{-6}$, $4.12{\times}10^{-7}$ respectively. Therefore, gamma radiation is effective in microorganism control from the perspective of disinfection and electron beam has the advantage of sterilizing with little DNA damage and bacterial mutation.
Blue-ice area is a glacial ice field in ice sheet, ice shelf and glaciers where snow ablation and sublimation is larger than snowfall. As the blue-ice area has large influences on the meteorite concentration mechanism and ice mass balance, it is required to quantify the concentration of blue-ice. We analyzed spectral reflectance characteristics of blue-ice, snow and cloud by using MODIS images obtained over blue-ice areas in McMurdo Dry Valleys, East Antarctica, from 2007 to 2012. We then developed Normalized Difference Blue-ice Index (NDBI) algorithm which quantifies the concentration of blue-ice. Snow and cloud have a high reflectance in visible and near-infrared (NIR) bands. Reflectance of blue-ice is high in blue band, while that lowers in the NIR band. NDBI is calculated by dividing the difference of reflectance in the blue and NIR bands by the sum of reflectances in the two bands so that NDBI = (Blue-NIR)/(Blue + NIR). NDBI calculated from the MODIS images showed that the blue-ice areas have values ranging from 0.2 to 0.5, depending on the exposure and concentration of blue-ice. It is obviously different from that of snow and cloud that has values less than 0.2 or rocks with negative values. The change of NDBI values in the blue-ice area has higher correlation with snow depth ($R^2=0.699$) than wind speed ($R^2=0.012$) or air temperature ($R^2=0.278$), all measured at a meteorological station installed in McMurdo Dry Valleys. As the snow depth increased, the NDBI value decreased, which suggests that snow depth can be estimated from NDBI values over blue-ice areas. The NDBI algorithm developed in this study will be useful for various polar research fields such as meteorite exploration, analysis of ice mass balance as well as the snow depth estimation.
