Effect of Light-Quality Control on Growth of Ledebouriella seseloides Grown in Plant Factory of an Artificial Light Type (인공광 식물공장내 광질 제어가 방풍나물 생장에 미치는 영향)

  • BACKGROUND: Plant factory system of an artificial light type using Light-Emitting Diodes (LEDs), fluorescent light, or metal halide lamp instead of sun light is an ultimated method for plant production without any pesticides regardless of seasonal changes. The plant factory is also completely isolated from outside environmental conditions such as a light, temperature, or humidity compared to conventional greenhouse. Light-environment control such as a quality or quantity in the plant factory system is essential for improving the growth and development of plant species. However, there was little report that the effects of various light qualities provided by LEDs on Ledebouriella seseloides growth under the plant factory system. METHODS AND RESULTS: Ledebouriella seseloides seedlings transplanted at urethane sponge were grown in the plant factory system of a horizontal type with LED artificial lights for 90 days. Yamazaki solution for hydroponic culture of the seedlings was regularly irrigated by the deep flow technique (DFT) system on the culture gutters. Electrical Conductivity (EC) and pH of the solution was recorded at 1.4 ds/m and 5.8 in average, respectively during the experimental period. Number of unfolded leaves, leaf length, shoot fresh and dry weight of the seedlings were three times measured in every 30 days after beginning of the experiment. Blue LEDs, red LEDs, and fluorescent lights inside the plant factory were used as light sources. Conventional fluorescent lamps were considered as a control. In all the treatment, light intensity was maintained at $100{\mu}mol/m^2/s$ on the culture bed. Fresh weight of the seedlings was 3.7 times greater in the treatment with the mixture radiation of fluorescent light and blue+red LEDs (1:3 in energy ratio; Treatment FLBR13) than in fluorescent light treatment (Treatment FL). In FLBR13 treatment, dry weight per seedling was two times greater than in FL or BR11 treatment of blue+red LEDs (1:3 in energy ratio; Treatment BR11) during the culture period. Increasing in number of unfolded leaves was also significantly affected by the FLBR13 treatment comparing with BR11 treatment. CONCLUSION(S): Hydroponic culture of Ledebouriella seseloides seedlings was successfully achieved in the plant factory system with mixture lights of blue, red LEDs and fluorescent lights. Shoot growth of the seedlings was significantly promoted by the FLBR13 with the mixture radiation of fluorescent light, blue, and red LEDs under 1:3 mixture ratio of blue and red LEDs during the experimental period compared to conventional light conditions.

Genetic Relationship among Sedum Species Based on Morphological Characteristics and RAPD Analysis (한국산 Sedum속 식물의 형태적 특성과 RAPD에 의한 유연관계 분석)

  • This study was conducted to investigate the potentiality of various Korean Sedum species as ornamental plants based on morphological characteristics and to analyze the genetic relationship among the Sedum species. S. kamtschaticum and S. takesimense possessing splendour flowercluster with yellow color could be suggested for garden plant, S. routundifolium having pink flower-clusters with round leaf shape for pot flower or garden plant and S. sarmentosum, S. polystichoides and S. oryzifolium with creeping stem and low plant height for ground cover plant or floral carpet. Eighteen oligonucleotide random primers were used to amplify genomic DNA of Sedum species using polymerase chain reaction (PCR). Ninety five polymorphic bands among 125 different DNA fragments in the range of 224 to 3,675 base pairs were obtained from RAPD analysis. Similarity matrix of RAPD profiles was generated by coefficient value of variation, and the data were subjected to be cluster analysis. Fifteen lines of Sedum species analyzed were classified into 3 groups with the similarity coefficient value of 0.418, and 12 groups with the value of 0.328. RAPD results showed similar trends as the morphological characteristics of the plants.

Effects of Planting Density on Growth and Yield of Vegetable Soybean Varieties (파종밀도가 풋콩 품종의 생육 및 수량에 미치는 영향)

  • The objective of experiment was to investigate the effects of planting density on growth and yield of vegetable soybean, and to clarify the optimum planting density of vegetable soybean in the middle west region of Korea. The field experiment with 4 levels of planting density was carried out at Yesan area in $2005{\sim}2006$. The days from seeding to flowering and the days from seeding to harvesting and lodging were not significantly different among planting distance. The stem length was increased as planting distance was shortened but the number of node, branch, pod per branch, pod per individual, weight of stem and pod, one hundred pod weight and rate of 2+3 seed per pod were decreased as planting density was increased. The size of vegetable soybeans was not significantly different among planting distance, but the harvest index of vegetable soybean was decreased as planting distance was shortened. Yield of vegetable soybean was increased as planting distance was decreased. However, the approriate densities for stem and pod weight per a plant, number of pod per a branch and the vegetable soybean yield of 2+3 seed per pod were different from that density. The optimal planting distance of varieties was $60{\sim}25\;cm$ in Sunheukkong and Ilpumgeomjeongkong and was $60{\sim}35\;cm$ in Galmikong.

Effect of Benzyladenopurine Concentration in Soaking Solution on Growth of Mungbean Sprouts (침종액 중 BA 농도에 따른 숙주나물의 생장)

  • Benzyladenopurine (BA), a synthetic chemical commonly used for bean sprout culture, should be minimized for wellbeing foods or not be applied. The study was done to check the effect of different BA concentrations treated during 5 hour imbibition on growth and morphological characters of mungbean sprouts. The mungbean seeds of 3 cultivars (Keumseongnogdu, Owoolnogdu, and Zhong Lu 1) were imbibed for 5 hours in the solutions with different BA concentrations (0, 25, 50, 75, and 100 ppm) before 3 hour aeration. On the 6th day after culture, the mungbean sprouts were classified by 4 categories on the base of hypocotyl length; > 7cm, 4 to 7cm, < 4cm, and non-germination, and their morphological characters, fresh and dry weights were measured. Regardless of cultivars the composition rate of hypocotyls of longer than 7cm was decreased with increased BA concentration over 50 ppm while the reverse result was true in the rate of shorter than 4cm. In the rate of 4 to 7cm. cv. Owoolnogdu showed the highest rate in its 50 ppm concentration although cv. Keumseongnogdu and Zhong Lu 1 showed similar result to the above two rates. Formation rate and its number of lateral roots were largely changed around 50 ppm concentration but the roots was not formed in over its 75 ppm concentration. Hypocotyl and root lengths of all the cultivars were shortened with increased BA concentration. In the diameter of middle part of hypocotyl, 3 cultivars showed nearly the same responses as the rate of 4 to 7cm hypocotyls. Hypocotyl and total fresh weights per sprout were heavier in BA treated sprouts than in no treated ones but the weights of the former sprouts were not influenced by its different concentrations.

Effects of Aeration Period and Temperature after Imbibition on Growth of Mungbean Sprouts (침종 이후의 Aeration 기간과 온도에 따른 숙주나물의 생장)

  • Lateral roots formed on mungbean sprouts should lower their quality. The study was carried out to clarify the effects of aeration periods (AP; 1, 2, 3, and 4 hours) and temperature (AT; 20, 30, and $40^{\circ}C$) after 5 hour seed imbibition into 50 ppm benzyladenopurine (BA) solution on growth and morphological characters of mungbean (cv. Keumseongnogdu, Owoolnogdu and Zhong Lu 1) sprouts. On the 6th day, the mungbean sprouts were sorted by 4 categories on the base of hypocotyl lengths; > 7cm, 4 to 7cm, < 4cm, and non-germination to calculate their composition rates, number of lateral roots, lengths of hypocotyl and root, diameters at middle and upper parts of hypocotyl, fraction fresh and dry weights were measured. AP more affected growth of the cultivars than AT showing little effect on them. In the composition rate of the above 4 categories, cv. Zhong Lu 1 had the highest rate in longer than 7cm but nearly the same rate in AP treatments. Rates of longer than 4cm hypocotyls in cv. Keumseongnogdu, Owoolnogdu were increased with longer AP but their rates of shorter than 4cm showed reverse response to the former. Formation rate and number of lateral roots per sprout were decreased with longer AP, showing more severe decrement when delayed 3 to 4 hour AP. Upper part of hypocotyls and roots were more thickened and shortened in longer AP, respectively. Total fresh weights had no significant difference between AP treatments while hypocotyl fresh weights were increased with longer AP. It was concluded that in mungbean sprout culture aeration from BA treatment to the first watering permitted at least 4 hours.

Effects of Temperature and Shading on Germination and Early Growth in Pimpinella brachycarpa (온도와 차광이 참나물 발아 및 초기 생장 특성에 미치는 영향)

  • This study was carried out to determine the effects of temperature and shading on germination and early growth of Pimpinella brachycarpa. Experiment was performed by temperature controls ($5^{\circ}C$, $10^{\circ}C$, $15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$, and $30^{\circ}C$) and shading treatments (control, 35% shading, 50% shading, and 75% shading). Seed pre-treatment before the germination experiment was carried out by temperature (with low temperature and wetting treatments for 0day, 20days, and 40days). Seeds of P. brachycarpa were, 20days of seed pre-treatment, germinated well at $5^{\circ}C$ and the more temperature went up, the more germination rate went down. As a result of surveying shading treatments, the height, leaf area, leaf length, leaf width etc were the highest (7.3cm, $12.8cm^2$, 5.4cm, and 2.3cm, respectively) under 50% shading. Specially, the root was grown well under 35% and 50% shading. Fresh weight of shoot (leaves+stem) was the highest under 50% and 75% shading and dry weight of shoot (leaves+stem) was the highest under 35% and 50% shading. As a result of surveying the whole experiment, it is concluded that the production of P. brachycarpa seedling is more effective under 50% shading.

Protease Activities in Tenderizing Effect of Vegetables used as Cooking Material (조리용 채소의 단백분해효소 활성 및 연육효과)

  • Protease activities were measured in vegetables used as cooking material or a side dish. Proteases of green pepper (Kwari and Chungyang), perilla leaves, soybean sprout and mungbean sprout were showed high activities. Especially, protease in soybean sprout was the highest activity among them. After dialysis, remaining activities in mungbean sprout, green pepper (Kwari and Chungyang) and perilla leaves were 12, 23, 45% and 37%. In the results of thawing after freezing the proteases, remaining activities in sprout of mungbean and soybean were 100% and 65%. Protease in soybean sprout was showed higher activity and stability than others. Proteolytic effects of soybean sprout on myofibrillar and sarcoplasmic protein were showed higher than stroma protein. In SDS-PAGE, myosin heavy chain, actin and tropomyosin were hydrolyzed with increasing time. Protease activity was rapidly increased with increasing meat concentration in a early proteolysis reaction, but was slightly increased in later.

Effects of Different NaCl Concentrations on the Growth of Suaedu asparagoides, Suaeda maritima, and Salicornia herbacea (염분농도에 따른 나문재, 해홍나물, 퉁퉁마디의 생장반응)

  • To investigate the effect of NaCl concentration on plant growth in three halophytes, Suaeda asparagoides, Suaeda maritima, and Salicornia herbacea, and to select afforestation species for reclaimed land along the shore, this experiment was conducted by ebb and flow system added fertilize of 1/2 Hoagland solution and halophytes were cultured for 4 month treated by 0, 50, 200, 400 mM NaCl, respectively. The shoot height was increased with 0 and 50 mM NaCl treatment of Suaeda asparagoides and Suaeda maritima and decreased with 200, 400 mM NaCl treatment, whereas them of Salicornia herbacea was increased by 200 mM NaCl treatment. The higher NaCl concentrations, the lower fresh and dry weight in Suaeda asparagoides. In case Suaeda maritima and Salicornia herbacea, fresh and dry weight was increased by 200 mM NaCl treatment. The higher NaCl concentrations, the lower chlorophyll contents in shoot of all treatment. The proline contents of 400 mM treatment in Suaeda maritima was higher than them of other treatments. The shoot water potential was generally lower in 50mM as compared to the control in Suaeda asparagoides, Suaeda maritima, significantly.

Flora of Mt. Samyeong (Yanggu-gun, Gangwon-do) in DMZ Area of Korea (DMZ 접경지역 인근 사명산(양구군, 강원도) 일대의 식물상)

  • This study was conducted to elucidate the distribution of vascular plants and their value of Mt. Samyeong. The vascular plants that were collected 10 times from October 2013 to July 2015 consisted of a total of 538 taxa; 92 families, 308genera, 466 species, 3 subspecies, 62 varieties and 7 forms. The observed rare plant species as designated by Korea Forest Service were 24 taxa. Endemic species were 25 taxa. 28 taxa were recorded as naturalized plants and their naturalized rate and urban index were noted as 5.3% and 8.7%. Among the surveyed 538 taxa, edible, medicinal, pasturing, ornamental, timber, dye, fiber and industrial plants included 220 taxa, 195 taxa, 167 taxa, 65 taxa, 25 taxa, 8 taxa, 6 taxa and 3taxa, respectively.

The Effects of Several Halophytes on Insulin Resistance in Otsuka Long-evans Tokushima Fatty Rats (OLETF 쥐에서 칠면초와 세발나물의 인슐린 저항성 개선 효과)

  • We evaluated preventive effects of Suaeda japonica (SJ) and Spergularia marina Griseb (SMG) on the insulin resistance in Otsuka Long-Evans Tokushima Fatty (OLETF) rats. The 10-week old OLETF rats were fed diets containing 3% (w/w) SJ and SMG for 18 weeks. Fasting blood glucose levels in SJ and SMG groups, measured using the oral glucose tolerance test, were lower than that of the control rats. The SMG group showed significantly lower levels of insulin, glycated hemoglobin, triglyceride, and total cholesterol than the control group. In addition, these levels were relatively lower in the SJ group than those in the control rats. The SJ and SMG groups had relatively lower protein levels of nuclear factor-kappa B (NF-${\kappa}B$) p65 in adipose tissue and serine phosphorylated insulin receptor substrate 1 (IRS-1) in skeletal muscle than the control group. These results suggest that SJ and SMG prevent insulin resistance and SMG in particular reduces blood triglyceride and total cholesterol levels.
