Control of Aflatoxin and Characteristics of the Quality in Doenjang(soybean paste) Prepared with Antifungal Bacteria (길항미생물에 의한 된장 중 아플라톡신 제어 및 그 품질특성)

  • In oder to acquire microbial agents that can be utilized for control of aflatoxin produced by Aspergillus. flavus and Asp. parasiticus, antifungal bacteria were isolated. Antifungal bacteria was identified as Bacillus spp. based on morphology and physico-biochemical characteristics. Amount of aflatoxin $B_1$ from Doenjang(soybean paste) prepared with Asp. flavus, Asp. parasiticus, antifungal bacteria(Bacillus sp.), or mixture of Asp. flavus and Asp. parasiticus was 27.2 ppb, 30.3 ppb, 3.4 ppb, and 3.7 ppb, respectively. Aflatoxin $B_1$ was not detected from Doenjang(control) and Doenjang prepared with antifungal bacteria. Content and compositions of free sugars, fatty acid, organic acid and free amino acid in Doenjang prepared with Asp. flavus and Asp. parasiticus, antifungal bacteria and mixture of Asp. flavus and Asp. parasiticus were not significantly different. For volatile flavor compounds of Doenjang prepared with antifungal bacteria, 2-pentyl furan and butanoic acid were disappeared or reduced, while octadecene compounds were produced. However, those of Doenjang prepared with Asp. flavus or Asp. parasiticus and Doenjang(control) were not significantly different. These results suggested that the antifungal bacteria(Bacillus sp.) inhibited production of aflatoxin and that antifungal bacteria did not effect the quality of Doenjang.

  • Fifty-three actinomycetes antagonistic to Phytophthora capsici and Magnaporthe grisea were isolated from rhizosphere soils in six pepper-growing areas and ashore soils. Thirty-two antagonistic actinomycetes, showing inhibition zone larger than 5 mm, were classified into 20 groups according to their colony morphology and color. The antagonistic activity against P. capsici greatly varied, which showed inhibition zone sizes in the ranges from 5.7 to 17.5 mm on V-8 juice agar and from 2.5 to 17 mm on tryptic soy agar. The antagonistic activity of some actinomycetes tested was remarkably different between the two test media. The antagonists showed a relatively broad antifungal spectrum, but their antibacterial activity was negligible, except for Pseudomonas solanacearum. Butanol extracts of culture filtrates from antagonistic actinomycetes inhibited mycelial growth of P. capsici and M. grisea, thereby confirming strongly antibiotic production in culture. Culture filtrates of some antagonistic actinomycetes completely inhibited Phytophthora blight in pepper plants.

Inhibition of Aflatoxin-producing Fungi with Antifungal Compound Produced by Bacillus subtilis (Bacillus subtilis가 생산하는 길항물질에 의한 아플라톡신 생성균의 억제)

  • An antifungal bacterium was isolated to inhibit of the growth of Asp. flavus and Asp. parasiticus, and its antifungal compounds were purified from lyophilized culture broth using chromatographic methods. Antifungal bacterium #19 which was shown a higher inhibitory activity on the growth of aflatoxin producing fungi was identified as Bacillus subtilis. The purified antifungal compound(1 mg) was demonstrated strong antifungal activity against the aflatoxin producing fungi.

Isolation and Utilization of Antagonistic Pseudomonas fluorescens from Soils for the Protection of Soybean Sprouts Rot (콩나물 부패병 방제를 위해 토양으로부터 분리한 길항균 Pseudomonas fluorescens의 이용)

  • Thirty-three bacterial and fungal strains were isolated from the rotten soybeans and soybean sprouts to isolate pathogenic microorganisms which cause soybean sprouts rot during soybean sprouts cultivation. In pathogenicity tests of the isolates on soybean sprouts, two isolates(K-17 and K-28) caused soybean sprouts rot and were identified as Erwinia carotovora and Fusarium sp., respectively. To isolate antagonists aganist K-17 and K-28 pathogens, bacteria were isolated from various soybean-cultivated soils and screened by the inhibition zone method. A bacterial isolate(J-232) which inhibited growth of both pathogens was identified as Pseudomonas fluorescens and further examined. The culture filtrate of P. fluorescens J-232 (dilution rate of 500 times) inhibited the growth of Erwinia carotovora K-17 and Fusarium sp. K-28 both on potato dextrose agar medium and on soybean sprouts cultivated in vessel. The development of soybean sprouts rots was observed during cultivation by inoculation of soybean seeds with culture filtrate of both pathogens. The combined inoculation of soybean seeds with culture filtrate of antagonistic bacterium and that of pathogens prevented soybean sprouts rot, and the growth of soybean sprouts was similar to that of control. The soybean sprouts inoculated with antagonists culture filtrate alone did not develop soybean sprouts rot, and the growth of the seedlings was shown to be slightly promoted as compared with that of control.

  • This study was carried out to clarify the effects of antifungal Streptomyces sp. isolated from the soil grown kiwifruit on the growth inhibition of fruit rot (Botryosphaeria dothidea) infected in kiwi fruit plants in the southwestern districts of Jeonnam. Two hundred and fifty microorganisms were isolated and examined into the antifungal activity against Botryosphaeria dothidea. We screened and isolated six bacterial strains which have a strong inhibition against Botryosphaeria dothidea. And the best antifungal strain designated as the strain #120 showing 96.0% antifungal activity against Botryosphaeria dothidea was finally selected. The strain #120 was identified as Streptomyces sp. #120 based on its morphological, physiological, biochemical and chemotaxonomic characteristics.

Biological Control of Blue Mold by Microorganism (잿빛 곰팡이병의 미생물학적 제어)

  • In order to screen the antagonistic bacteria which inhibit the growth of the plant pathogen, Penicillum expansum, we isolated an effective bacterial strain and investigated into the antifungal activity of the antagonist and it's identification. The eleven strains of bacteria which strongly inhibited P. expansum were isolated from the nature, and the best antagonistic bacterial strain designated as KB22, was selected. The antagonistic strain KB22 was identified to be the genus Bacillus subtilis based on morphological and biochemical characterization, The KB22 showed 55.9% of antifungal activity against the growth of P. erpansum. By the treatment of the culture broth and the heat treated culture filtrate of it, the B. subtilis KB22 showed 90% and 15% of antifungal activity, respectively.

  • The availability of Bacillus polimyxa 'HB 26-5' as a biological control agent was investigated. The antagonistic bacteria Bacillus polymyxa 'HB 26-5' grew well on the media at pH 7.0 and the optimum growth temperature was $25^{\circ}C$. The pH of the media changed to weak acid(pH 6.1~6.5) at the beginning of incubation, but to weak alkali(pH 7.8~8.2) at 7days after incubation. The best carrier to enhance colonization of the bacteria were the mixture of rice bran and peat, or rice bran and kaoline, in those formulation the density of the bacteria was changed slightly, though the density was beginning to decrease 3 weeks after application at field. In view of the physical characteristics of the formulation for the density maintenance during storage such as the hardness and the size, the best one was the formulation consisted of sodium alginate 2%, kaolin 15% and rice bran 3%.

Isolation of Antifungal Bacterial Strain Bacillus sp. against Gray Mold infected in Kiwi Fruits and its Disease Control (참다래 잿빛곰팡이 병원균에 대한 길항균 Bacillus sp. 분리와 병해 억제 작용)

  • This study was carried out to identity the effects of antifungal bacteria isolated from the soil grown kiwi fruit plants on the growth inhibition of Botrytis cinerea causing gray mold in kiwi fruit plants in the southern districts of Jeonnam. Two hundred and fifty antagonistic microorganisms were isolated and examined into the antifungal activity against Botrytis cinerea. We screened and isolated four bacterial strains which strongly inhibited Botrytis cinerea from the soil grown kiwi fruit plants. And the best antifungal bacterial strain which called CHO 163 was finally selected. Antagonistic microorganism CHO 163 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. Bacillus sp. CHO 163 showed 86.9% of antifungal activity against Botrytis cinerea. By the bacterialization of culture broth and heated filtrates of culture broth, Bacillus sp. CHO 163 showed almost all of antagonistic activity against Botrytis cinerea. And we also confirmed that in vitro the treatment of Bacillus sp. CHO 163 cultured by SD+B+P broth efficiently controled the growth of Botrytis cinerea causing gray mold in kiwi fruit plants.

  • The antagonistic microorganisms against Salmonella gallinarum causing fowl typhoid were isolated from the gut of wild pheasant. The isolated L19, L33, L50 strains were showed the characteristics of isolated Gram negative, rods, catalase positive and oxidase negative. Finally, all strains were identified as Sphingomonas sanguis by $Biolog^{\circledR}$ system. The optimal carbon sources of Sphingomonas sanguis L19, L33 and L50 for the these growth ~ere glucose, saccharose, and fructose respectively. But the optimal carbon sources of S. sanguis L19,L33, L50 for the antagonistic material production were maltose, galactose, and saccharose respectively. The optimal nitrogen sources of S. sanguis L19, L33, L50 for the growth were yeast extract, yeast extract, and $NH_4H_2PO_4$ respectively. But the optimal nitrogen sources of S. sanguis L19, L33 and L50 for the antagonistic material production were $(NH_4)_2SO_4$ urea, $(NH_4)_2S_2O_8$ espectively.
