Classification of Cordyceps spp. by Morphological Characteristics and Protein Banding Pattern (동충하초(冬蟲夏草)(Cordyceps) 속균의 형태적인 특징과 단백질 Pattern에 의한 계통 분류)

  • Ten species of Cordyceps species were collected throughout Kangwon province including Chuncheon Dongsanmyun KNU forest experiment from June to September, 1993. Collected Cordyceps species were identified as Cordyceps militaris, C. roseostromata, C. kyushuensis, C. scarabaeicola, Phytocordyceps ninchukiospora, C. nutans, Paecilomyces tenuipes, C. sphecocephala, Hymenostilbe odonatae, Torrubiella sp.. C. militaris, type species of Cordyceps species, was mainly formed on pupae of Lepidoptera and found after the rainy season around July. Fruiting body of C. roseostromata was morphologically similar to those of C. militaris, but relatively small in size and they were also found on lawn or pupa of Lepidoptera. Fruiting body of C. scarabaeicola was found on adult Scarabaeidae specifically and collect fruiting bodies of C. kyushuensis were on larva of moth. C. nutans and C. sphecocephala had host specificity on Hemiptera and Hymenoptera, respectively. Each species formed elliptical fertile part attach to the slim and carneous stalk and they were collected the most in specimen number through whole season of the summer. Ascospore of Phytocordyceps ninchukiospora on seed was characterized by two viable, multiseptate, fusiform units linked end-to-end by a long, filiform connective. Paecilomyces tenuipes, imperfect stage of the genus Cordyceps is multi-infective fungi that attack all stages of all groups of insects. Hymenostilbe odonatae attacks only adult Odonata and Torrubiella sp. formed on spider was difficult to collect because it was found the back side of leaf. As results of cultural test PDA medium showed the best mycelial growth. In the experiment of effect of the acidity inside of the media, C. militaris was good on pH 5, C. nutans and Phytocordyceps ninchukiospora were good on pH 6 and Paecilomyces tenuipes was on pH 7 and C. scarabaeicola was on pH 9. All isolates tested showed the best mycelial growth at $20^{\circ}C$. Morphologically similar isolates were used to analyze protein banding pattern among and within species. As a result, C. militaris, C. roseostromata and C. kyushuensis were clustered into close species and C. scarabaeicola and Phytocordyceps ninchukiospora were relatively distant from those species.

  • We surveyed on the host plants of Fungus gnat, Bradysia app. and found 21 species in the greenhouse and field. These are as follows: Cucumis sativus L., Cucumis melo L., Citrullus lanatus T., Cucurbita moschata F., Lycopersicon esculentum M., Capsicum annuum L (Pepper), Capsicum annuum L (Paprika), Lillium longiflorum T., Dianthus caryophyllus L., Rosa hybrida H., Gerbera jamesonii B., Chrysanthemum morifolium R, Phalaenopsis schilleriane R., Gladiolus grandiflours H., Zingiber officinale R., Cnidium officinale M., Canavalia gladiata DC., Angelica utilis M., Polygonatum odoratum D., Pinus densiflora S., and Pinus thunbergii P. Fungus gnat larvae cause damages to the root and promote decay and wilt by feeding on the roots and burrowing in plant tissue.

  • 기주식물 (토끼풀, Trifolium repens L.)에 기생하는 실새삼(Cuscuta australis R. Brown)의 흡기내 목부의 개체발생 과정을 광학현미경으로 조사하였다. 흡기의 목부 분화를 암시하는 최초의 해부학적 특징이, 흡기 자체내에서가 아니라, 그 흡기가 형성되어 있는 실새삼 줄기에서 나타났다. 즉 실새삼 줄기의 중심주의 유관속 내부 및 유관속 사이에서 세포분열 활성이 관찰되었다. 이 분열로부터 유도된 세포들은 짙게 염색된 세포질과 뚜렷한 핵을 갖고 있었다. 흡기가 기주조직에 침입하여 생장함에 따라, 실새삼 줄기의 중심주에서 시작된 세포분열 활성은 유관속 사이를 거쳐서 흡기의 기부를 향하여 구정적으로 확장되었다. 이와 동시에, 실새삼 줄기의 목부에 인접해 있던, 짙은 세포질 밀도를 갖는 세포들이 목부요소로 분화하기 시작하였다. 결국, 이미 형성되었던 짙게 염색된 세포들을 따라서 흡기의 목부는 구정적으로 분화하였다. 실새삼 줄기의 중심주에서 일어나는 유조직 세포들의 분열활성 및 이로부터 흡기의 목부 분화를 구정적으로 유도할 수 있는 가능성에 대하여 논의하였다.

  • Steinemematid and heterorhabditid nematodes were not effective to control the piU bug, Armadillidium vulgare although these nematodes were able to infect pill bugs. Steinernema carpocapsae Pocheon strain and S. glaseri Dongrae strain were more effective than S. carpocapsae AU strain or Heterorhabditis bacteriophora. Nematode concentration was more important factor than host density to develop infectivity.

  • Percent penetration on a specific rice variety was more affected by blast fungus isolate or predisposition temperature than by temperature and isolate combinations. A susceptible variety tested remained continuously susceptible regardless of whether the variety was grown at different temperature regimes and exposed to isolates/races with differences in pathogenicity and virulence. The expression of virulence by a particular blast fungus isolate race was observed to be changed by subjecting rice host plants to different predisposing temperature conditions prior to inoculation.

  • The relationships between the mean degree of hyphal growth in the leaf sheath and percent penetration in the leaf epidermis within the same predisposing temperature regime were highly significant. A positive relationship was found between degree of hyphal growth in the leaf sheath and Percent Penetration in the leaf epidermis.

Effect of Harvest Time and Infective Juvenile Size of the Entomopathogenic Nematode, Steinernema arenarium, on Pathogenicity, Development, and Propagation (곤충병원성선충, Steinernema arenarium의 수확시기와 유충 체장이 병원성과 발육 및 증식에 미치는 영향)

  • The size of infective $Steinernema$ $arenarium$ juveniles is variable and ranges from 724 to 1408 ${\mu}m$. Effects of harvest time and infective juvenile size on pathogenicity, development, and reproduction were examined in the last instar of the great wax moth, $Galleria$ $mellonella$. Harvest time of infective juveniles (IJs) of $S.$ $arenarium$ affected pathogenicity. IJs harvested at the 10th day from trapping were more pathogenic than those harvested the 3rd day from trapping. Mortality of $G$. mellonella also depending on harvest time, $i.e$, 100% died within 48h when IJs were harvested at the 10th day, without relation to size. However, mortality was 40% in the small size group (SSG) compared with 18% in the large size group (LSG) within 48h when IJs were harvested at the 3rd day. Establishment of $S.$ $arenarium$ within the host was different depending on IJ size. The number of established IJs was 1.8 in the SSG, 3.3 in the LSG, and 3.2 in the mixed size group (MSG) when IJs were harvested at the 3rd day, and 5.3 in the SSG, 7.4 in the LSG, and 7.6 in the MSG when IJs were harvested at the 10th day. The length of the female adult was 7,070.5 ${\mu}m$ in the SSG and 7,893.9 ${\mu}m$ in the LSG and that of the male was 1,460.5 ${\mu}m$ in the SSG and 1,688.2 ${\mu}m$ in the LSG when IJs were harvested at the 3rd day. The length of the female adult was 7,573.6 ${\mu}m$ in the SSG and 8,305.4 ${\mu}m$ in the LSG and that of the male adult was 1,733.4 ${\mu}m$ in the SSG and 1,794.4 ${\mu}m$ in the LSG when IJs were harvested at the 10th day. Harvest time and size of IJs did not influence numbers of progeny or size of IJS.

  • Tropical fire ant, Solenopsis geminata (Fabricius) is distributed over the tropical and temperate areas. It is a polyphagous pest attacking to various plant and causes the pain to the people doing outdoor work. So it is known for a hygienic pest to human being, especially the people doing outdoor activities. It can cause severe pain to human, even with just a bite. Of particular concern is the possibility of its settlement in this country, if it invade to the farmlands and/or forests of Jeju Island and southern part in Korea. It is listed as a regulated pest in Plant Quarantine Act of Korea. This ant is usually imported through a ship and early settled in the port area where it is already distributed. Recently its distribution is rapidly spreading into Southeast Asia and southern part of China. In Korea, it was detected the one time in 1990s, but was detected the ten times from 2000 to 2006 by an imported plant quarantine process. Therefore, we would like to provide some information on the species, including morphology, distribution, host plant, damaged symptom and its general bionomics, which will be useful data for plant quarantine process and control.

An Improved Collecting Method of the Infective Juveniles of the Entomopathogenic Nematode, Steinernema carpocapsae Weiser (감염태 곤충병원선충(Steinernema carpocapsae Weiser)의 효과적 회수법)

  • We report here an improved collecting method of the infective juveniles multiplied in the host insect with the entomopathogenic nematodes, Steinernema carpocapsae Weiser. The specific characteristics of this method involved the opening of the host insect hemocoel after the population of their infective juveniles reached at the maximum (6 days at 25$\^{C}$ after nematode treatment to nonimmunized host insects) to facilitate the escape of the multiplied nematodes. It also used 'Baermann funnel'method to select the infective juveniles effectively. This improved 'Baermann funnel'method was compared with a traditional collecting method, which was characterized with a combination of untreated host insects and 'White trap'collecting method, in both yield and pathogenicity of the collected infective juveniles to the fifth instar larvae of beet armyworm, Spodoptera exigua (H bner). More than 95% of the nematode populations collected by the two methods represented the morphological infective juveniles. To prove the nematodes to be infective juveniles functionally, pathogenicity and infective activity were compared in the nematodes collected by the two methods. They were not different in both pathogenicities and infective activities which were measured by the numbers of nematodes penetrated into the hemocoel of the insect hosts after exposure for the specific times to the same dote of infective juveniles. Significant difference between two collecting methods was found in the total yields of the infective juveniles per host insect About 50,000 infective juveniles per infected fifth instar larva of S. exigua after 6 day incubation at 25$\^{C}$ were collected only for 2 days by the improved 'Baermann funnel'method, while about 20,000 infective juveniles per host were collected for 10 days by the classical 'White trap'method.

Newly Introduced Insect Pest, American Serpentine Leafminer, Liriomyza trifolii (Burgess) (Diptera: Agromyzidae) in Korea (침입해충 아메리카잎굴파리(Liriomyza trigolii (Burgess))의 발생 보고)

  • The American serpentine leafminer (ASL), Liriomyza trifolii (Burgess), was fist found from a vinyl house planting gerbera in Kwangju area on January 26, 1994. To learn its distribution and damage pattern on crop plants, a nationwide survey was carried out at 293 vinyl houses from 25 cities/counties in six provinces. Its damage was found at 22 vinyl houses in Kwangju and Chinju areas. The range of leaves damaged by the ASL was 5-100% on gerbera, 40-70% on chrysanthemum, 20% on cherry-tomato, and 5-20% on celery. Host crop plants included gerbera, chrysanthemum, celery, cherry-tomato, watermelon, and pumpkin.
