Responses of Soil Chemical Properties and Microbiota to Elevated Temperature under Flooded Conditions (상승온도에 의한 담수토양의 미생물상 및 화학성 변화)

  • BACKGROUND: Our study aims to investigate the impact of temperature on the abundance and structure of soil microbial community in a temperature gradient tunnel.METHODS AND RESULTS: To investigate the interaction between temperature and input of C and N, rice straw and urea were applied to the study plots, respectively. We also studied the impact of plants by comparing plots cultivated with rice and unplanted plots. Soil microbial response was measured using the phospholipid fatty acid (PLFA) analysis. Soil chemical properties, including pH and ammonia and phosphate concentrations were influenced by warming and material addition. Microbial PLFA was partially influenced by material inputs, and actinomycetes PLFA was decreased by warming. In cultivated rice plots, an increase in the carbon to nitrogen ratio illustrated the effect of plant on microbiota caused by carbon addition through the root residues. Results from the principal component analysis of PLFA data showed that warmed and control plots applied with rice straw could be separated by principal component analysis.CONCLUSION: Our results suggest that plant influence both the microbial community structure and abundance, and temperature change has a minimal impact on soil microorganisms in flooded soil.

  • Various treaments such heating, acid tenting, acid healing, alkaline treating, acid+alkaline renting were attempted to investigate their effects on molar ratio, chemical composition, DTA and specific surface area(SSA) of natural zeolite poder. Molar ratio, Si to AI. of natural zeolite was 4.78, which represented high silica type. Composition of natural zeolite showed that $SiO_2$ was 66.34% $Al_2O_3$ was 13.89%, $Fe_2O_2$ was 1.55% X-ray diffraction showed that main component of natural zeolite was mordenite and clinoptliolite. Differential Thermal Analysis and Thermogravimetry curve of natural zeolite was showed to peak of endothermic peak at $80^{\circ}C$ and it means to the peak of dehydrate reaction, but recristalization was not formed below at $1,000^{\circ}C$. Weight loss during calcination was 16% at $1,000^{\circ}C$. Thermal treatments on SSA of natural zolite powder decreased from $75.2m^2/g\;to\;2.1m^2/g$. In contrast chemical treatments on SSA showed to increase to $300.2m^2/g$(1 N HCl treating), $54.9m^2/g$(1 N NaOH) and $90.9m^2/g$(HCl+NaOH)tudy, it could be proposed to employ acid tret method as packaging materisls for MA packaging.

Enhanced Production of Astaxanthin by Archaea Chaperonin in Escherichia coli (대장균에서 고세균 샤페론을 이용한 아스타잔틴 생산능 향상을 위한 연구)

  • The aim of this study is to increase production of carotenoids in recombinant Escherichia coli by Archaea chaperonin. The carotenoids are a widely distributed class of structurally and functionally diverse yellow, orange, and red natural pigments. These pigments are synthesized in bacteria, algae, fungi, and plants, and have been widely used as a feed supplement from poultry rearing to aquaculture. Carotenoids also exhibit diverse biological properties, such as strong antioxidant and antitumor activities, and enhancement of immune responses. In the microbial world, carotenoids are present in both anoxygenic and oxygenic photosynthetic bacteria and algae and in many fungi. We have previously reported cloning and functional analysis of the carotenoid biosynthesis genes from Paracoccus haeundaensis. The carotenogenic gene cluster involved in astaxanthin production contained seven carotenogenic genes (crtE, crtB, crtI, crtY, crtZ, crtW and crtX genes) and recombinant Escherichia coli harboring seven carotenogenic genes from Paracoccus haeundaensis produced 400 μg/g dry cell weight (DCW) of astaxanthin. In order to increase production of astaxanthin, we have co-expressed chaperone genes (ApCpnA and ApCpnB) in recombinant Escherichia coli harboring the astaxanthin biosynthesis genes. This engineered Escherichia coli strain containing both chaperone gene and astaxanthin biosynthesis gene cluster produced 890 μg/g DCW of astaxanthin, resulting 2-fold increased production of astaxanthin.

Establishment of Preparation Method of Dua-Chungkukjang (두아 청국장의 제조 방법 확립)

  • To increase the functional properties of black soybean, we prepared the Chungkukjang with black bean at $40^{\circ}C$ after its germination according to the time course. The black soybean was soaked for 3 hr and dried three times with 2 hr interval. The germination ratio reached 50% for 12 hr germination and 80% for 24 hr, respectively. The optimum temperature of the germination was $18^{\circ}C$. The general composition of the germinated black soybean were not different from the control black soybean. During germination and growth, protease activity decreased slightly, whereas glucoamylase activity increased. On the other hand, content of isoflavone was increased to two times when the germinated leaf was 20 mm length. Absolute amount of the isoflavone was about 260 ${\mu}g$/g. When we estimated the sensory test of the Dua-chungkukjang, the color and mucoid formation was not different from traditional chungkukjang but bad flavor was decreased compared to traditional chungkukjang. And the same result was detected in the Dua-chungkukjang soup.

Inhibitory effect of Koreinsis chinensis leaves extract on proinflammatory responses in lipopolysaccharide-induced Raw 264.7 cells (Lipopolysaccharide로 유도된 Raw 264.7 cell에서 잣 잎(Koreinsis chinensis L.) 추출물의 Pro-inflammatory 억제 효과)

  • Hyaluronidase inhibitory activity as inflammatory factor of Koreinsis chinensis leaf ethanol extract was showed higher inhibitory activity than water extract. 29.5% inhibitory activity was shown at concentration of $200{\mu}g/mL$ phenolics. Lipopolysaccharide (LPS)-stimulated Raw 264.7 cells were treated with different concentrations ($5-25{\mu}g/mL$) of Koreinsis chinensis leaf extract and the amount of nitric oxide (NO) was determined; LPS-treated cells produced 3 times more NO than non-LPS treated cells. Moreover, the NO production in cells treated with Koreinsis chinensis leaf extract showed inhibitory effect in a concentration-dependent manner. Due to the stimulant-induced NO production is regulated by the inducible nitric oxide synthase (iNOS), we determined the iNOS protein level to elucidate the mechanism by which the NO production was inhibited. It was reduced by 40% with a Koreinsis chinensis leaf extract concentration of $25{\mu}g/mL$ and identified iNOS inhibition in dose-dependent manner. The prostaglandin $E_2$ production in cells treated with Koreinsis chinensis leaf extract was reduced by 26.2% at concentration of $25{\mu}g/mL$. The protein expression of cyclooxygenase-2 in LPS-treated Raw 264.7 cells was inhibited by 64% at $25{\mu}g/mL$ of Koreinsis chinensis leaf extract. Koreinsis chinensis leaf extract had a concentration-dependent inhibitory effect on the production of tumor necrosis factor-${\alpha}$ and interleukin-6 as pro-inflammatory cytokine in LPS-treated Raw 264.7 cells at $25{\mu}g/mL$ of Koreinsis chinensis leaf extract. Their levels were decreased by 61.7 and 62% respectively.

  • 급속한 도시화 및 지구온난화로 인한 집중호우로 홍수피해가 해마다 증가하고 있다. 홍수피해를 최소화하기 위하여 4대강 중심의 홍수예경보시스템이 구축되는 등 다양한 제도적 장치가 마련되고 있으나 중소하천이 분포되어 있는 도시유역에서의 홍수예측기능은 부족한 실정이다. 본 연구에서는 중소 도시하천에 적용 가능한 실시간 도시홍수예측서비스 시스템(Realtime Urban Flood Forecasting Service, U-FFS)을 개발하였다. 경기도 성남에 위치한 탄천을 대상유역으로 선정하고 실시간 강우 및 수위관측소를 설치하여 수문데이타를 수집하였으며 이를 바탕으로 수위예측모형을 구축하였다. 모형구축에는 이미 국내외 학계에서 그 정확도가 입증된 바 있는 Data-driven 모델의 일종인 ANFIS(Adaptive Neuro-Fuzzy Inference System)를 이용하였다. 개발된 수위예측모형은 지정된 시간에 자동으로 작동 가능한 실행파일로 프로그래밍되어 최종적으로 홍수예측 웹서비스와 연동된다. U-FFS는 집중호우 발생 시 최종 유출구의 30분, 1시간, 2시간 후의 수위 예측값을 웹 상을 통해 제공함으로써 언제 어디서나 홍수예측 정보를 누구나 손쉽게 획득할 수 있는 장점이 있다. 시범운영 결과, 30분 및 1시간 후의 수위 예측은 정확도가 매우 뛰어났으며 2시간 후의 수위 예측의 정확성은 다소 떨어지는 것으로 확인되었으나 전반적인 홍수예측 판단에는 무리가 없을 것으로 예상된다. 본 시스템의 홍수예측모형은 생성 및 수정이 간편하여 그 활용성이 매우 높을 것으로 기대된다. 특히 안전함을 지향하는 각종 U-City나 홍수피해가 빈번한 도시유역에 적용하면 기존 시스템과 차별화된 실시간 홍수예측 서비스가 가능해져 홍수피해를 최소화할 수 있을 것이다. 취수구 직경 D의 3.3배를 벗어나지 않는다는 결과를 도출할 수 있었다.링 목적으로 사용될 수 있다. 본 연구에서 개발한 영상수위계는 한강홍수통제소 관할의 전류, 청담대교 등 4개소 낙동강 홍수통제소 2개소, 지자체 등에 적용되었으며, 적용 결과 비교적 안정적이면서 정확하게 수위를 측정하는 것으로 나타났다. 한편 기존 CCD 카메라 이외에 CCTV를 이용한 영상수위계를 개발하여 영상의 화질 개선뿐 아니라 하천화상 감시 기능을 강화하였다.소류의 섭취율은 높았다. 집단간의 상관도를 보면 교육별로 김치, 장아찌, 콩이 각각 p>0.5 수준에서 유의한 차가 없었고, 나머지는 유의한 차가 있었다. 연령별로는 멸치가 유의한 차가 없었고(p>0.5), 수입별로는 콩이 유의한 차가 없었다(p>0.5). 4. 영양지식(營養知識) 검토 가정생활(家庭生活)에 필요(必要)한 일반적(一般的)인 영양지식(營養知識)은 대체적으로 낮은 편이었다. 어린이 영양, 편식의 해로움, 비만증의 해로움, 임신부 그리고 수유부 영양에 대하여는 일반적으로 알고 있다고 하였으며, 그다음으로 이유기 영양, 어린이 발육에 필요한 식품, 식품과 영양소와의 관계, 우유의 성분, 노인영양에 대하여 잘 알고 있는 비율이 낮았으며, 인체의 영양소, 식단작성여부, 간식의 이론, 식품감별법에 대하여는 가장 낮은 비율을 나타냈다. 각 영양지식은 교육정도가 높을수록 영양지식이 높았고, 교육별 집단간의 유의한 차가 나타났다.   • Biological activities and application of Isodon japonicus H. were investigated. In the physiological activities, the electron donating ability (EDA) was 66.3% in 100 ppm and SOD-like activity was as high as 85.0% in 1,000 ppm with gradual increase. As for the inhibitory effect of xanthine oxidase, it was 70.0% in 1,000 ppm and as low as 40.0% in 500 ppm; also, as for the inhibitory effect of tyrosinase, it was as low as 20.5% below 1,000 ppm. The tyrosinase inhibition effect related to skin whitening function showed 30.0% at 1,000 ppm level or below, indicating a relatively low effect. As for the result of measuring the lipid oxidation, all the concentrations of medical ion treatments showed anti-acidification ability; also, as for the metal ion blocking effects against the lipid oxidation promoting factors $(Fe^{2+}\;and\;Cu^{2+})$, $Fe^{2+}$ was better than $Cu^{2+}$ and all concentrations of medical ion treatments was 60.0% in 100 ppm. Also, the clear zone against various bacteria at 0.5 and 1.0 mg/disc was clearly shown. When it was applied into a normal skin-softener, it was safe, showing its potential as a natural material of cosmetics.

  • This paper describe on the transmitting/receiving functions and the monitoring effects for the telemetry system which is designed to monitor the environmental condition of a culturing fishery ground inside a bay. The telemetry experiments were carried out by the telemetry system which constructed with the sea water temperature/salinity measuring sensor and telesounder at culturing fishery ground located in the coast of Sangyang-Myon, Kyongsangnam-Do province from October, 1995 to May, 1996. The results of this experiment showed that the developed telemetry system could be used for monitoring the sea water temperature/salinity and the distribution of fish in culturing fishery ground.

  • 이동 컴퓨팅 환경에서 다수의 이동 클라이언트들에게 데이터를 전달할 때는 클라이언트들의 동시 데이터 접근을 지원하는 브로드캐스트 기법을 사용하면 무선 채널 대역폭의 협소함과 클라이언트 측의 에너지 제약과 같은 단점을 해결할 수 있다. 기존 기법들은 클라이언트의 데이터에 대한 접근빈도(access frequencies)와 클라이언트의 에너지 효율(energy efficiency)을 동시에 고려하지 않았다. 따라서 원하는 데이터가 올 때까지 계속해서 채널을 들어야 함으로 인해 에너지 소비를 많이 하거나, 데이터를 얻을 때까지 추가한 많은 양의 정보에 따른 지연이 발생하는 단점이 있다. 본 논문에서는 클라이언트의 에너지 절약을 위한 tuning time을 최소화하고 실제 데이터를 얻을 때까지 소요되는 access time의 효율을 높이기 위해 데이터의 접근빈도와 에너지 효율을 동시에 고려하는 HIDAF: Hybrid Index considering Data Access Frequencies 기법을 제한한다. 제안하는 기법은 트리기반 기법과 해싱기반 기법을 함께 적용하여 구성한 인덱스를 브로드캐스트 주기에 배치한다. HIDAF 기법은 데이터 접근빈도를 고려한 트리기반 인덱스를 배치함으로써 데이터를 얻기 위한 클라이언트들의 평균 access time을 줄일 수 있고, 이러한 인덱스에 해싱기반 기법을 추가함으로써 클라이언트의 에너지 효율을 최소화하는 새로운 브로드캐스팅 기법이다. HIDAF 기법은 브로드캐스트 추기에 데이터의 접근빈도를 고려한 인덱스를 교차로 추가하여 핫 데이터에 대한 클라이언트들의 access time을 줄임으로써 전체 사용자에 대한 평균 access time을 최소화하는 동시에 클라이언트들의 제한된 에너지 소비를 최소화하는데 목적이 있다. 제안기법에 대한 평가는 수학적 분석을 통해 HIDAF 기법과 기존의 브로드캐스트 기법의 성능을 비교 분석한다.하였으나 사료효율은 증진시켰으며, 후자(사양, 사료)와의 상호작용은 나타나지 않았다. 이상의 결과는 거세비육돈에서 1) androgen과 estrogen은 공히 자발적인 사료섭취와 등지방 침적을 억제하고 IGF-I 분비를 증가시키며, 2) 성선스테로이드호르몬의 이 같은 성장에 미치는 효과의 일부는 IGF-I을 통해 매개될 수도 있을을 시사한다. 약 $70 {\~} 90\%$의 phenoxyethanol이 유상에 존재하였다. 또한, 미생물에 대한 항균력도 phenoxyethanol이 수상에 많이 존재할수록 증가하는 경향을 나타내었다. 따라서, 제형 내 oil tomposition을 변화시킴으로써 phenoxyethanol의 사용량을 줄일 수 있을 뿐만 아니라, 피부 투과를 감소시켜 보다 피부 자극이 적은 저자극 방부시스템 개발이 가능하리라 보여 진다. 첨가하여 제조한 curd yoghurt는 저장성과 관능적인 면에서 우수한 상품적 가치가 인정되는 새로운 기능성 신제품의 개발에 기여할 수 있을 것으로 사료되었다. 여자의 경우 0.8이상이 되어서 심혈관계 질환의 위험 범위에 속하는 수준이었다. 삼두근의 두겹 두께는 남녀 각각 $20.2\pm8.58cm,\;22.2\pm4.40mm$으로 남녀간에 유의한 차이는 없었다. 조사대상자의 식습관 상태는 전체 대상자의 $84.4\%$가 대부분이 하루 세끼 식사를 규칙적으로 하고 있었으며 식사속도는 허겁지겁 빨리 섭취하는 경우가 남자는 $31.0\%$, 여자는 $21.4\%$로 나타났고 이들을 제외한 나머지 사람들은 보통 속도 혹은 충분한 시간을 가지고 식사를 하였다. 평소 식사량은 조금 적게 혹은 적당하게 섭취하는 사람이 대부분이었으며 남자가 여자보다는 배부르게 먹는 경 향이 유의적으로 높았다(p<0.05). 식사는 혼자 하는 경우가 남자

Comparison of Gingerols in Various Fractions and the Antioxidant Effects of Supercritical Fluid Extracts from Ginger (생강 분획의 gingerols 분석 및 초임계 추출물의 항산화 효과)

  • Ginger (Zingiber officinale Rosc.) is a well known edible plant that is traditionally used to treat various illnesses related to inflammation and oxidative stress. Steam distillated ginger oil or water extract are mainly used for related products; however, it is unclear whether these fractions contain most of the bioactive compounds or the highest efficacy. This investigated the antioxidant effects of extracts prepared by supercritical fluid extraction (SFE). 6-Gingerol was the most abundant component in hexane fraction of ethanol extract from ginger. The antioxidative properties of SFE oil and Marc ethanol fractions were demonstrated using the 2,20-diphenylpicrylhydrazyl (DPPH) and 2,2'-azinobis 3-ethyl benzothiazoline-6-sulfonic acid (ABTS) radicals scavenging assays. A clear correlation was observed between total polyphenol contents and $RC_{50}$ values in the Marc ethanol fraction. These results indicate that not only SFE oil but the marc after SFE could be good sources for the food industry.
