Properties of Powder and Fluorescence as a Function of Oxygen Partial Pressure in ZnO : Zn System Prepared by Glycine Nitrate Process (GNP 방식으로 제초한 ZnO : Zn의 산소분압에 따른 분말특성 및 형광특성)

  • 저 전압용 형광체는 최근에 활발히 연구가 진행되고 있으며 가장 대표적인 형광체가 ZnO Zn 녹색 형광체이다. ZnO : Zn 형광체는 자체발광형 형광체로써 ZnO을 환원분위기 하에서 열처리를 함으로써 얻을 수 있다. 본 연구에서는 자발착화 연소반응법(Glycine Nitrate Process)을 이용하여 ZnO : Zn 분말을 합성하고 형광특성 및 분말특성을 알아보았다. 출발물질로는 Zn Nitrate와 Glycine을 이용하였고 자발연소 반웅이 발생하는데 적절한 글리신의 양을 확인하기 위해서 글리신과 양이온의 비를 변화시키며 ZnO를 합성하였다. 그리고 Zn Excess가 생겨난 앙과 그에 따른 형광특성을 관찰하기 위해 $N_2$ 분위기 에서 각기 50$0^{\circ}C$, 75$0^{\circ}C$, 95$0^{\circ}C$의 온도에서 열처리를 행하였다. 제조된 ZnO 분말의 입자형태와 결정상 태는 SEM과 XRD를 이용하여 분석하였고 TG-DTA를 측정하여 열처리온도에 따른 질량감소(Zn excess)를 관찰하였다. 또 Particle size analyzer로 분말의 크기를 알아보았고 형광체로써의 발광특성을 살펴보기 위해 PL을 이용하여 발광피크를 관찰하였다.

Transmethylase 억제제(TMI)의 활성 및 작용기전 연구개발

  • TMI는 알라닌과 글리신이 풍부한 18개의 아미노산으로 이루어져 분자량이 약 1,400Da인 물질이었으며, 가수분해 효소 처리후 억제력의 변화를 보이지 않았고, Porcine liver, spleen, testis에서 정제한 PM II에 대하여 TMI가 비상경적 억제작용을 하는 것으로 나타났으며, Ki value는 각각 300 nM, 250 nM 297 nM이었다. 그리고 돼지 간장에서 부분정제한 PM I, PM II, PM III, phospholipid methytransferase에 대한 정제된 TMI의 영향을 조사해 보면 각각 33%, 47%, 31%, 35%의 억제효과를 나타내었다.

  • Several Pt(II) complexes containing glycine (or glycino anion) and styrene have been prepared, and their structures have been confirmed by infrared spectroscopy. The results confirm the structure of chloro(glycino)(styrene)platinum(II) in which the nitrogen atom of the chelated glycino anion is coordinated to the platinum in the trans position to styrene (N-trans isomer).

Enhancing the Alginate Degrading Activity of Streptomyces sp. Strain M3 Alginate Lyase by Mutation (Streptomyces sp. M3 알긴산분해효소의 돌연변이에 의한 활성증대)

  • A polyguluronate-specific lyase from Streptomyces sp. strain M3 has been previously cloned and characterized. In this study, the M3 alginate lyase gene in the pColdI vector was mutated by site-directed mutagenesis and random mutagenesis to enhance the alginate degrading activity. Six mutants were obtained: Ser25Arg, Phe99Leu, Asp142Asn, Val163Ala, Lys191Glu, and Gly194Cys. Phe99Leu and Lys191Glu mutants completely lost their alginate lyase activity, whereas the alginate degrading activity of Gly194Cys mutant increased by nearly 10 fold. The 3-D protein structure of M3 alginate lyase, which was constructed using the Swiss-Model automodeler, was also compared to the crystal structure of another alginate lyase. A mutated glycine residue was positioned between Gly193 and Tyr195 of the C-terminal conserved sequence, YFKAGXYXQ. A phenylalanine residue (at position 99) and a glycine residue (at position 194) mutated in this study were distant from the active site, but the degrading activity was strongly affected by their mutation.

Inhibition of Enzymatic Browning of Crown Daisy by Maillard Reaction Products (마이얄반응생성물(Maillard reaction product)에 의한 쑥갓의 효소적갈면 억제)

  • The study was conducted to investigate the effect of Maillard reaction products (MRPs) on enzymatic browning of crown daisy (Chrysanthmum coronarium var. spatiosum). The MRPs prepared by heating various amino acid and sugar at $90^{\circ}C$ caused a strong inhibitory effect on crown daisy polyphenol oxidase (PPO, ${\sigma}$-diphenol oxygen oxidoreductase, EC 1.10.3.1). As the reaction time of the solution containing glycine and glucose increased at $90^{\circ}C$, the production of MRPs was increased, whereas the amounts of glycine and glucose were decreased. Accordingly, the inhibitory effect of crown daisy PPO activity by MRPs was increased as the amounts of synthesized MRPs were increased. The MRPs synthesized from the various amino acids and sugars significantly reduced the PPO activity, particularly MRPs prepared by glutamine and xylose. The Michealis-Menten constant value ($K_m$) of crown daisy PPO with catechol as a substrate was 22.0 mM, and MRPs were a noncompetitive inhibitor against crown daisy PPO.

Theoretical Studies on Conformation of Cyclic Hexapeptides and Blocking for the Cation Transport by 5,5-diphenylhydantoin (고리형 헥사펩티드의 형태 및 5,5-디페닐하이덴토인에 의한 양이온 전달방해에 관한 이론적 연구)

  • The conformations of hexapeptides, their complexation with alkali cations and the inhibition of the cation transport by 5,5-diphenylhydantoin(DPH) were studied theoretically using ECEPP/2 and MM2 force field. Several low energy conformations of uncomplexed cyclic hexapepides are obtained, and they adopt compact conformations in which most amide hydrogens form intramolecular hydrogen bond to amide carbonyl oxygens. The complexation energy of the peptide with $Na^+$ ion and DPH is -60 kcaal/mol and -18 kcal/mol, respectively. However, no suitable cavity to bind metal cation exists for the local minima of the peptide, and the internal energy of the uncomplexed hexapeptide having cavity is higher than that of the uncomplexed global minimum of this work by 10 kcal/mol. Also, one of the most important amino acid residue to bind DPH is Glycine, and this can explain experimental observation that the replacement of Gly by Sarcosine (N-methyl Glycine) reduce the inhibition ability of the cation transport.

  • Glycine residue in N-X-glycylphenylalanine methyl ester(X = trifluoroacetyl or acetyl) was converted into aspartic acid derivative by a photoalkylation reaction. The reaction was induced with 350 nm lamp using a combination of diacetyl/di-t-butyl peroxide (DBP) as the photoinitiator, and acetic anhydride as the alkylating agent. In the thermal reaction with DBP and acetic anhydride, the same alkylation reaction of the dipeptide was observed. From this thermal alkylation reaction the photoalkylation reaction is also thought to undergo via free radical mechanism.

고속 저응력 무전해 Cu 도금액 개발

  • 본 연구는 고속 저응력 무전해 Cu 도금액 개발에 관한 것으로 환원제로 포름알데히드와 차아인산나트륨을 혼합하여 개발하였다. 이때 도금액에 차아인산나트륨이 혼합될 경우 도금속도는 증가되나 도금액의 안정성은 감소하여 착화제로 글리신을 혼합하였다. 일정량의 착화제가 포함된 도금액에 차아인산 나트륨의 첨가량을 변화시켜 개발된 도금액의 도금속도를 관찰한 결과($7.2{\mu}m/hr{\rightarrow}12.5{\mu}m/hr$)로 도금속도는 급격히 증가한 것을 확인할 수 있었다. 또한 개발된 도금액의 응력값은 ($-150Mpa{\rightarrow}-50{\sim}10Mpa$)로 급격히 감소하는 것을 확인하였다.
