Initiation of spore germination in filamentous fungi such as Aspergillus nidulans and Botrytis cinerea requires the presence of nutrients. In this study, involvement of sugar sensing machinery was suggested in the germination of A. nidulans spores. Germination did not occur when the spores of A. nidulans were incubated in distilled water, whereas they were successfully germinated in the presence of 5% glucose with a germination rate of over 98% after 6hr incubation. Similar results were obtained when the spores were incubated in the presence of various sugars such as fructose, sucrose, and starch. Interestingly, spore germination was not observed in the presence of D-arabinose, whereas L-arabinose could induce germination as determined by the formation of germ tubes, indicating the presence of sugar sensing machinery that distinguish between the enantiomers of sugars. This inference was further supported by a decrease in germination rate (less than 25%) upon treatment of spores with trypsin. Subsequent MALDI-TOF mass spectrometry analysis of the surface proteins of spores identified ten proteins among which eight were involved in sugar metabolism. Taken together, our results suggest that spore germination in A. nidulans is initiated by the interaction of sugars with sugar binding proteins on the surface of spores.
전기화학발광(ECL) 시스템은 높은 감도와 넓은 선형동작영역을 가지므로 분석화학에서 넓게 연구 되고 있다. 그러나 포토멀티플러, 광섬유와 플루오르메터로 구성된 ECL 검출 시스템은 크기가 크고, 전력소모가 많으며, 고가인 단점이 있다. 따라서 이러한 단점을 보완하기 위해 p+n 포토다이오드를 이용한 마이크로 ECL시스템을 제작 과산화수소 농도를 50uM${\sim}$10mM 범위에서 측정하였고, 글루코오스는 1mM${\sim}$20mM 농도범위에서 측정하였다. 따라서 마이크로 ECL 시스템이 바이오센서 혹은 바이오 분석기로써의 가능성을 확인 하였다.
FRIR-ATR spectrometry has been used to monitor the aqueous reactions of compounds without distinct chromophores in ultraviolet and visible regions. For example, hydrolysis reactions of ${\alpha}$-cyclodextrin and ${\gamma}$-cyclodextrin in acidic aqueous solution were studied. FTIR-ATR method has been used for the monitoring of cyclodextrin hydrolysis in 1.0 M. 0.5 M, and 0.1 M HCl solutions, respectively. We also found that the hydrolysis of ${\alpha}$-cyclodextrin produced glucose, but the hydrolysis of ${\gamma}$-cyclodextrin proceeded further to give more fragmented products than glucose.
A differential refractive index detector was used for monitoring glucose contents in textile processing solutions. The sensitivity of the device was high enough to measure 0.05% aqueous glucose solution that could not be measured by normal refractive index measurement. The device was set to monitor glucose concentration continuously in real time by measuring differential refractive index and calibrated by standard glucose solutions in a range of 0.1 to 1.0%. The possibility of industrial application of the device was demonstrated by real-time monitoring of glucose concentration in textile processing solutions such as desizing bath and cellulase treatment bath. Both of solutions contained glucose as a major degraded product. The device would be able to control the weight loss of cellulosic fiber during cellulase treatment since the amount of degraded products in a processing bath is proportional to its weight loss.
The effect of diacylglycerol on glucose release was studied by using 1,2-dioctanoyl-sn-glycerol ($diC_8$), a cell permeable diacylglycerol, in perfused rat liver. The glucose release was increased by $diC_8(50\;{\mu}M$), and the effect was depended on calcium ions. The increment of glucose release by $diC_8(50\;{\mu}M$) was inhibited by indomethacin ($50\;{\mu}M$); the amount of glucose release was almost the same with that of control group. Arachidonic acid($200\;{\mu}M$) also increased glucose release and the release was inhibited by indomethacin. There was no synergistic effect on glucose release by the combination of $diC_8(50\;{\mu}M$) and phenylephrine($10\;{\mu}M$).
This study was described for measuring clinically relevant levels of glucose in undiluted plasma and whole blood by near-infrared (NIR) spectroscopy. Result from an initial measurement of major blood components powder was over-lapped the absorption bands of glucose at 1500-1600 nm. However, the NIR data of blood components were clearly separated by principle component analysis (PCA) space. By the use of partial least squares (PLS) regression, glucose concentrations in undiluted plasma and whole blood could be determined with standard errors of prediction (SEP) of 15 mg/dl and 76 mg/dl, respectively. Although these blood components possessed strong absorption bands that overlapped with the absorption bands of glucose, successful calibration models could be carried out.

The main aim of this study was to investigate the potential of wood charcoal on removing furan compounds (5-hydroxymethylfurfural (5-HMF), furfural) known as fermentation inhibitors in sugar hydrolysates obtained from supercritical water treatment of lignocellulosic biomass. For this aim, model hydrolysate was prepared, and removal rates of sugars or furan compounds depending on wood charcoal concentration and treatment time were calculated and analyzed in comparison with the case of activated carbon. 0.5, 1, 2, 4, 8, or 12% (w/v) of wood charcoal or activated carbon was loaded into the model hydrolysate, containing glucose, xylose, 5-HMF, and furfural, and treatment was conducted for 1, 3, 6, 12, or 24 h. After treatment, removal rates of 5-HMF and furfural gradually increased as wood charcoal concentration or treatment time increased, and over 95% of 5-HMF and furfural were removed at 8% of wood charcoal concentration and 3 h of treatment time, while the loss of sugars (< 2%) was hardly observed. On the other hand, in the case of activated carbon treatment, removal rates of 5-HMF and furfural were over 95% at mild condition (activated carbon concentration: 8%, treatment time: 1 h), but over 10% of glucose and xylose were removed. Therefore, considering sugar production and further process applied sugar, the wood charcoal treatment of sugar hydrolysate was more effective for removing furan compounds and maintaining the sugar yield.
A simple, fast and reproducible quantitative analysis method for sugar concentration composed in oligosaccharide mixture was developed. Two glass TLC plates were prepared per sample. After dipping one plate into the copper bicinchoninate reagent and the other plate into 5% sulfuric acid solution, both plates were baked in microwave oven until sugar spots were developed or the surface temperature of TLC plate becomes 60 to 70 $^{\circ}C$. The corrective factor values [F value =(the value of total sugar concentration converted as glucose unit/the value of reducing sugar concentration converted as glucose unit)/(polymerization degree of sugar)] of different molecular weight sugars were determined. Within the concentration of 0.25∼1.0 $\mu\textrm{g}$ in each sample loaded, the fructose-F (corrective factor value of fructose) was 0.45, yet for the higher concentration (2.5∼7.5 $\mu\textrm{g}$) fructose-F was 1.0. In case of glucose, in the range of 0.5∼7.5 $\mu\textrm{g}$, glucose-F was same as fructose-F, 1.0. However, as the molecular weight of sugar was increased, the F values were decreased in both maltodextrin and isomaltodextrin oligosaccharides in 0.5∼7.5 $\mu\textrm{g}$ of each sample loaded. Interestingly, F values were equal for the same molecular weight sugars, although the structures were different from each other. Using F value of each sugar, we could determine and compare the exact total sugar concentration of different molecular weight maltooligosaccharide and isomaltooligosaccharide. We also could determine if the unknown sugar was a reducing or non-reducing compound by using optimized TLC with microwave oven method.

Long term hydrogen production was investigated in an anaerobic sequencing batch reactor (ASBR) using mixed microflora. Glucose (about 8,250 mg/L) was used as a substrate for the ASBR operation under the condition of pH 5.5 and $37^{\circ}C$ with mixing at 150 rpm. The experiment was carried out over a period of 160 days. Hydrogen yield was 0.8mol $H_2/mol$ glucose with F/M ratio 2 at initial operation period. The hydrogen yield reached to maximum 2.6 mol $H_2/mol$ glucose at 80th day operation. However decreased hydrogen yield was observed after 80 days operation and eventually no hydrogen yield. Although well-known hydrogen producer Clostridium sp. was detected in the reactor by PCR-DGGE analysis, changed reactor operation was the major reason of the decreased hydrogen production, such as low F/M ratio of 0.5 and high propionic acid concentration 2,130 mg/L. Consequently the long period operation resulted in MLSS accumulation and then low F/M ration stimulating propionic acid formation which consumes hydrogen produced in the reactor.
Thermotropic liquid crystalline behavior of a homologous series of penta-O-4-{4'-(cyanophenylazo)phenoxy}alkyl-D-glucopyranoses(CAGETn, n = 2~10, the number of methylene units in the spacer) has been investigated. The CAGETn with n of 2 and 7 exhibited enantiotropic nematic phases whereas other derivatives showed monotropic nematic phases. This is the first report of glucose derivatives that form thermotropic nematic phases. The isotropic-nematic transition temperatures ($T_{iNS}$) of CAGETns and their entropy variation at $T_{iN}$ showed the odd-even effect as a function of n. This behavior was rationalized in terms of the change in the average shape of the side chains as the parity of the spacer is varied. This rationalization also accounts for the observed variation of nematic-crystalline phase transition temperatures ($T_{NkS}$) and associoated entropy change at $T_{Nk}$. The entropy change at $T_{iN}$ or $T_{Nk}$ reaches a mininum at n = 3, before it increases again for n = 4. This may be attributed to the difference in the arrangement of the side groups. The mesophase properties of CAGETns were entirely different from those reported for partially or fully alkylated glucopyranoses. This result suggests that the degree of substitution and chemical structure of the substituents play an important role in the formation of the mesophase structures in the liquid crystals.
