Effects of Culture Conditions on Organogenesis in Gladiolus 'Topaz' Callus (글라디올러스 'Topaz' 캘러스의 기관형성에 미치는 배양 조건의 영향)

  • This study was carried out to establish improved techniques on organogenesis from callus culture of Gladiolus. Organogenesis from the callus was effective in the half strength of MS solid medium without 2,4-D at 15 $^{\circ}C$ under 24 hours of daylength. Formation of adventitious root was most effective in the liquid shaking culture, and adventitious shoot induction was effective in the liquid stationary culture. From these results, we could find optimal culture conditions for redifferentiation from callus, in addition, liquid shaking culture revealed as more useful when compared with that of solid culture method for the redifferentiation of callus in Gladiolus `Topaz'.

'Arirang', Early-flowering Pure White Gladiolus with Purple Core (조생 순백색 자주 화심 글라디올러스 '아리랑')

  • 'Arirang', an early-flowering pure white gladiolus with purple core, was released by the National Institute of Horticultural & Herbal Science (NIHHS) in 1998. The selection was derived from the cross between the progenies of the early flowering dark orange 'Arianne' and bright red domestic cultivars 'Hongkwang'. The breeding was conducted in 1993 and finally selected in 1998 after five (5) years (1994 to 1998) of experimentation. 'Arirang' is a cut flower with bright white color petals and purple core which exhibited early flowering and vigor without stem bending. These can be cultivated in areas or regions with strong winds during summer. 'Arirang' has a very firm growth form and sturdy branches which do not bend when planted in the open fields. Likewise, the plant had high tolerance to flooding and resistance hence could grow vigorously in open areas. It is a fast- growing plant which exhibits high corm production rate. 'Arirang' had a 10.5 cm wide flower and 140 cm plant height. During summer, the average number of days to flowering was recorded at 75 days of 'Arirang'. Top dressing must be conducted once or twice during corm production to enhance corm quality and quantity.

  • Erwinia carotovora subsp. carotovora, Pseudomonas viridiflava and Pseudomonas marginalis pv. marginalis were tested for their pathogenicity to 35 kinds of domestic flowering plants. Among them, the following domestic flowering plants showed clear symptoms. 1. Erwinia carotovora subsp. carotovora : (Needle inoculation). Carnation(Dianthus catgophylius L.), madagascar periwinkle(Vinca rosea L.), flower gentle(Amaranthus tricolor L.), snapdragon(Antirrhinum majus L.), chrysanthemum(Chrysanthemum morifolium Ram.), mexiacan ageratum(Ageratum houstonianum Mill), china aster(Callistephus chinensis), youth and old age(Zinnia elegans Jacq.), common nasturtium(Tropaeorum majus L.), scarlet sage(Salvia splendens F.), dahlia(Dahia hybrida), pot marigold(Calendula officinalis L.), begonia treevine(Cissus dicolor Blume), cosmos(Cosmos bipinnatus Cav.), globe amaranth(Gomphrena globosa L.), black eyed susan(Thumbergia alata Bojer), common gypsophila(Gypsophila elegans Bieb.), ghent gladiolus(Gladiolus gandavensis Van.), indian shot(Canna orchiodes Bailey), iris(Iris nertschinskia Lodd), cyclamen primula(Dodecatheon meadia L.), scarlet kafir lily(Clivia miniata Regel.), flowering cabbage(Brassica oleracea L.). (Spray inoculation). Common nasturtium, ghent gladiolus, indian shot, cyclamen primula. 3. Pseudomonas marginalis pv. marginalis : (Needle inoculation). Carnation, madagascar periwinkle, flower gentle, snapdragon, cockscomb, mexican ageratum, youth and old age, common nasturitium, common petunia, sweet william(Dianthus barbatus L.), pot marigold, begonia treevine, cosmos, common gypsophila, ghent gladiolus, indian shot, iris, cyclamen primula, scarlet kafir-Lily, flowering cabbage. (Spray inoculation). Common nasturtium, ghent gladiolus, indian shot, cyclamen primula.

Proliferation of Gladiolus 'Topaz' Callus by Liquid Shaking Culture (액체진탕배양에 의한 글라디올러스 'Topaz' 캘러스의 증식)

  • This study was performed to enhance the proliferation rate of Gladiolus 'Topaz' callus. The callus was induced from the cermet tissue explants on MS solid medium with 10 mg/L 2,4-D. In the case of liquid shaking culture, proliferation of the callus was effective in MS medium with 0.05 mg/L 2,4-D at 2$0^{\circ}C$ under 16 hours daylength and in a 100 mL Erlenmeyer flask containing 20 mL of the liquid medium and at 75 rpm in rotation speed of the horizontal shaking culture. Furthermore the callus was also able to be subcultured in the same liquid medium.

  • This study aimed to identify leaf sheath function through the investigation of effect of removal times and removed number of leaf sheath on growth and flowering in gladiolus. The leaf sheath influenced on the growth and flowering regardless of light condition at early growth stages. When total leaf sheath were removed in early growth stages, flower stalk length and spike length remarkably decreased. Whereas days of flowering were little different by leaf sheath removal. Also when removed numbers of leaf sheath were 1, 2 and 3, the growth and flowering were little different compared to control. But when removed numbers of leaf sheath were 4, flower stalk length decreased above 25cm compared to others treatment, spike length and number of flower decreased and stem diameter became thick abnormally.

  • 'Duo', a late-flowering white gladiolus with pink core, was released by the National Institute of Horticultural & Herbal Science (NIHHS) in 2000. The selection was derived from the cross between the progenies of the early flowering dark pink 'True Love' and bright pink domestic cultivars 'Madame Valdiek'. The breeding was conducted in 1996 and selected finally in 2000 after four years (1997 to 2000) of experimentation. 'Duo' is a cut flower with bright white color petals and Pink core which exhibited late flowering and vigor without stem bending. It can be cultivated in areas or regions with strong winds during summer. 'Duo' has a very firm growth form and sturdy branches which do not bend when planted in the open fields. Likewise, the plant showed high tolerance to flooding and resistance, hence, could grow vigorously in open areas. It is a fast- growing plant which exhibits high corm production rate. 'Duo' had a 12.2 cm wide flower and 150 cm plant height. During summer, the average number of days to flowering was recorded at 75 days of 'Duo'. Top dressing must be conducted once or twice during corm production to enhance corm quality and quantity.

Characterization of Tobacco rattle virus(TRV-K) isolated in Korea (한국에서 분리한 Tobacco rattle virus(TRV-K)의 특성)

  • Tobacco rattle virus(TRV) was detected from Gladiolus hybridus, Crocus spp. and Narcissus spp. leaves show-ing notched or stripe on the leaf and malformation symptoms collected from Daegu and Kyungbuk province by electron microscopy (EM), immunosorbent electron microscopy (ISEM) and host range study. Direct negative staining method by EM showed rigid rod long particles 170~200$\times$22 nm and rigid rod short particles 40~114$\times$22 m. TRV-K isolated from G. hybridus propagated with Nicotiana tabacum. TRV coat protein(CP) gene was amplified using specific oligonucleotide primer by RT-PCR. Sequence analysis of amplified CP gene showed 99.5% nucleotide similarity to TRV-ORY.

The Preference Rate of Spider Mites (Tetranychus urticae and T. kanzawai) on the Cut-Flowers (절화류에 대한 점박이응애와 차응애의 기주 선호성)

  • The preference ratio of spider mites (Tetranychus urticae and T. kanrawai) on the chrysanthemum, Gypsophila paniculata, Carnation and Rose was high, but Lilium, Calla, Startis and Gladiolus was low. Field adaptability of Tetranychus kanzawai on the chrysanthemum variety was high in Yellowment variety but low in five other varieties. That of T. urticae line was high on 9818-1, 9819-5, 9820-6 chrysanthemum lines but low on other six lines . Preference ratio of T. urticae was 82.8% higher on upper than on lower surfaces in the field.

The Viruses in Gladiolus hybridus cultivated in Korea 2. Broad Bean Wilt Virus, Cucumber Mosaic Virus and Tobacco Rattle Virus (한국산 글라디올러스에 발생하는 바이러스)

  • Gladioli (Gladiolus hybridus) showing flower colour breaking leaf mosaics, notched leaf, and dwarfing or lack of visible symptoms were collected from gladioli growing areas in Taegu and Kyungpook province, Korea. The three viruses isolated from the naturally infected gladioli were identified as broad bean wilt virus (BBWV), cucumber mosaic virus (CMV), and tobacco rattle virus (TRV) by their host range, immunosorbent electron microscopy (ISEM), enzyme-linked immunosorbent assay (ELISA), direct tissue blotting immunoassay (DTBIA), and intracellural symptoms. By DTBIA and ISEM, TRV was detected in gladiolus showing notched leaf, while CMV was frequently detected in gladioli with dwarfing, color breaking and malformation of flowers. BBWV was also often detected in many symptomless gladiolus plants, but TRV was detected in notched-leaf of gladiolus. Electron microcopic examination of negatively stained preparations showed that BBWV and CMV are spherical particles of 28 nm and 30 nm in diameter, and TRV is rigid rod-shaped particles of 40∼200 nm in length. The rigid rodshaped virus particles reacted positively with TRV antiserum in ISEM and DTBIA, respectively.

The Viruses in Gladiolus hybridus cultivated in Korea 1. Bean Yellow Mosaic Virus and Clover Yellow Vein Virus (한국산 글라디올러스에 발생하는 바이러스)

  • Gladioli (Gladiolus hybridus) showing flower colour breaking, leaf mosaics, necrotic fleck, and dwarfing or lack of visible symptoms were collected from gladioli growing areas in Taegu and Kyungpook province, Korea. The two viruses isolated from the naturally infected gladioli were identified as ban yellow mosaic virus (BYMV) and clover yellow vein virus (CIYVV) by their host range, immunosorbent electron microscopy (ISEM), enzyme-linked immunosorbent assay (ELISA), direct tissue blotting immunoassay (DTBIA) and intracellural symptoms. In ultrathin sections of BYMV and CIYVV infected tissues, laminated aggregate-type inclusions, cytopalsmic bodies and nuclear inclusions as well as filamentous virus particles were observed in the cytoplasm of parenchyma cells. By DTBIA and ISEM, BYMV was detected in all tested gladiolus plants showing severe or mild mosaic symptoms, whereas CIYVV were mainly detected from those of mild mosaic symptoms. BYMV is the most prevalent in commercial gladioli and present major production problems. Detection sensitivity of BYMV and CIYVV in crude sap of infected gladiolus leaves by ISEM was about twice compared with ELISA. In a comparison of ELISA, ISEM, DTBIA, BYMV was detected in same degree by DTBIA in samples where sap extracts were positive in both ELISA and ISEM. DTBIA provides a specific, rapid, and simple tool for large-scale diagnosis of BYMV.
