Studies on the Root Nodule Formation of Several Leading Soybean Varieties (주요대두품종의 근류형 성성에 관한 연구)

  • 15 soybean cultivars were tested with the sandculture to investigate the affinity of cultivars to the nodule bacteria, Rhyzobium Japonicum #23, and the effects of inoculation on the vegetative growth of cultivars. The amount of nodule formation until the flowering stage was significantly different among the cultivars. The cultivar Haman showed the best nodule formation of 122.2 nodules per plant, while the cultivar Busuk showed the least amount of nodule formation. The inoculation of the nodule bacteria resulted in an increased plant growth and especially the chlorophyll content was significantly improved by inoculation of the nodule bacteria.

Role of Lectins in Host Plant-Rhizobium Interactions (근류균과 숙주식물의 상호작용에 관한 렉틴의 역할)

  • Experiments were carried out to elucidate the specific interactions between host plant, Phaseolus vulgaris, and symbiotic bacteria, Rhizobium Phaseoli. Purified P. vulgaris lectins and six species of cultured Rhizobium were subjected to agglutination test. Lectins from bean and R. phaseoli showed relatively high agglutination activity indicating that host plant lectins recognize carbohydrate moieties on the compatible Rhizobium cell surface. The specific carbohydrate receptors for binding of the lectins on the cell surface of R. phaseoli were found as mannose and galactose. The minimum concentration of sugars for the inhibition was 6.25mM. The lectin content of cultured plant roots was measured after germination and was maximum in 5-day seedlings. The nodulation was competitively inhibited by lectins for the plants cultured with Rhizobium cells. By immunochemical studies, there was some relationship in antigenic determinants between R. phaseoli and R. japonicum but no relationships were observed with other Rhizobium species. The results suggest that the infection by rhizobia to the roots of leguminous plants may be caused by the specific interaction of lectins with rhizobia.

  • To investigate the effects of seed treatments and liming on nodulation and growth of lucerne(Medicago sativa L.) in an upland yellow-brown earth in N.Z. the five levels of seed treatments(Uninoculated, Inoculated, Inoculated plus lime pelleted, Inoculated plus 50/50 phosphate/dolmite pelleted, Inoculated plus ta1c pelleted), and 2 levels of lime applications (0, 25kg/10a) were compared and their interactions were observed, and then the following results were obtained. 1. In oder to obtain the effective nodulation of lucerne on acid soil it is usually necessary to apply lime and inoculate the seed. 2. The coating of inoculated lucerne seed with adequate materials will promote the nodulation. 3. The pelleting of lucerne seed may be a of reducing the need for the heavy liming. 4. The finely grounded lime was the best to use origin upland yellow-brown earth. 5. When the seed were not inoculated or pelleted with inferior material, liming is very effective for the nodulation and growth of lucerne. 6. The pelleting seed will reduce the laborious work and expenditure.

  • Generally, excessive soil water stress in vegetative growth stage inhibits the growth of soybeans. Leaf area expansion of the plant during excessive soil water stress was only half and the respiration of roots was much diminished compared with the plant none water stress. When excessive soil water stress to the soybeans was continued for 7 days, outer epidermis and vascular system of tap root were severely cracked, more than thirty-five percent of nodule was died and the bacteroid layers of alive nodule were disintegrated.

  • Five soybean varieties of two early maturing Karikei73 and SS 79168, and three late maturing Tohoku76, Baegunkong and Jangbaegkong were used and evaluated in the study. Of the varieties examined, Karikei73 was characterized by the delayed leaf senescence. The varieties were planted in the pots of 1/3500 a filled with volcanic ash soil at the experimental fields of the National Institute of Agrobiological Resources in Japan. Major agronomic characteristics including the activity of nitrogen fixation for root nodules during the grain filling period were measured. Measurements during the stages were followed by the stage of development descriptions for soybeans made by Fehr and Carviness (1977). The acetylene reducing activity (ARA) per dry weight of root nodule measured using acetylene reduction assays was the highest at R4-R4.5 with decreasing trends thereafter for the early matruing varieties, while it continuously increased up to R6 but decreased rapidly thereafter for the late maturing varieties. The dry weights of root nodules and all parts of the host plant at each stage checked were greater in the late maturing varieties being the same in ARA per pot.

  • In order to obtain a basic information related to the utility of Sinorhizobium fredii for field soybean, the effectiveness and competitiveness of nine fast-growing S. fredii strains including TAL 1871 etc. were examined on eleven Korean soybean(Glycin max L.) cultivars. Nine S. fredii strains all modulated soybean cv. Kwangkyo, but did not Danyeobkong. The averaged shoot dry weight by S. fredii strain TAL 1781, TAL 1840, TAL 1899 marked only about 53% as compared to that by Bradyrhizobium japonicum strain YCK 213 and USDA 110. The shoot dry weight by S. fredii strains, of which TAL 1781 was the most effective in that, was increased by coinoculation with B. japonicum YCK 213 rather than with B. japonicum USDA 110. Nodule occupancy by S. fredii ranged from 8.3 to 26.7% in coinoculation with B. japonicum YCK 213, but did from 10.0 to 30.0% with B. japonicum USDA 110. These results indicated that S. fredii strains were inferior effective and competitive to B. japonicum strains.

Effects of Herbicide Application on Growth and the Nodulation in Soybean (제초제 처리가 콩의 생육 및 근류형성에 미치는 영향)

  • Present study was conducted to reveal the effects of the herbicides, Lasso and Devrinol, on the soybean growth and the nodulation in field condition. Emergence rate was reduced positively in proportion to increase in the concentration of herbicides regardless of the herbicidal difference and it was significantly reduced even in the recommended concentration as compared to untreated plot, showing marked abnormal symptom on seedlings. Plant height, fresh weight of the plant, number of internodes, branches, pods, seeds per plant and 100-seed weight were reduced with increase in the concentration of herbicides and were highly significant in difference between the untreated plot and double concentration plot, eventhough most were nonsignificant in difference from the recommended concentration. Nodulation was significantly decreased with increase in the concentration of herbicides. The reduction was remarkably different with soybean varieties and consistently appeared from three weeks to six weeks after sawing. Significant correlation was realized between the reduction of nodulation and the agronomic characters of soybean and it was considered that the reduction of nodulation by misapplication of the herbicides might be a causal factor for decrease in soybean yield.

  • To evaluate Rhizobium meliloti BALSAC, a strain selected from Canada as an acid tolerant one, and ordinary lucerne inoculant in acid condition, lucerne (Medicago sativa L. cv. Wairau) was inoculated and/or pelleted in the laboratory, and grown for two months in an acid soil (Lismore silt loam, pH 5.4) with three levels of lime in the, glasshouse. The results of controlled (noninoculated), nitrogen fertilized, laboratorial and commercial inoculated seeds were compared to give the following conclusions: 1. There was no significant difference in the top and root dry matter yields between two Rhizobium strains. However, Balsac inoculant showed higher single nodule dry matter weight and relatively higher number of larger nodules than the ordinary inoculant. 2. Lime application increased dry matter yields of plants and nodules, and the number of nodules per pot and the increase of nodules on the lateral roots in both inoculants. Lime application also caused an evenly distribution of nodules on the root by showing an increase of nodules mainly on the lateral roots. 3. Fertilizer nitrogen without inoculant slightly increased the nodulation percentage, the nodule dry matter weight per nodule and the relative proportion of larger nodules. 4. Commercially inoculated and pelleted seed showed less consistent results. 5. Relatively larger variations in measuring nodule characteristics was discussed and concluded that extreme cares should be given to reduce the variation.

  • This experiment was carried out to study the effects of nitrogen concentration of cultural solution, Rhizobium inoculation, and planting density on the growth and yield of soybean cultivars, Hwanggeumkong, Jangbaegkong, Paldalkong, Clark, and non-nodulation isoline of Clark. Rhizobium inoculation increased the stem length, particularly in Hwanggeumkong, Jangbaegkong, and decreased it significantly in non-nodulation Clark. Stem length was increased by the increase in nitrogen fertilization by the 195ppm level and decreased by the increase in plant population density. Rhizobium inoculation also increased the shoot dry weight, but significantly decreased it in non-nodulation Clark. As nitrogen concentration in the cultural solution increased the shoot dry weight decreased in Jangbaegkong and paldalkong. However, the shoot dry weight was decreased by the increase in plant population density. Rhizobium inoculation and the increase in nitrogen concentration of cultural solution increased the ratio of shoot dry weight to root weight. The Rhizobium inoculation and the increase in nitrogen concentration of cultural solution increased the grain yield per pot in Hwanggeumkong and paldolkong, While non-nodulating Clark showed significant decrease in grain yield. Grain yield per pot was also increased by the increase of plant population density. Grain yield was significantly correlated with shoot dry weight, nodule number, and nitrogen content of the soybean plants. The correlation between nitrogen contents of the soybean plants and stem length, shoot dry weight, and nodulation was significant. The allantoin-N content in stem was also significantly correlated with nodulation.

  • These experiment were conducted to investigate the seasonal changes of nodule formation and $N_2$-fixing activitues of soybean root nodules which grown in pot on five cultivars in different soils. The results obtained were as follows; 1. It was found that the development of nodules starts from 20 days after planting and continues to increase up to 50-60 days after planting. 2. $N_2$-fixing activities were different with cultivars of soybean. The trend was high in order H-25 > Suweon 63 > Hill > Yugwoo 3# > Gwanggyo 3. Rhizobium japonicum isolated from different soybean cultivars showed different acetylene reducing activities in vitro ; high in order A62-1 > Suweon 63 > Hill > Gwanggyo > Yugwoo 3#.
