Biological Control of Lettuce Sclerotinia Rot by Bacillus subtilis GG95 (길항미생물 Bacillus subtilis GG95를 이용한 상추 균핵병의 생물학적 방제)

  • Sclerotinia sclerotiorum, a plant pathogenic fungus, can cause serious yield and quality losses in the winter lettuce field. For biological control of S. sclerotiorum, soil-born microorganisms that inhibit the mycelia growth of S. sclerotiorum and Fusarium oxysporum were isolated from diseased soil. Among the isolates, bacterial isolate, GG95, which was identified as Bacillus subtilis according to the morphological, physiological characteristics and by 16S rRNA similarity, showed the highest level of inhibitory activity. The growth conditions for B. subtilis GG95 were optimized in TSB media (pH 7) by culturing at $28^{\circ}C$ for 24 hrs. Maltose or fructose and peptone were selected as the best carbon and nitrogen sources, respectively. Greenhouse experiment was performed to test effectiveness of B. subtilis GG95 in the control sclerotinia rot. Drench application ($1{\times}10^8cfu/mL$, 3 times) of the bacterial culture broth to lettuce showed an effectiveness value of 88%, suggesting that B. subtilis GG95 would be a promising biocontrol agent for control of sclerotinia rot.

Occurrence of White Rot on Alliaceous Vegetable Crops (백합과 채소작물에서의 흑색썩음균핵병 발생)

  • 1995년 4월, 국내 백합과 채소작물 4개 재배지역에서 흑백썩음균핵병 발생을 조사하였다. 이 병은 마늘에서 최고 85%까지 가장 심하게 발생하였으며, 파에서는 매우 적게 발생하였다. 쪽파와 양파에서는 흑색썩음균핵병이 국내에서는 처음으로 발견되었으며, 발생율은 각각 최고 10%와 30%에 이르렀다. 이 병의 원인균은 배양적 및 형태적 특성에 E라 Sclerotium cepivorum Berk로 동정되었다. 병원성검정 결과, 이 균은 상기한 4종의 백합과 채소작물에 병원성을 나타냈다.

Activities of the Hydrolytic Enzymes Produced by Plant Pathogenic Fungi, Sclerotium rolfsii, Sclerotinia Sclerotinia and Sclerotiorum, and Helminthosporium sigmoideum var. irregulare (수종의 식물병원균(흰비단병균$\cdot$균핵병균 및 좀검은 균핵병균)이 생산하는 가수분해효소의 활성)

  • Activities of various hydrolytic enzymes produced by three plant pathogenic fungi, Sclerotium rolfsii Sacc., Sclerotinia sclerotiorum (Lieb.) deBary and Helminthosporium sigmoideum var. irregulare Crallery et Tullius, were measured. Activties and amounts of the enzymes in mycelia, cultural filtrates, and sclerotia(except of sclerotia of H. sigmoideum var. irregulare) were estimated at various pH levels in order to find out optimal pH for their enzymatic activities. Enzymes such as cellulase (ex), invertase, xylanase, $\beta-amylase$, polymethylgalacturonase, polygalacturonase, phosphatase and protease were estimated. Culture solution for production of enzymes was prepared by adding of 10g, D-glucose, 1.3g $NH_4NO_3,\; 0.5g\; MgSO_4,\;7H_2O,\; and\; 1.0g\; KH_2PO_4$ into 1 liter of potato decoction plus 2ml of micro element solution consisting of 0.2mg. Fe, 0.2mg Zn, and 0.1mg Mn as the sulphates into 1 liter of distilled water. All tested mycelia and cultural filtrates were obtained from the cultures incubarted in previous solution for ten days at $25^{\circ}C$, and sclerotia were harvested from PDA plates of 3. days old, The crude enzyme solutions were prepared according to the method of Miyazaki etal. Ten days after incubation, activities of Cx produced by Scl. sclerotiorum were higher than those of the other fung and each of Cx from three fungi showed different pH optima, such as S. rolfsii and Scl. schlerotiorum in acid side (around pH 3.0), H. sigmoideum var. irregulare in neutral side (around pH 6.3). Invertase activities of S. rolfsii were 20 times higher than those of the other fungi in all samples. All tested fungi, however, showed no significant difference between the enzymatic activities of their cultural filtrate and mycelia and the activities in sclerotia of S. rolfsii and Scl. sclerotiorum were hardly recognized. There were multiple peaks on the xylanase activity curves of three fungi in terms of pH values. High activities of the xylanase were revealed in sclerotia of S. rolfsii and Scl. sclerotiorum, and in mycelia of H. sigmoideum var. irregulare. The highest activities of $\beta-amylase$ were shown both in mycelia and cultural filtrate of H. sigmoideum var. irregulae among the tested fungi, and their optimal pH was 6.2 in both mycelia and cultural filtrate. In the S. rofsii and Sel. sclerotiorum, however, the activities of cultural filtrates were higher than those of the other fungi, and optimal pH was 3.0 and 6.2 for cultural filtrate and both mycelia and sclerotia, respectively. Activities of PMG were high in cultural filtrates of all tested fungi, especially in Scl. sclerotiorum and H. sigmoideum var. irregulare. Mycelia of themalso showed the considerable activities. Optimal pH for enzymatic activities were variable with thekind of fungi or with the samples measured. The highest activities of PG were presented by mycelia of S. rolfsii and Scl. sclerotiorum. $9.l\mu /min.\; and\; 9.5\mu g/min.$, respectively. Optimal pH for activity of PG in mycelia was around 4.5 in S. rolfsii and around 3.0 in Scl. sclerotiorum. Phosphatase of S. rolfsii and Scl. sclerotiorum was more active in acid side (optimal PH3. 5) and that of H. sigmoideum var. irregulare showed one peak each in acid, neutral and alkaline side. But the highest peak was at pH 9.5. Protease of all tested fungi was more active at pH 10.0, especially that of the cultural filtrate of H. sigmoideum var. irregualre.

Chemical Management Strategies for Popcorn Disease and Mulberry Sucker on Fruit-Producing Mulberry (오디 생산용 뽕나무에 발생하는 균핵병과 뽕나무이의 약제 방제체계)

  • From our field investigation from 2017 to 2018, five diseases and four insect pests have been identified as the primary biotic stressors of fruit-producing mulberry, which include popcorn disease and mulberry sucker, respectively. In this study, we examined the relative control effects of selected agro-chemicals against the popcorn disease and mulberry sucker. Our systemic treatment methods to simultaneously control the popcorn disease and the mulberry sucker indicated that an integrated control method showed a superior result with the control efficacy of 89.3%, while a conventional control method resulted in 66.7%. As a result, we concluded that it is much efficient to control both disease and insect pest in mid-April while sequentially applying chemicals only for the popcorn disease from February. By considering the ecological aspects of the popcorn disease and mulberry sucker, this systemic treatment will be able to reduce the labor of growers required for the control.

Occurrence of Sclerotinia Rot on Perilla Caused by Sclerotinia sclerotiorum under Structured Cultivation Condition (Sclerotinia sclerotiorum에 의한 시설재배 들깨 균핵병 발생특성)

  • Sclerotinia rot caused by Sclerotinia sclerotiorum frequently causes serious and unpredicable yield losses of the leaves of perilla growing under structured cultivation as vegetable in Korea. Temperature for mycelial growth ranged from 5 to $30^{\circ}C$ with optimum temperature d $20^{\circ}C$. Sclerotia were formed fewer at low temperature, but their dry weight was heavier than that at high temperature. The apothecia were formed from the sclerotia that buried up to 3 cm soil depth at $15^{\circ}C$ in moisture condition. The incidence of perilla Sclerotinia rot caused by S. sclerotiorum was observed throughout the growing season at greenhouse. The occurrence of this disease was especially severe from January to February of low temperature period. The average incidence rates of this disease was up to 15%. The significant occurrence of this disease was showed mainly in the continuous cropping field for more than five years. The incidence of this disease increased according to the increase of continuous cropping year. The incidence rates of this disease reached up to 20% in the continuous cropping field for ten years. Also it was firstly investigated the natural infection caused by S. sclerotiorum on weed plants, Gnaphalium affine and Mazus pumilus in farmer's field. The casual fungus showed pathogenicity on 11 weed plants species tested, and more severe pathogenicity on G. affine, Latuca indica and Ixeris dentata included in the family Compositae. This result suggests that effective crop rotation and weed eradication can be the method for organic control of perilla sclerotinia rot, and sudden outbreaks of this disease on perilla growing under structure after paddy rice call be explained by the presence of weed hosts.

Biological control of Paraconiothyrium minitans CM2 on Lettuce Sclerotinia Rot Caused by Sclerotinia sclerotiorum (Paraconiothyrium minitans CM2의 상추 균핵병균(Sclerotinia sclerotiorum)에 대한 생물적 방제)

  • A mycoparasite, Paraconiothyrium minitans CM2 was selected for biological control of sclerotinia rot of lettuce caused by Sclerotinia sclerotiorum. The experiment was carried out in a lettuce greenhouse in Yangpyeong from March to April.. When lettuce sclerotinia rot showed in the early stage of occurrence, Conidial suspension of the mycoparasite was weekly treated once to three times onto soil surface around lettuce plants. Incidence of sclerotinia rot in the once-application plot of the mycoparasite ($1{\times}10^7$ spores/$m{\ell}$) and in the benomyl(WP)-treated plot was 11.0% and 2.7%, respectively, whereas that of control was 31.0%. Incidence of twice- and three-application plots of the isolate was 7.9% and 12.8%, respectively. For increasing the effect of the mycoparasite, the experiment for the timing of application of P. minitans CM2 was carried out in a lettuce greenhouse in Yangpyeong and Suwon. Control efficacy against lettuce sclerotinia rot in the soil-drenching plots of P. minitans CM2 ($5{\times}10^6$ spores/$m{\ell}$) in the planting was 75.3~84.7%, and control effect by treatment of the isolate at the pot drenching+the soil-drenching plots in the early stage of disease occurrence was 63.8~58.0%. As the results, P. minitans CM2 could be a prospective biofungicide for biological control of sclerotinia rot of lettuce.

Studies on the Identification of Pythium spp. and Sclerotial Fungi isolated from Rice plants in Korea (II) (수도(水稻)에 관여(關與)하는 Pythium spp와 균핵병균류(菌核病菌類)의 분류동정(分類同定)에 관(關)한 연구(硏究)(II))

  • Four species of Pythium and two species of Sclerotium previously not recorded in Korea during 1976 and Leptosphaeria salvinii which previosly reported but reidentified. Pythium aristosporm Vantery, Pythium sp. and Pythium myriotylum Drechsler were isolated from diseased rice seedlings and from green withered rice plants and Pythium irreglare Buisman was isolated from paddy soil. Three species of Pythium except P. irregulare grew well at $40^{\circ}C$ on Potato dextrose agar and were confirmed as highly pathogenic but P. irregulare showed low pathogenicity on Yushin variety of rice. Sclerotium hydrophilum and Sclerotium oryzae-sativae appeared to be weakly pathogenic, but Leptosphaeria salvinii was confirmed as a highly pathogenic. Ordinally the two species of Sclerotium grew and produced many sclerotia on dead sheath and stems of rice. There are still some problems to clarify and reconsider in regard to the pathogenicity of the sclerotial fungi because their populations were so very high in paddy fields, but their role might be wound parasite.

Biological Control of Sclerotinia sclerotiorum in Lettuce Using Antagonistic Bacteria (길항세균을 이용한 상추 균핵병의 생물학적 방제)

  • To isolate antagonistic bacteria against sclerotinia rot of lettuce, caused by Sclerotinia sclerotiorum, soil samples were collected from the diseased greenhouse field in Namyangju city, Gyeong-gi province from 2007 to 2008. A total of 196 bacterial isolates were isolated using serial dilution method. In dual culture assay in vitro, 26 isolates showed more than 80% of inhibition rates of mycelial growth of S. sclerotiorum. Based on 16S rDNA sequence analysis, the 26 isolates were identified as Bacillus megaterium, B. cereus, B. subtilis, Arthrobacter nicotianae, A. ramosus, Pseudomonas filiscindens, Stenotrophomonas maltophilia, Brevibacterium frigoritolerans and Sphingobacterium faecium. The 26 isolates inhibited the mycelial growth of S. sclerotiorum up to 80% and the sclerotial germination 0-100%. In the greenhouse pot test of ten isolates conducted in summer, 2 isolates B. megaterium (DK6) and B. cereus (C210) showed control efficacy on sclerotia viability of S. sclerotiorum, 20% and 35%, respectively. In the greenhouse pot test in winter, the disease incidence of the control group was 80%, whereas those of 9 isolates among 26 were approximately 20%. From the result, the 9 isolates are expected as potentially antagonistic bacteria for biological control of sclerotinia rot of lettuce caused by S. sclerotiorum.

Effect of Crop Rotation Cultivation on the Suppression of Garlic White Rot Caused by Sclerotium cepivorum (전작물 재배에 의한 마늘 흑색썩음균핵병 억제효과)

  • This study was conducted to evaluate the effect of crop rotation cultivation on the suppression of garlic white rot caused by Sclerotium cepivorum in the mini plot ($2^*1^*$ 0.5 m). Six crops, soybean, sesame, mung bean, squash, crotalaria and spring onion, were previously transplanted in the mini-plots infested with S. cepivorum before garlics were planted. After cultivation of the previous crops, garlic was sown in the mini-plot. Non-cultivation plots and non-infested plots with white rot pathogen were used as control. The effect of crop rotation cultivation on the suppression of garlic white rot was evaluated by investigating comparatively the disease incidence (the percentage of infected plants) and yields. As a results, infection rate of garlic white rot was recorded lower in the non-infested plot, crotalaria and soybean cultivation than in the plot of the other crop cultivation. Especially when squash was previously cultivated and garlics were planted in 2013, infection rate of garlic white was recorded the highest score. In 2014, the infection rate of garlic white were low in the garlic on soybean, crotalaria and spring onion treatment whereas it was high in squash treatment, as well. In 2013, garlic yield was the highest in no inoculation plot, followed by crotalaria, soybean, no crop cultivation, sesami, mungbean, squash cultivation plot. In 2014, the yield in the plot of crotalaria and soybean was much higher than that in no inoculation plot. Based on above-described results, it is considered that soybean-garlic and crotalaria-garlic cultivation system can be good crop rotation systems to control garlic white rot.

Biological Control of Paraconiothyrium minitans S134 on Garlic White Rot Caused by Sclerotium cepivorum (Paraconiothyrium minitans S134의 마늘흑색썩음균핵병에 대한 생물적 방제)

  • Sclerotium cepivorum is a causal agent of white rot disease on different plants including Allium species such as garlic. A mycoparasite, Paraconiothyrium minitans S134 was selected for biological control of sclerotinia rot of garlic caused by S. cepivorum. The experiment was carried out in a garlic field in Taean from October in 2011 to June in 2012. Spore suspension of the mycoparasite was treated twice onto soil surface around garlic plants in sowing in 2011 and late Feb. in 2012, and disease rating was made June in 2012. Incidence of white rot in the twice-application plot of the mycoparasite ($5{\times}10^6$ spores/mL) and in the fluquinconazole (WP)-treated plot was 6.8% and 0.4%, respectively, whereas that of control was 19.5%. As the results, P. minitans S134 could be a prospective biofungicide for biological control of white rot of garlic.
