Investigation of Indicator Kriging for Evaluating Proper Rock Mass Classification based on Electrical Resistivity and RMR Correlation Analysis (RMR과 전기비저항의 상관성 해석에 기초하여 지시크리깅을 적용한 최적 암반 분류 기법 고찰)

  • In this study geostatistical technique using indicator kriging was performed to evaluate the optimal rock mass classification by integrating the various geophysical information such as borehole data and geophysical data. To get the optimal kriging result, it is necessary to devise the suitable technique to integrate the hard (borehole) and soft (geophysical) data effectively. Also, the model parameters of the variogram must be determined as a priori procedure. Iterative non-linear inversion method was implemented to determine the model parameters of theoretical variogram. To verify the algorithm, behaviour of object function and precision of convergence were investigated, revealing that gradient of the range is extremely small. This algorithm for the field data was applied to a mountainous area planned for a large-scale tunneling construction. As for a soft data, resistivity information from AMT survey is incorporated with RMR information from borehole data, a sort of hard data. Finally, RMR profiles were constructed and attempted to be interpreted at the tunnel elevation and the upper 1D level.

Evaluation of Uncertainty Sources in Temperature Measurement Using Platinum Resistance Thermometer Caused by Temperature Gradient in Furnace and Sealed-type Freezing Point Cells (전기로 및 봉입형 응고점 셀 내의 온도구배가 미치는 표준백금저항온도계 온도측정의 불확도 요소 평가)

  • In the international temperature scale of 1990 (ITS-90), standard platinum resistance thermometer (SPRT) is a defining standard thermometer used in the temperature range from 13.8033 K to $961^{\circ}C$. Uncertainty of SPRT is about several mK and uncertainty of defining fixed points of the ITS-90 which is used for calibrating SPRT is about several tenth of mK. Above $0^{\circ}C$. the defining fixed points are gallium melting point and indium, tin, zinc, aluminium and silver freezing points which are all realized using an electric furnace or a liquid bath. To realize freezing point of tin ($231.928^{\circ}C$) and zinc ($419.527^{\cir}C$), two 3-zone furnaces which have 3 electric heaters were manufactured. Temperature gradient of the constructed furnaces were tested. Uncertainty caused by temperature gradient of furnace and immersion effect of SPRT in the sealed-type freezing point cells were evaluated 0.038 mK for tin freezing point and 0.036 mK for zinc freezing point.

Performance Analysis of a Deep Vertical Closed-Loop Heat Exchanger through Thermal Response Test and Thermal Resistance Analysis (열응답 실험 및 열저항 해석을 통한 장심도 수직밀폐형 지중열교환기의 성능 분석)

  • Due to the limited areal space for installation, borehole heat exchangers (BHEs) at depths deeper than 300 m are considered for geothermal heating and cooling in the urban area. The deep vertical closed-loop BHEs are unconventional due to the depth and the range of the typical installation depth is between 100 and 200 m in Korea. The BHE in the study consists of 50A (outer diameter 50 mm, SDR 11) PE U-tube pipe in a 150 mm diameter borehole with the depth of 300 m. In order to compensate the buoyancy caused by the low density of PE pipe ($0.94{\sim}0.96g/cm^3$) in the borehole filled with ground water, 10 weight band sets (4.6 kg/set) were attached to the bottom of U-tube. A thermal response test (TRT) and fundamental basic surveys on the thermophysical characteristics of the ground were conducted. Ground temperature measures around $15^{\circ}C$ from the surface to 100 m, and the geothermal gradient represents $1.9^{\circ}C/100m$ below 100 m. The TRT was conducted for 48 hours with 17.5 kW heat injection, 28.65 l/min at a circulation fluid flow rate indicates an average temperature difference $8.9^{\circ}C$ between inlet and outlet circulation fluid. The estimated thermophysical parameters are 3.0 W/mk of ground thermal conductivity and 0.104 mk/W of borehole thermal resistance. In the stepwise evaluation of TRT, the ground thermal conductivity was calculated at the standard deviation of 0.16 after the initial 13 hours. The sensitivity analysis on the borehole thermal resistance was also conducted with respect to the PE pipe diameter and the thermal conductivity of backfill material. The borehole thermal resistivity slightly decreased with the increase of the two parameters.

  • In general, the behavior of moisture transmission is estimated by vapor permeability or vapor transmission resistance, but its values obtained by experiments do not have great adaptability for practical situations because of changes in the experimental conditions. This fact is why only theoretical discussions have advanced. Thus, the fundamental study of the moisture transmission phenomenon has been treated lightly. Here, as the first step toward the basic research of moisture transmission, the amount of moisture transmission and the moisture distribution in specimens were investigated. The experiment was conducted in a steady state, and the moisture distribution was measured by slicing and weighing the specimens. From the examination of the vapor transmission resistance, the phenomenon of moisture transmission was dealt with devide the moisture transfer on the wood surface and moisture diffusion in wood. The following results were obtained. 1) The phenomenon of moisture transmission should be approached by its division into moisture transfer on the wood surface and moisture diffusion in the wood because the positive values of vapor transmission resistance exist in the extrapolation of thickness 0mm. 2) The distribution of moisture in wood can be illustrated by two straight lines intersecting at the point of nine percent moisture content : namely, diffusion coefficients have two constant values at moisture contents below and above nine percent. The shape of the distribution curve of moisture content is similar irrespective of the wood thickness. On the other hand, when the moisture contents on both sides was more than nine percent, the distribution of the moisture content could be illustrated by one straight lines. 3) The amount of moisture movement is determined by the moisture gradient in wood. 4) Coefficients of the moisture transfer depend on the thickness of the specimens.

  • Vortical systems are considered a main feature to sustain turbulence in a boundary layer through interaction. Such turbulent structures result in frictional drag and erosion or vibration in engineering applications. Research for controlling turbulent flow has been actively carried out, but in order to show the effect of vortices in a turbulent boundary layer, it is necessary to clarify the mechanism by which turbulent energy is transferred. For this purpose, it is convenient to demonstrate and capture phenomena in a laminar boundary layer. Therefore, in this study, the interactions of disturbed flow around a hemisphere on a flat plate in laminar flow were analyzed. In other words, a street of hairpin vortices was generated following a wake region formed after flow separation occurred over a hemisphere. Necklace vortices surrounding the hemisphere also appeared due to a strong adverse pressure gradient that brought high momentum fluid into the wake region thereby leading to an increase in the frequency of hairpin vortices. To mitigate the effect of these necklace vortices, local suction control was applied through a hole in front of the hemisphere. Flow visualization was recorded to qualitatively determine flow modifications, and hot-film measurements quantitatively supported conclusions on how much the power of the hairpin vortices was reduced by local wall suction.

Effects of Drill Widths and Nitrogen Application Levels in Early Spring on the Growth Characteristics and Seed Productivity of Italian Ryegrass (Lolium multifloum Lam.) (휴폭 및 이른 봄 질소시용이 이탈리안 라이그라스의 생육특성과 종자수량에 미치는 영향)

  • The effects of drill widths and nitrogen application levels on the growth characteristics and seed productivity of Italian ryegrasss(Lolium multiflorum Lam.) were evaluated. The experimental design was a split-plot arrangement with three replications. The main-plots were three drill widths of 15, 30 and 45cm, and subplots were three nitrogen application rates of 50, 75 and 100 kg/㏊ in early spring. Among the growth characteristics, lodging tolerance appeared to be highest in the treatment including 45cm of drill width and 50kg/㏊ of nitrogen application compared to other treatments. The numbers of stem per one square meter tended to be increased more in drill width of 15cm than in that of 30 or 45cm, but other growth characteristics were similar among treatments. In seed yield components, the numbers of spike per one square meter were getting increased according as drill width was narrower, but spike length and 1,000 seed weight were longer and heavier. respectively. The rate of ripened pains was highest in treatment including 30cm of drill width and 50kg/㏊ of nitrogen application. Maximum seed yield was also obtained in same treatment.

Effects of Nitrogen Application on the Patterns of Amino Acids, Nitrogen Contents and Growth Response of Four Legume Plants under Saline Conditions (염분 환경하에서 4종 콩과식물의 생장, 아미노산 및 질소함량에 미치는 질소원의 영향)

  • Four legume plants showed better growth by the external nitrogen supply rather than the symbiotic nitrogen fixation only under salt additions. In case of Glycine max and Phaseolus angularis, total nitrogen contents decreased by high salinity level but their amino acid levels significantly increased with the increase of salt treatments and indicated high soluble-/insoluble-N ratios. Cassia tora and Albizzia julibrissin contained less amino acids than G. max and P. angularis but total N (esp. insoluble N fraction) increased with higher salt levels. Asparagine occurred as a main amino acid especially in G. max and P. angularis and can be seen as potential N-storage form in these plants. It might be play an important role for the osmoregulation mechanism under the saline condition. Meanwhile, to investigate what kinds of nitrogen sources are effective for overcoming salt stress on soybean plants, various N forms and concentrations (NH₄NO₃-N, NO₃-N, NH₄NO₃-N; 2.5 and 5 mM) were additionally supplied to the salt gradient medium. Soybean plants treated with NH₄NO₃-N showed the best growth up to 40 mM NaCl and NO₃- fed plants indicated good growth even at 80 mM NaCl treatments. Contrary to NH₄NO₃- and NO₃- fed plants, NH₄/sup +/- fed plants showed remarkable growth reduction and died by 40 and 80 mM NaCl treatments after the first harvest (15th day). Consequently, these results suggest that salt excluding and resistant capacities of soybean plants under NaCl treatments are increased in order of NH₄ - N, control, NO₃- N and NH₄NO₃- N depending on N concentration except NH₄- N treatments.

Characterizing Responses of Biological Trait and Functional Diversity of Benthic Macroinvertebrates to Environmental Variables to Develop Aquatic Ecosystem Health Assessment Index (환경변이에 대한 저서성 대형무척추동물의 생물학적 형질과 기능적 다양성 분석: 수생태계 건강성 평가 관점에서)

  • The biological indices based on the community structure with species richness and/or abundance are commonly used to assess aquatic ecosystem health. Meanwhile, recently functional traits-based approach is considered in ecosystem health assessment to reflect ecosystem functioning. In this study, we developed a database of biological traits for 136 taxa consisting of major stream insects (Ephemeroptera, Plecoptera, Trichoptera, Coleoptera, and Odonata) collected at Korean streams on the nationwide scale. In addition, we obtained environmental variables in five categories (geography, climate, land use, hydrology and physicochemistry) measured at each sampling site. We evaluated the relationships between community indices based on taxonomic diversity and functional diversity estimated from biological traits. We classified sampling sites based on similarities of their environmental variables and evaluated relations between clusters of sampling sites and diversity indices and biological traits. Our results showed that functional diversity was highly correlated with Shannon diversity index and species richness. The six clusters of sampling sites defined by a hierarchical cluster analysis reflected differences of their environmental variables. Samples in cluster 1 were mostly from high altitude areas, whereas samples in cluster 6 were from lowland areas. Non-metric multidimensional scaling (NMDS) displayed similar patterns with cluster analysis and presented variation of taxonomic diversity and functional diversity. Based on NMDS and community-weighted mean trait value matrix, species in clusters 1-3 displayed the resistance strategy in the life history strategy to the environmental variables whereas species in clusters 4-6 presented the resilience strategy. These results suggest that functional diversity can complement the biological monitoring assessment based on taxonomic diversity and can be used as biological monitoring assessment tool reflecting changes of ecosystem functioning responding to environmental changes.

Combined Effect of Ganciclovir and Vidarabine on the Replication, DNA Synthesis, and Gene Expression of Acyclovir-resistant Herpes Simplex Virus (Acyclovir저항성 Herpes Simplex Virus의 복제, DNA합성 및 형질 발현에 미치는 Ganciclovir 및 Vidarabine의 병용효과에 관한 연구)

  • Combined effects of ganciclovir (GCV) and vidarabine (ara-A) on the replication, DNA synthesis, and gene expression of wild type-1 herpes simplex virus (HSV-1) and three acyclovir (ACV)-resistant HSV-1 mutants were studied. These mutants include a virus expressing no thymidine kinase $(ACV^r)$, a virus expressing thymidine kinase with altered substrate specificity $(IUdR^r)$, and a mutant expressing altered DNA polymerase $(PAA^r5)$. GCV, an agent activated by herpesvirus specific thymidine kinase, showed potent antiviral activity against the wild type HSV-1(KOS) and DNA polymerase mutant $(PAA^r5)$. The ACV-resistant mutants with thymidine kinase gene $(ACV^r\;and\;IUdR^r)$ were resistant to GCV. All tested wild type HSV-1 or ACV-resistant HSV-1 mutants did not display resistance to vidarabine (are-A). Combined GCV and ara-A showed potentiating synergistic antiviral activity against wild type KOS and $PAA^r5$, and showed subadditive combnined ativiral activity against thymidine kinase mutants. Combined GCV and ara-A more significantly inhibited the viral DNA synthesis in wild type KOS and $PAA^r5-infected$ cells to a greater extent than either agent alone, but the synergism was not determined in $ACV^r$ or $IUdR^r-infected$ cells. These data clearly indicate that combined GCV and ara-A therapy might be useful for the treatment of infections caused by wild type HSV-1 or ACV-resistant HSV-1 with DNA polymerase mutation. ACV-resistant viruses with the mutation in thymidine kinase gene are also, resistant to GCV, but susecptible to ara-A, indicating that ara-A would the drug of choice for the treatment of ACV-resistant HSV-1 which does not express thymidine kinase or expresses thymidine kinase with altered substrate specificity. While the synthesis of viral ${\alpha}-proteins$ of wild type HSV-1 was not affected by ACV, GCV, ara-A, or combined GCV and ara-A, the synthesis of ${\beta}-proteins$ was slightly but significantly increased at the later stage of viral infection by the antiviral agents. The synthesis of ${\gamma}-proteins$ of wild type HSV- 1 was significantly inhibited by ACV, GCV, ara-A, and combined GCV and ara-A. Combined GCV $(5-{\mu}M)$ and ara-A $(100-{\mu}M)$ also significantly altered the expression of viral ${\beta}-and$ ${\gamma}-proteins$, of which efffct was similar to that of GCV $(10-{\mu}M)$ alone. Although ACV at the concentration of $10-{\mu}M$ did not alter the expression of ${\alpha}-$, ${\beta}-$, and ${\gamma}-proteins$ of ACV-resistant $PAA^r5$, GCV and ara-A significantly alter the epression of ${\beta}-and$ ${\gamma}-proteins$, not ${\alpha}-protein$, as same manner as they altered the expression of those proteins in cells inffcted with wild type HSV-1. Combined GCV $(5-{\mu}M)$ and ara-A $(100-{\mu}M)$ altered the expression ${\beta}-and$ ${\gamma}-proteins$ in $PAA^r5$ infected cells, and the effect of combined regimen was comparable of that of GCV $(10-{\mu}M)$. These data indicate that the alteration in the expression of ${\beta}-and$ ${\gamma}-proteins$ in wild type HSV-1 or $PAA^r5$ infected cells could be more significantly affected by combined GCV and are-A than individual GCV or ara-A. In view of the fact that (a) viral ${\alpha}-$, ${\beta}-$, and ${\gamma}-proteins$ are synthesized in a cascade manner; (b) ${\beta}-proteins$ are essential for the synthesis of viral DNA; (c) the synthesis of ${\beta}-proteins$ are inhibited by ${\gamma}-proteins$; and (d) most ${\gamma}-proteins$ are made from the newly synthesized progeny virus, it is suggested that GCV and ara-A, alone or in combination, primarily inhibit the synthesis of viral DNA, and by doing so might exhibit their antiherpetic activity. The alteration in viral protein synthesis in the presence of tested antiviral agents could result from the alteration in viral DNA synthesis. From the present study, it can be concluded that (a) combined GCV and ara-A therapy would be beneficial for the control of inffctions caused by wild type HSV-1 or ACV-resistant DNA polymerase mutants; (b) the combined synergistic activity of GCV and ara-A is due to further decrease in the viral DNA by the combined regimen; (c) ara-A is the drug of choice for the infection caused by ACV-resistant HSV-1 with thymidine kinase mutation; and (d) the alteration in viral protein synthesis by GCV and ars-A, alone or in combination, is mostly due to the decreased synthesis of viral DAN.
