광합성 세균의 Carotenoid 생합성

수소생성 광합성 세균 Rhodopseudomonas strain K-7의 색소 생성능에 대한 연구

  • 광합성 세균의 수소생성 기작에 대한 연구의 일환으로 광합성 세균에서 생성되는 색소의 성분을 조사하였다. 분리한 광합성 세균 K-7은 수소생성능이 뛰어난 균주로 조사된 홍색 비유황세균으로서 생리, 형태 및 배양학적 조사에 의하여 Rhodopseudomonas spheroides로 분류하였다. Type culture인 Rhodopseudomonas spheroides NCIB 8253과 비교 연구하였으며, SEM(Scanning Electron Microscope)하에서 기존균주와 분리균주의 형태학적인 특징을 확인하였다. 군 동정의 주요열쇠가 되는 색소성분을 조사한 결과, 균주 K-7에서 추출된 carotenoids로는 spheroidene의 산화형태인 OH-spheroidenone이 주성분이었고, Neurosporene, Lycopene, Anhydro- rhodovibrin, Rhodovibrin등이 동정되었다.

Comparison of Pigment Content and Photosynthetic Capacity of Quercus mongolica Stands at Namsan, Seoul and Paekcheoksan, Kangwondo (서울 남산과 강원도 백적산의 신갈나무 숲에서 엽록소 함량 및 광합성능의 비교)

  • To elucidate effects of air pollutants on ecophysiological properties of two Quercus mongolica stands at different levels of air pollution, photosynthetic capacity and chloroplast pigment were measured in leaves of Q. mongolica. Photosynthetic capacity, photosynthetic pigment content and SLA (specific leaf area) of Q. mongolica leaves in polluted area were lower than those in nonpolluted area. Also, the chlorosis and mottling in some of leaves were observed in polluted area after August. Except for total carotenoid (P<0.05) the correlations between photosynthetic capacity and pigment content were not significant. Photosynthetic capacity of Q. mongolica leaves in polluted area was decreased after July.

  • The light harvesting pigment proteins of dinoflagellates exhibit essentially 100% efficient energy transfer from carotenoid (peridinin) to chlorophyll a within the antenna pigment complexes. The high efficiency of solar energy harvesing (particularly blue light) for photosynthesis in dinoflagellates is attributable to the unique molecular topography of peridinin and chlorophyll e within the protein crevice. The mechanisms of energy transfer from carotenoids to chlorophyll in higher plants have also been discussed in comparison with the dinoflagellate antenna pigment complexes. As an example of solar energy harvesting, particularly red light, for photosynthesis in algae, the molecular topography and energy transfer in the photosynthetic accessory pigment protein, Chroomonas phycocyanin, have also been discussed.

Studies on the leaf Color and Absorbance in Visible Range of Rice Plant (벼 엽색과 가시부의 흡광도와의 관계)

  • The rice leaf color, which was one of main factors to determine application time of fertilizers, was analyzed in visible region, while the distinction of photosynthetic pigments controlling the rice leaf color was separated by T.L.C. The light absorption peaks of ethyl ether extracts of rice leaf blades were occurred in 413, 432, 453 and 662nm. Especially, the maximal light absorption was 432nm in blue and 662nm in red region. The both light absorption ratios (blue to red region) was not greatly different among rice ecotypes, whereas decreasing tendency in increased fertilization was observed. There was a significant negative correlation between the both light absorption ratios and the values of color chart, and the content of chlorophyll in rice leaf blades. The composition ratio of chorophyll was larger than that of carotenoid at heading stage, and the carotenoid was larger than the chlorophyll at 30 days after heading. The composition ratio of chlorophyll among photosynthetic pigments according to rice ecotypes, Japonica x Indica cultivars were larger than Japonica cultivars at heading stage, and Japonica cultivars were larger than Japonica x Indica cultivars at 30 days after heading, while carotene was vice versa.

  • 식물체에서 일어나는 유전자 발현의 광조절 현상은 매우 복잡하다. 이는 각 유전자들이 조직에 따라 질적, 양적, 시간적 측면에서 빛에 대한 반응에 커다란 차이점을 보이고 있기 때문이다. 따라서 식물체를 대상으로 광조절 기작을 연구하는 데는 현상의 복합성과 실험과정의 기술적, 시간적, 경제적 제약이 많기 때문에 이에 관한 연구가 아직 초보적 수준에 머물고 있다. 광합성 세균 중에서 purple bacteria나 green bacteria와는 달리 식물성 광합성을 하며, 식물체보다 세포구조가 훨씬 간단한 cyanobacteria는 유전자 발현의 광조절 기작을 연구하는데 간단, 명료한 'model'로서 기대되는 바 크다. 따라서 이 글에서는 cyanobacteria의 광계와 광합성 색소인 chlorophyll의 생합성 과정을 중심으로 광조절 현상에 대한 최근 연구 동향과 전망을 살펴보고자 한다.

A Survey of Plastid Crystals and Microtubules in Flowering Plants (꽃피는식물 색소체 내 결정구조와 미세소관의 발달양상 조사 연구)

  • The plastid inclusion has long been known to exist in leaves of numerous plant species, especially in those of flowering plants. Among the inclusions, crystalline bodies are the most frequently distinguished structures of the foliar plastids, however, microtubules and phytoferritins are also reported occasionally. The crystalline inclusions vary in shape, and are located either in the stroma or within intrathylakoidal spaces, whereas microtubules and phytoferritins are more uniform in shape and are formed in the stroma. In crystalline structures, the composing elements exhibit a lattice pattern and/or paralleled tubules that are either bounded by membranes or exist without membrane enclosing. Other types of inclusions have not been shown to be enclosed by any membranous structures. According to the current survey, the plastid inclusion, with the exception of phytoferritins, has been shown to exhibit a crystalline or tubular pattern, and has been reported in more than 56 species of various families. Their occurrence is not restricted to any photosynthetic pathway, but is found to be randomly distributed among C-3, C-4 and CAM species, without phylogenetic relationships. The progress in plastid inclusion research reveals more information about the function and complexity, but the need for characterizing the 3-D structure of the crystalline inclusions also has been acknowledged in previous studies. A 3-D characterization would utilize tilting and tomography of serial sections with appropriate image processing that would provide valuable information on the sub-structures of the crystalline inclusions. In fact, recent studies performed on 3-D reconstruction of the plastid inclusions revealed important information about their comprising elements. In this article, the crystals and microtubules that have been reported in various types of plastids have been reviewed, with special consideration given to their possible sub-cellular function within the plastids.

Study on the Biosynthetic Characteristics of Photosynthetic Pigments in Dodder(Cuscuta australis R. Br.) Plant (실새삼의 광합성색소 생합성특성에 관한 연구)

  • This study was conducted to investigate whether 1) photosynthetic pigments(chlorophylls and carotenoids) are formed in dodder plant(Cuscuta australis), 2) there are any characteristics in the pigment biosynthesis, compared to that of other normal plants, and 3) dodder responds to some herbicides having target site on chloroplast. 1. Chlorophyll content of dodder tendrill grown under a natural daylight was 9 times and 50 times lower than that of field bindweed stem and leaf, respectively. 2. The photosynthetic pigment contents varied in different tissues, being higher in a apical region than in a lower region of seedling or tendrill. Chlorophyll wasn't almost observed below the 4th internode from the upper. 3. Pigment contents were greatly dependent on light intensity so that there were 4 to 6 times difference among light conditions. When the shoot containning low pigment contents under natural light, was incubated in growth chamber with various light intensities, the pigment contents were increased by 3 times of initial contents at about 97${\mu}E$ $m^{-2}s^{-1}$PAR. While the change in pigment contents was not observed at above 450${\mu}E$ $m^{-2}s^{-1}$PAR 4. Exogenous supply of 5mM 5-aminolevulinic acid increased protochlorophyllide by 7 times and 1.4 times in the etiolated shoot from field bindweed rhizome and in dodder stem, respectively, showing that dodder relatively has a low response to 5-aminolevulinic acid. 5. Pigment loss was observed in the treatment of paraquat, norflurazon, oxyfluorfen and diuron, and protoporphyrin IX was accumulated by oxyfluorfen as in normal plants Based on above results, several chracteristics of pigment biosynthesis in dodder seem to be summerized as follows. Photosynthetic pigment biosynthesis in Cuscuta australis runs even in low level. The pigment contents is differentially distributed in different regions and their contents seem significantly to be controlled by light intensities. Especially, chloroplast rapidly tends to degenerate with the development of tissue. Some herbicides having target site on chloroplast induce damage to dodder stem but are unlikely to control it well in field, except paraquat, due to low chloroplast activity and parasitic mode of nutrition.

Isolation of Photosynthetic Bacterium, Rhodopseudomonas palustris JK-1 and Researches on IAA and Carotenoid Production (광합성세균 Rhodopseudomonas palustis 분리 및 IAA와 Carotenoid 생성에 관한 연구)

  • The JK-1 isolate which was the best producer of indole-3-acetic acid and carotenoid among the 388 strains isolated from 28 wetlands in Jeju, was identified to be Rhodopseudomonas palustirs belongs to a typical group of non sulfur purple bacteria based on 16S sRNA sequencing. This study investigated the effect of different cultural conditions of pH, temperature, agitation, light and aeration on growth, IAA and carotenoid production of photosynthetic bacterium JK-1 for optimization of IAA and carotenoid production. It was found that growth, IAA, carotenoid, and bacteriochlorophyll production with light (3,000~3,500 Lux) and agitation (100 rpm) showed better results than those with dark/static or dark/agitation (100 rpm) in anaerobic conditions. The optimal pH, temperature and agitation speed for cell growth were 7, $30^{\circ}C$, 150 rpm, for IAA production were 9, $30^{\circ}C$, 150rpm and for carotenoid production were 6, $25^{\circ}C$, 50 rpm, cultured for 72 h under anaerobic light, respectively. The growth and IAA production were high in aerobic culture compared with anaerocic culture, whereas carotenoid and bacteriochlorophyll content were decreased extremely in aerobic condition (0.5~1 vvm). Subsequently, the optimal culture conditions for JK-1 were selected with pH 7, $30^{\circ}C$ and 100 rpm under anaerobic light and the effect on plant growth was tested by pot assay. Inoculation of JK-1 with 3% (v/v) level caused increase in shoot and root dry weigh that varied from 20%~58% to 40%~28% in young radish in camparison to uninoculated treatment at 50 days of growth. The study suggests that the JK-1 isolate may serve as efficient biofertilizer inoculants to promote plant growth.
