Effects of Mashing Proportion of Soybean to Salt Brine on Kanjang(soy sauce) Quality (메주의 담금비율이 간장의 품질에 미치는 영향)

  • This work was carried out to investigate the effects of mashing proportion of meju(as wet weight basis of raw soybean) to 20% salt brine on the chemical compositions and sensory characteristics of kanjang. By increasing the salt brine proportion in the kanjang mash, total nitrogen, TCA soluble nitrogen and total free amino acid content in kanjang were decreased accordingly, but although pH values and organic acids content were not different noticeably, the major organic acids in kanjang, lactic acid, pyroglutamic acid and acetic acid were increased up to the mashing proportion of 1:2 from 1:1 and pH values decreased up to that of 1:2.5. Residual sugar content in 1:1 kanjang was exceptionally high. The major free amino acids in kanjang were glutamic acid, lysine, alanine and leucine. The acceptability scores of kanjang tested by sensory evaluation were decreased in inverse proportion to the salt brine proportion of kanjang mash, but those of kanjang samples with $1:1{\sim}1:2.5$ mashing proportions were not significantly different but that of 1:3 different from 1:1 statistically at 5% level. The major chemical compositions, significantly effective to the acceptability of kanjang, were found to be glutamic acid and free amino acid by statistical analysis.

Effects of Soaking Conditions on the Manufacture of Calcium Enriched Rice (침지조건이 칼슘 강화미 제조에 미치는 영향)

  • In order to investigate the transfer rate of calcium into rice and the effect of soaking conditions-temperature (20, 30, 40, 50 and $60^{\circ}C$, $X_1$), calcium concentration of soaking solution (0, 250, 500, 750 and 1000 mg/L, $X_2$) and soaking time (5, 10, 15, 20 and 25 min, $X_3$), moisture absorption rate, hardness and calcium content of rice after soaking were analyzed. When the soaking temperature and time were increased the moisture absorption rate also increased (p<0.01). Quadratic and interaction effects on the moisture absorption rate among variables were highly significant, within 5%. The predictive model for moisture absorption rate $(Y_1)$ was as follows: $Y=16.14+0.78X_1+1.24X_3-0.23X_1{^2}+0.14X_2{^2}-0.42X_3X_1-0.38X_3{^2}\;(R^2=0.9123)$. Hardness was decreased as soaking time and temperature increased. Calcium contents of soaked rice increased as temperature, the calcium concentration of soaking solution and soaking time were increased (p<0.01). Also the interactive effect between soaking time and calcium concentration of soaking solution was high. The predictive model for calcium content $(Y_3)$ was as follows: $Y_3=470.80+110.87X_1+123.47X_2+52.29X_3-7.42X_1{^2}-14.87X_2{^2}-9.49X_3X_2-20.57X_3{^2}\;(R^2=0.9840)$. From the results in this study, if adults and children take 400 g of calcium enriched rice with 600 mg/kg and $700{\sim}900\;mg/kg$, respectively, they can expect to take more calcium than the recommended intake without other calcium reinforcing agents.

Effect of Whole Powder and Extracts of Gastrodiae Rhizoma on Serum Lipids and Body Fat in Rats Fed High-Fat Diet (천마 분말, 에탄올 및 열수추출물이 고지방식이를 급여한 흰쥐의 혈청지질과 체지방에 미치는 영향)

  • Effects of Gastrodiae Rhizoma on serum lipids and body fat in rats fed high-fat diet supplemented with 10% (w/w) lard during 11-week experimental period were investigated. Ninety-six male rats were randomly divided into eight groups: A, negative control (lard 10%); B, positive control (lard 10%+rat chow+5 brix water extract); C, lard 10%+1% G. Rhizoma powder; D, lard 10%+5% G. Rhizoma powder; E, lard 10%+2 brix 50% ethanol extract; F, lard 10%+10 brix 50% ethanol extract; G, lard 10%+2 brix water extract; H, lard 10%+10 brix water extract. Body weight gain, although slightly higher in groups A, B, and G, did not differ significantly among dietary groups. Weights of liver, spleen, kidney, and testis were significantly higher among dietary groups. Serum total cholesterol and triglyceride concentrations were markedly higher in both control groups than group H (p<0.05). Serum HDL concentration was significantly higher in group E, and lower in group A (p<0.05). Serum LDL+VLDL concentration was significantly lower in groups E, G, and H (p<0.05). Serum testosterone level was slightly higher in groups E, D, and H. Blood glucose level was significantly higher in groups B and G, whereas lower in group H (p<0.05). Epididymal fat pad (EFP) was markedly higher in control groups than G. Rhizoma diet-fed groups (p<0.05). The results indicated that decrease in serum lipids (TC, TG, LDL+VLDL) and body fat, and increase in serum HDL and testosterone levels were higher in groups E and H extracts than groups fed G. Rhizoma powder.

An Improved Method for Determination of Catechin and Its Derivatives in Extract and Oil of Grape Seeds (포도씨유 및 추출물의 카테킨류 측정방법 개선)

  • An improved method in place of a conventional vanillin spectroscopic method (CVSM) was developed for determination of catechin and its derivatives in extract and oil of grape seeds. For the CVSM, grape seed extracts had relatively high catechin content in the range of $17{\sim}43%$ (g/100g of extract), while grape seed oils had relatively smaller catechin content in the range of $30{\sim}40mg%$ (mg/100g of oil). For the improved vanillin spectroscopic method (IVSM) using a polyamide cartridge, catechin content of grape seed extracts was in the range of $4.0{\sim}7.5%$, while that of grape seed oils was below 5 ppm. Meanwhile, the quantities of catechin and its derivatives were determined by HPLC in the grape seed extracts and oils. Four major catechins [(+)-catechin, procyanidin B2, (-)-epicatechin, and epicatechin gallate] were detected from grape seed extracts, and the ranges of concentrations were as follows: (+)-catechin, $1.35{\sim}2.60%$; procyanidin $B_2$, $0.77{\sim}1.36%$; (-)-epicatechin, $2.35{\sim}4.59%$; (-)-epicatechin gallate, $0.06{\sim}0.30%$. In contrast, four catechins were barely detectable in the grape seed oils. The reproducibility of four major catechins in grape seed extracts, given as coefficient of variation, was below 5%, and the recovery close to above 95%. The achieved detection level of four catechins was $1{\sim}5\;ppm$. Additionally, the contents of catechin compositions in grape seed extract were also determined by HPLC in relation to different cultivars and producing areas. Thus, HPLC method and IVSM using polyamide cartridge can be used as alternative to CVSM for determination of catechin and its derivatives in extract and oil of grape seeds.

Effects of Additives on the Improvement of Frozen Dough Quality (첨가물이 냉동반죽의 품질향상에 미치는 영향)

  • This study was carried out to reduce the loss of frozen dough quality during frozen storage. Using response surface method, ascorbic acid 160.4 ppm, L-cysteine 63.1 ppm, and SSL 0.6% were found to be optimum, with xanthan gum 0.3% (formula A) and Ultra tex-3 5% (formula B) added as cryoprotectants. During frozen storage at $-20^{\circ}C$, control rapidly deteriorated after 4 weeks, while formulas A and B showed slight deterioration with immutable quality after 10 weeks.

Quality Changes and Pasteurization Effects of Citrus Fruit Juice by High Voltage Pulsed Electric Fields (PEF) treatment (고전압 펄스 전기장 처리에 의한 감귤주스의 품질변화)

  • A non-thermal pasteurization technology, high Pulsed Electric Field (PEF) has been thought to be a new alternative processing technology instead of heating. The objective of this study was to examine and compare the effect of PEF and High Temperature Short Time (HTST) treatments on the physicochemical, microbiological and sensory characteristics of citrus juices. Total sugar and titratable acidity values of fresh citrus juice and two treatments were not significantly different each other at p<0.05. The concentration of vitamin C in fresh citrus juice $(31.2{\pm}0.59\;mg%)$ was not significantly different with the value of PEF treatment $(29.4{\pm}0.75\;mg%)$ but was significantly higher than the value of HTST treatment $(27.4{\pm}0.75\;mg%)$. The color values (L, a, and b) in PEF treatment were significantly lower than the fresh citrus juice, but were higher than the values of HTST treatment. Both total bacterial cell counts $(6.65\;{\pm}\;0.08\;log_{10}(cfu/mL))$ and yeast counts $(7.79{\pm}0.07\;log_{10}(cfu/mL))$ in fresh citrus juice were significantly reduced by PEF $(1.39{\pm}0.14,\;2.42{\pm}0.1\;log_{10}(cfu/mL))$ as well as HTST treatment (0, 0). PE activity of fresh citrus juice $(1.3{\pm}0.12\;units/mL)$ was significantly reduced by PEF treatment $(0.11{\pm}0.01\;units/mL)$ and was totally inactivated by HTST treatment. Sensory evaluation scores in flavor, taste and overall acceptability between the fresh and PEF treated citrus juices $(7.2{\sim}7.5)$ were not significantly different but the values of HTST treatment $(5.1{\sim}5.8)$ were lower than others. Consequently, PEF treatment is thought to be a good alternative pasteurization method for fresh citrus juice to HTST treatment due to its strong pasteurization effect, reduced destruction of nutrients and good sensory characteristics.

  • This experiment was made to select the optimum thickness of the polyethylene (P.E) film for Cheongdo Bansi and Sagoksi in the P.E film storage kept at $0^{\circ}C$. The experimental plots were divided into 4 plots by film thickness (0.04, 0.06, 0.08 and 0.10mm) and those were subdivided into 3 plots by fruits number (3, 10 and 50 persimmons) in each film bags. We investigated five experimental items; the change of loss of weight, firmness, titratable acidity, sugar contents and soluble tannin contents. 1. In the changes of loss of weight, the plot of packing in 0.04mm P.E. film bag with 50 persimmons were more retarded than other plots in Cheongdo Bansi, and packing in 0.08mm with 10 persimmons, 0.04 mm with 50 persimmons were more retarded than other plots in Sagoksi. 2. In the change of softening, the plot of packing in 0.04 mm with 50 persimmons were more retarded than other plots in Cheongdo Bansi and Sagkai. 3. In the changes of titratable acidity, the plot of packing in 0.04 mm with 50 persimmons were more slightly decreased than other plots in Cheongdo Bansu also in Sagoksi, packing in 0.06 mm with 10 persimmons were the same results. 4. In the changes of soluble tannin contents, the plots of packing in 0.06 mm with 10 persimmons, 0.04 mm with 50 perimmons were more ratarded in Chenongdo Bansi, also in Sagoksi, packing in 0.04 mm with 10 persimmons 50 persimmons were the same results. 5. In the changes of soluble tannin contents, the plots of packing in 0.04mm with 3 and 10 persimmons were more slowly decreased than other plots in Cheongdo Bansi and Sagoksi, on. the other hand, pcaking in 0.04mm with 50 persimmins in Cheongdo Bansi and Sagoksi, had not astringent taste at 120 days in storage. Judging through the upper results, the most desirable storage conditions for Cheongdo Bansi and Sagoksi were to pack in P.E film bag of 0.04mm with 50 persimmons.

Drying of Rough Rice by Solar Collectors (태양(太陽) 열(熱 )집열기(集熱機)를 이용(利用)한 벼의 건조(乾燥)에 관(關)한 연구(硏究))

  • The flat-plate and tubular soar collectors were designed and constructed for drying the rough rice, and the performance of the collectors and drying effect were investigated when rough rice was packed in grain bin connected to collectors. Average-monthly radiation on a horizontal surface based on bright sunshine in Daejeon area during 1978 was the highest as $16,814\;KJ/m^2{\cdot}day$ in May and the lowest as $4,254\;KJ/m^2{\cdot}day$ in December, and significane was not recognized between the calculated and recorded values. The thermal effciency of collectors were increased as radiation increased during drying period and the average thermal effciency of flat-plate and tubular collectors in 11 to 12 o'clock a.m were 28.12 and 16.75%, respectively. The average inlet temperature of grain bin at 12 o'clock was shown as 20.02 at control 40.5 at grain bin connected to tubular collector and $55.1^{\circ}C$ at grain bin connected to flat-plate collector. In 25 cm rough rice depth in grain bin, tim taken for drying from initial moisture content at 27.4 to decrease upto 17.0% (14.5 % on wet basis) were 32 in control, 18 in grain bin connected to tubular collector and 11 hrs to flat-plate collector, and grain depth influenced drying rate remarkably. In the view point of drying characteristics, drying pattern showed initially falling-rate to constant-rate period finally.

Chungkook-jang Koji Fermentation with Rice Straw (볏짚을 이용(利用)한 청국장 제조(製造)에 관(關)한 연구(硏究))

  • Chungkook-jang Koji was fermented with rice straw at $40^{\circ}C\;and\;50^{\circ}C$ for 72 hours. The changes of proximate composition, pH, titrable acidity, nitrogen compounds, protease activity and free-amino acids during the fermentation were investigated. Moisture, lipid and protein contents remained essentially unchanged during the fermentation. The pH was gradually increased from 6.4 to 7.46 and 7.82 at $40^{\circ}C\;and\;50^{\circ}C$, respectively, after 72 hour fermentation. Amino type and water soluble nitrogen increased as fermentation progressed. however, the former slightly decreased after 60 hour fermentation. Chungkook-jang fermented at $40^{\circ}C$ showed somewhat higher protease activity than $50^{\circ}C$. However, protease activity at both fermentation temperatures showed the same trend; that is, it increased until 48 hour fermentation and thereafter decrease. Free amino acid content of Chung-kook-jang after 72 hour fermentation at $40^{\circ}C$ was 6 times greater than that of the steamed soybean, while it was 2.5 times greater at $50^{\circ}C$. Based on these results. it seems that the optimum fermentation conditions for Chungkook-jang were $40^{\circ}C$ and 72 hours.

Studies on the Changes of Lipids during Soybean Koji Preparation for Daenjang Fermentation in Model System (된장 발효(醱酵) 중 콩 Koji 제조(製造)과정에 있어서 지질성분(脂質成分)의 변화(變化)에 관한 연구(硏究))

  • The studies are conducted on the changes of soybean lipids in terms of physicochemical characteristics, lipid classess and fatty acid composition during the fermentation process of soybean Koji preparation for daenjang (soybean paste) in a model system using cooked soybean inoculated by Aspergillus oryzae. The total lipids contents were increased during soybean Koji preparation, generally iodine values decreased but acid values increased. Total lipids of soybean Koji consisted of about 90.6% neutral lipids, 7.6% phospholipids and 1.8%, glycolipids indicating that phospholipids contents of soybean Koji was increased when compared to those of cooked soybean. The major components of nonpolar lipids in soybean Koji were free fatty acids(39.6%) and triglycerids(29.2%). Free fatty acids increased as the triglycerides decreased during soybean Koji preparation by the hydrolysis of lipase action. The major components of polar lipids in soybean Koji were phosphatidyl choline and phosphatidyl ethanolamine. Differences were observed in the composition of the polar lipids of cooked soybean and soybean Koji. A little changes also occurred in fatty acid compositions of total lipids, triglycerides and free fatty acids fractions in soybean Koji preparation. Especially a considerable increase of linoleic acid in free fatty acid fraction was observed in soybean Koji.
