Optimization of Conditions for the Microencapsulation of ${\alpha}-Tocopherol$ and Its Storage Stability (${\alpha}-Tocopherol$ 미세캡슐화의 최적화 및 저장안정성 규명)

  • We have produced the microcapsule composed of ${\alpha}-tocopherol$ as a core material (Cm) and the gelatinized polysaccharide as a wall material (Wm). Firstly, we have developed a simple, sensitive, and quantitative analysis method of the microencapsulation product using 5% cupric acetate pyridine solution. We could then optimize all the conditions for the microencapsulation process such as the ratio of [Cm] to [Wm], the temperature of dispersion fluid, and the emulsifier concentration using response surface methodology (RSM). As for the microencapsulation of ${\alpha}-tocopherol$, the regression model equation for the yield of microencapsulation (YM, %) to the change of an independent variable could be predicted as follows : YM=99.77-1.76([Cm]:[Wm])-1.72$([Cm]\;:\;[Wm])^2$. From the ridge of maximum response, the optimum conditions for the microencapsulation of ${\alpha}-tocopherol$ were able to be determined as the ratio of [Cm] to [Wm] of 4.6:5.4(w/w), the emulsifier concentration of 0.49%, and dispersion fluid temperature of $25.5^{\circ}C$, respectively. Finally, the microcapsules produced under the optimal conditions were applied for the analysis of storage stability. The optimal conditions for the storage were found to be the values of pH 9.0 and $25{\sim}35^{\circ}C$. And the storage stability of the microcapsules containing ${\alpha}-tocopherol$ were higher than 99% for a week at pH 9.0 and $25^{\circ}C$.

An Investigation of the Hazards Associated with Cucumber and Hot Pepper Cultivation Areas to Establish a Good Agricultural Practices (GAP) Model (오이와 고추생산 환경에서의 GAP 모델 개발을 위한 위해요소 조사)

  • To analyze the hazards associated with cucumber and hot pepper cultivation areas, a total of 72 samples were obtained and tested to detect the presence of biological (sanitary indicative, pathogenic bacteria and fungi) and chemical hazards (heavy metals and pesticide residues). The levels of sanitary indicative bacteria (aerobic plate counts and coliforms) and fungi were ND-7.2 and ND-4.8 log CFU/(g, mL, hand, or $100cm^2$) in cucumber cultivation areas, and ND-6.8 and 0.4-5.3 log CFU/(g, mL, hand, or $100cm^2$) in hot pepper cultivation areas. More specifically, the soil of hot pepper cultivation areas was contaminated with coliforms at a maximum level of 5.6 log CFU/g. Staphylococcus aureus was detected only in glove samples at a level of 1.4 log CFU/$100cm^2$ and Bacillus cereus was detected in the majority of samples at a level of ND-4.8 log CFU/(g, mL, hand, or $100cm^2$). Listeria monocytogenes, Escherichia coli O157:H7, and Salmonella spp. were not detected. Heavy metal (Zn, Cu, Ni, Pb, and Hg) chemical hazards were detected at levels lower than the regulation limit. Residual insecticides were not detected in cucumbers; however, hexaconazole was detected at a level of 0.016 mg/kg (maximum residue limit: 0.3 mg/kg) in hot peppers.

Control of Microorganisms in School Refectories and Kitchens Using Ozone Water and Ozone Gas (오존수 및 오존가스를 이용한 학교급식에서의 미생물 제어)

  • This study was performed to determine optimum approaches for control of microorganisms in school refectories and kitchens. A reduction of more than 5.0 log CFU/mL was noted for Escherichia coli O157:H7, Salmonella Typhimurium, Listeria monocytogenes, Staphylococcus aureus, and Streptococcus mutans after treatment with 5 ppm ozone water for 60 s. Treatment of different vegetables with ozone water for 5 min showed bactericidal effects with 2-4 log reduction of viable cell number; the bactericidal effects differed according to the kinds of vegetables. The viable cell number on kitchen apparatus and tableware was not detected by ozone water treatment for 60 s. In addition, the count of the bacteria floating in the air in refectories and kitchens decreased 2.0 log CFU/1,000 L air to 0-1 CFU/1,000 L air on treatment with 45 ppm ozone gas for 12 hr. Therefore, ozone water and ozone gas may be good candidates as antimicrobial agents that can be used to improve sanitary conditions in school refectories and kitchens.

Neuronal Cell Protective Effect of New Green Extract against H2O2-induced Oxidative Stress and Analysis of Bioactive Compounds (과산화수소(H2O2)로 유도된 산화 스트레스에 대한 뉴그린 추출물의 신경세포 보호효과 및 생리활성물질 분석)

  • In vitro antioxidant activities and neuronal cell protective effects of the ethyl acetate fraction of a new green extract (Brassica oleracea var. botytis aut italiana) against $H_2O_2$-induced oxidative stress were investigated, and its industrial feasibility was evaluated. The extract showed the highest contents of total phenolic compounds among other extracts as well as a 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity and malondialdehyde (MDA) inhibitory effect. This extract not only decreased the intracellular reactive oxygen (ROS) level but also protected the neuronal cells against $H_2O_2$-induced oxidative stress. On analysis using gas chromatograph-mass spectrometry, the following phenolic compounds were identified: quinic acid, ferulic acid, and caffeic acid. Collectively, these results suggest that this new green extract could contain functional substances that would help prevent the risk of neurodegenerative disease.

Characteristics of gaeryangmerou wine deacidified by a malic acid-degrading yeast (사과산 분해 효모에 의한 개량머루주의 감산 특성)

  • The effect of malic acid-degrading yeast on the quality of Gaeryangmerou (Vitis spp.) wine obtained from grapes commonly used for making wine in Korea was investigated. Alcoholic fermentation was carried out at $25^{\circ}C$, for 14 days. A malic acid-degrading yeast was used as the experimental sample and a commercial yeast, Fermivin, was used as a control. The fermentation process for the experimental yeast lasted 2-3 days longer than that of Fermivin. The pH and the volatile acid content of the wine were 3.94 and 244.20 mg/L, respectively. The total acid content was 0.86% for the experimental group and 0.94% for the control group. The contents of malic acid and succinic acid in the experimental group were significantly lower than that of the control group (245.61, 50.18 mg% for experimental group versus 302.44, 68.39 mg% for control group, respectively). In contrast, lactic acid content was slightly higher in the experimental group. As expected, the main volatile flavor compounds of Gaeryangmerou wine varied with the yeast used for fermentation and were determined to be isoamyl alcohol, ${\beta}$-phenethyl alcohol, 2-methyl-1-propanol, and diacetyl for the experimental wine.

Protective effects of Aruncus dioicus var. kamtschaticus extract against hyperglycemic-induced neurotoxicity (포도당 처리로 유도된 뇌신경세포 독성에 대한 눈개승마 추출물의 보호효과)

  • To assess the physiological effects of Aruncus dioicus var. kamtschaticus extract on cytoxicity of a neuronal cell line, antioxidant activity, and neuroprotection against intensive glucose-induced oxidative stress were quantitated. Compared to the other fractions, the ethyl acetate fraction of Aruncus dioicus var. kamtschaticus (EFAD) showed the highest total phenolics and flavonoids. The 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) assay and malondialdehyde inhibitory effect test confirmed the superior antioxidant activity of EFAD. Moreover, EFAD also decreased the intracellular ROS level and suppressed neuronal cell death against intensive glucose- or $H_2O_2$-induced oxidative stress. Additionally, assessment of ${\alpha}$-glucosidase and acetylcholinesterase inhibitory activities revealed that EFAD was an effective inhibitor of ${\alpha}$-glucosidase and acetylcholinesterase. Finally, high-performance liquid chromatography analysis identified caffeic acid as the main ingredient of EFAD. Overall, these results suggest that the EFAD is a good natural source of biological compounds that counteract hyperglycemic neuronal defects.

The Brassica rapa Tissue-specific EST Database (배추의 조직 특이적 발현유전자 데이터베이스)

  • Brassica rapa is an A genome model species for Brassica crop genetics, genomics, and breeding. With the completion of sequencing the B. rapa genome, functional analysis of the genome is forthcoming issue. The expressed sequence tags are fundamental resources supporting annotation and functional analysis of the genome including identification of tissue-specific genes and promoters. As of July 2011, 147,217 ESTs from 39 cDNA libraries of B. rapa are reported in the public database. However, little information can be retrieved from the sequences due to lack of organized databases. To leverage the sequence information and to maximize the use of publicly-available EST collections, the Brassica rapa tissue-specific EST database (BrTED) is developed. BrTED includes sequence information of 23,962 unigenes assembled by StackPack program. The unigene set is used as a query unit for various analyses such as BLAST against TAIR gene model, functional annotation using MIPS and UniProt, gene ontology analysis, and prediction of tissue-specific unigene sets based on statistics test. The database is composed of two main units, EST sequence processing and information retrieving unit and tissue-specific expression profile analysis unit. Information and data in both units are tightly inter-connected to each other using a web based browsing system. RT-PCR evaluation of 29 selected unigene sets successfully amplified amplicons from the target tissues of B. rapa. BrTED provided here allows the user to identify and analyze the expression of genes of interest and aid efforts to interpret the B. rapa genome through functional genomics. In addition, it can be used as a public resource in providing reference information to study the genus Brassica and other closely related crop crucifer plants.

Development of Root Media Containing Carbonized and Expanded Rice Hull for Container Cultivation of Horticultural Crops (팽연왕겨와 훈탄을 포함한 원예작물 용기재배용 혼합상토의 개발)

  • Objective of this research was to develop root media containing expanded rice hull (ERH) and carbonized rice hull (CRH). To achieve this, the physico chemical properties of two materials were analysed and blended with peatmoss (PM) or coir dust (CD) with various ratio. Based on the physical properties of the blended materials, 4 root media were selected for future experiment. After the analysis of pH and EC of the selected root media, the kinds and amount of pre-planting nutrient charge fertilizers (PNCF) incorporated into each root medium were varied, and then, final chemical properties of the root media were analysed. The total porosity (TP), container capacity (CC), and air-filled porosity (AFP) were 81.3%. 39.9%, and 41.4% in ERH and 77.6%, 64.1%, and 13.5% in CRH, respectively. The percentage of easily available water (EAW, from CC to 4.90 kPa tension) and buffering water (BW, 4.91-9.81 kPa tension) were 11.37% and 5.27%, in ERH and 17.26% and 14.28% in CRH, respectively. The pH of ERH was 7.1, but it was extremely high in CRH such as 11.2. The EC and CEC were $1.31dS{\cdot}m^{-1}$ and $12.1meq{\cdot}100g^{-1}$ in ERH and $6.53dS{\cdot}m^{-1}$ and CEC 7.79 $meq{\cdot}100g^{-1}$ in CRH, respectively. The ranges of TP, CC and AFP in 4 selected media (PM + ERH, 6:4, v/v; CD + ERH, 8:2; PM + CRH, 7:3; CD+CRH 6:4) were 89.2-90.3%, 67.3-81.8%, and 8.3-21.9%, respectively. The pHs and ECs in root media containing peatmoss such as PM + ERH (6:4) and PM + CRH (7:3) were 4.0-4.3 and $0.33-0.365dS{\cdot}m^{-1}$, whereas those of CD + CRH were 7.4-7.9 and $1.282dS{\cdot}m^{-1}$. The pHs and ECs, however, analysed before and after the incorporation of PNCF in each root medium were not significant different. This result indicated that the incorporated fertilizers in PNCF to adjust medium pH did not dissolve enough to influence medium pH, but it is very normal in root media containing dolomitic lime and sulfur powder in adjusting pH. The Information obtained in this study may facilitate an effective formulation of root media containing rice hulls.

Environmental Assessment of the Shihwa Lake by using the Benthic Pollution Index (저서오염지수(BPI)를 이용한 시화호 환경평가)

  • In order to assess the ecological changes induced by organic pollutants of the Shihwa Lake, BPI (Benthic Pollution Index) based on the benthic faunal community was employed. It was modified from Infaunal Trophic Index (ITI), and recommended as a pollution detecting method for the environmental assessment. The BPI values were calculated from the benthos data, which were collected for three terms: in 1980, before the Shihwa Lake was built up; in 1994-1997, which the Shihwa Lake was completely isolated from the outer seawater; in 1997-1999, after inflow of the outer seawater. Since the Shihwa Dike was constructed in February 1994, the pollution intensity of the lake had been increased from the narrow and inner part of the former Gyeonggi Bay and spread fast along the coast line of the Shihwa Lake. Then, in 1996 it showed the very high BPI levels all around the Lake. This serious polluted condition had been lasted till 1997, when the inflow of the seawater was begun. In 1998, from the nearest part of the Shihwa Gate, the BPI levels gradually became low in most area of the Lake, except its inner and narrow part. These greatly lowered BPI levels mean that the seawater inflow could be assumed to affect positively in the lake. Furthermore, BPI gave the same results from the other environmental assessment based on the abundance and the species richness of macrobenthic community. It shows that BPI could be useful as an effective method to assess the marine environment and evaluate the status of environmental conditions.

Relationship between Water Temperature and Oxygen Consumption Rate of the Black Scraper Thamnaconus modestus (말쥐치, Thamnaconus modestus의 산소소비율과 수온의 관계)

  • The black scraper Thamnaconus modestus was a commercially important fish species in the 1980s, but suddenly its commercial significance decreased in the 1990s mainly due to continuous overfishing. Recently, in order to reverse the depleted stocks of the black scraper and help the species recover, seed production technology has emerged. This has led to the farming of the black scraper in several parts of the southern coast of Korea. Since detailed research on its metabolism in relation to water temperature has been scanty, this was the investigative focus of the present study. The standard metabolism rates of the black scraper (9-10 months old, total length=$22.6{\pm}0.8cm$, wet weight=$140.3{\pm}13.9g$) were measured at seven different water temperature settings (12, 15, 17, 20, 23, 26, $28^{\circ}C$) to understand the relationship between metabolism and water temperature. Relationships between water temperature (WT) and oxygen consumption rate (OCR) were obtained as SOCR (weight-specific oxygen consumption rate) = 0.0117WT - 0.0135 ($r^2=0.9351$) and IOCR (oxygen consumption rate per individual) = 1.8160WT - 5.4007 ($r^2$ = 9428). The $Q_{10}$ (temperature sensitivity), an indicator of the sensitivity of biological function to temperature, was analyzed. In our experiment, when the water temperature increased, the $Q_{10}$ value decreased. The $Q_{10}$ value was 6.27 in waters where the temperature ranged from $12-15^{\circ}C$ and this was much higher than the values obtained in waters where temperatures ranged between (1) $15-23^{\circ}C$ and (2) $23-28^{\circ}C$. Consequently, it was shown that the black scraper is a warm water species and inhabiting waters in the temperature range from $15-28^{\circ}C$ is deemed appropriate.
