Laminin Expression in the Rat Lung Development (흰쥐 폐 발생시 Laminin의 발현에 대한 연구)

  • Laminin, a kind of multidomain glycoproteins, is mainly localized in the basement membranes of various tissues. It is known that laminin plays an important part in mammalian lung morphogenesis. The authors have undertaken this study to investigate the changes in the distribution of laminin, and to find out cells which synthesize laminin during the organogenesis and differentiation of the lung. The fetal and neoantal rats (Sprague-Dawley strain) were used as experimental animals. The immunohisto-chemical methods were employed for detection of laminin within the developing lung tissue and the immunegold cytochemical methods were performed for detection of cells which synthesize laminin according to each stage of development. The results are as follows; 1. During fetal life, strong immunoreactivity for laminin is maintained in the basement membranes of the blood vessels and the bronchioles, the extracellular matrix of the mesenchyme, and basal lamina of the alveolar septum in the fetal rat lung. 2. After birth, laminin immunoreactivity at the alveolar septum is gradually reduced. 3. During fetal life, laminin is mainly detected within the cytoplasm of the mesenchymal cells, the endothelial cells of blood vessels and the fibroblasts in fetal rat lung. 4. According to the differentiation of type I and type II pneumocyte after birth, laminin is detected within cytoplasm of the type I pneumocytes, type II pneumocytes and fibroblasts. It is consequently suggested that laminin is largely expressed in the developing lung and laminin may be also synthesized by the type II pneumonocytes at early newborn stages.

Analysis of Surface Properties of PVC Thin Film according to Addition of Non-solvent to PVC-THF Solution (PVC-THF 용액에 비용매 첨가에 따른 PVC 박막의 표면 특성 분석)

  • The effect of the addition of a polyvinylchloride (PVC) non-solvent to a PVC-tetrahydrofuran (THF) solution on the surface properties of the PVC thin film was analyzed. The non-solvents used were composed of alcohol-based and non-alcoholic ones. Surface morphologies of PVC thin films according to the addition of the non-solvent were compared. In addition, the hydrophobic properties relying on the surface characteristics were compared. The micro-bubbles generated in the preparation of PVC-THF solution affected the surface morphology of the thin film. In order to implement the normal surface physical properties of the coating thin film at the relatively high concentration of PVC-THF solution, the selection of appropriate drying method was required. When an alcohol-based non-solvent was added, a PVC thin film having a granular porous surface was obtained and exhibited super hydrophobic properties. The volume ratio of the PVC-THF solution to the non-solvent affects the surface shape of the coating thin film. The larger the amount of non-solvent was added, the more advantageous it was to form a super hydrophobic PVC thin film.

  • To provide basic information on the integumentary system of the blenny, Pholis nebulosa, ultrastructures of epidermal and dermal layer were examined by means of the light and transmission electron microscope. The skin of the blenny consisted of epidermal and dermal layer. Epidermal layer consisted of supporting cell and unicellular gland. The supporting cells were classified into superficial cell, intermediated cell and basal cell, and the gland cells were classified into mucous secretory cell and club cell. The cytoplasm of supporting cells was divided into cortex and medullar part. In the cortex and medullar part, microfilaments and cell organelles were well developed, respectively. Superficial cell of epidermal layer was cuboidal and contained nucleus of horseshoe shape. Intermediated cell had a nucleus of irregular form and the electron density was higher than the other supporting cells, Basal cell was columnar, but nucleus was situated in the upper cytoplasm. Cell organelles of the basal cell were poor than the other supporting cells, but membrane interdigitations were well developed. The cytoplasm of mucous secretory cell had a well-developed ovoid secretory granules, which reacted to red with AB-PAS reaction. The club cell had a we31-developed round secretory granules and endoplasmic reticulum. figment cells were classified into two type. The one contained pigment granules of electron dense, and the other contained reflecting platelets. The cytoplasm of fibrocyte had n well developed rough endoplasmic reticulum.

Ethyl Acetate Extract of Korean Rice Wine Lees Inhibits IgE-Mediated Degranulation in Rat Basophilic Leukemia RBL-2H3 Cells and Passive Cutaneous Anaphylaxis in Mice (주박 에틸아세테이트 추출물의 항알러지 효과)

  • Mast cells, the central effector cells involved in the allergic response, release histamine, arachidonic acid, and proinflammatory cytokines. We investigated the effect of the ethyl acetate fraction (EA), derived from Korean rice wine lees, on RBL-2H3 cell degranulation and passive cutaneous anaphylaxis in an animal model. The EA fraction suppressed the release of beta-hexosaminidase, a marker of degranulation, and the mRNA expression of interleukin-3 (IL-3) and IL-13. EA also successfully suppressed the passive cutaneous anaphylaxis (PCA) reaction in mice in a dose-dependent manner. These results suggest that EA can inhibit mast cell degranulation through the inhibition of IL-3 and IL-13 mRNA expression, and that EA may potentially serve as an anti-allergic agent.

ULTRASTRUCTUAL ANALYSIS OF THE FIBROUS LAYER OF CONDYLE IN THE RAT TEMPOROMANDIBULAR JOINT WITH AGEING (가령에 따른 흰쥐 하악과두 섬유층의 미세구조 및 교원원섬유의 변화)

  • The fibrous layer of mandibular condyle of the neonatal, 1-, 7-, 14-, 27-, 55-days and 1 year old rats were examined in the electron microscope with particular attention to the ultrastructure and diameter of collagen fibrils. In the 1-day rats, most of the cells of the fibrous layer were undifferentiated mesenchymal cells and fibroblasts with rough a little developed rough endoplasmic reticulum(RER) and golgi apparatus(GA). In 7-, 17 and 27-days old rats, most of the fibroblast showed well developed GA and RER with widely distended cisternae containing granular materials. In many of these cells contained intracytoplasmic filaments among the cytoplamic organelle. In 55-day and 1-year old rats, three types of cells were observed, ie, cells containing well developed cytoplasmic organelle presumed to be involved in the collagen fibril synthesis, cells showing well developed lysosomes, golgi apparatus, mitochondria and short cytoplasmic process presumed to be involved in the active resorption of the injured collagen fibrils or cellular debris, cells containing many intracytoplasmic filaments and a little organelle presumed to be cells of inactive state. The average diameters of collagen fibrils were similar in 1- and 7-day old rats as $38.48{\pm}3.81nm$, $38.06{\pm}3.86nm$. That was thickest in 14 days old rats as $50.21{\pm}3.93nm$ among experimental groups. They were gradually thinner in 27-, 55-day rats as $40.05{\pm}2.52nm$, $43.63{\pm}1.20nm$ and thinnest in 1-year old rats as $37.38{\pm}2.17nm$. The distribution pattern of diameters of collagen fibrils were unimordal with peak of 30-60nm in rats from 1-day to 17-day old. With aging from 27-day to 1 year old rats, collagen fibril diameters showed wide distribution pattern and percentage of thin collagen fibrils increased. These results may show the functional adaptation of fibrous layer of mandibular condyle to the increased mechanical forces with aging.

Isolation and Identification of a Marine Bacterium Producing an Immunostimulant (면역증강물질을 생산하는 해양미생물의 분리 및 동정)

  • 면역증강물질을 생산하는 균주를 해수로부터 분리하여 동정한 결과 크림색의 둥근 점질성의 집략을 나타내었으며, Gram 음성의 간산형이고, 미약한 운동성을 가지며 catalase 양성과 oxidase 음성 반응을 보였다. 균주를 6시간 , 20시간, 72시간, 및 144시간 뱅양하여 전자 현미경사에서 관찰한 결과 20시간 배양한 영양세포는 1.25~0.6$\times$0.6$\mu\textrm{m}$ 크기의 간균 형태를 갖추었으며 시간이 흐를수록 세포내 관립의 밀도가 증가하면서 세포형태가 다형태성으로 변하였다. 세포내 과립의 존재를 확인하기 위해 sudan black B로 염색한 결과 양성으로 polyhy-droxy butyrate로 예상되었다. 생육을 위한 최적 온도는 3$0^{\circ}C$, pH는 3.0~10.0이었으며, 호기성균이었다. D-Glu-cose, D-mannose, D-mannitiol. insitol, maltose 등의 당과 L-asparagine, L-glutamate 등의 아미노산을 이용하였고 특시, O/F test에서 glucose와 maltose를 산화 하였다. 이상과 같은 결과로 해양세균 유래인 Pseudo-mallei group의 Burkholderia 속으로 확인되었음로 본 균주를 Burkholderia sp. IS-203으로 명명하였다.

  • Neutropenia is a major dose-limiting factor in cancer chemotherapy diminishing its usefulness and increase patient's susceptibility to infectious disease. Some recombinant human granulocyte colony stimulating factors(rhG-CSFs) are in use to reduce the risk of this serious side effect. In this study, we examined the pharmacological properties of DA-3030, a rhG-CSF expressed in E. coli. DA-3030 100 and $\mu\textrm{g}$/kg, i. v., had no significant effect on the central nervous, gastrointestinal system in mice and cardiovascular system in rabbits, but it slightly inhibited the spontaneous motility of isolated nonpregnant uterus in rats. It also had no influence on excretion of urinary electrolytes. DA-3030 administered for successive 3 days increased the blood WBC count in zymosan air pouch inflammed rats and in normal rats. These results indicate that DA-3030 has little side effects in animals.

  • PAF (Platelet-activating factor: 혈소판 활성화인자)는 1972년 Benveniste등에 의해 토끼의 호중구 배양 상청액중에서 발견되어 1979년 그 구조가 1-alkyl-2-acethyl-sn glycero-3-phosphocholine의 구조를 갖는 에테르형 인 지질임이 밝혀졌다. 그후 혈소판 이외 과립구, 단구나 macrophage, 혈관내피세포등 조직의 염증담당세포가 다양한 자극에 응하여 PAF를 생성됨이 보고되었다. PAF가 나타내는 대표적 활성으로는 혈소판, 호중구, 단구들의 활성화, 호중구의 유주 활성, 혈관투과성 항진, 혈압강하작용. 기관지 수축 등이 알려졌으며. 또한 염증, 알러지, 천식 endotoxin shock 등 여러질병에 직·간접적으로 관여함이 알려졌다. 이와같은 여러 생리 현상은 PAF의 특이적수용체를 개재하여 일어난다는 것이 밝혀졌다. 따라서 PAF의 다양한 질병의 관여가 밝혀짐으로서, PAF길항제의 개발이 활발히 진행되어왔다. 지금까지 PAF길항제의 개발은 PAF 구조 유사체. benzodiazepam유도체, thiazole유도체 등과 같은 합성품과 ginkolide, kadsurenone과 같은 천연물 유리의 것이 알려져 in vivo model에서도 그 효능이 확인되었다. 본 연구는 이와 같은 배경에서 20여 종의 생약에서 PAF 길항제를 검색하던 중 5종류의 생약에서 PAF 길항작용을 갖는 분획을 찾았기에 이에 보고한다.

고양이 시상하부의 뇌실옅핵과 시각교차위핵의 Vasopressin과 Oxytocin 신경원에 대한 면역조직화학적 연구

  • 고양이의 시상하부내 SON과 PVN에서 VP분비신경원과 OT분비신경원의 형태와 분포를 관찰하기 위해서 면역조직화학적 반응을 시행하였다. 광학 및 전자 현미경적 관찰에 의하면 VP분비신경원과 OT분비신경원의 형태 차이는 없었다. 이들의 형태는 방추형, 타원형 그리고 원형이었으며 크기는 25-40mm으로 나타났다. SON은 시삭위에 능형의 형태로 나타나고 OT분비신경원과 VP분비신경원의 비율은 1:2.4이며 PVN은 제3뇌실을 따라 삼각형 모양으로 나타나고 VP분비신경원과 OT분비신경원의 비율은 1:1.6으로 나타났다 전자현미경적 관찰에 의하면 VP분비신경원과 OT분비신경원에서 과립형질내세망, 사립체, 골지장치, 그리고 미세소관 등이 특히 잘 발달되어 있음을 관찰할 수 있었다.

Ultrastructural Observations of Glutamatergic Synaptic Components in the Basilar Pontine Nuclei of the Dog (개의 교핵내 glutamate성 연접 성분의 미세구조적 위치관찰)

  • The distribution of glutamatergic synaptic structures in the dog basilar pons was investigated at the ultrastructural level using monoclonal antibodies against fixative-modified glutamate. Electron-dense reaction product was densely localized at the perinuclear region in the neurenal somata and often observed along the microtubules located within the dendritic processes. One or more unlabelled axon terminals made asymmetric synaptic contacts with glutamate-immunoreactive dendritic profiles. In audition, reaction product was observed either within axonal processes surrounded by myelin sheath or axon terminals. Immunoreactive axon terminals made asymmetric synaptic contact either with unlabelled or labelled dendritic profiles. These observations provided an anatomic evidence of how this excitatory neural element might perform its function in a multisynaptic pathway involving glutamatergic afferents to the basilar pons, glutamate-immunoreactive pontocerebellar projection neurons, and the glutamate-positive granule cells of the cerebellar cortex.
