A seed pelleting technique was developed for easy handling of small sesame seeds (Chinbaeckkae) and for direct seeding in field. The mixture of pelleting material, binder(sodium alginate) and seeds were dropped in 100mM CaCl$_2$ solution. Charcoal was appeared as a best material among all to make pellets with the characters of hardness, breakdown, destruction rate and germination percentage. Zeolite formed the hardest pellet seeds among the materials and 1~3 seeds contained in a pellet on the treatment of 87g of sesame seeds mixed in 1$\ell$ CaCl$_2$ solution was estimated optimum rate of seed mixure with few appearance of seedless pellet. The moisture absorption rate was greater in the order of peat+charcoal > peat > charcoal > zeolite. Germination percentage of pelleted seeds with charcoal was the best and similar to that of common non-treated seeds. Germination percentage of pelleted seeds by sowing depth of exposing a quarter of pellet seeds on the surface of vermiculite germination test box and were 92% in charcoal treatment and was the best among all released materials.
The changes of protein bodies in endosperm cells of both seeds with red seed coat and indehiscent seeds of Panax ginseng C.A. Meyer have been investigated in relation to the embryo development. In the early stage of seeds with red seed coat, spherical spherosomes were distributed in endosperm cells. Protein bodies were formed from vacuoles containing the storage protein. Cell organelles were hardly observed in the cytoplasm. In the late stage of the seed with red seed coat, the endosperm was filled with spherosomes and protein bodies. The protein bodies consisted of amorphous inclusions with high electron density or proteinaceous matrix with even electron density. In the seed of in dehiscence, the protein body in endosperm cells contained globoids and protein crystalloids. The globoid of protein body had a electron dense materials. Umbiliform layer was formed between embryo and endosperm. The deformation patterns of endosperm cell wall and the cellulose microfibril were observed in endosperm cells near the umbiliform layer. Umbiliform layer consisted of lipid body and autolyzed cell debris. The protein body of endosperm cell near the umbiliform layer showed various degenerative patterns, and so electron density of proteinaceous matrix was gradually decreased.reased.
Accumulations of the storage proteins in protein storage vacuole and the differentiation of protein bodies from protein-filled ER in developing pea cotyledons have been investigated using conventional and immunoelectron microscopy. To improve the fixation quality, single cells separated enzymatically from sliced cotyledons were used. At early stages of seed development osmiophilic protein accumulates in rER lumen were observed quite often. This protein-filled ER cisternae were differentiated into cytoplasmic protein bodies at late stage by the process called terminal dilations which have been considered a principal route of the formation of cytoplasmic protein bodies somewhat later in seed maturation. Immunocytochemical labellings of the vicilin and legumin show that presence of vicilin on both of the cytoplasmic PB and PD, but limited presence of legumin only on the cytoplasmic PB at intermediate stage of seed development. Immunogold labellings of Bip, ER retention protein, were observed on the inner periphery of protein deposits in protein storage vacuole. This result was regarded that Bip can recognize and retrieve misfolded protein during active accumulation of storage protein to the PD in PSV.
Formation, cellular distribution and structural changes of storage lipid, and active site and cellular localization of lipase in endosperms and cotyledons of lipid-rich seeds such as Helianthus annuus, Ricinus communis and Pinus koraiensis, and in those of starch-rich seeds such as Pisum sativum and Zea mays were investigated in relation to the seed development by cytochemical methods. In endosperms and storage cotyledons of lipid- and starch-rich seeds after seed-gathering, there were widely distributed storage material which was composed of spherical protein bodies, spherosomes, and starch granules. But cellular organelles were hardly observed in the cytoplasm. Staining pattern of vesicles released from SER, and of low electron dense membraneous granules, which were perhaps at an early stage of spherosomes, were the same as in the spherosome. Electrondense granules released from RER were observed in the vicinity of plasma membrane. As a result of lipid staining, the spherosomes were more electron dense and were uniform as compared with the protein matrix within the protein body and cytoplasmic proteinaceous granules. The major component of the spherosome was determinated to be lipid. Spherosomes and vesicles containing SER-released materials showed the same as in the electron density. Lipase activity was especially strong in the inner region and on the surface of decomposed spherosomes and near the plasma membrane.mbrane.
Sesame was one of the economical crops in Korea. However, cultivation area of sesame has been decreasing rapidly due to the lack of mechanization for this crop and the opening of agricultural product market. Sesame seed is so small that ordinary seeder can not seed properly. In rural practice, farmers seed sesame with hand and do thinning after shoot emerges. Seeding and thinning in sesame cultivation take more than 40% of total labor To reduce labor in seeding and thinning, a pellet seed machine for sesame has been developed. The pellet seed machine is very simple in structure. It utilizes the chemical reaction between alginate solution and $CaCl_2$. 유전체 전장 연관분석 (GWAS)은 단일염기다형성(SNP)의 유전자형과 표현형 간의 통계적인 연관성을 분석함으로써 품종 선발용 SNP Marker 개발에 응용되고 있다. 본 연구에서는 Tano Red와 Ruby seedless 교배실생 278 계통을 대상으로 여러 과실 특성에 따른 관련 SNP를 동정함으로써 육종 선발에 필요한 DNA marker 개발에 필요한 기초 유전 자료를 얻고자 하였다. 한 계통 당 5~10개의 포도알을 선택하여 과립중, 과육탄성, 과피탄성, 과육경도, 과피경도, 과립당 종자갯수, 과립당 종자무게 및 인장강도를 측정하였다. 각 개체는 Genotyping by sequencing (GBS) 방법으로 Sequencing하여 Reference genome (Vitis vinifera PN40024 12X v2.)과 mapping 하였다. MAF (Minor allele frequency) >5%, Missing Data <30% 의 조건을 가진 SNPs 만 1차 선발하여 TASSEL과 GAPIT 프로그램으로 GWAS 분석을 하였다. Manhattan plot 결과 과립중 형질에서는 33개, 과립당 종자무게 25개와 인장강도에서는 20개의 통계학적으로 유의한 SNPs 가 선발되었고, 특이적으로 이들 모두 18번 염색체에서 발견되었다. 그러나 나머지 형질에서는 유의한 차이를 보이는 SNPs를 선발하지 못하였다. 과실의 인장강도는 수확 후 저장성과 유통과정에 영향을 미치기 때문에 Marker 개발을 통한 품종선별이 중요하다. 향후 이러한 특성과 본 연구를 통해 동정된 SNPs 의 상관관계를 구체적으로 연구하여 Marker 개발에 활용하고자 한다.
Immunoelectron microscopy of storage protein at early stage of seed development showed legumin was firstly accumulated protein in between endoplasmic reticulum (ER) cisternae, and these accumulates were differentiated into protein body (PB) by transformation at later stage. Thin sections of pea cotyledons during the later stages of seed maturation showed three morphologically different types of protein bodies. One of these, presented as rough-surfaced cisternae with terminal dilations, which contained protein deposits and were often found interdigitated between stacks of rough endoplasmic reticulum. Conventional electron microscopy at earlier stages of cotyledon development showed this protein body type initially developed from the rough ER. This transformation of endoplasmic reticulum into a protein body is believed to represent a new pathway of protein body development.
