  • 점박이송사리에 관한 해부학적 자료축적의 일환으로 부속지골격의 형댈 발생 및 골화 진전양상을 조사하였다. 각 부속지의 구성골은 전피성골이 연골성골보다 먼저 골화되었다. 부속지의 기조는 가슴지느러미, 구리지느러미, 등 및 됫지느러미, 그리고 배지느러미의 순으로 골화되었다. 골화 진전에 따라 초기 자어시 유함되어 있던 수직지느러미의 기골은 방사기골과 중담기골로, 가슴지느러미의 오초-견갑골은 오훼골과 견갑골로 분리, 분화되었다. 그러나 골화진전에 따른 구성 골간의 유합은 나타나지 않았다. 5가지 부속지의 골화 완성은 가슴지느러미, 꼬리지느러미, 등지느러미, 됫지느러미 그리고 배지느러미의 순으로 이루어졌다.

  • 의약품등의 배아(태자) 독성물질에 의해서 유도될 수 있는 태자체중의 감소와 골화지연은 실험동물을 이용한 생식독성 시험에 있어서 자궁내 발육지연 (intrautering growth retardation)으로 평가된다. 태자체중은 측정이 매우 용이한 변수에 속하지만 골화지연의 평가에는 유해인자 (noxa) 의 작용시 기형이 유발되지 않는 골격부위가 가장 적합한데, 어느 골격부위가 발육지연 효과의 평가를 위하여 가장 적합한지를 알아내기 위해서는 태자체중과 골화의 상관성 조사가 필수적이다. 연구결과 골격형성은 골격부위에 따라 큰 차이를 보였는데, 크게는 거의 골화가 종료된 부위 (예: 흉골) 와 골화가 진행증인 부위 (예: 천미추) 로 구분되었고 주요 골격부위의 골화와 태자체중의 관계를 조사한 바, 골격부위에 따라서 상이한 상관관계를 나타내었다. 그런데 본 연구에서 나타난 바로는 ICR 마우스에 있어서 천미추의 골화짐 수가 태자제중과 가장 높은 상관성을 나타내었다. 또한 천미추의 변이계수는 타 골격부위에 비해서 그다지 높지 않았다. 따라서 ICR 마우스를 이용한 생식독성시험에 있어서 천미추의 골화 진행도는 의약품등에 의해 유도될 수 있는 발육지연 효과의 평가를 위한 가장 적합한 지표라고 판단된다.

The Role of Cartilage Canals in Osteogenesis and Growth of the Vertebrae in the Human Fetuses (인태아 척추 골화과정에서 연골관의 역할)

  • To investigate a role of cartilage canals in osteogenesis and growth of the vertebrae, in human fetuses ranging from 50 mm to 260 mm crown rump length were studied by electron microscopy. The initial appearance of cartilage canals of the vertebral body was observed at 60 mm fetus. In 80 mm fetus, primary ossification center in the vertebral body was first noted. The vertebral body showed calcified chondrocytes surrounded by a tone of hypertrophied chondrocytes and deep canals which terminated in calcified matrix. Most hypertrophied chondrocytes in the centrum showed in various stage of degeneration in disorderly arrangement. At the blind end of deep canal, osteogenic cells, osteoblasts and chondroclasts were observed. Resorption of unmineralized cartilage septa was undertaken by perivascular cells within cartilage canals. The ruffled border of the chondroclast was restricted to resorption site of calcified cartilagenous matrix. The periosteal bone formation was followed by the appearance of primary center of the centrum at 120 mm fetus. The osteoblasts of the perichondrium started to lay down a thin membranous bony lamella on the outer surface of the osseous trabeculae of the centrum. The processes of bone formation in the vertebral bodies were found to possess morphological similarities to that occurring at secondary center of the epiphysis of a long bone. These results indicate that the connective tissue cells within the cartilage canals proliferate and differentiate into osteoblasts at the site of endochondral ossification of the vertebrae.

Osteological Development of the Larvae and Juveniles of Takifugu pardalis (Teleostei: Tetraodontodae) (졸복 (Takifugu pardalis) 자치어의 골격발달)

  • Fertilized eggs of Takifugu pardalis (Temminck et Schlegel) were collected on the coast of Jook-do in Tongyong, Korea, from March 1997 to June 1999, and hatched and reared in the laboratory to investigate osteological development of the larve and juveniles. The newly hatched larvae attained a mean of $3.13{\pm}0.05mm$ in total length (TL). Ossification first began in the parasphenoid, premaxillary and dentary. At 5 days after hatching, the postlarvae attained a mean of $3.82{\pm}0.03mm$ in TL, and their sphenotic, prefrontal, preopercle, opercle, maxillary, and articular were ossified. At 15 days, the postlarvae attained a mean of $7.84{\pm}0.06mm$ in TL, their nasal and posttemporal bones were ossified and the hyoid arch completed ossification. Ossification of all bones was completed in the juveniles (mean = $10.21{\pm}0.06mm$ in TL) at 21 days.

  • Osteolgical development of Korean striped bitterling, Acheilognathus yamatsutae (Cyprinidae), were investigated using cartilage and bone staining. The parasphenoid, dentary, pterygoid, ceratohyal, branchial arch and parts of the caudal skeleton were formed first as cartilaginous elements at $6.82{\pm}0.08mm$ total length (TL). Formation of frontal, parietal, and maxillary were notable at $7.76{\pm}0.09mm$ TL, and the proximal radial, scapula,and coracoid in the pectoral girdle were developed at this stage. At the same time, the neural and hemal spine were present in the caudal vertebra. The opercle and branchiostegals were observed at $9.68{\pm}0,14mm$, and the posttemporal was formed at $12.9{\pm}40.64mm$ TL, respectively. Ossification in the parasphenoid, pharyngeal bone, dentary, premaxillary, maxillary, and opercle began at about $9.68{\pm}0.14mm$ TL. The cleithrum, supracleithrum, urostyle, and caudal fin were calcified at this stage. The vertebral column was formed and ossified at $11.52{\pm}0.13mm$ TL, and the frontal, preopercle, subopercle, and hyomandibular were ossified at $15.30{\pm}0.68mm$ TL. This fish was late in developing the skeletal formation and ossification as compared with other fishes.

  • The skeletal development of the larvae and juvenile of red marbled rockfish, Sebasticus tertius (Barsukov et Chen) was studied based on individuals that were discharged and reared in the laboratory from April to May 1997. In 8 days after bearing, the postlarvae attained 4.42 mm in total length (TL), and its parasphenoid, premaxillary, maxillary, and clavicle were ossified for the first time at this stage. In 15 days after bearing, the postlarvae attained 5.23 mm in TL, and its pterotic, basioccipital, exoccipital, opercle, and preopercle were ossified, with one spine on the each cranium and preopercle. In 27 days after bearing, the postlarvae attained 8.81 mm in TL, its vertebra were posteriorly ossified to the 15th centrum, and five spines were formed on the preopercle. In 39 days after bearing, the juveniles attained 14.21 mm in TL, and the all bones were almost completed at this stage.

  • Skeletal development in the oblong rockfish, Sebastes oblongus, was studied based on extensive larval rearing conditions from December 2007 to March 2008. Newly-hatched larvae lacked osteological elements. After 3 days of bearing, jaw bones were ossified almost simultaneously with the frontal, parietal, clavicle, opercle, preopercle and branchiostegal rays at 8.0 mm average total length (TL). Ossification of the opercular was completed by 12.3 mm and the full complement of ossified elements of cranium and pectoral girdle were completed by 16.2 mm. Ossification of the cartilaginous caudal complex began to at 9.8 mm, and completely ossified by 18.0 mm. The fusing of the first and second, and the third and fourth hypurals initially occurred by 10.8 mm, and their fusion was finally completed at 18.0 mm. Notochord flexion occurred and formed an individual centrum by 8.5 mm and 10.8 mm, respectively, and all 26 centra were ossified by 13.2 mm. The preorbital bone began to ossify on the anterior region of eye at 10.8 mm, and the $1^{st}$ suborbital bone appeared ossified on the lower of eye by 12.3 mm, and all elements were ossified at 27.5 mm. Finally, after 71 days of bearing, the juveniles became 27.5 mm, and ossification was completed at this stage.

Early Life History and Spawning Behavior of the Gobiid Fish, Luciogobius guttatus Gill (미끈망둑, Luciogobius guttatus Gill의 산란습성(産卵習性)및 초기생활사(初期生活史))

  • The gobiid fish, Luciogobius guttalus Gill has an anguilliform with some blackish and reddish brown color in life. It grows up to 90mm in total length. The specimens have been collected from several localities in the southern parts of Korea and Hokkaido, Japan. During the ebb tide, the fish was found in high level of intertidal zone exposed to the air among pebbles in the hollows and slopes of rocks. There are also some other small gobiid fishes comprising 3 species of relative gobies and 1 species of blennioid fish. A total of 5 egg masses were collected from the coast of Haeundae in April to May 1990. Each egg mass was deposited in one layer on the underside of a stone embedded in pebbles and guarded by the male parent. The eggs are club-shaped ranging from 2.71 to 2.80mm in long axis and from 0.65 to 0.74mm in short axis. The eggs were hatched in 98 hours after incubatied at the temperature varying from 19.5 to $25.5^{\circ}C$The newly hatched larvae were from 3.85 to 4.00mm in total length with 35~36 myomeres. In eleven days after hatching, total length reached 5.50mm. The part of the fin-fold of the future dorsal and anal fins became high. In sixteen days after hatching, the lavae averaged 6.20mm in total length and the caudal notochord flex at $45^{\circ}$. The larvae reached the juvenile stage in 48~50 days after hatching and attained 12.80~14.00mm in total length, and all fin-rays was formed. Ossification of the cranium took place at 5.50mm of mean total length in parasphenoid and basioccipital. Ossification of the visceral skeleton occurred in areas where active movements of bones are required, notalbly in the parts of feeding and respiration. Vertebrae began to develop from the anterior end to ossify posteriorly. Neural and haemal spines of vertebrae ossified always prior to the corresponding centra. When larvae reached to about 6.60mm in mean total length (17~18 days after hatching), jaw bones were more repidly ossified than vertebrae and cranium. Ossification of all bones nearly completed when the larvae reached to 13.40mm in mean total length (47~50 days after hatching).

  • Samples were obtained from broodstok in May, 2010, while naturally fertilized embryos were maintained, and the process of skeletal development was observed from larvae and juvenile. Prelarvae immediately after hatching showed an average total length of $6.85{\pm}0.63mm$ (n =5), Premaxillary and dentary were ossified, parasphenoid was ossified in the cranium, and centrum and caudal bone did not ossify. Prelarvae showed ossification with maxillary, articular, and epihyal, and branchiostegal rays of hyoid arch were ossified at 2 to 3 days after hatching with an average total length of $7.25{\pm}0.28mm$ (n=5). The vertebrae began to ossify in the direction of the tail, and neural spine began to ossify above the ossified vertebra. Postlarvae showed ossification of lateral ethmoid, alisphenoid, parietal, and caudal skeleton in the cranium when the average total length was $9.00{\pm}1.53mm$ (n=5) in 6 days after hatching. At 40 days after hatching, postlarvae ossified maxillary in the cranium, and ossified endopterygoid and ectopterygoid, etc. in the palate, when the average length of $23.3{\pm}0.28mm$ (n=5). At 120 days after hatching, with the average length was $37.5{\pm}2.83mm$ (n=5), caudal skeleton had one additional epural bone ossification, resulting in ossification of a total of 3 epural bone to complete ossification of all spicules.
