Seven Newly Recorded Species of the Genus Centistes Haliday (Hymenoptera: Braconidae: Euphorinae) from Korea (한국산 Centistes Haliday속 (벌목: 고치벌과: 잎벌레고치벌아과)에 대한 보고)

  • Seven species of the genus Centistes (Hymenoptera: Braconidae: Euphorinae) are reported for the first time from Korea: C. (Ancylocentrus) ater (Nees), C. (A.) mucri Belokobylskij, C. (A.) paupella (Shenefelt), C. (A.) rufus Chen and Achterberg, C. (Centistes) pumilio Belokobylskij, C. (C.) tsherskii Belokobylskij, and C. (Syrrhizus) delusorius (Förster). Diagnosis, distribution, host information and photographs are given for each species.

  • We report new host records of four ichneumonids from Korea. They were discovered from the pupae of Apatura metis Frever (Lepidoptera: Nymphalidae), Allotraeus sphaerioninus Bates (Coleoptera: Cerambycidae) and Ivela auripes (Butler) (Lepidoptera: Lymantriidae) respectively. Diagnoses, illustrations, host records of the four species, and a list of ichneumon species parasitizing of I. auripes are provided.

  • A study was carried out to investigate the morphological characteristics and life histories of H. morosa and H. diomPhalia during the period from 1984 to 1986. The head capsule width and bOdy length of H. Morosa larvae were slightly greater than those of H. diomPhalia. Otherwise, the two species looked very similar and were hard to be differentiated from each other in all life stages. H. morosa adults emerged in late June and laid eggs in soil during the period between mid July and mid August. Larvae developed by September to the last(3rd) instar which overwintered to pupate in late May. H. diomPhalia adults emerged from mid August but remained under the soil until next spring after which females laid eggs in soil from late May to early July. Larvae developed in most cases to the last instar by September and overwintered thereafter. Unlike H. morosa, they resumed feeding next April and began to pupate in late July. The egg periods of H. morosa and H. diomPhalia did not significantly differ but the larval and pupal periods of the latter were longer than those of the former.

Animal species identification by co-amplification of hypervariable region 1 (HV1) and cytochrome b in mitochondrial DNA

  • Mitochondrial DNA (mt DNA) sequence analysis has been a useful tool for species identification of animals and human individuals. Two hypervariable regions (HV1 and HV2) in control region of mitochondrial genome were analyzed for human individual identification. In case of animal species identification, several genes on mt DNA such as cytochrome b (cytb), RNAs, cytochrome oxidases (CO) were used. In this study, co-amplification of HV1 and cytb was carried out in order to check the contamination of animal DNA and to verify the human DNA. The primer sets used in PCR were H15997/L16236 for HV1 and H14724/L15149 for cytb. PCR products for HV1 and cytb were 239 bp and 425 bp, respectively. The appearance of two bands on agarose gel implied the DNA came from human, however the single band of cytb gene represented the non-human animal DNA.

Rearing and Innoculation Condition of Nd hybrid Silkworms for Paecilomyces tenuipes Production (Paecilomyces tenuipes 생산을 위한 나용교잡종 누에 사육 및 균 접종 조건)

  • To shorten the labor cost for the production of the fungus, Paecilomyces tenuipies, uncocooning mutant hybrid silkworms as a host was used. The yield of P. tenuipies was reduced when Nd (uncocooning mutant) was used as a host because Nd was smaller than the hybrid Baegokjam that has been used as a typical host. To increase the host weight, Nd was crossed with Jam l23 or Jam 124, and resulted in not only greater weighter but also higher yield of P. tenuipies. The optimum temperature for rearing Nd hybrid silkworms was $3^{\circ}C$ higher than standard temperature of rearing silkworms to increase the naked pupation rate. The optimum time for the infection of P. tenuipies was 12 h at $28.5^{\circ}C$ and 95% R.H., when $1{\times}10^7spores/ml$ of P. tenuipies were sprayed on the surface of the silkworm. The conditions were applicable to both spring-rearing and autumn-rearing.

  • Studies on Pathogenicity of Nosema bombycis Naegeli are summarized as follows: 1. The mortality of the parents, Jam 103 and Jam 104, is remarkbly higher than that of the hybrid, Jam 103$\times$Jam 104, whereas there is no difference in the mortality between the parents. 3. In the mortality of the pathogen-concentration, it is increased in order of the following concentrations inoculated, 10$^{8}$ , 10$^{7}$ , 10$^{8}$ and 10$^{5}$ /ml. 3. In the mortality of each instar, it is high in order of 5th, 4th, 3rd, and 2nd instar. 4. In the interaction between the mortalities of the varieties and the concentrations, 1) The mortality shows no differences between the parents and the hybrid in the high concentration of 10$^{8}$ /ml. 2) The mortality of the hybrid is lower than that of the parents in the low concentration of 10$^{5}$ /ml, whereas no difference is found between the parents. 3) The interaction appears at the same level in the middle concentration of 10$^{6}$ /ml to the parents and of 10$^{7}$ /ml to the hybrid. 5. It was pointed out that active immunity depends upon the volume of antigen injection, immunizing period, and injection intervals. In this experiment, it is noticed that the optimum volume of injection is above 20ml and D is the best one of the four treatment (A.B.C.D.). 6. The immune sera indicate such a 12,800 high titer in the indirect method can be obtained from the D immunizing method. Silkworm tissues and N. bombycis spores show self-fluorescence, but it is able to distinguish it from the F.I.T.C. by using the U.V. filter. 7. The midgut epithelium is examined to be the first site of the tissues which are penetrated into and multiplied by the inoculation of the pathogen per os.

Study on a pair of small spot of nonsexual gland hind of the larvae in Bombyx mori L. (I) (유충의 생식후원선이 아닌 한 쌍의 소반점에 관한 조사연구 (1))

  • External features of silkworms are not evident during stage of larvae and pupae like in the stage of imagines. In general, therefore, sexing of larvae has been depended on Herold's gland of males or Ishiwata' sexual spots (Anterior sexual spots and posterior sexual spots) of a female 2-3 days after of the 5th age since it is relatively easy to classify in this period. Sometimes. however, we made mistake of sexing since a pair of small spot around the abdomen of 9th abdominal of males and posterior sexual spot of females was found at the same position. This study has been conducted in order to obtain essential points of classifing the above it will be summarized as follows: A. Existence of the males small spots on 9th abdominal segment were varied by races. Through observation races by races the more larvae which had the small spots were found in chinese races than the Japanese, and 57 percent of the Chinese orgin larvae of the small spot were picked among total counted males. B. The small spots were more or less roundly shaped just as posterior sexual spots of females, and these spots were colored slightly even the colors were somewhat different by races. Milky color was seen in the case of the posterior sexual spots. The small spots were observed smaller than the posterior sexual spots. It is difficult classify the sexual spots during stage of mature larvae, but it was easily classified small spots by the naked eye even in the same stage. The small spots were found at the same site of the posterior sexual spots. C. After sexing by based on the small spots and Herold's gland, the sexing through purpae bodies were carried out again to confirm its correctness. Average 5. 2 percent of more opposite sex were found in the latter. This figures supposed to be caused the location of the small spots coincide with the posterior sexual spots. D. The moths, no disorder were found in daily activities and their generative functions depend on the existence of the small spot and thickness of its color. E. It is presumed that existence of the small spots were caused by races, heredity nature, voltinism, moulting, and etc.

Study on Spring Cocoon Crops with the Leaf Produced in the Mulberry Field close to the Totacco Field (개량 Mulching 담배밭 부근뽕잎이 춘잠작에 미치는 영향에 관한 연구)

  • The studies are to know how much cocoon crops is damaged by the stained leaf with nicotine produced from the tobacco field cultivated in mulching system in spring season and by residual nicotine in autumn season. Furthermore, the new knowledges are to make both industries keep up with their development. In spring season mulberry Held is located higher on the West-North of tobacco held below 20 degrees of slope and with 36 per cent of East-South wind and 18 per cent of South wind blowing from tobacco fold to the mulberry fold. In addition, silkworm larvae are fed with the mulberry leaf produced in the different plots placing by the different distances, l0m, 25m, 50m, 80m, and loom far from the tobacco Held as a control and it is also considered that narcotic larvae including the dead larvae are not observed. On the other hand, it is noted that better leaf quality and abundant growth of mulberry tree is produced from the mulberry fold closer to the tobacco field and with a low slope. 1) Maximum weight of larval body at the 5th stage is damaged by the stained leaf with the nicotine up to 25m far from the tobacco held. 2) The larvae fed with the mulberry leaf in mulberry Held up to 25m far from the tobacco fold produce small number of the fresh cocoons per 1 liter. 3) Low single cocoon weight and low cocoon shell weight are produced by the poison damaged larvae fed with the mulberry. leaf up to 25m far from the tobacco field and weight of cocoon shell is damaged higher than the single cocoon weight. It is resulted in low percentage of cocoon shell. 4) Cocoon yield including the double cocoon from 10,000 larvae is decreased by the larvae fed with the stained leaf in the mulberry fold up to 25m far from the tobacco fold and 19 per cent of cocoon yield is decreased with 2.4kg of cocoon yield in l0m plot and with 2.5kg of cocoon yield in 25m plot at the first season and at the 2nd season with 1.8kg o( cocoon yield in l0m plot and with 11.5kg of cocoon yield in 25m plot, 11 per cent and 9 per cent of cocoon yield including double cocoon from 10,000 larvae is decreased, as compared with the control, respectively. With these results, it is observed that nicotine damage is occurred to the silkworm larvae if the larvae are fed with the leaf in the mulberry Held within 25m-50m far from the tobacco field.
