A Study on the Effect of the Dendropanax Mobifera Extract on Anti-Hypertensive (황칠나무 추출물이 고혈압에 미치는 영향에 관한 연구)

  • This study examined the effects of an extract of Dendropanax morbifera on blood pressure, Angiotensin II, Angiotensin Converting Enzyme, Aldosterone, and lipid levels of spontaneously hypertensive rats. The groups were as follows: Control group, Hypertension control group, Water extract treated group, Ethanol extract treated group, n- hexane fraction treated group, Ethyl acetate fraction treated group, n- butanol fraction treated group, and Water fraction treated group. The blood pressure, and Angiotensin II, and Angiotensin Converting Enzyme, and Aldosterone levels were lower in the Ethyl acetate fraction treated group than in the hypertension control group. The change in blood pressure was lower in the Water extract treated group, Ethyl acetate fraction treated group, Ethanol extract treated group, n- hexane fraction treated group, and n- butanol fraction treated group than the hypertension control group. The concentration of Angiotensin II was lower in the Ethyl acetate fraction treated group, Ethanol extract treated group, n- hexane fraction treated group, and n- butanol fraction treated group than the hypertension control group(p<0.05). The level of Angiotensin Converting Enzyme was lower in the Ethyl acetate fraction treated group, Ethanol extract treated group, n- hexane fraction treated group, and n- butanol fraction treated group than the hypertension control group. The concentration of Aldosterone was lower in the Ethyl acetate fraction treated group, and n- butanol fraction treated group than the hypertension control group(p<0.05). In addition, the concentration lower in the Water extract treated group, Ethanol extract treated group, n- hexane fraction treated group than the hypertension control group. Overall, the effect of the Dendropanax morbifera extracts might be useful as anti-hypertensive, and functional food agents.

Effects of the Hizikia Mineral Salts on Blood Pressure and Mineral Metabolisms in Spontaneously Hypertensive Rats and Normotensive Rats (톳유래의 무기성분강화염이 본태성 고혈압쥐와 정상쥐의 혈압과 미네랄대사에 미치는 영향)

  • This study was conducted to evaluate the effects of the salts fortified with Hizikia component on blood pressure and mineral composition in spontaneously hypertensive rats (SHR/NCrj) and normotensive rats (WKY/NCrj). The two species rats were assigned to three groups and were fed with drinking water to which Hizikia salts were added for 6 weeks. The final blood pressures (BP) among groups of SHR were increased to 12, 4, and 21%, respectively. In terms of urine and fecal minerals, the $Na^+$ contents in two species were significantly higher in the control than in the salt groups, and vice versa with regard to the $ K^+$ contents. The amount of excretion mineral was higher in urine than in feces. The $Na^+$, $Ca^{++}$ and $Mg^{++}$ contents of the liver in SHR were higher in the control than in the salt groups (p<0.05). The $Na^+$ content of the kidney in SHR was higher in the control than in the salt groups, but the content was shown an opposite trend in WKY. The $K^+$ contents were higher in the low salt group (p<0.05). These results may indicate that the salts fortified with the Hizikia component may be useful in lowering systolic blood pressure owing to the maintenance of positive mineral metabolism.

Effect of Sodium Intake on Responses of Blood Pressure, Renin-Aldosterone and Renal Excretions to Atrial Natriuretic Peptide in Spontaneously Hypertensive Rats (소금 섭취량을 달리한 정상 및 고혈압쥐에서 Atrial Natriuretic Peptide가 혈압, Renin-Aldosterone 및 신배설에 미치는 영향)

  • Effects of atrial natriuretic peptide (ANP) on blood pressure, plasma lenin activity, aldosterone and renal excretion were compared in conscious spontaneously hypertensive rats (SHR) and normotensive Wistar rats fed low, medium or high sodium diet (2, 10, 25 mmol NaCl/100g diet) for 6 weeks. ANP infusion (380 ng/kg/min for 20 min) produced reductions in blood pressure, plasma renin activity, and aldosterone level, but marked increases in hematocrit, urine flow, and excretions of sodium and potassium. The low sodium group showed a significantly enhanced aldosterone lowering effect of ANP than the high sodium group. However, three salt groups showed no difference in effects of ANP on blood pressure, plasma renin activity, hematocrit and diuresis. Natriuretic response to ANP was significantly greater in the high salt-than in the low sait-SHR, but was not different between the Wistar salt groups. There were strain differences in effects of ANP: SHR showed greater responses of blood pressure and natriuresis than Wistar rats. Above results indicate that aldosterone-lowering and natriuretic effects of ANP were modifed by different dietary sodium intakes. However, blood pressure- and renin-lowering, or diuretic effects of ANP were not affected by dietary sodium intakes. The mechanisms whereby dietary sodium intakes alter the effects of ANP in the pathogenesis of hypertension are not clear.

Antihypertensive Activity of Korean Medicinal Plants Against Okamoto-SHR (I) (오카모토 고혈압쥐에 대한 한국산생약의 항고혈압작용 (I))

  • Six medicinal plants described in Dong-Ee-Bo-Gam as to be useful for hypertension treatment were evaluated their antihypertensive activity against Okamoto-SHR, an animal model of spontaneous hypertension. Ethanol extracts of plant samples were prepared and were p.o. administered once daily for 20 days. Average blood pressures of rats were measured during two weeks periods before drug administration. Then blood pressure of each rat was measured every three days during drug administration period. As a positive control, propranolol, ${\beta}-blocker$ was used and the blood pressures of test group rats were compared with those of negative and positive control group rats. Among 6 plant extracts, Acanthopanax sessiliflorum and Atractylodes japonica (alba) extracts appeared to exhibit antihypertensive activity.

Effect of Dietary Ca and Na Levels on Blood Pressure and Mineral Metabolism in Spontaneously Hypertensive Rats Fed High Fat Diet (고지방식을 섭취한 본태성 고혈압쥐에서 칼슘과 소디움 섭취가 혈압과 무기질 대사에 미치는 영향)

  • This study explored the effect of dietary levels of Na and Ca on spontaneously hypertensive rats (SHR). SHR were randomly divided into 5 groups and fed a high fat/cholesterol diet containing three levels of Na (0.05, 0.1, 1.5%) and Ca (0.1, 0.5, 1.5%) for 9 weeks. Body weight gain was not influenced by dietary intake but water intake significantly increased in high Na supplementation. Systolic blood pressure was not influenced by dietary Na and Ca levels but was decreased by dietary low Na/high Ca levels at 9 weeks. Angiotensin-II level was affected by dietary Na level but not by Ca levels. Plasma Ca, Mg, K and Na levels were in the normal range regardless of dietary Na and Ca levels. Weight, and K and Na contents of the heart and kidney were not significantly different among those with different dietary Na and Ca levels. Ca and Mg contents of the heart and kidney were significantly higher in the normal Na/normal Ca group. Ca and Mg in the feces were higher in those with high Ca intake. Na in the feces was higher in those with high Na intake. Therefore, Na and Ca had different mechanisms in the hypertension/hyperlipidemia models, respectively. And we suggested that Mg must be supplemented when Ca intake was high because Mg excretion was increased by Ca supplementation.

Brown Rice Phytosterol Improves Hypertension and Lipid Metabolism in Spontaneously Hypertensive Rats (본태성 고혈압쥐(SHR)에서 현미 식물성스테롤의 혈압 및 지질 대사 개선 효과)

  • This study was conducted to investigate the effects of brown rice and brown rice phytosterol on blood pressure and lipid levels in spontaneously hypertension rats (SHR). SHR were grouped according to blood pressure and fed either a control diet or experimental diets containing 50% brown rice powder or 5% brown rice phytosterol for 3 weeks. Body weight gain and epididymal fat weight were significantly reduced in the brown rice powder and brown rice phytosterol groups compared to control. Brown rice and brown rice phytosterol diets suppressed age-dependent increases in systolic blood pressure compared to control. In addition, brown rice and brown rice phytosterol diets decreased total lipid, triglyceride, and cholesterol levels in the liver, whereas serum HDL cholesterol increased. Lastly, brown rice phytosterol reduced TBARS contents in the kidney. These results suggest that brown rice and brown rice phytosterol exert antihypertensive effects that improve lipid metabolism in SHR.

중등 정도 고혈압인 사람에서, 정어리 근육 가수분해물로부터 유도된 단쇄 펩타이드인 Valyl-Tyrosine의 항고혈압 효과

  • 본 연구는 선천선고혈압쥐(SHR)에서와 마찬가지로, 높은 정상혈압 및 중등 정도의 본태성고혈압 환자에서 Valyl-Tyrosine(VY)의 항고혈압 효과 여부를 결정하기 위해 수행되었다. 29명의 지원자에 대하여 무작위 이중맹검 위약검사를 실시하였으며, 3 mg의 VY를 포함하는 100-ml 음료와 100ml-위약 음료를 조제하여 사용하였다. 연구대상은 VY (남16/여1, 45.5${\pm}$3.2세, 146.4${\pm}$2.3/90.5${\pm}$1.8 mmHg) 집단과 위약(P) (남11/여1, 48.8${\pm}$3.0 세, 145.5${\pm}$2.4/92.3${\pm}$1.8 mmHg) 집단으로 나누었다. 대조(C)기간 3주 째에 VY-음료 또는 P-음료를 하루 2번씩 투여하기 시작하여 4주 간의 실험(E)기간(BP)은 매주 측정되었으며, 아침에 앉은 자세에서 측정하였다. 혈액 표본은 C기간과 E기간의 마지막 날 채취하였다. VY집단에서, 수축기(S) 및 이완기(D) 혈압의 감소는 첫째 주에 각각 9.7 및 5.3 mmHg (P < 0.001)이었으며, E기간의 시작에 따른 넷째 주에는 각각 9.3 및 5.2 mmHg (P < 0.001)이었다. P 집단에서는 SBP와 DBP 모두 변하지 않았다. VY 집단의 혈압은 회복기간의 끝까지 점진적으로 증가하였다. VY집단에서, 혈장 안지오텐신(Ang) I과 VY의 농도는 현저히 증가한 반면 Ang II와 알도스테론은 VY 투여 후에 현저히 감소하였다. VY는 SHR에서와 마찬가지로 중등 정도의 고혈압 환자에서도 Ang I-전환효소 억제를 통하여 현저한 항고혈압 효과를 가지는 것으로 보이며, 어떤 부작용도 검출되지 않았다.

  • As a folkloric medicine, Siegesbeckia pubescens has been used for treatment of brain stroke and hypertension. Diterpene compound, 16,17-dihydroxy 16-${\beta}-(-)$ kaurane-19-oic acid, was isolated from the plant. Its potential antihypertensive activity was evaluated againstan animal model of hypertension, Okamoto-SHR. When diterpene compound with dose of 50mg/kg/day was orally administered, it exhibited mild antihypertensive activity comparable with propranolol administration dose of 75mg/kg/day.

Effects of Gastrodia Rhizoma Extracts on Global Coronary Circulation in Rats (천마 Extracts가 백서의 국소적 관상순환기능에 미치는 영향)

  • Gastrodia (G) Rhizoma has been used clinically as an oriental herbal medicine with sedative, anticonvulsive, and depressor effects. The present study tested effects of G. Rhizoma extracts on the coronary circulation and myocardial oxygen consumption in perfused rat hearts. Sprague Dawley rats (SD) and spontaneously hypertensive rats (SHR) were employed as experimental animals and nonworking Langendorff heart perfusion technique introduced for heart experiments. G. Rhizoma extracts were prepared from grinding G. Rhizoma into powder, extracting in water and 50% ethanol for 4 or 16 hr and diluting with Krebs-Henseleit bicarbonate perfusion buffer to be 70%. Hearts were perfused with bicarbonate buffer oxygenated with 95% $O_{2}:$ 5% $CO_{2}$ at constant coronary perfusion pressure of $90cmH_{2}O$. The diluted extracts were infused into coronary arteries in a concentration of $1{\sim}5\;{\mu}M$ for $7{\sim}8 min. While in SD water- or ethanol-extracts of G. Rhizoma extracted for 16 hr increased coronary perfusate flow (CPF) and decreased coronary vascular resistance (CVR), ethanol-extracts in SHR produced coronary vasoconstriction associated with enhanced CVR. G. Rhizoma extracts-induced increase in CPF reduced myocardial oxygen extraction, and thus myocardial oxygen consumption ($MVO_{2}$) remained at that observed prior to infusion of extracts. In SD and SHR 16 hr-water-extracts markedly altered coronary venous effluent pH and $Pco_{2}$ and evoked metabolic acidosis, which could be a coronary vasodilator mechanism decreasing CVR. In this study, the extracts decreasing CVR in SD and SHR did not augment the lactate production. Therefore, although the effects of the extracts on cardiac function and coronary circulation depended on solvents and duration for extraction, the 16hr-water-extracts, at least, exhibited coronary vasodilation in SD and SHR. Conversely, ethanol-extracts constricted coronary arteries in SHR. G. Rhizoma extracts-induced vasodilation might be due to the metabolic acidosis rather than due to the increased lactate production. The results indicate that G. Rhizoma extracts obtained from proper extracting procedures can be used as a safe and clinically applicable herbal medicine in the cardiovascular diseases such as coronary artery disease and hypertension for vasodilatory and antihypertensive actions.

Effect of Heat Treatment of Garlic Added Diet on the Blood of Spontaneously Hypertension Rat (처리법을 달리한 마늘 첨가식이가 자발성 고혈압쥐의 혈액에 미치는 영향)

  • It has been known that garlic, one of the essential ingredients of spices in Korean food, has a hypotensive effect. The following experiments were done to compare the effect of heat treatment of garlic on change in blood pressure. We selected SHR(Spontaneously Hypertension Rat) for experimental animals since, in the case of human beings, 85~90% of high blood pressure is in hereditary origin. Animals were divided into 3 groups, control group(no garlic), 3% raw garlic group and 3% heated garlic group. Serum was analyzed for lipid concentration, and plasma for prothrombin time and fi-brinogen concentration. The effects of heat treatment of garlic were as follow. There was no significant differences in body weight gain and feed efficiency ratio except that feed intake of 3% heated garlic-fed group was significantly lower than that of control group and 3% raw garlic-fed group. Garlic-fed groups, in contrast to the control group, showed significant difference in cholesterol content in pro-thrombin time and in fibrinogen concentration. Taken together, hypotensive effects of garlic on high blood pressure were significant. Regardless of heat treatment both heated garlic and raw garlic showed hypotensive effects.
