Effects of Aerobic Exercise on Body Weight Decrease (운동에 대한 체중감량의 효과)

  • 이 연구의 목적은 운동이 고혈압쥐의 체중감량에 어떠한 효과가 있는가를 밝히고자 함이다. 실험동물은 수컷 고혈압쥐로 일정기간 실험실 적응 후 실험군과 대조군으로 분류하였다. 이후 실험기간동안 체중의 변화를 관찰하였다. 실험결과 집단간 체중의 차이는 유의하지 않았고(p>.05), 집단간과 집단내의 시간에 따른 상호작용은 유의 (p<.05)하게 나타났다. 결론적으로 8주간의 유산소운동은 고혈압쥐의 체중을 감량시키는 효과가 있다.

  • 이미 발표된, 혈압에 미치는 인삼의 효과에 관한 연구들의 보고는 서로 상반된 견해들이 많이 있다. 인삼에는 혈압상승작용과 혈압을 내려주는 효과가 모두 있다고 보고하였다. 본 저자들은 고혈압 쥐에 미치는 인삼의 급${\cdot}$만성효과를 연구하기 위해 일련의 2가지 실험을 하였으며, 실험재료로 사용한 홍삼은 일한홍삼주식회사에서 구입하였다. (실험 1) 홍삼분말을 일반 쥐사료에 혼합하여 (5g/kg), 임의로 사료를 섭취하게 하여 11주간 쥐를 사육하였으며, 하루에 섭취한 인삼의 양은 체중 kg당 $250{\~}750$mg이었다. 홍삼분말의 투여는 DON쥐, SHR쥐, 그리고 한개의 신장을 협자시킨 고혈압 쥐의 경우 혈압에 영향을 미치지 않았다. 홍삼분말의 투여는 DOC 쥐에 있어 혈압을 약간 상승시켰다. (실험 2) 홍삼분말을 체중 kg당 350mg씩 경구투여한 후 복대동맥에 카뉼제를 삽입하여 혈압을 24시간 직접 측정하였다. 그 결과 DOC쥐와 SHRSP쥐에 있어 혈압에 영향이 없었다. 이 실험에서는 마취를 시키지 않았으며, 자유로이 풀어 놓았다. 실험 1과 같이 매일 체중 kg당 $300{\~}500$mg씩 11주간 홍삼분말을 만성적으로 투여하였을 때, SHRSP쥐의 혈압에 영향이 없었다. DOC 고혈압 쥐의 경우 약간의 혈압 강하가 있었으며, DOC 고혈압의 정도는 (실험 2)에서 약간 낮았다.

Differential Function of EDRF in Systemic Arterial and Pulmonary Arterial System of Renal Hypertensive Rats (신성 고혈압쥐의 전신성 동맥계와 폐동맥계에 대한 EDRF 기능의 차이)

  • To investigate the endothelium dependent vascular reactivity of the systemic arterial and the pulmonary arterial system in acute renal hypertensive rats of 2-kidney, 1-ligation type (RHRs), acetylcholine (ACh)-induced vasodilation and depressor effects were evaluated in isolated arteries and in vivo, respectively, in the presence and absence of functional endothelium. ACh $(10^{-5}\;M)$ relaxed the intact thoracic aortas from RHRs and normotensive rats (NRs), but the effect was significantly smaller for those from RHRs (34 and 86%, respectively, p<0.01). ACh-induced vasodilation was completely abolished after removal of endothelial cell or pretreatment with EDRF inhibitors, L-NAME and MB, indicative of its dependence on intact endothelial or EDRF function. ACh also induced vasorelaxation of the intact pulmonary arteries from RHRs and NRs; however, unlike the effects on the thorcic aorta, no significant difference in amplitude was noted between two groups. ACh $(0.1{\sim}10\;{\mu}g/kg,\;i.v.)$ reduced mean systemic arterial pressure in anesthetized RHRs and in NRs to the similar magnitude (% change: 39 and 46% at $10\;{\mu}g/kg$, respectively) and these hypotensive effects were significantly decreased after pretreatment with L-NAME (30 mg/kg, i.v.). Deprssor effects of ACh on mean pulmonary arterial pressure were similar in RHRs and NRs with and without pretreatment of L-NAME. However, in both NRs and RHRs, the depressor effects of ACh on mean pulmonary arterial pressure were significantly reduced compared with those for mean systemic arterial pressure, and the increment of mean pulmonary arterial pressure noted after L-NAME $(0.1{\mu}100\;mg/kg,\;i.v.)$ was significantly smaller than that for mean systemic arterial pressure. These results indicate that in RHRs the endothelial cell function was impaired, at least in part, in systemic arterial system, but not in pulmonary arterial system, and both ACh-evoked and basal release of EDRF was less in the pulmonary arterial system than in systemic arterial system of both NRs and RHRs.

Effects of Germinated-Buckwheat on Blood Pressure , Plasma Glucose and Lipid levels of Spontaneously Hypertensive Rats (발아메밀이 본태성 고혈압쥐의 혈압, 혈당 및 혈중 지질수준에 미치는 영향)

  • The purpose of this study was to investigate the effect of germinated buckwheat on blood pressure, plasma glucose and lipid levels of spontaneously hypertensive rats(SHR). The male and female SHR were divided into two groups. After feeding the germinated buckwheat diet(50%, w/w) ad libitum for 6 weeks, the weight gain of the germinated buckwheat group in male rats was higher than those of control group. Systolic blood pressure of the germinated buckwheat group in male rats was significantly decreased at 6 weeks by 3%. The fasting glucose levels of the germinated buckwheat group had a tendency to be lower compared with those of the control group. Total cholesterol, HDL-cholesterol and HTR of female SHR fed the germinated buckwheat diet were significantly increased compared to the control diet. Atherogenic index of the germinated buckwheat group was decreased in males(ns) and females(p<0.05) than those of the control group. From these results the germinated buckwheat might be useful for anti-hypertensive and anti-hyperlipidemic agents.

Na, K-ATPase Activity in the Aged Erythrocytes of Hypertensive Rats (고혈압쥐 노화 적혈구에서의 Na, K-ATPase에 관한 연구)

  • To study the age dependent change of Na, K-ATPase in the erythrocyte of hypertensive rat, 1-kidey 1-clip hypertensive rat was made by the removal of right kidney and partial ligation of left renal artery. After 4 weeks, aged erythrocyte fraction was separated by density gradient centrifugation, and Na, K-ATPase activity and $^3H-ouabain$ binding with ghost cell membrane and ouabain sensitive Rb-uptake with whole cell were measured. 1) In the hypertensive rats, blood pressure was significantly increased to 165.5/119.0 mmHg (systolic/diastolic). Mean corpuscular volume and membrane protein(mg) per $10^9RBC$ were decreased and hemoglobin content was increased in the aged erythrocyte. 2) Na, K-ATPase activity in the solution containing 110 mM NaCl and 10 mM KCI, was decreased in hypertensive rat, and decreased in aged erythrocyte of both group. 3) Ouabain sensitive Rb-uptake by low RbCl concentration(4 mM) was slightly decreased in aged erythrocyte compared to that in young erythrocyte of each group, but slightly increased in young erythrocyte in hypertensive rat compared to that in normotensive rat. 4) Ouabain sensitive Rb-uptake by high RbCl concentration(16 mM) was decreased about 30% to 50 % in aged erythrocyte in both group. And in hypertensive rat, especially in young erythrocyte it was significantly decreased compared to that in normotensive rats. 5) $^3H-ouabain$ binding at 0.13 or $1{\times}10^-6M$ ouabain concentration was slightly decreased in aged erythrocyte of normotensive rat, and significantly decreased in aged erythrocyte of hypertensive rats. 6) $^3H-ouabain$ binding at 6 or $64{\times}10^-6M$ ouabain concentration is slightly decreased in aged erythrocyte of both group, but significantly decreased in young and aged erythrocyte of hypertensive rats compared to that of normotensive rats. The present results suggest that (1) in the young erythrocyte of hypertensive rat, the alterations of Na-pump activity that slightly increased in weak stimulation and inhibited in strong stimulation, may be related to increased molecular activity and the decrease in the number of low affinity site without change in high affinity site, (2) in the aged erythrocyte of normotensive rat, inhibited Na-pump may be related to the change in molecular activity of pump. (3) And in the aged erythrocyte of hypertensive rat, it may be related to the decrease in the number of high and low affinity site as well as the change in molecular activity

Hypotensive Effect of Germinated Brown Rice on Spontaneously Hypertensive Rats (발아현미의 본태성 고혈압쥐에 대한 혈압강하 효과)

  • To investigate the hypotensive effect of germinated brown rice, spontaneously hypertensive rats were randomly divided into 4 groups and fed with experimental diets for 6 weeks; control group fed with standard diet, rice group fed with diet containing 50% rice, brown rice group fed with diet containing 50% brown rice and germinated brown rice group fed with diet containing 50% germinated brown rice. Body weight gain and FER (food efficiency ratio) of germinated brown rice group, $86.3\;{\pm}\;11.1\;g$ and $10.4\;{\pm}\;1.4%$, were significantly lower than those of other groups. Systolic blood pressure of germinated brown rice group after feeding for 6 weeks was $169.2\;{\pm}\;6.2\;mmHg$, which was significantly lower than those of other groups and decreased largely compared to that of beginning stage. And blood triglyceride of germinated brown rice group, $138.4\;{\pm}\;29.5\;mg/dL$, was significantly lower than those of other groups, but all groups didn't s show significantly difference in total and HDL-cholesterol of blood. These results suggested that germ mated brown rice had hypotensive effect on spontaneously hypotensive rats.

Effect of hypertention falling of functional GABA green tea (기능성 GABA차의 고혈압 강하효과)

  • GABA tea which was increased of GABA by anaerobic conditions in green tea leaves) was fed in the diet to spontaneously hypertensive rats(SHR). The mean systolic blood pressure of SHR fed a diet containing 1.5% GABA tea after 3 weeks was not different with zero day as 177.3mmHg. However, 1.5% green tea and control after 3 weeks were more increased 4.2 percents and 10 percents than in zero day. The plasma levels of aldosterone of SHR fed a diat containing 1.5% GABA green tea after 3 weeks was more decreased 2.5 times than in water treatment.

Hypoxia Induced Expression of Vascular Endothelial Growth Factor in Rat Pulmonary Artery Smooth Muscle Cells (쥐의 폐동맥 평활근 세포에서 저산소에 의한 Vascular Endothelial Growth Factor의 발현)

  • Purpose : Pulmonary vascular hypertension is a common problem in congenital heart disease, the most common cardiac condition in childhood. However, the mechanisms responsible for this pathologic change, treatment, and prevention are poorly understood. Therefore, we studied the gene expression of vascular endothelial growth factor(VEGF) by using a hypoxic model of the pulmonary artery smooth muscle cells. Methods : The main pulmonary artery and its proximal branches of a 6 wk old Fischer rat were excised. They were cut into multiple small pieces and suspended in DMEM medium supplemented with 20% fetal bovine serum and incubated in 5% $CO_2$-95% air atmosphere. The smooth muscle cells were confirmed by immunostaining with smooth muscle myosin and ${\alpha}$-smooth muscle actin antibodies. The VEGF gene expression in the hypoxic group was compared with the one in control the group as well as the one in the starved group by RT-PCR and Northern blot hybridization. Results : There was no statistically significant difference among the control, hypoxic and starved groups. Conclusion : There are few studies of pulmonary vascular hypertension at the molecular level in Korea. Therefore, we studied the expression of VEGF gene in hypoxic pulmonary vascular smooth muscle cells. Further studies will be needed to find the difference between newly born and adult rats, or human and rat pulmonary vascular smooth muscle cells in gene expression. We hope that the study will lead to a better understanding of pulmonary vascular hypertension.

The Effects of Exercise Training on Cardiac eNOS, ET-1 mRNA and Skeletal Muscle eNOS Protein Level in SHR (지구성 운동이 본태성 고혈압 쥐 심장근의 eNOS, ET-1 mRNA와 골격근 eNOS 단백질 발현에 미치는 영향)

  • In the present study, all of the treadmill exercise-trained SHR expressed clear adaptive changes such as reduced resting heart rate and blood pressures, LPOA, homocysteine Therefore, treadmill exercise was sufficient to induce physiological adaptation in the SHR. Endurance training is known to induce physiological cardiac hypertrophy, while hypertension induces patho logical cardiac hypertrophy that increases cardiomyocyte apoptosis. The pathological adaptation to pressure overload has also been associated with a further increase in the expression of several marker genes including cardiomyocyte ET-1 in the SHR, but not in the exercise-trained SHR. Additionally, there is an increase in the endothelial nitricoxide synthases (eNOS) protein expression of soleus, gastrocnemius, and extensor digitorum longus muscle in the exercise-trained SHR but not in the SHR in the present study. Thus, compared to pathological adaptation to pressure overload, physiological adaptation to exercise training is associated with distinct alterations in cardiac and molecular phenotypes. based on these results, exercise training improves hypertension by cardiovascular regulating genes and hemodynamic parameters.
