Alginate-Titanium hydroxide에 의한 L-Lysine 생산 융합균주의 고정화 및 연속생산에의 적용

  • L-Lysine 생성균인 Corynebacterium glutamicum의 동종간 융합주 RS 99를 담체인 Alginate와 여기에 TiCl$_4$로부터 제조된 Titanium hydroxide를 혼합하여 각각 고정하고 이들의 Gel strength, 활성도 및 회분식 발효조건을 비교하였다. 그 결과 Alginate-Titanium hydroxide를 담체로 선정하여 고정화 C, glutamicum 융합주의 재사용성 및 안정성을 검토하였으며, Continuous-Flow Stirred-Tank Reactor를 구성하여 L-Lysine 의 연속발효를 시도하였다.

Development of Magnetically Separable Immobilized Trypsin (자석에 의해 분리가 가능한 고정화 Trypsin 개발)

  • Magnetically separable immobilized trypsin was developed and their biocatalytic activity was evaluated for the different immobilization media. The activity, recyclability, pH effect, and stability of immobilized enzymes were evaluated for the different supporting media. The biocatalytic activity of immobilized trypsin was highest with magnetically separable polyaniline (PAMP), and Vm and Km of PAMP were 0.169 mM/min and 0.263 mM respectively. With increasedpH, the biocatalytic activity increased for all supporting materials used. Immobilized enzymes were recycled and recycle activities were over 90% of their original activity after ten times reuse. The operational stabilities of enzymes were greatly improved with enzyme immobilization.

Production of 5균-GMP by Immobilized 5균-GMP Producing Fusant RC102 (5균-GMP 생산 융합균주 RC102의 고정화에 의한 5균-GMP 생산)

  • The effective production of 5'-GMP(5'-Guanylic acid) by immobilized 5'-GMP producing fusant RC102(intergeneric protoplast fusion between Brevibacterium ammoniagenes ATCC21263 and Corynebacterium glutamicum ATCC21171) was investigated. The Fusant RC102 was immobilized by entrapping in -carrageenan, agar, polyacrylamide or Ca-alginate. 3% k-carrageenan was selected as the most suitable matrix. In the production of 5'-GMP using the immobilized whole cells of fusant RC102, the optimum conditions were $32^{\circ}C$, pH 8.0, $30\mu\textrm{g}/L\;of\;Mn^{2+},\;1{\times}10^{-6}%\;of\;Zn^{2+}$. In order to use fermentation medium containing CSL(Corn Steep Liquor) plentiful in $Mn^{2+}$, the optimum conditions of penicillin G, D-cycloserine and POESA(polyoxyethylene stearylamine) for production of 5'-GMP were 0.8unit/ml, 0.8unit/ml, 0.8unit/ml and 5mg/ml, respectively. Cationic surfactant, POESA was effective and superior to the antibiotics, penicillin G or D-cyloserine in 5'-GMP productivity. The condinuous fermentation using immobilized fusant RC102 showed that 5'-GMP productivity was stable for more than 15 days.

Immobilization of $\beta$-glucosidase and properties of Immobilized Enzyme ($\beta$-glucosidase의 고정화와 효소 반응특성)

  • $\beta$-glucosidase derived from Aspergillus niger was immobilized by (1) covalent linkage on chitin and chitosan with glutaraldehyde, (2) adsorption on DEAE-cellulose and Amberite IRA93 after succinylation, and (3) entrapment on alginate and polyacrylamide gels with various cross linking agents. The retention yield of $\beta$-glucosidase immobilized on chitosan was 31.5% and operational stability was 69% after continuous operation at column reactor(5$0^{\circ}C$ at pH 4.8) for 15 days. The retention yield and operational stability were 24.7% and 60% respectively, in adsorption on Amberite IRA 93. On the other hand, the entrapment method by alginate and polyacrylamide gel was identified to be not appropriate due to the continuous elution of inlmobilized $\beta$-glucosidase. Optimum conditions for the immobilization on chitosan were also studied with optimum pH of 4.8 and glutaraldehyde concentration of 0.4%(w/v). The properties and stability of immobilized $\beta$-glucosidase are also investigted. The conversion yield of cellobiose to glucose was also analyzed using the column type enzyme reactor to evaluate the effectiveness of immobilized enzyme.

The Removal of Styrene using Immobilized Microorganisms in Hydrogel Beads (미생물 고정화 복합고분자담체를 이용한 Styrene 제거)

  • As an alternative for the traditional materials packed in biofilters treating gaseous VOCs, a novel packing material has been developed and tested. In the packing material(named as Hydrogel Bead, HB), pollutant-degrading microorganisms were immobilized in hydrogel consisted of alginate, polyvinyl alcohol(PVA), and powdered activated carbon. A closed-bottle study showed that the HB rapidly removed gaseous styrene without the losses of adsorption and biodegradation capacity. Biofilter column experiments using the HBs also demonstrated that greater than 95% of removal efficiencies were found at an inlet styrene loading rate of $245g/m^3/hr$, which was higher biofilter performance than other elimination capacity reported earlier. Furthermore, when the inlet styrene concentration increased stepwise, the adsorption played an important role in overall styrene removals. The absorbed styrene was found to be biodegraded in the following low inlet loading condition. Consequently, the new HB material is able to successfully minimize the drawbacks of activated carbon(necessity of regeneration) and biological processes(low removal capacity at dynamic loading conditions), and maximize the overall performance of biofilter systems treating VOCs.

The Removal of Organics, Nitrogen and Phosphorus in Loop Reactor Using Fluidized Media (유동상 담체를 이용한 Loop Reactor에서 유기물 및 질소, 인 제거)

  • This study was accomplished using Anaerobic/Anoxic/Oxic upflow packed-bed column reactors with fixed media and Loop Reactor with fluidized media instead of Oxic reactor. The objectives of this study was to investigate the characteristics of organics, nitrogen and phosphorus removal from sewage with the HRT. The average removal efficiencies of $BOD_5$ and SS increase as increasing the hydraulic retention time (HRT) until 16 h of the HRT, and they were constant over 16 h of the HRT. The removal efficiency of $BOD_5$ in case of packed-bed reactor and Loop Reactor was about 86.6% and 90.9% respectively at 16 h of the HRT. The removal efficiency of SS in packed-bed reactor and Loop Reactor was about 78.0% and 88.2% respectively at 16 h of the HRT. The average removal efficiencies of $COD_{Cr}$ and $COD_{Mn}$ showed similar trends as those of $BOD_5$ and SS. At the HRT of 16 h, the removal efficiency of $COD_{Cr}$ in case of packed-bed reactor and Loop Reactor was 63.5%, 75.2% and that of $COD_{Mn}$ was 60.7%, 73.6% respectively. The average removal efficiencies of T-N and T-P increase as increasing the HRT. The removal efficiencies of T-N and T-P in Loop Reactor were 33.6% and 54.5% respectively at 16 h of the HRT and T-N and T-P were better removed in Loop Reactor. From this result, it was found that the performance of Loop Reactor was much higher than the performance of packed-bed reactor and the optimum HRT was 16 h.

Penicillin Fermentation using a Carrier-supported Mycelial Growth (담체에 고정화된 균사체 증식을 이용한 페니실린의 발효)

  • A carrier-supported mycelial growth of Penicillium chrysogenum was applied to penicillin fermentation system. Among various materials tested, celite was found to be most effective for both spore adsorption and bioparticle development. Hyphal growth through pore matrices of the material showed strong anchorages and provided highly stable biofilm growths. When 5-10% celite was employed, both cell growth and penicillin production were observed to increase significantly comparing to the dispersed filamentous growth. Specific productivity of penicillin, however. was found to be kept almost constant at a value of 1,900 unit/g cell/hr. A semicontinuous fermentation in a fluidized-bed reactor. using the tarrier-supported biofilm growth, was conducted successfully although free mycelia appeared in the late phase of the fermentation made the reactor operation difficult. Control of the size of bioparticles was considered as a major operating factor to maintain the reactor productivity at a desired level.

Characteristics of Wastewater Treatment by the Continuous-Flow Fixed Biofilm Process Using Porous Fly Ash Carrier (다공성 석탄회 담체를 이용한 연속류식 고정 생물막 공정의 폐수 처리 특성)

  • We have prepared the porous carrier that has high capacity for immobilization of microorganisms and adsorption capacity for cation using fly ash in the previous work. In this study, we investigated the characteristics of piggery wastewater treatment in comparison with commercial carrier and conventional activated sludge process by continuous-flow fixed biofilm process in laboratory scale at the same experimental conditions to develop the biofilm process using porous fly ash carrier for wastewater treatment. As a result, the prepared fly ash carrier showed that removal efficiency of COD, TN and $NH_4{^+}-N$ items were 80%, 77% and 65%, respectively, which were higher efficiency than the commercial carrier and conventional activated sludge process. And the result of measurement for immobilized microorganisms after treatment showed higher capacity than the commercial carrier, and it was confirmed by SEM observation on fly ash carrier that the colony of microorganisms was stably formed.

The Effect of Fixed Media and Recycling Ratio on Nutrients Removal in a Pilot-Scale Wastewater Treatment Unit (고정식 담체 유무와 반송비에 따른 소규모 하수처리 시스템 내 영양염류 제거 특성)

  • The effect of recycling ratio and fixed media on nitrate and phosphate removal was investigated in a pilot-scale wastewater treatment unit using synthetic wastewater. Addition of fixed media increased nitrate removal from 45 to 58% while no noticeable change was observed for Chemical Oxygen Demand (COD) and phosphate removal (<5%). Nitrate removal efficiency also enhanced (Ca 7%) when the influent wastewater flow was doubled (2Q), however phosphate removal was decreased from 40.9 to 26.6% with the increasing recycling rate. The attached biomass analysis showed the presence of bacteria (73.4 $mg/cm^2$) on the surface of added media in anoxic reactor. Pseudomonas aeruginosa a common denitrifying bacterium dominated the bacterial growth (58%) in the anoxic reactor which was determined using Fluorescence In Situ Hybridization (FISH) analysis.

Inorganic Phosphate Solubilization by Immobilized Pantoea agglomerans under in vitro Conditions (고정화된 Pantoea agglomerans에 의한 난용성 인산의 가용화)

  • It is now widely accepted that immobilized microbial cells can overcome some of the problems associated with microbial survival stability, efficacy, storage, transportation and ease of application in agricultural environments. Pantoea agglomerans, a phosphate solubilizing bacterium, was immobilized in alginate, agar and gelatin carriers. All the three immobilfized carriers with bacterial cells of P. agglomerans were compared for solubilization of tricalcium phosphate in pure liquid cultures. While alginate beads were tested for phosphate solubilization on alternate days up to five days, agar beads and gelatin cubes were subjected for one time phosphate solubilization analysis after seven days. Both alginate and agar immobilized cells of P. agglomerans exhibited higher efficiency in increasing the solubilizaliun of tricalcium phosphate than gelatin immobilized cells. The culture filtrate of alginate bead inoculation treatment registered a rapid increase in soluble phosphate concentration upon incubation. A corresponding decrease in the pH of the medium was also observed in all the treatments.
