Vasorelaxing Mechanism of Crude Saponin of Korea Red Ginseng in the Resistance-sized Mesenteric Artery of Rat

  • It has been well known that Korea red ginseng has an antihypertensive effect. The antihypertensive effect may be due to its ability to change the peripheral resistance. Change of vascular tone in the resistance-sized artery contribute to the peripheral resistance, thereby regulate the blood pressure. Therefore, we investigated to clarify the vasorelaxing mechanism induced by crude saponin of Korea red ginseng in the resistance-sized mesenteric artery of rats. The resistance-sized mesenteric artery was isolated and cut into a ring. The ring segment was immersed in HEPES-buffered solution and its isometric tension was measured using myograph force-displacement transducer. Crude saponin of ginseng relaxed the mesenmetric arterial rings precontracted with norepinephrine (3$\mu$M) in dose-dependent manner (0.01 mg/㎖ -1 mg/㎖. The relaxation by crude saponin was smaller in endothelium-intact preparation than that in endothelium-denuded preparation. The contraction induced by A23187 or phorbol 12,13-dibutyrate was not affected by crude saponin of ginseng. The vasorelaxing effect of crude saponin of ginseng was significantly attenuated by the increase of the extracellular K$\^$+/ concentration. Crude saponin-induced vasorelaxation was not affected by tetraethylammonium (1 mM), glybenclamide (10$\mu$M), and 4-aminopyridine (0.1 mM) in these preparations. Ba$\^$2+/(10$\mu$M ∼100$\mu$M) markedly reduced the crude saponin-induced vasorelakation dose-dependently. From the above results, we suggest that crude saponin of ginseng may stimulate K$\^$+/ efflux and hyperpolarize the membrane, thereby cause the vasorelaxation in the resistance-sized mesenteric artery of rats.

  • 인삼(Panax ginseng C.A. Meyer)은 두릅나무과(Araliaceae)에 속하는 식물로 동아시아에서 2,000여년 전부터 보기(補氣)제로 사용된 중요한 약재이다. 홍삼은 인삼의 뿌리를 찐 것으로, 고려시대 이전부터 쪄서 말리는 가공법이 있었던 것으로 알려져 있다. 건강기능식품에서 말하는 홍삼제품은 홍삼을 원재료로 하여 이를 분말화하거나 물이나 주정으로 추출하여 농축 또는 발효하여 식용에 적합하며, 진세노사이드Rg1과 Rb1의 합이 0.8~34 mg/g이 되어야 한다. 2011년 한국의 건강기능식품 시장은 약 3조 6,000억원에 이르며, 홍삼시장은 1조 2,000억원에 이른다. 2007년 6,420억원이었던 홍삼시장이 5년만에 두배가 된 것이다. 홍삼시장이 성장하면서 한국인삼공사(정관장)와 농협중앙회(NH한삼인), 동원F&B(천지인), 웅진식품(다채움수) 등의 회사가 기초연구와 다양한 제품개발을 통하여 경쟁하고 있다. 한국인삼공사의 경우, 최근 식품의약품안전청으로부터 홍삼의 '항산화' 기능에 대하여 건강기능식품기능성원료 인정을 받았고, '홍삼정타브렛'을 출시하였으며 '홍이장군' 브랜드를 리뉴얼하였다. 농협중앙회에서는 이미지 향상을 위한 '지성이면 한삼인' 시리즈와 짜 먹는 '뽀로로 홍삼 젤리'를 출시하였고, 동원F&B은 '홍송조화건조'를 제조공정에 도입하였고, 캡슐형태의 제품을 출시하였으며, 최근에는 꼬마버스 타요를 활용하여 '천지인 꼬마버스 타요' 시리즈를 개발하였다. 웅진식품, 대상웰라이프 등에서도 사포닌이 흡수되기 좋게 만든 '발효홍삼' 제품을 출시하고 있다. 향후 홍삼시장의 지속적인 성장을 위해 기초연구와 소비자의 니즈에 맞는 국내시장만이 아닌 해외시장까지 나아갈 수 있는 특화된 제품 개발이 절실히 요구되는 시점이다.

A correlation of the modern scientific efficacy of Korean Red Ginseng with the legendary medicine for anti-aging and longevity (전설적 불로장생약과 고려홍삼의 현대 과학적 효능과의 연관성)

  • In this paper, through the mutual interpretation and verification of the ancient Korean history books with different origin that have been suspected as false documents, it proves that they could be logically real records and reveal that the substance of the legendary 'medicine for anti-aging and longevity', which also had been mentioned in Chinese old books, is Korean ginseng. Furthermore, with reference to the modern Y chromosomal map of the migratory routes of mankind corresponding to these routes recorded in 「Budoji」, the core history book, the formation of the four ethnic constitution groups (Sasang Constitution) based on the life style of each human group has been estimated. And the cause of Korean ginseng with fever problem for Southeast Asians is their pharmacogenomic constitution problem by protopanaxatriol (PPT) type ginsenosides in ginseng. It was resolved with over production of protopanaxdiol (PPD) type ginsenosides against PPT type in Korean red ginseng as historical or scientific point of view. In addition, by explaining that the processing method to Korean red ginseng could increase red ginseng acidic polysaccharides (RGAP), the RGAP, PPD type ginsenosides, and arginine which is originally abundant in Korean ginseng could increase the expression of the 'heat shock proteins' as a kind of chaperone in the body, this paper presents the theory allowing the scientific interpretation of the efficacy of Korean red ginseng as an 'adaptogen' or 'medicine for anti-aging and longevity'. Lastly, through the consideration of the growing environment of American ginseng and Korean ginseng, the differences are presented.

Effects of Crude Saponin and Saponin-free Fraction of Korea Red Ginseng on the Skin and Cerebral Blood Flow in the Rats (백서의 피부 및 뇌혈류에 미치는 고려홍삼 사포닌 및 비사포닌의 영향)

  • To study the effect of Korea red ginseng (KCG) on the skin and cerebral blood flow, we evaluated the change of skin perfusion rate and cerebral perfusion rate after the intravenous, intraperitoneal, and oral administration of crude saponin (CS) and saponin-free fraction (SFF) of KRG in the rats. The change of skin perfusion rate and cerebral perfusion rate was measured laser doppler flowmetry. The intravenous injections of CS or SFF of KRG and intraperitoneal injection of SFF of KRG did not change the relative skin and cerebral blood flow in the rats. When the rats were treated by the intraperitoneal injection of CS of KRG, relative cerebral blood flow was significantly increased with a time-dependent manner, however, relative skin blood flow was not influenced by the them. Oral administration of CS of KRG slightly increased skin blood flow in the rats. Also, the change of cerebral blood flow by transient bilateral carotid arterial clamp in the CS-treated rats was significantly decreased, compared with control groups. From the above results, it was suggested that Korea red ginseng have a increasing property of cerebral blood flow in the rats.

Quality Characteristics of Korean Red Ginsneg Powder on Pulverizing Methods (분쇄방법에 따른 고려홍삼분말의 품질특성)

  • In this study, cell cracker method as a non-collision method was evaluated for the possibility in manufacturing red ginseng powder. In color value of red ginseng powder, the L values indicating brightness were 68.18 for hammer mill (group A) and 72.08 for cell cracker (group B). The a values (redness) and b values (yellowness) were 4.21 26.56 for group A and 5.73, 28.36 for group B, respectively. As an extraction time increased, the absorbances at 420 nm of water extract of both groups were increased. In the loose bulk density, tapped bulk density and volume reduction radio indication the of powder in group A were less than those in group B. In both groups, the angles of side of red ginseng powder were shown less value than the angles of side of the powder. These two angles in group A were lets than those in group B. In sensory evaluation, the touchness between both groups was, not observed significant difference. In the contrary, the color and flavor, main factors affecting the overall quality of red ginseng powder product of group B were shown superior to those of group A. The overall preference of group B was shown higher than that of group A.

Bibliographic consideration on the efficacy and the origin of Korean ginseng (고려인삼의 유래 및 효능의 서지학적 고찰)

  • Korean ginseng (Panax ginseng) has been known as one of the representative special and healthful products originating from Korea for 4500 to 5000 years. The word of ginseng was first mentioned in JiJuZhang(急就章), written by ShiYou during the reign of King Yuah Di of the Chien Han Dynasty, China (33-48 BC). It has been known that wild Korean ginseng grows in Korean peninsula including Manchuria and the ginseng is found only between the $33^{rd}$ and $48^{th}$ parellels of north latitude. Since the times of three kingdom in Korea at 4-7 century, which is Kokuryo, Baekje and Shila, Korea has been the chief ginseng producing country. A large quantity of ginseng was exported from Korea to China for medicinal use at that times. That was written in SamGukSaGi(三國史記) by BuSik Kim of Koryeo Dynasty in Korea in 1145. The cultivation of Korean ginseng was also recorded in Bencaogangmu(本草綱目) written by LiShi Zen during the regin of the Ming Dynasty in 1596, China. The ginseng seedling, which was known as an original method invented by imitating the method of rice transplantation, appeared in the SeungJeongWon Ilgi(the diaries of the royal secretariat, 承政院日記), 1687 in the regin of King SukJong in Korea. It was suggesting that ginseng cultivation was firstly established in the early 1600s in Korea. On the other hand, red ginseng(written as 熟參) was reported firstly in GoRyeoDoGyeong(高麗圖經)(a record of personal experience in Korea, written in 1123) by SeoGung in Song Dynasty, China. The names of Pansam(written as 板蔘) and Pasam(written as 把蔘), which were the another types of red ginseng products, were came on in the JoSeon Dynasty Annals in 1552 and 1602, respectively. Although the term of red ginseng(Hongsam in Korean) was firstly appeared in the JoSeon Dynasty Annals in 1797, it is believed to have been developed a little earlier periods from the King Jungjong(1506~1545) to the King SeonJo(1567~1608) in Korea. Then, the Korean red ginseng has begun production on a large scale in SamJeong Department of NaeJangWon(內藏院 蔘政課) in the Korean Empire(大韓帝國) in 1899. More detailed records about red ginseng production method were written in the SohoDanag Miscellany(韶濩堂集) by Taekyoung Kim at 1916 year in Korea. On the while, the efficacy of ginseng was first recorded in Shennongbencaojing(神農本草經) written in China(BC 83-96) and the efficacy has been continuously inherited.

Antioxidant Effects of Raw Ginseng, Soft Red Ginseng, and Red Ginseng Sap (수삼, 연질 홍삼, 수액 홍삼의 항산화 효과)

  • Korean ginseng (Panax ginseng) generally has a good safety profile and contains many bioactive substances, such as ginsenosides or panaxosides. Korean red ginseng might help to stabilize the sympathetic nervous system and improve cognition in individuals. Soft red ginseng is produced by new processing technology. This study focused on investigating whether soft red ginseng produced under the new processing technology reduces or improves the existing antioxidant effects. No significant difference in 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) scavenging activity was found between soft red ginseng and ready-made red ginseng (p<0.05). Soft red ginseng extract showed higher 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and hydroxyl radical (OH) scavenging activity than other ginseng extracts. OH scavenging activity was significantly different across three groups (raw ginseng, soft red ginseng, and red ginseng sap) (p<0.05). Nitric oxide (NO) scavenging activity was also significantly different among raw ginseng, soft red ginseng, and purchased red ginseng liquid products (p<0.05). Many calcium crystals appeared on the electron microscope in soft red ginseng. Magnesium and potassium showed no significant difference between soft red ginseng and hard red ginseng. The extract of soft red ginseng scavenged different free radicals efficiently due to the presence of DPPH and OH and may help treat free radical-induced diseases.

Comparison of the Content of Saponin and Mineral Component in Korean Red Ginseng and Other Red Ginseng (한국산 및 외국산 홍삼의 사포닌 및 무기물 성분 비교)

  • This study was carried out to compare the contents of saponins and minerals component in Korean Red Ginseng (Heaven, Earth, Good grade), North Korean Red Ginseng (Heaven, Earth, Good grade), Japanese Red Ginseng (Oonju, Sinju 1, 2, 3 grade, respectively) and Chinese Red Ginseng (Seokju, Gilim, 1, 2, 3 grade, respectively). Crude saponin contents were different on according to the grade and cultivation area, and was 3.05-3.76% in Korean Red Ginseng, 2.09-3.21% in North Korean Red Ginseng, 2.82-3.71% in Chinese Seokju Red Ginseng, 2.72-3.62% in Chinese Gilim Red Ginseng, 2.11-2.44% in Japanese Oonju Red Ginseng, 2.18-2.87% in Japanese Sinju Red Ginseng, and the amount of ginsenoside-Rb1, -Re, -Rg$\_$1/ in Korean Red Ginseng were higher than those of North Korean, Chinese and Japanese Red Gingsen. The contents of mineral components were similar, but La, Na and Sn component in Korean Red Ginseng showed the higher amount than those of other Red Ginsengs.

CHEMICAL STUDIES ON CRUDE DRUG PROCESSING RED GINSENG AND WHITE GINSENG (생약에 관한 화학구조-홍삼 및 백삼-)

  • 생약의 화학적 특성에 대한 계속적인 연구가 이루어짐에 따라 우리는 홍삼 및 백삼의 화학성분을 상대적으로 규명하였다. 홍삼은 극성이 약한 분획에서 5개의 새로운 배당체(20R-ginsenoside $Rg_{2},\;Rh_{1};20R$, 20S-ginsenoside $Rg_{3}; ginsenoside\;Rh_{2}$와 새로운 아세칠렌 화합물(Panaxytriol)을 함유하는 특징적인 성분들이 gins - enoside Rh1, Rg2와 함께 분리되었다. ginsenoside Rh2는 배양된 종양세포에 대해 세포독소 효과를 보여주었다. 백삼은 수용성 분획에서 특징적인 성분이 있는 것으로 밝혀졌으며, 여기에서 malonly-ginsenosides Rb1, Rb2, Rc 및 Rd로 명명된 새로운 배당체 성분이 분리되었다. Malona-ginsenosides는 백삼에서는 주요한 배당체이지만, 홍삼에서는 검출되지 않았다.

Flavor Characteristics of Korean Red Ginseng (한국홍삼의 향 특성)

  • The flavor characteristics of the Korean red ginseng were compared with those of the Chinese red ginseng organoleptically. The Korean red ginseng had strong sweety and roasted odor while the Chinese red ginseng had strong earthy, woody and hay-like odor. The sweety odor and the roasted odor of the Korean red ginseng occurred in the neutral subfraction of the ether-soluble fraction of its headspace volatiles.
