  • In August 2020, necrotic ringspots on leaves were observed on 20 from 143 Plantago asiatica plants in open fields in Eumseong, Chungcheongbuk-do. Eight symptomatic Plantago asiatica plants were subjected to investigation on viral infection by reverse transcription and polymerase chain reaction. Impatiens necrotic spot virus, tomato spotted wilt virus and cucumber mosaic virus were detected from the symptomatic plants. Two impatiens necrotic spot virus (INSV) isolates ('INSV-plantain kr1' and '-plantain kr5') were sequenced and analyzed by comparing L genomic segment, nucleoprotein (N) gene and non-structural protein S (NSs) gene sequences. The nucleotide sequence of 'INSV-plantain kr1' isolate (MW114834) was most closely related to that of a 'Phalaenopsis' isolate (GQ336991) from China in the L genomic segment. 'INSV-plantain kr1' and '-plantain kr5' isolates shared the highest identities with those from 'Pepe' isolate (LC384872) and 'J' isolate (AB109100) in the NSs gene, respectively, and with that from 'YSMi-SH' isolate (FN400773) in the N gene. Phylogenetic analysis based on L genomic segment grouped the INSV-plantain kr1 isolate together with isolates from Korea (LC384870), China (GU112505, GQ336991), and Italy (DQ425094). This is the first report on INSV in P. asiatica from Korea.

  • The green alga Chlamydomonas reinhardtii, a unicellular haploid eukaryote, has long been used by researchers and industries as a cell factory to produce high value-added microalgae substances using genetic modification. Microalga K01, presumed to be Chlamydomonas, was isolated from 12 freshwater samples from the Chungcheong and Jeolla regions to replace C. reinhardtii, an introduced species currently used in most basic and industrial research. The isolated K01 strain was identified as C. reinhardtii through morphological and phylogenetic studies of the 18S rDNA gene sequence (NCBI accession number KC166137). The growth and lipid content of the isolated C. reinhardtii K01 were compared with three wild and four mutant strains in TAP medium, and it was found that the K01 strain could produce 1.74×107 cells/ml by the third day of culture. The growth rate of C. reinhardtii K01 was 1.5 times faster than UTEX2244, which showed the highest number of cells (1.20×107 cells/ml) among the compared strains. The lipid content of the isolated C. reinhardtii K01 (20.67%) was similar to those of the wild strains, although the fatty acid oleate C18:1 was not detected in the isolated strain but was identified in the seven others. The cell density of the isolated strain increased to 0.87 g/l during a six-day culture in BG11 medium, where nitrate (NaNO3) was introduced as a nitrogen source, while the seven acquired strains showed almost no cell proliferation.

  • The seven mitochondrial DNA regions (ND2, ND5, ND4, ND4L, ND6, ND1 and 12SrRNA) of Haliotis discus hannai were examined to estimate the availability as a genetic marker for the study of population genetic. The region with the highest genetic variation was ND4 (Haplotype diversity = 1.0000, Nucleotide diversity = 0.0108). On the other hand, ND2 and ND1 regions have significantly appeared genetic divergence between clusters (divergence of 90% and 87%). Also, pairwise $F_{ST}$ between clusters within ND2 and ND1 regions showed high values; 0.4061 (P = 0.0000), 0.4805 (P = 0.0000) respectively. Therefore we can infer that it is the most efficient and accurate way to analyze the region of ND4 with the highest variation in addition to the regions of ND2 and ND1, which formed clusters with high bootstrap value, for study of population genetic structure in this species.

  • A specimen of porous-head eelpout Bothrocara hollandi (Zoarcidae: Perciformes) caught from the East Sea was found to harbour a myxosporean parasite. Numerous whitish pseudocysts were scattered throughout the body musculature of this individual specimen. Fresh myxosporean spores were found from the squashed pseudocysts under light microscopy. They were subspherical in frontal view with a length of $11.9(11.0{\sim}13.5){\mu}m$, width of $11.6(10.7{\sim}13.6){\mu}m$, and thickness of $7.8(6.9{\sim}8.8){\mu}m$. Two polar capsules were almost equally pyriform with a length of $4.4(3.2{\sim}5.3){\mu}m$ and width of $3.3(2.4{\sim}4.2){\mu}m$. Morphometric and host ecology analysis revealed that this myxosporean parasite could be identified as Myxobolus aeglefini Auerbach 1906. Phylogenetic analysis based on 18S rDNA sequences also revealed that M. aeglefini was clustered with M. albi and M. groenlandicus in the same branch, sharing 97.7% and 96.9% sequence similarities with M. albi and M. groenlandicus, respectively.

  • A new heat shock protein 70 was identified in red-spotted grouper (Epinephelus akaara) based on an expression analysis. The cDNA of red-spotted grouper Hsp70 (designated RgHsp70) was cloned by the rapid amplification of cDNA ends (RACE) techniques. The full-length of RgHsp70 cDNA was 2,152 bp, consisting of a 5'-terminal untranslated region (UTR) of 105 bp, a 3'-terminal UTR of 274 bp, and an open reading frame (ORF) of 1,773 bp that encode a polypeptide of 590 amino acids with a theoretical molecular weight of 64.9 kDa and an estimated isoelectric point of 5.2. Multiple alignment and phylogenetic analyses revealed that the RgHsp70 gene shares a high similarity with other Hsp70 fish genes. RgHsp70 contained all three classical Hsp70 family signatures. The results indicated the RgHsp70 is a member of the heat shock protein 70 family. RgHsp70 mRNA was predominately expressed in the liver, with reduced expression noted in the head-kidney tissues. The expression analysis of different water temperatures (21, 18, 15 and $12^{\circ}C$) for sampled livers revealed that expression gradually increased at $12^{\circ}C$ compared to $21^{\circ}C$. In this study, the effects of water temperature lowering on the physiological conditions were investigated, and the results revealed that novel RgHsp70 may be an important molecule involved in stress responses.

  • Several microorganisms in particular Bacillus subtilis group have been isolated from diverse places such as soils and the gastrointestinal tract of ruminants etc., and used as biocontrol agent against various plant pathogens and utilized as plant growth promoting agents. Among them, Bacillus is well known as one of the most useful bacteria for biocontrol and plant growth promotion. Bacterium GH1-13 was isolated from a reclaimed paddy field in Wando Island and identified as Bacillus velezensis using phylogenetic analysis on the basis of 16S rRNA and gyrB gene. It was confirmed that GH1-13 produced indole acetic acid (IAA) associated with promoted growth of rice root. GH1-13 showed characteristics of antagonization against the main pathogen of rice as well as diverse pathogenic fungi. GH1-13 had biosynthetic genes, bacillomycin, bacilycin, fengycin, iturin, and surfactin which are considered to be associated closely with inhibition of growth of pathogenic fungi and bacteria. This study showed that GH1-13 could be used as a multifunctional agent for biocontrol and growth promotion of crop.

  • In this study, an attempt has been made to analyze the morphology of Cephalopods distributed in Korea and collected samples from South-East Asian countries including Thailand, Indonesia, Vietnam, and China. A phylogenetic analysis was performed using the mitochondrial gene, Cytochrome c oxidase subunit I (COI) to understand the genetic divergences of the species and validate their origins. For achieving the objectives, samples were collected directly from Thailand Hat Yai, Songkhla, Indonesia Medan, Vietnam Ho Chi Minh, and Vung Tau in August 2015 and from China in September 2015. A total of 23 species of Cephalopods were identified falling under three orders, four familyies and nine genus. The species were distributed under Order: Octopoda (1 family, 3 genus, and 9 species), Order: Sepiolioda (1 family, 2 genus, and 8 species), and Order Teuthoidea (2 family, 4 genus, and 6 species). 23 species which is 1 family 3 genus 9 species in Octopoda, 1 family 2 genus 8 species in Sepiolioda, 2 family 4 genus 6 species in Teuthoidea. Phylogenetic analysis using COI gene was conducted for 18 species. For the remaining 5 species sequencing results showed severe variation and hence were not considered further. The COI phylogenetic analysis for the 18 species of Cephalopods were found consistent with the morphological identification. The excluded species will be subjected for a further detailed analysis.

  • The shrimp fishery for Penaeus japonicus on the Koje-Do coast commences in May and continues to October. The best catches are made twice a year, namely June and September. Modal carapace length for the female is $51\~55\;mm$ in May, $51\~60\;mm$ in June occupying about 80 percent of the total catch. There is conspicuous mode in July and $46\~50\;mm$ in August. Modal carapace length for the male is $41\~45\;mm$ in May, $46\~50\;mm$ in June, $41\~45\;mm$ in July and August. The stock of p. japonicus can be divided into two groups, namely, spring and fall groups. Among samples of p. japonicus during the period from May through August the sex ratio consisted of about 33 percent females and 67 percent males, showing wide departure from a 1:1 ratio. The relationships between carapace length ($\iota$), body length (L) and body weight (W) are expressed by the following formulae : $$L=2.9418{\iota}+2.0166$$, $$W:1.449{\iota}^{2.2858}$$ (Mayssample) $$L=2.5551{\iota}+4.2986$$, $$W=1.528{\iota}^{2.2595}$$ (June sample) $$L=2.6738{\iota}+3.4037$$, $$W=1.341{\iota}^{2.3598}$$ (July sample) The relatlonship between carapace length ($\iota$) and ovary weight (W) is shown as follows: $W=2.695\times10^{-3}\iota^{4.2973}$ for May, June and July Samples.

  • Morphometric and genetic analyses on six populations of the two subspecies of Cobitis koreensis were performed to investigate the geographic variation and to clarify their taxonomic status. No significant differences between subspecies were found. Puan population of C. k. pumilus tends to be smaller in numbers of cross bars and dorsal spots, than those of C. k. koreensis populations with no statistical significance. Average degree of genic variation among six populations of the two subspecies was A=1.5, P =37.3%, HD=0.053, and HG=0.097. Marginal populations of Samchog (C. k. koreensis) and Puan(C. k. pumilus) were least variable: A= 1.3, P=23.8%, HD=0.0l0, HG=0.043, and A= 1.2, P=19.0%, HD=0.029, HG=0.078 respectively. The difference between HD and HG in all populations may indicate a patchness distribution between demes within each population. Genetic similarities between the two subspecies were, on the average, S=0.894, and this value correspond to the average genetic similarities among five populations of C. k. koreensis (5=0.899). Chongup population of C. k. koreensis and Puan population of C. k. pumilus was most similar genetically (5 =0.931). The results of morphometric and genetic analysis in this study indicate that C. k. pumilus is not a valid subspecies and should be considered as a population of C. koreensis.
