The 1,874 rice lines were selected from 3,000 Ds insertional mutant pool by Basta herbicide treatment and were surveyed for trait variation and molecular characteristics of genes knocked out by Ds insertion. Compared with "Donjin", an original japonica cultivar used for transformation, Ds insertion mutant pool showed large variation in major agronomic traits including tiller, panicle, and heading etc. Southern blot analysis demonstrated that these lines on the average had two Ds copies in Donjin genome, resulting in 38.4% of one copy, 32.5% of two copies, 16.7% of three copies, and 11.3% of over four copies. GUS analysis showed that 3.9% of lines (73/1,860) had tissue-specific expression in leaves, nodal parts, floral organs such as stigma and pollen, and roots. Data set obtained from agricultural trait variation and molecular characteristics for individual Ds insertional lines would provide researchers with more information for understanding the function of unknown rice genes controlling economically important traits.
'Anyu', a perilla (Perilla frutescens (L.) Britton) cultivar for edible seed was developed by the Department of Functional Crop Science, National Institute of Crop Science, RDA in 2008. It was originated from the cross between a pedigree of YCPL1824 as a female and SF9225-3-1 as a male parent in 1998. 'Anyu' can be characterized by purple flower color and brown grain color. 'Anyu' showed semi-dwarft type with 120 cm height, and has lodging tolerance. Maturing date of 'Anyu' was September 24, which is 15 days faster than that of 'Saeyeopsil'. The early maturity of this new cultivar showed a great advantage to various planting system with other crops. This new cultivar has high oil content (45%) and high linolenic acid in the fatty acid composition. The yield potential of 'Anyu' was about 0.9 ton/1ha in the regional yield trial.
'Punghan' (Avena sativa L.), a winter oat for forage use, was developed by the breeding team at the Department of Rice and Winter Cereal Crop, NICS, RDA in 2008. It was derived from a cross between 'Beltsville 61-150 (IT133501)' and $F_1$ between 'Sikyonggwiri' and 'PA202-210'. A oat cultivar from USA, 'Beltsville 61-150', has a high cold tolerance, while the $F_1$ between 'Sikyonggwiri' and 'PA202-210' has early heading and high yielding with large-size grain. Subsequent enerations followed by the cross were handled in bulk and pedigree selection programs at Suwon and Yeoncheon, respectively. A line, 'SO99013-B-YB-31', was selected for earliness and good agronomic characteristics and designated as a line name of 'Gwiri 60'. The line 'Gwiri 60' was subsequently evaluated for winter hardiness, earliness, and forage yield in five locations, Yesan, Cheongwon, Iksan, Kimjae, and Jeju, from 2006 to 2008 and finally named as 'Punghan'. Over 3 years, the average forage dry matter yield of 'Punghan' harvested at milk-ripe stage was $14.5\;ton\;ha^{-1}$, compared with $13.6\;ton\;ha^{-1}$ of check cultivar 'Samhan'. 'Punghan' is about 2 days earlier in heading date than check cultivar. 'Punghan' is adapted primarily for winter planting use in the mid-southern regions of Korean peninsula.
A new small red bean cultivar, 'Geomguseul', was artificially crossed between 'Chilbo' and 'SA8412-3-1-4-3-3-2-3' in 1998, fixed excellent agronomic characters by pedigree breeding method, and selected for the further trials with the name of 'Miryang 10'. It was prominent and showed good result, such as high grain quality, lodging resistance, and high-antioxidant activity in the regional adaptation yield trials (RYT) for three years from 2009 to 2011 and released for the public consumption as the name of 'SA9801-24-4-3-1' in 2011. 'Geomguseul' has a semi-determinate growth habit, yellow flower, green embryonic axis, black seed coat, white hilum, and small spherical seed (12.2 grams per 100 seeds). The average yield of 'Geomguseul' was 2.01 MT/ha in the regional yield trials (RYT) carried out for three years from 2009 to 2011 which was a little higher than that (1.98 MT/ha) of the check variety, 'Chungju'.
Several major agronomic traits of 4 isogenic lines for tillering(Kindred, Morex)were investigated under the different seeding rates and seedling mortality grown in the pots. The normal-tillering phenotypes had higher grain yield(46%) and leaf area index(56%) than their uniculm isogenic counterparts. The assortment rate(69%) and 1,000 grain weight(27%) in uniculm type were greater than the normal type. Uniculm plants were earlier in heading(4∼8days) and maturing(2∼3 days) dates compared with their counterparts. The grain yield was fitted to linear regression, ranged 0.659g to 0.5g per spike number. On increasing the seedling mortality rates, the grains per spike and 1,000 grain weight were slightly decreased, but the grain yield per plot was decreased markedly because of decrease of spikes per plot. The assortment rate, leaf photosynthetic capacity and reproductive tiller number were increased with increase of seedling mortality. The grain yield was largely dependent on the number of spikes per plot on occurrence of seedling mortality. For the uniculm genotypes to be introduced and bred into a present leading variety, the isogenic lines were precisely tested in the open field for a couple of years.
This study was conducted in order to breed Japonica salt tolerant rice and select salinity tolerant lines by establishing reliable and practical method of screening voluminous materials. Salinized nutrient solution by adding NaCl was effective compared with the salinized nutrient solution by adding 0.3%, 0.6% and 0.7% of diluted sea water. There was no different visual score of salinity injury between salinization using tap water and distilled water. Seedling salinity tolerant lines between region and order by year were showed very stable and reproducible results, 3~4.2 of visual score at Gyehwado-substation and International Rice Research Institute (IRRI, Philippines) in 2002, 2003 and 2004. Heading date of 6 selected seedling salt tolerant lines showed a range of 16. Aug.~21. Aug. and delayed 2~6 days than that of Donjinbyeo. Percentage of ripened grain and yield of milled rice in 6 lines was lower, 52.2~14.7% and 50~5%, respectively than those of Donjinbyeo.

A FA (Lilium formolongi $\times$ Lilium asiatic hybrid) intersectional hybrid lily cultivar 'Orange Crown' was developed in 2007 at National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA) of Korea. The cross and immature embryo rescue was conducted between female parents 'Migreen' (Lilium formolongi 'Raizan' $\times$ Lilium asiatic hybrid 'A61') and male parent 'A01-187' (L. asiatic 'A96-28' $\times$ 'Sanzio') by cut style pollination method at Suwon in 2001. It was preliminarily selected as 'FA04-27' in 2004. Multiplication, bulb production and characteristic tests were conducted from 2004 to 2007. The evaluation of characteristics and preference were surveyed at a lily flower show of NIHHS at Taean in 2007. 'Orange Crown' flowers in the late of June and grows more than 183 cm stem length. Flowers bloom upward-facing, a little spotted and orange (RHS, O24A) petals. The pollen of 'Orange Crown' was sterile.
Over 7 individual rice (Oryza sativa L.) plants per a line were sowed and sampled by pooled sampling method for genomic DNA extraction. The 5,400 flanking sequence tags (FSTs) were analysed by adaptor PCR and direct sequencing. FST analysis showed that the intragenic FSTs, the intergenic FSTs, and the original insertional sequences including hot spot covered 48.1% (2,597), 25.6% (1,383), and 25% (1,350), respectively. The 2,597 intragenic FSTs were used for genotyping to determine whether they are heterozygous or homozygous, and 1,393 core lines were selected. Among them, 422 knockout genes were distributed on chromosome 3, while 56 - 157 intragenic FSTs scattered on other chromosomes. Among 1,393 FSTs, known genes such as transcription factor covered 59.4% (827), while unknown genes such as expressed protein covered 40.6% (566). RT-PCR indicated that some core lines had no expression or decreased expression level in their knockout genes. It means that core lines are very useful knockout lines for functional genomic studies.
In previous studies, we reported QTLs for grain weight (GW), qGW3 and for spikelets per panicle (SPP), qSPP3 linked to RM60 on chromosome 3 using advanced backcross lines derived from a cross between Oryza sativa ssp. Indica cv. Milyang 23 and O. glaberrima. The O. glaberrima alleles at this locus increased GW and spikelets per panicle in the Milyang 23 background. To further confirm and narrow down the position of the QTLs on chromosome 3, substitution mapping was performed using five lines containing the target O. glaberrima segment on chromosome 3. The size and position of the O. glaberrima segment on chromosome 3 were different in each line. These lines possessed 3-10 non-target O. glaberrima introgressions in the Milyang 23 background. These five lines were evaluated for seven agronomic traits including 1,000 grain weight and spikelets per panicle and also genotyped with seven SSR markers. Four lines were informative in delimiting the position of QTLs, qGW3 and qSPP3. Two lines with the O. glaberrima segment flanked by SSR markers, RM60 and RM523 displayed significantly higher values than Milyang 23 in GW and SPP whereas two lines without that O. glaberrima segment displayed no difference in GW and SPP compared to Milyang 23. The result indicates that two QTL, qGW3 and qSPP3 are located in the interval between RM60 and RM523 which are 1.2-Mb apart. Introgression lines having QTLs, qGW3 and qSPP3 would be useful materials not only to indentify the relationship between these two yield QTLs, but also to develop high yielding variety via marker-aided selection technology.

The study was performed to investigate the effect of a plug culture method in the early stage of a potato breeding program. The 105, 200 and 288-cell plug trays were used for raising potato seedlings. The % germination in plug trays ranged between 94 and 96%. Percent tuberization measured at 50 and 60, and 70 days after sowing was the greatest in 105-cell and 200-cell trays, respectively. However, tuberization was most delayed in 288-cell trays. Thus, it is considered that 105-cell tray is the most effective for the selection of early maturing plants. Plant maturity was determined by the morphological characteristics at the seedling stage of stolons cultured in plug trays. Most of seedlings with shorter stolons at seedling stage (1st generation) were the early-maturing, while most of seedlings with longer stolons were the late-maturing at the clonal stage (2nd generation). These results mean that the plant maturity in the early potato breeding program can be effectively grouped by observing the morphological characteristics of stolons after plug culture.
