An Analytical Validation of the GenesWellTM BCT Multigene Prognostic Test in Patients with Early Breast Cancer (조기 유방암 환자를 위한 다지표 예후 예측 검사 GenesWellTM BCT의 분석적 성능 시험)

  • GenesWell$^{TM}$ BCT is a 12-gene test suggesting the prognostic risk score (BCT Score) for distant metastasis within the first 10 years in early breast cancer patients with hormone receptor-positive, HER2-negative, and pN0~1 tumors. In this study, we validated the analytical performance of GenesWell$^{TM}$ BCT. Gene expression values were measured by a one-step, real-time qPCR, using RNA extracted from FFPE specimens of early breast cancer patients. Limit of Blank, Limit of Detection, and dynamic range for each of the 12 genes were assessed by serially diluted RNA pools. The analytical precision and specificity were evaluated by three different RNA samples representing low risk group, high risk group, and near-cutoff group in accordance with their BCT Scores. GenesWell$^{TM}$ BCT could detect gene expression of each of the 12 genes from less than $1ng/{\mu}L$ of RNA. Repeatability and reproducibility across multiple testing sites resulted in 100% and 98.3% consistencies of risk classification, respectively. Moreover, it was confirmed that the potential interference substances does not affect the risk classification of the test. The findings demonstrate that GenesWell$^{TM}$ BCT have high analytical performance with over 95% consistency for risk classification.

Environmental Geochemistry and Contamination Assessment of the Tohyun Mine Creek, Korea (토현광산 수계의 환경지구화학적 특성과 오염도 평가)

  • The pH values of the mine and surface water from the Tohyun mine creek were higher compared with those of groundwater, and 2nd round samples in same sites were even alkaline. The stream and mine waters belong to the characteristics of (Ca+Mg)-(SO$_4$) and (Ca+Mg)-(HCO$_3$) types, and groundwaters have to the (Ca+Mg+Na+K)-(HCO$_3$+SO$_4$) type. As the 2nd samples. concentrations of mostly anions are increasing compared with the forder samples. However, the mostly cation concentrations are decreasing. The hydrogeochemistry indicate that water quality is different chemical characteristics and evolution trends. The range of $\delta$D and $\delta$$^{18}$ valutes (relative to SMOW) in the waters are shown in -62.2 to -70.1$\textperthousand$, and -8.1 to -9.4$\textperthousand$. The values are plowed parallel to $\delta$D=8$\delta$$^{18}$ O+ (6$\pm$4). The d values of groundwater show 2.4, which is lower than the surface (5.2) and mine (7.6) waters. Strontium concentra titans range from 0.025 to 11.844 mg/$\ell$ in all kinds of water samples, but the groundwater has the highest contents The $^{87}$ Sr/$^{86}$ Sr ratios (0.7115 to 0.7129) show more lightened to the groundwater. The $\delta$$^{18}$ O value, Ca and Sr contents are decreased with $^{87}$ Sr/$^{86}$ Sr increasing, because it is support to the altitude effects of the sampling sites rather than a water-rock interaction of environmental isotope. Using computer code of WATEQ4F, saturation indices of albite, Quartz, gibssite and gypsum are calculated to be soluble. The calcite and dolomite show super saturation state, however, clay mineral species are plotted boundary between undersaturation and supersaturation. In the Tohyun mine creek, reaction materials with ore wastes arid precipitation have influence upon increasing EC and TDS of the waters independent of pH. The SO$_4$ concentrations in the mine water is 181.845 mg/$\ell$. This is abruptly increase in surface water and then detected 249.727 mg/$\ell$ in the groundwater. As a results of the calculated sulfate mineral solubilities, the sulfate ions became saturation states an above 150 mg/$\ell$ concentrations.

Vertical Distribution and Contamination of Trace Metals in Sediments Within Hoidong Reservoir (회동저수지 호저퇴적물의 미량원소 오염 및 수직적 분산특성)

  • In order to investigate the vertical variations and speciations of trace elements, and their correlations in Hoidong reservoir, sediment cores (21-41 cm below surface) and interstitial water samples were collected from five sampling locations. The total average concentrations of trace metals in sediment core samples were $232{\pm}30.8mg/kg$ for Zn, $119{\pm}272mg/kg$ for Cu, $58.4{\pm}4.1mg/kg$ for Pb, $15.7{\pm}3.3mg/kg$ for Ni and $1.6{\pm}0.3mg/kg$ for Cd. The total concentrations of trace metals in core sediments generally decreased toward the center of the Hoidong reservoir. The total concentrations of Mn, Pb and Zn decreased with depth for all the sample locations, while Cu and Fe concentrations increased. The trace metal concentrations of interstitial water sample were in the order of Fe>Mn>Cu>Zn, but Cd, Ni and Pb were not detected. The concentrations of Zn, Cu, Fe and Mn in the interstitial water samples showed a tendency of increasing toward the bottom of the core, suggesting a possible upward diffusion. This migration of trace metals may lead to their transfer to the sediment-water interface. These trace elements would be subsequently fixed onto amorphous Fe and Mn-oxides and carbonates in the topmost layer of sediment. Based on the $K_D$ values, the relative mobilities of the studied metals were in the order of Mn>Cu>Zn>Fe. Geochemical partitioning confirmed that surface enrichment by trace metals mainly resulted from a progressive increase of the concentrations in the fractions II and III. Copper, Fe, Mn and Zn concentrations of interstitial water were closely correlated with their exchangeable fractions of sediments.

Hydrochemistry and Distribution of Uranium and Radon in Groundwater of the Nonsan Area (논산지역 지하수중 우라늄과 라돈의 수리지질학적 특성과 정밀함량분포)

  • A total of 100 groundwater samples were collected from the Nonsan area and the behaviors of uranium and radon as natural radionuclides were investigated with respect to other physicochemical components in the groundwater in order to understand their occurrence, properties, and origins. Radionuclide levels were used to construct detailed concentration maps. The concentration of uranium ranges from 0 to 378 ${\mu}g/L$, with an average of 8.57 ${\mu}g/L$, standard deviation of 42.88 ${\mu}g/L$, and median of 0.56 ${\mu}g/L$. The correlation coefficient between uranium and radon is 0.42, whereas these radionuclides show no relation with other physicochemical components in groundwater. It is noteworthy that the uranium level in most samples (97% of the samples) is less than 30 ${\mu}g/L$, where the bedrock of the aquifer is granite or complex rocks located along the boundary between granite and metamorphic rocks. In the Okcheon metamorphic belt, the uranium concentration of most groundwater is less than 1 ${\mu}g/L$. Radon levels varies from 128 to 9,140 pCi/L, with an average of 2,186 pCi/L, standard deviation of 1,725 pCi/L, and median of 1,805 pCi/L. High radon levels (> 4,000 pCi/L) are most common in regions of Jurassic granite, whereas low radon areas are found in regions of sedimentary rock. In conclusion, the distribution and occurrence of radionuclides are intimately related to the basic geological characteristics of the rocks in which the radiogenic minerals are primarily contained.

Truncation Artifact Reduction Using Weighted Normalization Method in Prototype R/F Chest Digital Tomosynthesis (CDT) System (프로토타입 R/F 흉부 디지털 단층영상합성장치 시스템에서 잘림 아티팩트 감소를 위한 가중 정규화 접근법에 대한 연구)

  • Chest digital tomosynthesis has become a practical imaging modality because it can solve the problem of anatomy overlapping in conventional chest radiography. However, because of both limited scan angle and finite-size detector, a portion of chest cannot be represented in some or all of the projection. These bring a discontinuity in intensity across the field of view boundaries in the reconstructed slices, which we refer to as the truncation artifacts. The purpose of this study was to reduce truncation artifacts using a weighted normalization approach and to investigate the performance of this approach for our prototype chest digital tomosynthesis system. The system source-to-image distance was 1100 mm, and the center of rotation of X-ray source was located on 100 mm above the detector surface. After obtaining 41 projection views with ${\pm}20^{\circ}$ degrees, tomosynthesis slices were reconstructed with the filtered back projection algorithm. For quantitative evaluation, peak signal to noise ratio and structure similarity index values were evaluated after reconstructing reference image using simulation, and mean value of specific direction values was evaluated using real data. Simulation results showed that the peak signal to noise ratio and structure similarity index was improved respectively. In the case of the experimental results showed that the effect of artifact in the mean value of specific direction of the reconstructed image was reduced. In conclusion, the weighted normalization method improves the quality of image by reducing truncation artifacts. These results suggested that weighted normalization method could improve the image quality of chest digital tomosynthesis.

Mineral Geochemistry of the Albite-Spodumene Pegmatite in the Boam Deposit, Uljin (울진 보암광산의 조장석-스포듀민 페그마타이트의 광물 지화학 조성 연구)

  • In this study, we investigated the mineral geochemistry of the albite-spodumene pegmatite, associated exogreisen, and wall rock from the Boam Li deposit, Wangpiri, Uljin, Gyeongsangbuk-do, South Korea. The paragenesis of the Boam Li deposit consists of two stages; the magmatic and endogreisen stages. In the magmatic stage, pegmatite dikes mainly composed of spodumene, albite, quartz, and K-feldspar intruded into the Janggun limestone formation. In the following endogreisen stage, the secondary fine-grained albite along with muscovite, apatite, beryl, CGM(columbite group mineral), microlite, and cassiterite were precipitated and partly replaced the magmatic stage minerals. Exogreisen composed of tourmaline, quartz, and muscovite develops along the contact between the pegmatite dike and wall rock. The Cs contents of beryl and muscovite and Ta/(Nb+Ta) ratio of CGM are higher in the endogreisen stage than the magmatic stage, suggesting the involvement of the more evolved melts in the greisenization than in the magmatic stage. Florine-rich and Cl-poor apatite infer that the parental magma is likely derived from metasedimentary rock (S-type granite). P2O5 contents of albite in the endogreisen stage are below the detection limit of EDS while those of albite in the magmatic stage are 0.28 wt.% on average. The lower P2O5 contents of the former albite can be attributed to apatite and microlite precipitation during the endogreisen stage. Calcium introduced from the adjacent Janggun formation may have induced apatite crystallization. The interaction between the pegmatite and Janggun limestone is consistent with the gradual increase in Ca and other divalent cations and decrease in Al from the core to the rim of tourmaline in the exogreisen.

Development of Cloud Detection Method Considering Radiometric Characteristics of Satellite Imagery (위성영상의 방사적 특성을 고려한 구름 탐지 방법 개발)

  • Clouds cause many difficult problems in observing land surface phenomena using optical satellites, such as national land observation, disaster response, and change detection. In addition, the presence of clouds affects not only the image processing stage but also the final data quality, so it is necessary to identify and remove them. Therefore, in this study, we developed a new cloud detection technique that automatically performs a series of processes to search and extract the pixels closest to the spectral pattern of clouds in satellite images, select the optimal threshold, and produce a cloud mask based on the threshold. The cloud detection technique largely consists of three steps. In the first step, the process of converting the Digital Number (DN) unit image into top-of-atmosphere reflectance units was performed. In the second step, preprocessing such as Hue-Value-Saturation (HSV) transformation, triangle thresholding, and maximum likelihood classification was applied using the top of the atmosphere reflectance image, and the threshold for generating the initial cloud mask was determined for each image. In the third post-processing step, the noise included in the initial cloud mask created was removed and the cloud boundaries and interior were improved. As experimental data for cloud detection, CAS500-1 L2G images acquired in the Korean Peninsula from April to November, which show the diversity of spatial and seasonal distribution of clouds, were used. To verify the performance of the proposed method, the results generated by a simple thresholding method were compared. As a result of the experiment, compared to the existing method, the proposed method was able to detect clouds more accurately by considering the radiometric characteristics of each image through the preprocessing process. In addition, the results showed that the influence of bright objects (panel roofs, concrete roads, sand, etc.) other than cloud objects was minimized. The proposed method showed more than 30% improved results(F1-score) compared to the existing method but showed limitations in certain images containing snow.
