Development and Preliminary Evaluation of a Leukocyte Removal Aptamer Filter (압타머를 이용한 백혈구제거필터의 개발 및 예비평가)

  • Background: Leukocyte reduction filters are widely used to prevent transfusion reactions caused by leukocytes in blood components. Commercial filters are not sufficient for removal of leukocytes for prevention of transfusion associated graft-versus-host disease; therefore, irradiation of blood components was performed using expensive equipment. Techniques using an aptamer substituted for antibody have been developed and are available in clinical areas. The purpose of this study is to develop the aptamer filter system and to evaluate its efficiency and the possibility of its clinical application. Methods: Aptamers targeted to CD45 were selected by the Postech Aptamer Initiative. The aptamer filter in which aptamers attached to beads were bound to leukocytes and removed by magnetic field was developed. Filtration of 14 units of leukoreduction-red blood provided by Korean Red Cross Blood Services was performed using aptamer filters. Leukocyte removal rate and red cell recovery rate were evaluated and bacterial culture was performed. Results: After filtration using the aptamer filters, 45.6% of leukocytes were additionally removed and the red cell recovery rate was 92.8%. No growth in the bacterial culture was observed. Conclusion: In order to apply the cell depletion technique utilizing an aptamer to blood filter system, we developed and evaluated the aptamer filter system. Through improvement of the binding efficiency of the aptamer and the filtering process, and application of the various aptamers for other different cells, we suggest that this technique can be applied in the clinical area, such as a substitution for the irradiation process for TAGVHD prevention.

Study on Bandwidth and Characteristic Impedance of CWP3DCS (Coplanar Waveguide Employing Periodic 3D Coupling Structures) for the Development of a Radio Communication FISoC (Fully-integrated System on Chip) Semiconductor Device (완전집적형 무선통신 SoC 반도체 소자 개발을 위한 주기적인 3차원 결합구조를 가지는 코프레너 선로에 대한 대역폭 및 임피던스 특성연구)

  • In this study, we investigated the characteristic impedance and bandwidth of CPW3DCS (coplanar waveguide employing periodic 3D coupling structures), and examined its potential for the development of a marine radio communication FISoC (fully-integrated system on chip) semiconductor device. To extract bandwidth and characteristic impedance of the CPW3DC, we induced a measurement-based equation reflecting measured insertion loss, and compared the measured results of the propagation constant β and characteristic impedance with the measured ones. According to the results of the comparison, the calculated results show a good agreement with the measured ones. Concretely, the propagation constant β and characteristic impedance exhibited an maximum error of 3.9% and 6.4%, respectively. According to the results of this study, in a range of LT = 30 ~ 150 ㎛ for the length of periodic structures, the CPW3DC exhibited a passband characteristic of 121 GHz, and a very small dependency of characteristic impedance on frequency. We could realize a low impedance transmission line with a characteristic impedance lower than 20 Ω by using CPW3DCS with a line width of 20 ㎛, which was highly reduced, compared with a 3mm line width of conventional transmission line with the same impedance. The characteristic impedance was easily adjusted by changing LT. The above results indicate that the CPW3DC can be usefully used for the development of a wireless communication FISoC (fully-integrated system on chip) semiconductor device. This is the first report of a study on the bandwidth of the CPW3DC.

  • As stage equipment export markets along with special lighting service lack the attraction for already globally established businesses, such markets can be viewed as an advantageous opportunity for SMEs as in general. In reality, global businesses tend to focus on large construction projects and this indicates relatively less substantial markets such as stage equipment and special lighting service export are more suitable for SME businesses. However, possible problems may be recognized as following; doubtful capabilities by such businesses to join in the vast and competitive global market and pursue manufacturing and service based export. This point is also supported by the fact that such in general SME businesses have substantially less experience in exporting products and services abroad. Realizing the distinctive features of the Korean economy, it is unarguable that every sector and area of global market must be regarded and monitored closely. Hence, it can be argued that there is an imminent need for establishment of supportive institution to assist export process of combination of stage equipments and special lighting service. This study emphasizes the need to improve export process of stage equipments, special lighting services as well as other related products and services which have been focused in domestic market only until now. Further, it also analyzed the potential prospect of such direction reconciling current crisis our manufacturing industry is facing. Even though it maybe regarded as one of the niche market for export of Korea in the short term view, stage equipment and special lighting service industry may rapidly grow as the global cultural industries have grown along with the increase of national income earnings overall. Due to such advantageous features, it can be expected that such industries will show strong growth in the near future. After analyzing the fact that Korea's plants (eg. powerplants) export sector is at its boom, there is a need to transform stage equipment and special lighting service export market into a primary market from a secondary(niche) market for SMEs. This study is viewed from the Korean economic and export sector aspect in the aim of seeking a solution to conquest our realistic limit in our export sector by developing a suitable export model. There have been cases of very few attempts to expand abroad by SMEs who have failed miserably due to their failure to adapt to foreign culture, practice and languages as well as substantial lack in experience in export marketing. Despite this, neglecting our manufacturing industry as it is which is showing its limit and problems is out of option therefore, it is imminent that we come up with an effective measure to address this problem and service export can be suggested as one of them. This study reveals manufacturing-service export model of stage equipment and special lighting service and its related areas is recognized as a field with a very strong future and furthermore, it is expected to bring synergy effects in manufacturing and services sector as well. Further, the operation strategy contains combination, composition and fusion(convergence) of manufacturing and service sectors which could derive various of export products which displays greater success probability or this export model. The outcome of this research is expected to become a useful source for enterprises related to such industry which are seeking a possible global expansion. Furthermore, it is also expected to become a catalyst which fastens the process of global expansion and not only that, we are firmly assured that this study will become an opportunity to improve our current policies and institutions related to this area's export market.

Remediation of Heavy Metal Contamination in OBOD Site with Soil Washing : Selection of Extractants (토양세척법에 의한 중금속오염 폐탄처리장 토양의 정화 : 세척액의 선정)

  • The efficiences of mineral acid (HCl), neutral salts ($CaCl_2$), and chelating agent (citric acid and $Na_2$-EDTA) were tested for extracting heavy metals from open burning and open detonation (OBOD) site soil. The extraction efficiencies of Cd, Cu, Pb and Zn from soil for various extractants were in the order of HCl > citric acid > $Na_2$-EDTA > $CaCl_2$, HCl (1.0 M) extracted effectively 82%, 86%, 80%, and 46% of initial total concentrations of Cd, Cu, Pb, and Zn, respectively. Significant negative correlations were observed between pH of extractant and amount of extracted heavy metals. Initially, examined heavy metals were predominantly bound to carbonate and Fe, Mn-oxide fraction. Though the significant amount of carbonate and Fe, Mn-oxide bounded metals were removed but a significant amount remained metals shifted to exchangeable (more mobile) fraction by HCl and citric acid extraction. The increased mobility of remaining metals could be problematic for water resources, thus careful management is needed to control the movement of heavy metals.

Skin Whitening and Skin Immune Activities of the Peptides Isolated from Crassostrea gigas by Ultrasonification Processes (초음파 복합처리를 통한 참굴 펩타이드의 피부미백 및 피부면역 활성)

  • In this study, peptides were isolated from Crassostrea gigas using an ultrasonification process at $40^{\circ}C$. The yield of the peptides was greater than 34%, and their cytotoxicity was found to be less than 22.8% against several cell lines that were treated with the extracts at a dose of 1.0 mg/mL. In addition, the tyrosinase inhibitory and melanin synthesis of the peptides isolated from Crassostrea gigas were also evaluated to determine if they could be used as a potential cosmetic agent. The peptides were found to significantly inhibit the melanin synthesis of the clone M-3 cell line by up to 62.7%. The inhibitory activities of the tyrosinase were observed 34.51% in ascorbic acid, 42.49% in extract with the ultrasonification at $40^{\circ}C$ and 35.37% in $40^{\circ}C$ extract at 1.0 mg/mL concentration, respectively. Finally, when samples were treated with the peptide extracts at a concentration of 0.6 mg/mL, PGE2 expression was significantly decreased. Taken together, these results indicate that Crassostrea gigas may be a source of cosmetic agents capable of improving physiological hyperpigmenting and immuno-modulating skin disorders.

Effect of Task-irrelevant Feature Information on Visual Short-term Recognition of Task-relevant Feature (기억자극의 과제 무관련 세부특징 정보가 과제 관련 세부특징에 대한 시각단기재인에 미치는 영향)

  • The summed-similarity model of visual short-term recognition proposes that the estimated amount of summed similarity between remembered items and a recognition probe determines recognition judgement decision (Kahan & Sekuler, 2002). This study examined the effect of a task-irrelevant location change on the recognition decision against two remembered Gabor gratings differing in their spatial frequencies. On each trial in Experiment, participants reported if two gratings displayed across the visual fields are the same or not as the probe grating displayed after about a second of memory delay. The probe grating would be the same as or different from the memory items (lure) by 1 or 4 JND units. The location of the probe would also vary randomly across the left and right visual field with respect to the location of the corresponding memory item. The participants were instructed to perform their recognition task exclusively to the spatial frequencies of the memory items and the probe while ignoring the potential location change of the probe. The results showed that false-recognition rates of the lure probe increased as the summed similarity between the memory items and the probe increased. The rates also further increased in the condition where the probe location was different from the location of the corresponding memory item compared to the condition where the probe location was the same. The increased false-recognition rates indicate that information stored into visual short-term memory is represented as a form of well-bound visual features rather than independent features.

A Possible Mechanism Related with Non-spinning Syndrome of Bombyx mori that Intimidates the Sericultural Industry in Northern Kyungbuk (경북 북부지역의 양잠산업에 피해를 주고 있는 누에(Bombyx mori) 미화용 기작에 관한 연구)

  • Non-spinning syndrome of Bombyx mori has been serious issue in sericulture industry near Kyungbuk area. This study was focused on the analysis of the mechanism and on screening candidate chemicals inducing the anti-metamorphosis of the silkworms. Rearing temperatures or initial body weight of the final instar larvae did not affect a normal larval to pupal metamorphosis of B. mori. However, pyriproxyfen (a juvenile hormone (JH) agonist) induced follicle patency significantly even at its 10$\^$-8/ M concentration and inhibited metamorphosis of B. mori in both developmental time and dose dependent manners. Pyriproxyfen induced JH esterase (JHE) activity and downregulated expression of JH binding protein of 5. mori. These results suggests that pyriproxyfen induced JHE activity as a JH agonist and that the elevated JHE activity degraded endogenous JH and resulted in JHBP gene expression. Based on the fact that the JH agonist induced follicle patency and inhibited metamorphosis of B. mori, follicle patency bioassay suggested that three commercial pesticides including simazine, molinate or alachlor were proved to give potent JH agonistic effect on B. mori. Further direct exposure experiments to these candidates are required to determine the chemicals responsible for the non-spinning syndrome of 8. mori.

Properties of Compressive Strength of Mortar Based on High-activated Blast Furnace Slag using the Slag by-product as an Activator (슬래그부산물을 자극제로 활용한 고활성 고로슬래그 미분말 모르타르의 압축강도 발현 특성)

  • Recently, many efforts related to the utilization of industrial by-products have been made to reduce carbon dioxide emissions in the construction industry. Of these various efforts, concrete incorporating ground granulated blast furnace slag (BFS) provides many advantages compared to conventional concrete, such as high long-term compressive strength, improved durability and economic benefits because of its latent hydraulic property, and low compressive strength at early curing age. This paper investigates the compressive strength of high-activated ground granulated blast furnace slag blended mortar with slag by-product S type(SBP-S). The results of the experiment revealed that incorporating high-activated ground granulated blast furnace slag would affect the compressive strength of mortar. It was found that increasing the Blaine fineness and replacement ratio of slag by-product S type shows high compressive strength of mortar at early curing age because of its high $SiO_2$ and CaO contents in the slag. It is confirmed that an increase of curing age does not affect the compressive strength of mortar made with slag by-product S type at a high curing temperature. Moreover, it is possible to develop and design concrete manufactured with high-activated ground granulated blast furnace slag as binder considering the acceleration curing conditions and mix proportions.

Computational Chemistry Study on Gas Hydrate Formation Using HFC & HCFC Refrigerants (R-134a, R-227ea, R-236fa, R-141b) (수소불화탄소 및 수소염화불화탄소 냉매(R-134a, R-227ea, R-236fa, R-141b)를 이용한 가스 하이드레이트 형성에 관한 계산화학적 해석)

  • Although the desalination technique using gas hydrate formation is at a development stage compared to the commercially well-established reverse osmosis (RO), it still draws attention because of its simplicity and moderate operational conditions especially when using refrigerants for guest gases. In this study, DFT (density functional theory)-based molecular modeling was employed to explain the energetics of the gas hydrate formation using HFC (hydrofluorocarbon) and HCFC (hydrochlorofluorocarbon) refrigerants. For guest gases, R-134a, R-227ea, R-236fa, and R-141b were selected and three cavity structures ($5^{12}$, $5^{12}6^2$, and $5^{12}6^4$) composed of water molecules were constructed. The geometries of guest gas, cavity, and cavity encapsulating guest gas were optimized by molecular modeling respectively and their located energies were then used for the calculation of binding energy between the guest gas and cavity. Finally, the comparison of binding energies was used to propose which refrigerant is more favorable for the gas hydrate formation energetically. In conclusion, R-236fa was the best choice in terms of thermodynamic spontaneity, less toxicity, and low solubility in water.

A component based framework for service development of ubiquitous healthcare (유비쿼터스 서비스 개발을 위한 컴포넌트 기반의 서비스 개발 프레임워크)

  • 유비궈터스 헬스케어는 단일화된 서비스가 아니라 다양한 기술들이 복합적으로 결합되어 운용되는 서비스이다. 따라서 서비스의 형태가 고정적이지 않고 매우 다양하게 나타난다. 하지만 실제로 차이가 발생하는 부분은 서비스의 구현에 관한 세부적 내용에서 나타나고, 서비스 운용을 위한 기본 구성요소에 있어서는 큰 차이가 없이 유사한 형태를 가진다. 그 결과 유비쿼터스 헬스케어 서비스 개발 과정에서는 실제 서비스의 구현 외의 통신과 데이터베이스의 이용, 메시지 전달과 같은 중복되는 항목에 대한 고려가 매번 이루어져야 한다. 이것은 개발 과정에 있어 불필요한 비용의 증가를 불러온다. 본 논문에서는 이와 같은 불필요한 비용을 감소시키며 서비스의 개발과 운용이 가능한 유비쿼터스 헬스케어 서비스의 제공을 위한 아키텍처와 서비스 개발을 위한 프레임워크를 제안한다. 제안하는 서비스 제공 아키덱처는 크게 이용자 단말, 유비궈터스 헬스케어 서비스 센터, 외부 기관으로 구성된다. 서비스 개발 프레임워크는 서버와 클라이언트 프레임워크로 구분된다. 서비스 개발 프레임워크는 서비스를 제공하는 서버에서 필요한 유비쿼터스 헬스케어 서비스의 공통 구성요소를 가진다. 서비스의 개발을 위해 우선 프로세스에 대한 정의를 수행하고, 정의된 내용에 따라 필요한 코드 템플릿을 결합하여 서비스의 초기 형태를 만들어낸다. 여기에 각 서비스가 필요로 하는 세부 사항을 작성하는 것으로 서비스의 개발을 수행하게 된다. 제안된 서비스 제공 아키텍처와 서비스 개발 프레임워크를 실제 적용해보기 위해 전림선비대증 환자 진료를 위한 시스템을 설계하고 구현하였다.JSHOP2 계획수립기내에 구현하였다. 계획 실행 방법으로는 주어진 강건한 계획에 대하여 행위들이 직접 실행하수 있도록 한다.며 용량에 의존하는 양상을 보였다. $H_2O_2$에 의해 유발(誘發)된 DNA의 손상은 catalase와 deferoxamine에 의해 억제되었지만 DPPD는 억제시키지 못했다. 배기음(排氣飮)은 $H_2O_2$에 의해 유발(誘發)된 ATP의 소실을 회복시켰다. 이러한 실험결과 $H_2O_2$에 의해 유발(誘發)된 세포(細胞)의 손상(損傷)은 지질(脂質)의 과산화(過酸化)와는 다른 독립적인 기전에 의해 일어남을 나타낸다. 결론 : 이러한 결과들로 볼 때 Caco-2 세포(細胞)에서 배기음(排氣飮)이 항산화작용(亢酸化作用)보다는 다른 기전을 통하여 Caco-2 세포안에서 산화제(酸化劑)에 의해 유발(誘發)된 세포(細胞)의 사망(死亡)와 DNA의 손상(損傷)을 방지할 수 있다는 것을 가리킨다. 따라서 본 연구(硏究)는 배기음(排氣飮)이 반응성산소기(反應性酸素基)에 의해 매개된 인체(人體) 위장관질환(胃腸管疾患)의 치료(治療)에 사용할 수 있을 가능성(可能性)이 있음을 제시하고 있다.에 이를 이용하여 유가배양시 기질을 공급하는 공정변수로 사용하였다 [8]. 생물학적인 폐수처리장치인 활성 슬러지법에서 미생물의 활성을 측정하는 방법은 아직 그다지 개발되어있지 않다. 본 연구에서는 슬러지의 주 구성원이 미생물인 점에 착안하여 침전시 슬러지층과 상등액의 온도차를 측정하여 대사열량의 발생량을 측정하고 슬러지의 활성을 측정할 수 있는 방법을
