Determination of Nitrite-scavenging Activity of Seaweed (해조류에 의한 아질산염 소거활성 분석-연구노트-)

  • Nitrite plays an important role in the formation of carcinogenic nitrosamine. In order to effectively inhibit the formation of carcinogenic nitrosamine in food and biological systems, scavenging of residual nitrite is necessary. In screening test of nitrite-scavenging effect of seaweed, The methanol extract from Phaeophyta was more effective than those from Rhodophyta and Chlorophyta. Among the brown algae family, Laminariaceae, A. cribrosum (90.4$\%$) and E. cava (80.1$\%$), E. stolonifera (79.0$\%$) that belong to genus Ecklonia showed marked nitrite-scavenging effect at pH 1.2. Furthermore, these algae showed potent reducing power and radical scavenging activity. The nitrite-scavenging effect was correlated with level of reducing power.

  • Ecklonia cava which is a kind of phaeophyta and belongs to Laminariales Alariaceae is a perennial seaweed distributed over Jeju and Dokdo islands. The purpose of this study evaluate antioxidant activities of Ecklonia cava crackers. Crackers were prepared with different measurements (0, 2, 4, 6, 8 g) of Ecklonia cava; indicated as S1, S2, S3, S4 and S5, and are used to examine and compare antioxidant activities, and quality characteristics. Ecklonia cava powder was used, after being extracted with 70% ethanol in hot water and done in freeze dehydration. To examine antioxidant activities of Ecklonia cava and cracker, DPPH radical scavenging activity, total phenolic and reducing power were tested. Total phenolic content was 36.86 mg GAE/g. DPPH radical scavenging activity and reducing power showed good vitality as amounts of Ecklonia cava powder increased (p<0.001). pH level decreased as amounts of power increased. L (lightness) values and b (yellowness) values decreased with an increased concentration of Ecklonia cava powder (p<0.001), whereas (redness) values didn't show tendency. S5 indicated the lowest spreadness. According to sensory evaluation, S4 (6 g) was preferred the most in terms of color, flavor, taste, appearance, texture, and overall palatability. Results suggest that Ecklonia cava powder has antioxidant effect and is useful as a functional food resource.

Effect of Ecklonia cava Extracts Supplementation on cognitive ability in mice (감태(甘苔)에서 추출한 Dieckol 성분이 쥐의 인지능력에 미치는 영향)

  • Objectives : The purpose of this study aimed to investigate that dieckol - isolated from Ecklonia cava - supplementation can improve cognitive ability in mice. Methods : 48-male mice(6 weeks old) were divided into four groups; High-Dieckol group(n=12), Low-Dieckol group(n=12), Placebo group(n=12), Control group(n=12) and they were administered orally 5 days per week for 4 weeks at the same time. We performed Morris water maze test, Passive avoidance test, Blood serotonin analysis. And there was examined on neurogenesis in dentate gyrus of hippocampus using 5-bromo-2'-deoxyuridine (BrdU) to label proliferating cells. Results : The results are as follows; As a Morris water maze results, Trial duration was significantly decreased in high dieckol group comparing to placebo group and control group. Distance to target was significantly decreased in high dieckol group and low dieckol group comparing to placebo group and control group. Mean speed was significantly low in high dieckol group comparing to low dieckol group, placebo group and control group. As a Passive avoidance test results, latency time was significantly long in high dieckol group comparing to low dieckol group, placebo group and control group. BrdU cell count was significantly high in high dieckol group comparing to low dieckol group, placebo group and control group. Conclusions : As a conclusion, it is considered that dieckol supplementation might improve learning and cognitive ability.

Effect of Dietary Supplementation with Alga (Hizikia fusiformis and Ecklonia cava) on the Non-specific Immune Responses of Parrot Fish Oplegnathus fasciatus (사료 내 해조류(톳, 감태) 첨가가 돌돔(Oplegnathus fasciatus) 치어의 비특이적 면역반응에 미치는 영향)

  • Two feeding trials were conducted to determine the effects of dietary supplementation with Hizikia fusiformis or Ecklonia cava on the non-specific immune responses of parrot fish Oplegnathus fasciatus. Fish were fed experimental diets to which H. fusiformis or E. cava powder were added to final concentrations of 0, 2, 4 and 6%, respectively. After feeding for two weeks, phagocytic activity was significantly higher in fish fed diets containing H. fusiformis, but not E. cava, than in fish fed the basal diet. Lysozyme activity was significantly increased in the fish fed diets containing 6% H. fusiformis and E. cava. Myeloperoxidase activity was also significantly higher in fish fed diets containing 2 and 4% H. fusiformis, as compared to the basal diet, but not in those fish fed E. cava. These two studies indicate that dietary supplementation with H. fusiformis or E. cava could enhance the innate immune responses of parrot fish during their growth stage.

  • We have reported the seasonal and portional variation in the composition of uronic acid of alginic acid from Ecklonia cava in the previous study. In the present paper, uronic acid block structure of alginic acid from Ecklonia cave was investigated, and some related properties such as viscosity, the dependence on temperature and substitution of metallic ion were also discussed. The proportion M block was highest amnog three blocks in both of frond and stipe. The average values of M and G block ratios were $51.6|%\;and\;32.3\%$ in the frond, whereas $45.7\%\;and\;38.6\%$ in the stipe, respectively. The proportions of alternating blocks in the frond and stipe were similar, and the values were $16.1\%$ in the frond and $15.6\%$ in the stipe. The viscosity of $1\%$ sodium alginate solution was almost inversely proportional to the M/G and/or M block ratio. Viscosities were marked maximum from summer to autumn and minimum winter to spring. In the value of lower M/G and M block ratio, the viscosity seemed to have a considerable dependence on temperature. The temperature dependence was more appearant in the alginic acid from the frond (M/G ratio, 0.95; M block, $54.2\%$) collected in July and the stipe (M/G ratio, 1.61; M block $47.6\%$) in April. The affinity with metallic ion appeared higher in order of $Pb^{2+}>Cu^{2+}>Co^{2+}>Zn^{2+}$. The amounts of metallic ion exchange of the $Pb^{2+},\;Cu^{2+},\;Co^{2+}\;and\;Zn^{2+}$ were ranged from 3.4. to 4.7, 2.5 to 3.2, 1.8 to 2.2 and 1.6 to 1.7 meq/(g. sodium alginate), respectively.

Development and Characterization of High Quality Salted Mackerel Using Enzyme Hydrolysates of Ecklonia cava (감태 효소 가소분해물을 이용한 고품질 간고등어의 개발 및 특성)

  • This study was conducted to develop and characterize of a high quality salted mackerel using enzymatic extracts of Ecklonia cava (EEC). In this study, potential antioxidative properties of EEC were evaluated by DPPH free radical scavenging activity, hydrogen peroxide scavenging activity, peroxide value, and fatty acid composition, and the antimicrobial properties were also measured by analysis for volatile basic nitrogen, pH, viable cells, Eschericia coli and biogenic amine. Compared to EEC-untreated salted mackerel, the salted mackerel with EEC was superior in antioxidative properties, while was negligible in the difference of antimicrobial properties. These results suggested that the high quality salted mackerel with antioxidative activity could be developed by treatment of EEC.

The Effects of Anti-Obesity on Enzyme-treated Ecklonia cava Extracts (효소 처리한 감태(Ecklonia cava) 분획물의 항비만 효과)

  • Obesity is a worldwide problem that contributes to serious diseases, including diabetes, hypertension, and atherosclerosis. Recently, much research has examined functional natural materials and their anti-obesity activity. This study investigated the effect of enzyme-treated Ecklonia cava extracts on mice fed a high fat diet. To test the anti-obesity effects of a diet containing the enzyme-treated E. cava extracts (EEc), C57BL/6NTacSam mice were divided into six groups : normal diet (ND), high-fat diet (HFD), high-fat with Garcinia extract diet (GHD), and three high-fat with EEc diet (EHD250, EHD500, and EHD1000) groups. After 9 weeks, body weight was increased significantly in the HFD group compared to all of the EHD groups, and the weights of the liver, perirenal fat and epididymal fat paralleled the increase in body weight. The serum GOT, GPT, triglyceride, total cholesterol, and LDL-cholesterol levels were lower in the EHD1000 group than in the HFD group. The glucose and leptin concentrations were lowest in the EHD1000 group and C/EBP family expression was decreased in a dose-dependent manner. These results indicate that E. cava extracts not only have anti-oxidation functions but also anti-obesity effects.

Interactive Effects of Increased Temperature and pCO2 Concentration on the Growth of a Brown Algae Ecklonia cava in the Sporophyte and Gametophyte Stages (갈조류 감태(Ecklonia cava)의 포자체와 배우체 생장에 영향을 주는 수온과 pCO2 농도의 상호작용)

  • To examine the effects of increased $CO_2$ concentration and seawater temperature on the photosynthesis and growth of forest forming Ecklonia cava (Laminariales, Phaeophyta), sporophytic discs and gametophytes were cultured under three $pCO_2$ concentrations (380, 750, 1000 ppm), four temperatures (5, 10, 15, $20^{\circ}C$ for sporophytes; 10, 15, 20, $25^{\circ}C$ for gametophytes), and two irradiance levels (40, $80{\mu}mol$ photons $m^{-2}s^{-1}$) for 5 days. Photosynthetic parameter values ($ETR_{max}$, $E_k$, and ${\alpha}$) were generally higher as sporophytic discs were grown under low temperature and increased $CO_2$ concentration at 750 ppm. However, photosynthesis of Ecklonia sporophytes was severely inhibited under a combination of high temperature ($20^{\circ}C$) and 1000 ppm $CO_2$ concentration at the two photon irradiance levels. The growth of gametophytes was maximal at the combination of 380 ppm (present seawater $CO_2$ concentration) and $25^{\circ}C$. Minimal growth of gametophytes occurred at enriched $pCO_2$ concentration levels (750, 1000 ppm) and high temperature of $25^{\circ}C$. The present results imply that climate change which is increasing seawater temperature and $pCO_2$ concentration might diminish Ecklonia cava kelp beds because of a reduction in recruitments caused by the growth inhibition of gametophytes at high $pCO_2$ concentration. In addition, the effects of increased temperature and $pCO_2$ concentration were different between generations - revealing an enhancement in the photosynthesis of sporophytes and a reduction in the growth of gametophytes.

Effects of Temperature and Light Intensity on the Gametophyte Fragment Growth of Ecklonia cava Kjellman (Laminariales, Phaeophyta) (대형 갈조류 감태(Ecklonia cava Kjellman) 배우체 단편의 생장에 미치는 온도와 조도의 영향)

  • We examined the effects of temperature and light intensity on gametophyte growth of Ecklonia cava Kjellman. The growth of female and male gametophytes was investigated before maturation. Gametophytes prepared from Munseom, Jeju, Korea in October 2014 were separated by sex and maintained under the following conditions: 15℃, 20 μmol·m-2·s-1, and a 12-h/12-h light/dark cycle. The experiments were conducted at five temperatures (10, 15, 20, 25, and 30℃) and three light intensities (5, 20, and 40 μmol·m-2·s-1). Daily growth rates were estimated by the increase in filamentous frond area. The optimal growth conditions for female gametophytes were 20℃ and 40 μmol·m-2·s-1; the optimal conditions for male gametophytes were 15℃ and 20 μmol·m-2·s-1. The relative growth rates (RGR) of female and male gametophytes decreased at 25℃ and 30℃, respectively. At 30℃, RGRs of gametophytes decreased by approximately 1.0% day-1. In addition, RGRs of male gametophytes were higher than those of female gametophytes. These results indicate that female gametophytes were more sensitive to temperature and light intensity than male gametophytes. Moreover, these results suggest that E. cava growth rates could decrease as a result of global warming.

Regeneration and Maturation Induction for the Free-living Gametophytes of Ecklonia cava Kjellman (Laminariales, Phaeophyta) (대형갈조류 감태(Ecklonia cava Kjellman) 유리배우체의 재생 및 성숙 유도)

  • Induction of regeneration and maturation for the free-living gametophytes of Ecklonia cava Kjellman was conducted under various temperature, irradiance and photoperiod conditions. Culture conditions for female or male gametophyte fragments were 4 temperature (5, 10, 15 and $20^{\circ}C$), 4 irradiance (5, 10, 20 and $40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and 3 photoperiod (14:10, 12:12 and 10:14 h L:D). Female and male gametophyte fragments were maintained in active regeneration without reaching sexual maturity under $5{\sim}10^{\circ}C$, $5{\sim}10{\mu}mol{\cdot}m-2{\cdot}s-1$, 12:12h or 10:14h (L:D) and $15-20^{\circ}C$, $20-40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 14:10h (L:D), respectively. Sexual maturation of female and male gametophytes facilitated under $15^{\circ}C$, $20-40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 14:10h (L:D) and $5-10^{\circ}C$, $5-10{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, 10:14h (L:D), respectively. These results provide basic informations to control the regeneration or maturation of the free-living gametophytes for artificial seed production of E. cava.
