Prevalence of Anaplasma sp. in Thrushes (Family Turdidae) in Jeju Island, Republic of Korea (제주도와 마라도내 지빠귀과 조류에서 Anaplasma spp. 감염 조사)

  • Anaplasmosis is a rickettsial zoonosis mediated by blood-sucking arthropods, such as ticks, flies, and mosquitos. Migratory birds are common hosts of ticks that are mediators of anaplasmosis, in particular, the tick infection rate in thrushes (family Turdidae) has been known to be high. The main purpose of this study is to survey the occurrence and prevalence of Anaplasma spp. from the migratory thrushes in Jeju island. We collected blood samples from 6 thrushes rescued at the Jeju Wildlife Rescue Center and from 34 wild-caught thrushes on Mara island which is a satellite island of Jeju. As a result, the nested PCR confirmed that seven out of 40 individuals (17.5%) were infected by Anaplasma spp. and all of them were identified as A. phagocytophilum based on sequences obtained from partial 16S rRNA. All the infected birds were on their northward migration in spring, our results suggest that the Turdidae family, which is a common and abundant migrant group passing through Jeju island, may act a role as active reservoir and disperser of A. phagocytophilum causing potential influx of the zoonotic pathogens from its wintering grounds in lower latitude to the mainland Korea as well as Jeju.

A ten-year retrospective study of bovine infectious disease agents occurred in Korea from 2010 to 2019 (최근 10년간 국내 소 질병 원인체에 관한 문헌적 고찰)

  • For estimating the prevalence of bovine infectious disease agents, the pathogens were classified as follows: the digestive disease agents, respiratory disease agents, reproductive disease agents, and tick-borne disease agents. This study covered 81 published papers regarding bovine infectious diseases in Korea that determined the presence of diverse pathogens or the antibodies elicited by the infectious agents in cattle from 2010 to 2019. In total, 59,504 cows were involved in the papers reporting the causative agents in their cases. The disease prevalence for the digestive, respiratory, reproductive, and tick-borne cases was 9.0%, 13.4%, 10.4%, and 7.8%, respectively. Bovine viral diarrhea virus, Escherichia coli, Mycobacterium avium subsp. paratuberculosis, and Eimeria spp were more significantly prevalent in the cows under one-year age than over one-year age. Bovine viral diarrhea virus, Escherichia coli, Mycobacterium avium subsp. paratuberculosis, and Anaplasma spp. were more significantly prevalent in Hanwoo than dairy cattle. Coxiella burnetii, Neospora caninum, and Theilieria spp. were more significantly prevalent in dairy cattle than Hanwoo. Tick-borne disease agents were more prevalent in cows grazing than the case in housing. Our analytic data obtained from this study emphasize the need for more studies on the occurrence of these pathogens according to the breed, age, and the region, to come up with bovine infectious disease control measures in Korea.

  • Korea which has not only beautiful scenery but long history in the world deserves to be a pride to all the koreans. It has prospered flourishing culture through the long history and preserved a lot of cultural properties all around the nation. Traditional and cultural properties should be preserved and developed induding those disappeared in the past. The economical competitiveness in provincial area is descending compared with that of international market places. The alternatives to save and promote provincial economy should be proposed and developed. The researches and studies which are active to make the traditional, cultural properties into sightseeing resources will contribute to save the provincial economy.

Flow Cytometric Characterization of Lymphocyte Subpopulations in the Korean Bovine (한우 혈액내 백혈구아군의 특성 및 말초혈액상)

  • Characterization of immune cell subpopulations in the bovine was performed using a direct immunofluorescence technique adaptable for routine and repeated monitoring. This whole blood procedure is faster and requires less volume than conventional density gradient isolation methods. Low inter-and intra-animal variations were seen in hematology parameters and in CD4, CD8 and CD2 lymphocyte subtypes. CD4 values were 30% of lymphocytes in male and 32% in females. Thriteen percent were CD8 in males and 13% in females. CD4: CD8 ratios were approximately two in both sexes. Fiffty three percent were CD2 in males and 54% in females. The mean RBC counts of peripheral blood were 7.20$\times10^6/{mm}^3$ for male cattle and 6.36$\times10^6/{mm}^3$ for females. The mean WBC counts were 8.09$\times10^3/{mm}^3$ for males and 7.09$\times10^3/{mm}^3$ for females. The percent of lymphocytes(63-65%) was higher than the percent of neutrophils(17-18%), the percent of eosinophils(11-15%), te percent of monocytes(4-5%), and percent of basophiles(1%).

Study on disease prevalence to Holstein calves reared in Chonnam area (전남 지방의 홀스타인 송아지의 질병 발생율 조사)

  • The prevalence of major calf disease was investigated in 117 Holstein dairy calves in Chonnam area. All of them were moved in the College experimental farm which is operated in intensive units. clinical signs were daily examined throughout two months after the introduction of the College farm. Among calves, 92 cases(78.6%) died in the two months after the introduction in it. Outbreaks of respiratory and alimentary diseases were their main causes of their fatality. The incidence of respiratory disorders during the full period of the experiment was up to 42.8%, and the alimentary diseases were occurred 35.9% of the herd. Most of the mortality was related with respiratory(59.9%) and alimentary(52.1%) pathogens. Also calf mortality by combined infection claimed 6.6% among 100 morbidity cases. Principle pathogens to cause mortality were Pasteurella spp(44.4%), E coli(29.9%), bovine viral diarrhea virus(16.2%), IBRV(12.0%), respectively. Viruses also played as an important role in increasing calf morbidity to secondary respiratory bacterial pathogens. Pasteurella infection combined with infectious bovine rhinotracheitis virus(11 cases), parainfluenza virus type-3(9 cases), or bovine respiratory syncytial virus(7 cases) was appeared as major pattern to mortality. colibacillosis in causing enteritis was concurrently infected with BVD(19 cases), bovine coronavirus infection(14 cases), salmonellosis(5 cases), coccidiosis(5 cases) and clostridial infection(4 cases). Ninty-two cases to death were appeared to have 100% neutralizing antibodies to BCV; Among them, 73.8% had the neutralizing antibody level higher than 64. Calves with neutralizing antibodies higher than 16 to BVDV were 50%. The cases with neutralizing antibody level lower than 8 to BEFV were 89.4% that means the necessity of appropriate vaccination.

Characterization of Phytase from Bacillus coagulans IDCC 1201 (Bacillus coagulans IDCC 1201이 생산하는 Phytase의 특성)

  • A native extracellular acid phosphatase, phytase (EC 3.1.3.8), from Bacillus coagulans IDCC 1201 (commercially known as Lactobacillus sporogenes) used as probiotics, was characterized. Though some strains of B. coagulans have been evaluated with regard to several health-promoting effects, it has not been reported to produce phytase. Partially purified phytase front the strain IDCC 1201 had a pH optimum of 4.0 and a temperature optimum of $50^{\circ}C$, respectively. The requirement for divalent cations was studied and cobalt ion remarkably increased the enzyme activity. The removal of metal ions from the enzyme by EDTA decreased activity below 50%. The enzyme activity depleted restored when the assay was performed in the presence of $Co^{2+}$. Also, $Co^{2+}$ is the most active stimulator and has unique activation effect at high temperature. The phytase was specific for sodium phytate and p-nitrophenylphosphate, which is different from other known Bacilli phytases. The putative amino acid sequences of the phytase from B. coagulans IDCC 1201 were very similar to that of the phytase from B. subtilis strain 168. Based on these data, we concluded that the phytase from B. coagulans IDCC 1201 is a $Co^{2+}$-dependent acid phosphatase. Therefore, the strain B. coagulans IDCC 1201 is thought to be a valuable addititive for livestocks as well as a beneficial probiotics for human.

  • Study the gene about economical characteristic of human disease or domestic animal is a matter of grave interest, preserve and elevation of gene of Korea cattle is key subject. Studies have been done on the gene of Korea cattle using EST based SNP map, but it is based on statistical model, therefore there are difference between real position and statistical position. These problems are solved using both EST_based SNP map and Gene on sequence by Lee et al. (2009b). We have used multifactor dimensionality reduction(MDR) method to study interaction effect of statistical model in general. But MDR method cannot be applied in all cases. It can be applied to the only case-control data. So, method is suggested E-MDR method using CART algorithm. Also we identified interaction effects of single nucleotide polymorphisms(SNPs) responsible for average daily gain(ADG) and marbling score(MS) using E-MDR method.

  • Veterinary drugs have been used to prevent livestock diseases for many years. In spite of having advantages, sometimes indiscriminate application, overdose and abuse may cause risk for human. Therefore, management and risk assessment of veterinary drugs become essential to ensure food safety. So the National Veterinary Research & Quarantine Service (NVRQS) impose on registration for use of veterinary drugs also Korea Food and Drug Administration (KFDA) fixed the maximum residue limits (MRLs) for veterinary drugs in food. Basically, KFDA set MRLs based on the veterinary drugs residual data. Moreover, KFDA estimates the theoretical maximum daily intake (TMDI) with food consumption data and population mean body weight. Recently, 116 veterinary drug MRLs including banned veterinary drugs have been established and other 32 veterinary drugs MRLs will be established within 2009. In 2010, MRLs among antibiotics and synthetic antibacterial agents in livestock fishery products (including milk and eggs) and honey (including royal jelly and propolis) which have not been set in Korea Food Code and Codex Alimentarius Commission (CAC) will be regulated as uniform limit (0.03 mg/kg). In future, veterinary drugs will be controlled strictly to strengthen public health by improving analytical method.

Study on gross finding of lung lesions and causative pathogens of porcine respiratory disease complex from slaughtered pigs in Incheon (인천지역 도축돈에서 돼지호흡기질병복합감염증의 육안적 폐병변과 원인체에 관한 연구)

  • The purpose of this study was to investigate association with gross lesions and causative pathogens of porcine respiratory disease complex (PRDC) including porcine circovirus type 2 (PCV2), porcine reproductive and respiratory syndrome virus (PRRSV), swine influenza virus (SIV), Mycoplasma hyopneumoniae (MH), Pasteurella multocida (PM), Actinobacillus pleuropneumoniae (APP), Haemophilus parasuis (HP) in slaughtered pigs. A total of 1,200 lung samples were collected randomly from slaughtered pigs in Korea during August of 2010 through July of 2011. The gross lesions were classified according to the six stages (0, 1~10, 11~20, 21~30, 31~40 and ${\geq}41$, unit=%) and 48 samples from each stage were selected to detect viral and bacterial pathogens. The results according to the six stages were 100 (8.3%), 259 (21.6%), 326 (27.2%), 213 (17.8%), 144 (12.0%) and 158 (13.2%) cases, respectively. Prevalence of pneumonia according to season was 87.0~96.7% and the highest prevalence was in spring. In detection of pathogens by PCR, 53 samples were not detected any causative pathogens of PRDC. PCV2, PRRSV, SIV, MH, PM, APP serotype 2, APP serotype 5 and HP were positive in 45.5%, 12.5%, 10.4%, 60.1%, 1.7%, 13.9%, 12.2% and 15.6%, respectively. In co-infection, PCV2-MH was the most detected causative pathogens of PRDC. The detection rate of PCV2 and PRRSV was the highest in spring, of SIV, MH and HP was in winter. The detection rate of APP-2 and APP-5 had no seasonal prevalence. The more severe gross lesions increased, the higher the detection rate showed.

Recently epidemiological survey of the viral diseases of broiler chickens in Jeonbuk province from 2005 to 2007 (최근 3년간 (2005-2007년) 전북지역 육계의 주요 바이러스성 질병 발생추이 분석)

  • Recently, the major viral diseases, Newcastle disease (ND), infectious bronchitis (IB), low pathogenic avian influenza (LPAI), avian pneumovirus infection (APV), Marek's disease (MD) and infectious bursal disease (IBD), have led to huge economic losses in chicken industry of Korea. To evaluate prevalence of the major viral disease infections in broiler breeder and broiler farms, epidemiological survey has been conducted in Jeonbuk province from 2005 to 2007 by serological ELISA test for APV, PCR for MD, and RT-PCR for ND, IB, LPAI and IBD, respectively. A total of 424 cases was submitted to our laboratory for diagnosis of the major viral disease from broiler breeder and broiler farms in the above period. The diagnosed results were analysed for the detection rate of infections on basis of years, seasons and ages, respectively. This study was showed that the detection rates of ND and APV were considerably high for every years regardless of seasons and ages in both broiler breeder and commercial broiler. In comparison with detection rates of ND and APV, IB and LPAI were lower but detected around 10% for every years. Especially, detection rate of IB was significantly high in commercial broiler than in broiler breeder. Therefore, to minimize economic losses for broiler breeder and broiler farms, it will need for effective countermeasures to decrease detection rate of the viral respiratory diseases. Although the detection rates of MD and IBD were gradually decreased from 2005 to 2007 in both broiler breeder and commercial broiler, it will continually make an effort about disease control for increasing productivity in chicken industry.
