Hydrogeochemistry and Origin of $CO_2$ and Noble Gases in the Dalki Carbonate Waters of the Chungsong Area (청송 달기탄산약수의 수리지화학과 탄산 및 영족기체 기원)

  • Hydrochemical analyses, carbon isotopic (${\delta}^{13}C_{DIC}$) analyses, and noble gas isotopic ($^3He/^4He$ and $^4He/^{20}Ne$) analyses of the Dalki carbonate waters in the Chungsong area were carried out to elucidate their hydrochemical composition and to determine the source of $CO_2$ gas and noble gases. The carbonate waters have a pH of between 5.93 and 6.33, and an electrical conductivity 1950 to $3030{\mu}S/cm$. The chemical composition of all carbonate waters was Ca(Mg)-$HCO_3$, with a high Na content. The contents of Fe, Mn, and As in some carbonate waters exceed the limit stipulated for drinking water. The concentrations of major ions are slightly higher than those reported previously. The ${\delta}^{13}C_{DIC}$ values range from -6.70‰ to -4.47‰, indicating that the carbon originated from a deep-seated source. The $^3He/^4He$ and $^4He/^{20}Ne$ ratios vary from $7.67{\times}10^{-6}$ to $8.38{\times}10^{-6}$ and from 21.32 to 725.7, respectively. On the $^3He/^4He$ versus $^4He/^{20}Ne$ diagram, the noble gas isotope ratios plot in the field of a deep-seated source, such as mantle or magma. We therefore conclude that $CO_2$ gas and noble gas in the Dalki carbonate waters originated from a deep-seated source, rather than an inorganic $CO_2$ origin as suggested in a previous study.

Origin and Hydrochemical Characteristics of Natural Carbonated Water at Seoqwipo, Jeju Island (제주도 서귀포지역 천연탄산수의 기원과 수리화학특성)

  • In this study, geochemical composition, CFCs (Chlorofluorocarbons), ${\delta}^{18}O$, ${\delta}D$, ${\delta}^{13}C$ isotopes and noble gases isotopes (He, Ne) were analyzed to determine their recharge age, source of $CO_2$ gas and noble gases of carbonated hot spring water and carbonated-water samples collected in the Seoqwipo of the Jeju. The pH of the carbonated waters ranges from 6.21 to 6.84, and the high electrical conductivity range ($1,928{\sim}4,720{\mu}S/cm$). Their chemical composition is classified as $Mg(Ca,\;Na)-HCO_3$ and $Na(Ca,\;Mg)-HCO_3$ types. As a result of the calculation of groundwater age using CFCs concentrations as an environmental tracer, the carbonated water and groundwater were estimated to be about 47.5~57.2 years and about 30.3~49.5 years, respectively. The ${\delta}^{13}C$ values of carbonated water range from -1.77 to -7.27‰, and are plotted on thr deep-seated field or the mixing field of the deep-seated and inorganic origin. Noble gases isotopic ($^3He/^4He$, $^4He/^{20}Ne$) ratio shows that helium gas of carbonated hot waters comes from deep-seated magma origin.

  • Hydrochemical and carbon isotopic (${\delta}^{13}C_{DIC}$) analyses of 11 water samples, and noble gas isotopic analyses of 8 water samples collected in the Kyeoungbuk and Kangwon areas of Korea were performed to determine their hydrochemical characteristics and to interpret the source of noble gases and $CO_2$ gas in the water. The carbonated mineral waters are weakly acidic (PH = 5.59-6.04), and electrical conductivity ranges from 302 to $864\;{\mu}S/cm$. The chemical composition of all the water samples is Ca-$HCO_3$ type. The high contents of Fe and Mn exceed the safe limits for drinking water. The ${\delta}^{13}C_{DIC}$ values of the samples range from -5.30‰ to -2.84‰, indicating that the carbon is supplied mainly from a deep-seated source and to a lesser degree from an inorganic carbonate source. The $^3He/^4He$ ratios of the samples range from $1.51{\times}10^{-6}$ to $6.45{\times}10^{-6}$. The samples plot into three groups on a $^3He/^4He$ versus $^4He/^{20}Ne$ diagram: the deep-seated field (e.g., a mantle source), the atmospheric field, and the air-mantle mixing field. A wide range of $^4He/^{20}Ne$ ratios is observed ($0.036{\times}10^{-6}$ to $1.76{\times}10^{-6}$), indicating that while radiogenic $^4He$ is dominant in these water samples, mantle-origin He is also present. The supply of $CO_2$ gas and noble gases from a deep-seated source to carbonated waters is inferred to be controlled by geological structures such as faults and geological boundaries.

Optimization of a protocol for the production of transgenic lily plants via particle bombardment (유전자총 실험조건 최적화를 통한 형질전환 백합 식물체 생산)

  • Transgenic lily plants have been obtained after particle bombardment, using PDS-1000/He system and scale explants of lilies, followed by PPT (D-L-phosphinothricin) selection. In this study, scales of the lily plants cv. 'red flame' were bombarded with a plasmid containing the bar gene as a selectable marker, and the AtSIZ gene as a gene of interest, showing salt tolerance and drought tolerance respectively, and both being driven by the CaMV 35S promoter. For optimization of a protocol, factors which optimized and showed a high transformation efficiency under following conditions, were considered: a bombardment pressure of 1100 psi, a target distance of 6 cm and $1.0{\mu}m$ of gold particle, and 24-h pre-culture and post-culture on MS medium containing 0.2 M sorbitol and 0.2 M mannitol as osmoticum agents. After bombardment, all the bombarded scales of lily were transferred to MS medium without selective agents, for a week. Subsequently, these bombarded scales were transferred to a selection MS medium containing 10 mg/l PPT, and incubated for a month for further selection, after which they were cultured for another 4-8 weeks with a 4-week subculture regime on the same selection medium. After transferring into hormone-free MS medium, the PPT-resistant scales with shoots were successfully rooted and regenerated into plantlets. PCR analysis revealed that the surviving putatively transformed plantlets indicated the presence of both the bar gene and the AtSIZ gene. In conclusion, when 100 scales of lily cv. Red flame are bombarded, this study produced approximately 17-18 transgenic plantlets with an optimized bombardment protocol. The protocol described here can contribute to the breeding program of lilies.

High temperature properties of surface-modified Hastelloy X alloy (표면처리에 따른 Hastelloy X 합금의 고온물성)

  • Surface treatments and their effects on high temperature properties for the Hastelloy X, which is a promising candidate alloy for high temperature heat-transport system, have been evaluated. For TiAlN and $Al_2O_3$ overlay coatings, the two different PVD (physical vapor deposition) methods using an arc discharge and a sputtering, were applied, respectively. In addition, a different surface treatment method of the diffusion coating by a pack cementation of Al (aluminiding) was also adopted in this study. To achieve enhanced thermal oxidation resistance at $1000^{\circ}C$ by suppressing the inhomogeneous formation of thick $Cr_2O_3$ crust at the surface region, a study for the surface modification methods on the morphological and structural properties of Hastelloy X substrates has been conducted. The structural and compositional properties of each sample were characterized before and after heat-treatment at $1000^{\circ}C$ under air and He environment. The results showed that the Al diffusion coating showed the more enhanced high temperature properties than the overlay coatings such as the suppressed thick $Cr_2O_3$ crust formation and lower wear loss.

Hydrochemistry and Noble Gas Isotopes of Groundwaters around the Fault Zones (단층대 지하수의 수리화학 및 노블가스 동위원소 특성)

  • The chemical composition and noble gas isotopes of 10 deep groundwater samples were analyzed to know the circulation of groundwaters in the Yangsan fault and the Gampo fault. The chemical types of groundwaters show the $Ca-HCO_3$ type and $Ca-SO_4(Cl)$ type, and show indistinct relationship with geology. Noble gas isotopic data of most groundwaters were plotted along the air-crust mixing line on $^3He/^4He$ vs. $4^He/^{20}Ne$ diagram, and show dominant $^3He$ of air origin except one sample that shows helium mixing of crust origin. This indicates that groundwater actively circulates along fault, and fault could not play an role of upward pathway of a deep-seated helium gas. A comparatively high $^4He$ indicates that groundwater flows in an aquifer assuring relatively enough water-rock interaction.

Geochemical Origins and Occurrences of Natural Radioactive Materials in Borehole Groundwater in the Goesan Area (괴산지역 시추공 지하수의 자연방사성물질 산출특성과 지화학적 기원)

  • The origins and varieties of natural radioactive materials, including uranium and radon-222, were examined in a drilled borehole extending to a depth of 120 m below the surface in the Goesan area. In addition to core samples, eight groundwater samples were collected at different depths, using a double packer system and bailer, and their geochemical characteristics were determined. Most of the rock samples from the drilled core consisted of granite porphyry, with sedimentary rocks (slate, carbonate, or lime-silicates) and pegmatite occurring in certain sections. The pH of samples varied from 7.8 to 8.4, and the groundwater was of a Na-$HCO_3$type. Uranium and thorium concentrations in the core were < 0.2-14.8 ppm and 0.56-45.0 ppm, respectively. Observations by microscope and an electron probe microanalyzer (EPMA) showed that the mineral containing the natural radioactive materials was monazite contained in biotite crystals. The uranium, which substituted for major elements in the monazite, appeared to have dissolved and been released into the groundwater in a shear zone. Concentrations of Radon-222 in the borehole showed no close relationship with levels of uranium. The isotopes of noble gases, such as helium and neon, would be useful for analyzing the origins and characteristics of the natural radioactive materials.

The Flow Behavior Characteristics of Methane with Phase Change at Low Heat Flux (저열유속에서 상변화를 수반하는 메탄의 유동거동특성)

  • A liquefied natural gas(LNG) in cryogenic liquid is converted back into gaseous form for distribution to residential and industrial consumers. In this re-gasification process, LNG supplies a plenty of cold thermal energy about $83.7{\times}10^4kJ/kg$. The LNG cold thermal energy is utilized for the re-liquefaction process of cryogenic fluids such as Nitrogen, Hydrogen and Helium, and ice manufacturing process and air-conditioning system in some advanced countries. Therefore, it is also necessary to establish the recovery systems of the LNG cold thermal energy around Incheon, Pyungtaek and Tongyung LNG import terminals in our country. Methane is used as working fluid in this paper, which is the major component of LNG over 85 % by volume, in order to investigate the flow behavior characteristics of LNG with phase change at low heat flux. This paper presents the effects of pipe diameters, pipe inclinations and saturation pressures on the flow boundaries of methane flowing in a cryogenic heat exchanger tube, together with those of nitrogen, propane, R11 and R134a. The outcomes obtained from this theoretical researches are also compared with previous experimental data. It was also found that the effect of pipe inclination on the methane flow boundaries was significant.

Analysis of Volatile Flavor Compounds in Cow's Milk by Purge & Trap Method (Purge & Trap 법에 의한 국내산 우유 중의 휘발성 향기성분 분석)

  • Purge & Trap method was applied to perform more simple and rapid detection for analysis of volatile flavor compounds in milk. Maximal sampling of 30 mL milk for glass flask sparger was treated by He gas purging for 2 hours. Reported major volatile compounds were detected by GC-MS after 2 hours absorption and desorbed from Purge & Trap equipped with Tenax trap. Volatile flavor compounds were analyzed by Purge & Trap and GC-MS to investigate the changes of flavor components in milk between raw and deodorized milk. Fourteen volatile compounds including acetaldehyde, ethanol, 2-propanone, dimethyl sulfide, isobutanal, 3-methyl 2-butanone, 2-butanone, 3-methyl butanal, pentanal, 3-hydroxy-2-butanone, methyl disulfide, hexanal, and 2 others were detected. Six compounds such as ethanol, dimethyl sulfide, pentanal, 3-hydroxy-2-butanone, and methyl disulfide were completely eliminated after deodorization treatment. Four compounds such as 3-methyl 2-butanone, 2-butanone, 3-methyl butanal, and an unknown compound 81 (M/sup +/) were also decreased after raw milk was deodorized. The other four compounds such as acetaldehyde, 2-propanone, hexanal, and an unknown compound (M/sup +/) were not decreased.

Anti-inflammatory effect of non-thermal atmospheric pressure plasma for periodontitis treatment: in vitro pilot study (치주염 치료를 위한 저온상압 플라즈마의 항염효과: 예비 실험)

  • Purpose: The purpose of this study was to evaluate the anti-inflammatory effects of non-thermal atmospheric pressure plasma (NTP) on human gingival fibroblasts (HGFs) for clinical application of periodontal treatment. Materials and Methods: HGFs were treated with Porphyromonas gingivalis (Pg) lipopolysaccharide (LPS). Customized NTP device was developed for periodontal in vitro study. Cell viability was evaluated with cell counting kit-8. The levels of inflammatory cytokines, including interleukin (IL)-8 and 6, were determined by enzyme-linked immunosorbent assay. Results: When NTP was applied, the cell viability did not change significantly, and there was no difference for 6 h and 24h. When Pg LPS was treated to HGFs, the secretion of IL-8 and IL-6 was increased compared to the control group. But when the NTP was applied, the secretion of them was significantly decreased. Conclusion: NTP did not affect cell viability of HGFs. And it inhibited the LPS-induced production of IL-8 and IL-6.
