  • 살모넬라 갈리나륨(Salmonella gallinarum)이 원인체인 가금티프스는 모든 품종의 닭에서는 물론이고 어린 일령의 병아리에서부터 산란중인 성계에 이르기까지 전 일령의 닭에서 높은 폐사를 일으키는 세균성 질병으로 우리나라뿐만 아니라 세계적으로 발생되고 있는 질병이다.

Isolation, Identification and Cultural Condition of the Antagonistic Microorganism Against Salmonella gallinarum Causing Fowl Typhoid (가금티브스균 Salmonella gallinarum의 생육을 저해하는 길항미생물의 선발 및 동정)

  • Diarrllea and death of chicken have been brought about by fowl typhoid caused by Salmonella gallinarum, which causes a great loss of chicken farms. For the development of the probiotic which can control a fowl typhoid of S. gallinarum without any adverse effect of commercial existing antibiotics, we isolated antagonistic intestinal bacteria against S. gallinarum from a bowel of the chicken which was pastured in a chicken farm of Gumi, Kyoungbuk. An Y3 strain which had a strong antagonistic ability to S. gallinarum was selected as a candidate of chicken probiotic microorganism among isolated strains. It was identified as a Bacillus amyloliuefaciens by 98% similarity by the result of cultural, physiological, biochemical test and Biolog system$(Microlog^{TM} 4.0)$, and named as Bacillus amyloliquefaciens Y3. The strain showed the strongest antagonistic activity and a good growth at pH 5-9, $37^{\circ}C.$

  • The antagonistic microorganisms against Salmonella gallinarum causing fowl typhoid were isolated from the gut of wild pheasant. The isolated L19, L33, L50 strains were showed the characteristics of isolated Gram negative, rods, catalase positive and oxidase negative. Finally, all strains were identified as Sphingomonas sanguis by $Biolog^{\circledR}$ system. The optimal carbon sources of Sphingomonas sanguis L19, L33 and L50 for the these growth ~ere glucose, saccharose, and fructose respectively. But the optimal carbon sources of S. sanguis L19,L33, L50 for the antagonistic material production were maltose, galactose, and saccharose respectively. The optimal nitrogen sources of S. sanguis L19, L33, L50 for the growth were yeast extract, yeast extract, and $NH_4H_2PO_4$ respectively. But the optimal nitrogen sources of S. sanguis L19, L33 and L50 for the antagonistic material production were $(NH_4)_2SO_4$ urea, $(NH_4)_2S_2O_8$ espectively.

Suggestions for a better inspection method according to the occurrence of fowl typhoid in broiler breeders (육용종계 가금티프스 발생에 따른 검사방법 개선 방안)

  • Pullorum disease and fowl typhoid are septicemic avian diseases transmitting through egg by transovarian infections. A series of tests has been performing in breeding flocks of chickens and test plans for proper inspection have been modified by government veterinary institute to control of such diseases. To improve inspection plans, different test methods were compared using fowl typhoid positive samples from a poultry farm located in Jeonbuk state in 2012. Based on first inspection, 11 samples among total 200 samples were positive by rapid slide agglutination (RSA) test and 7 samples among RSA positive samples were finally diagnosed as Salmonella Gallinarum infection by ELISA, bacterial isolation, PCR, and histopathologic examination. In the second inspection, 20 samples among total 100 samples were positive by RSA test. Among RSA positive ones, 19 samples were positive by ELISA, S. Gallinarm were successfully isolated in 3 samples, and 16 samples were positive by PCR in the cecal tonsils where were not successful for bacterial isolation. Based on histopathologic examination, severe inflammation in the 13 cecal tonsils and infiltration of lymphocytes and heterophils in the 11 livers were observed. Therefore, we suggest that bacterial isolation, PCR, and histopathologic examination methods in the third inspection need to be further used in various tissues for correct diagnosis and for final eradication of pullorum disease and fowl typhoid in breeding flocks of chickens.

Examination of Economic Performance of Egg Type Breeder Lines for the Selection of Fowl-Typhoid Resistant Breed (가금티푸스 저항성 계통 선발을 위한 산란종계의 경제능력 검정에 관한 연구)

  • A preliminary experiment was conducted to produce fowl-typhoid resistant egg type breeds, using three brown egg layer breeders (ISA, Hyline, and Lohman) and two white egg layer breeders (ISA and Lohman). Various economic parameters were measured from day-old to mature stages using parent stocks of each breed. No significant difference in viabilities of mature hens from 34 to 66 wk of ages was found between brown(93.3%) and white(93.5%) egg layer breeders (P>0.05). Also, there was no difference in viabilities of mature cockerels. Although not significant, the mature body weights of brown egg layer breeders tended to be heavier than those of the white egg layer breeders both in hens and cockerels. No difference in feed intakes of mature brown and white hens were detected. Ages of sexual maturity of white and brown egg layer hens were 147.7 and 140.5 d, respectively (P<0.05). Egg production of white egg layer hens during 23-66 weeks age was significantly higher than their counterpart (81.4% vs. 75.0%). Comparisons of Haugh Unit, egg shape index, yolk color, and egg weight between brown and white eggs were not significantly different; however, the brown eggshells were thicker and stronger than those of white eggs (P<0.05).
