Electrophoresis of Proteins in the Tissues from Witches' - Broom infected Jujube Tree (빗자루병(病)에 감염(感染)된 대추나무 조직내(組織內) 단백질(蛋白質)의 전기영동(電氣泳動))

  • Protein profiles of healthy and witches' broom (mycoplasma) infected jujube plants (Zizyphus jujuba) were investigated by 2-30% linear gradient polyacrylamide gel electrophoresis. Distinct differences in band patterns between healthy and infected samples were observed. Gels from samples of healthy leaves showed a characteristic protein band in the 50kd-range, which was not detected in infected leaves. Band with 25kd was more distinct in the infected leaves, whereas band with 198kd was more apparent in the healthy leaves. Healthy-looking leaves in the infected samples demonstrated the characteristic band of 50kd with less intensity showing intermediate pattern between healthy leaves and infected leaves. In contrast with leaf samples, gels from infected stem samples showed a characteristic band in the 335kd-range which was absent in healthy stem samples.

Evaluation of Early Generation of Crosses for Incorporation of Resistance to Bacterial Spot into Sweet Pepper (감미종(甘味種) 고추에 더뎅이병(病) 저항성(抵抗性)을 도입(導入)하기 위한 교잡(交雜) 초기세대(初期世代) 검정(檢定))

  • A leading sweet pepper cultivar, Keystone Resistant Giant #3, was crossed with a line with resistance to Xanthomonas campestris pv. vesicatoria, PI271322, for incorporation of the resistance and to study the inheritance of resistance to the disease. Seedlings of parents, $F_1$, $F_2$ and backcross populations of a cross between Keystone Resistant Giant #3 and PI271322 were inoculated with race 1 and race 3 of Xanthamonas campestris pv. vesicatoria by infiltrating bacterial suspension into abaxial side of leaves. PI271322 was carrying a gene ($Bs_3$) for hypersensitive resistance to race 1 of X. c. pv. vesicatoria and also a component of non-hypersensitive resistance to race 3 inheriting in a quantitative mode. Correlation coefficient between disease indices to race 1 and race 3 of non-hypersensitive plants in $F_2$ population was highly significant, thus indicating that a component of non-hypersensitive general resistance in PI271322 acted consistently to both races. Individual plants with resistance to both race 1 and race 3 of X. c. pv. vesicatoria were selected and a breeding program for incorporation of the resistance is continued.

An Outbreak of Ibaraki Disease in Korea (한국(韓國)에서의 이바라기 병(病)(Ibaraki disease)의 발생)

  • Epizootics and sporadic outbreaks of the cattle disease characterized by fever, stomatitis and dysphagia were observed in Korea in September through November, 1982. The number of cattle showed typical symptoms of the disease was estimated to 73 and these animals were concentrated in Gyongido province, the central district of Korea. In other districts around Gyongido province the disease tended to be sporadic occurrence. The main lesions of the disease were defined as edema, hemorrhage, degeneration and necrosis of the oral mucosa and musculatures of the esophagus and laryngopharyngeal area, and abomasum. Of these lesions hyaline degeneration and hemorrhage of the striated muscle of the esophagus and laryngopharyngeal area were the most striking changes of the disease and rated to be the lesion of diagnostic significance. The serum samples of the cattle with typical symptoms showed significant positive titer of the antibodies against Ibaraki virus. On these clinical, epidemiological, pathological and serological findings of the disease it was diagnosed as Ibaraki disease presenting the only report on the epizootics in the countries other than Japan.

Comparative Study on the Long-Acting Thyroid Stimulator in Graves' Disease (Graves 병(病)에 있어 Long-Acting Thyroid Stimulator(LATS)에 관(關)한 연구(硏究))

  • In order to study the role of LATS in the pathogenesis of the Graves' disease, the serum activity of the LATS was determined by the bioassay of the modified McKenzie method. The subjects examined in the study consisted of 76 individuals including 12 cases of normal control, 54 cases with typical Graves' disease and 10 cases of chronic thyroiditis. The data observed in the patients with the Graves' disease were analyzed in comparison with the clinical features, laboratory findings, and responsiveness to the treatment. The results obtained are as follows: 1. None of the subjects which did not have the Graves' disease showed a positive LATS activity, except one case with the chronic thyroiditis. 2. Twenty-two out of the 54 cases with the Graves' disease showed positive results for LATS(40.7%). The positivity was significantly higher in the patients who had been treated with anti. thyroid regimen but still showed hyperthyroidism than in the patients who had not been treated. 3. The activity of LATS was gradually decreased or even became absent as the hyperthyroidism was corrected after the treatment. 4. No significant difference was noticed in age & sex between the positive and negative groups of LATS. 5. There was no evidence of significant correlation between the LATS activity and clinical features.

  • The first outbreak of Aujeszky's disease(AD) was identified from piggery located at the southern part of Korea in July, 1987. This piggery suffered from a significant economic loss caused by unexpected piglet mortality and reproductive failure. Etiologic viral agents were isolated from tonsil and spleen of the infected piglets, and the isolates produced a typical cytopathic effects of herpesvirus with giant cell formation when inoculated in many different cells. Subsequently the field isolates were characterized as suid herpesvirus I by cross-neutralization test and indirect fluorescence assay utilizing specific monoclonal antibody, and were proved to be a pathogenic strain of AD virus(ADV).

  • A simple test using latex-agglutination was developed to detect serologically tobacco mosaic virus (TMV), soybean mosaic virus (SoyMV), and barley stripe mosaic virus (BSMV) from infected Plants. Latex spheres ( 0.81 $\mu$, Difco) were adsorbed with immuno globulin purified by electrophoresis from crude antiserum against viruses. The antibody- sensitized latex suspension was mixed with sap from virus -infected leaves in a glass capillary tube (inner diam. 1mm $\times$ 100 mm length) The mixture, after agitation, was observed under a stereo microscope at low magnification (X20 - X4O), to examine the reaction between antigen (Virus) and its antibody. Flocculation occurred when the reaction was positive.

  • The medicinal plants of 33 species with symptoms of mosaic, necrosis, line pattern, malformation (fine leaf), stripe, and ring spot infected by cucumber mosaic virus spontaneously, were collected in Korea and Japan. Among them, 2 mosaic diseases of portulaca oleracea L. and Celocia argentea L., 2 necrotic mosaic diseases of Aristolochia debilis Sieb. and Tetragonia exponsa Murr., 6 mottle diseases of Cassia torosa Cav., Bupleurum falcantum L., Angelica acutioloba Kitagawa, A. keiskei Koidz., Peucedanum japonicum Thunb., and Foeniculum vulgare Mill., and ringspot disease of Basella rubra L., were named newly in these studies.

  • The pathogenic bacteria isolated from the rotted fruits and tubers of red pepper, tomato, eggplant and potato which were collected from farms of Chungnam province and the markets of Seoul, Pusan, Taejon, Chonju, Iri, Weonju, Jecheon, Chungju and Cheonan in Korea. These isolates were divided into ninety-eight isolates of Enoinia sp. and seven isolates of Pseudomonas spp. on the basis of their taxonomic characteristics. Among seven isolates of Pseudomonas spp., six isolates were identified as P. viridiflava and two were P. marginalis and ninety-eight isolates of Erwinia sp. as E. carotovora. subsp. carotovora on the basis of bacteriological properties.

  • Mycoplasma-like organisms (MLO) were identified from three plants. The disease symptoms show stunting, yellowing and witche's broom on Cnidium officinale Makino collected from Ulreung Island, Bupleurum falcatum L. collected from Jangsu, and Plantago asiatica L. collected from Daegwanreung. The particles of MLO were observed in the phloem tissues by a Hitachi Hu-11E electron microscope. C. officinale infected with MLO was frequently observed in Ulreung Island.
