Molecular Cloning and Characterization of CDNA Encoding Immunoglobulin Heavy and Light chain Variable Regions from Four Chicken Monoclonal Antibodies Specific to Surface Antigens of Intestinal Parasite, Eimeria acervulina

  • We have developed four chicken hybridomas secreting monoclonal antibodies to induce a protective immune response against the chicken disease avian coccidiosis, caused by the intestinal parasite Eimeria acervulina. Huwever, since the amount of antibodies secreted from these hybridomas is too low or sometimes they lost their ability to produce antibodies, the hybridoma method is not satisfactory in the production of large amounts of chicken monoclonal antibodies. To bypass these problems, we applied the antibody engineering technology using polymerase chain reaction. We cloned and determined the sequences of variable domains of the four chicken monoclonal antibodies, namely, 2-1, 5D11, 13C8 and 8C3. The sequences comparison to germline sequences skewed that the gene con version mechanism might contribute to developing diversification of heavy and λ-light chains in chicken antibodies. Several pseudogene families regarded as donors in gene conversion were identified at each framework region and the complementarily determining region of λ-light chains. In addition, as expected, numerous changes of nucleotide sequences such as nucleotide substitution, insertion and deletion were found predominantly in complementarity determining regions, which are likely to be somatic hypermutations as a result of affinity maturation in antibody-producing cells.

중학생의 성장 발육과 기생충 보유 현황에 관한 조사 연구

  • A survey was conducted on 2,321 Chung Rhang Middle School boys to check out their physical growth, development and the number of parasite carriers during the period from June 16. to June 23. 1970 As the results of this study, the following conclusions were revealed. A) Physical Growth and Development State. 1. Direct curve in physical growth and development was observed during the period from 13 to 16 years of age. 2. The age and quantity of maximum growth and development. Height; 13~14 years of age 6.1cm Body weight; 14~15 years of age 5.1kg Chest circumference; 13~14 years of age 9.9cm Sitting height; 14~15 years of age 3.4cm 3. Comparison with Japanese students Most Korean students are shorter than Japanese students, though the standard of height became higher than before. The standard of body weight is visibly lower than Japanese students. The standard of chest circumference of the students of age 14 is higher than counter part of Japanese students and the all boys are lower than Japanese students except at the age of 14. The standard of the sitting heights of the Korean students are growing steadily, but every students are shorter than Japanese students. 4. The greatest differences in physical growth and development were shown in 13~14 years of age. 5. The students of 13~15 years of age developed their bodies most highly according to index of Rohrer. 6. Body weight and chest circumference have not grown enough compared with height. The development of sitting height was made highly and the development is steadily growing as they grow old. B. The Number of Parasite Carriers The number of parasite carriers was shown 79.4 per cent among all the student checked out. The number of trichuris was shown 79.7 per cent. The number of parasite carriers was shown higher per cent as the grow old.

Status of intestinal parasites infection among primary school children in Kampongcham, Cambodia

  • A survey was made to find the extent of intestinal parasite infection in Kampongcham, Cambodia in February 2002. A total of 251 fecal specimens were collected from Tonlebat primary school children and examined by formalin-ether sedimentation technique. The overall infection rate of intestinal parasite was 54.2% (males, 57.3%; females, 50.8%). The infection rate of intestinal helminths by the species were as follows: Ascaris lumbricoides 26.3%, Echinostoma sp. 15.6%, hookworm 6.4%, Opisthorchis sp. 4.0%, Rhabditis sp. 2.4%, and Trichuris trichiura 0.4%. The infection rate of intestinal protozoa were as follows: E. coli 7.6%, G. lamblia 3.2%, I. butschlii 3.2%, and E. histolytica 0.8%. More than two different kinds of parasites were found in 16.7% of the stool samples. All the children infected were treated with albendazole, praziquantel and metronidazole according to parasite species. The results showed that intestinal parasites are highly endemic in this area.

Development of Lateral Flow Immunoassay for Antigen Detection in Human Angiostrongylus cantonensis Infection

  • Angiostrongyliasis is difficult to be diagnosed for the reason that no ideal method can be used. Serologic tests require specific equipment and are not always available in poverty-stricken zone and are time-consuming. A lateral flow immunoassay (LFIA) may be useful for angiostrongyliasis control. We established a LFIA for the diagnosis of angiostrongyliasis based on 2 monoclonal antibodies (mAbs) against antigens of Angiostrongylus cantonensis adults. The sensitivity and specificity were 91.1% and 100% in LFIA, while those of commercial ELISA kit was 97.8% and 86.3%, respectively. Youden index was 0.91 in LFIA and 0.84 in commercial ELISA kit. LFIA showed detection limit of 1 ng/ml of A. cantonensis ES antigens. This LFIA was simple, rapid, highly sensitive and specific, which opened an alternative approach for the diagnosis of human angiostrongyliasis.

  • Calreticulin (CALR), a multifunctional protein thoroughly researched in mammals, comprises N-, P-, and C-domain and has roles in calcium homeostasis, chaperoning, clearance of apoptotic cells, cell adhesion, and also angiogenesis. In this study, the spatial and temporal expression patterns of the Opisthorchis viverrini CALR gene were analyzed, and calcium-binding and chaperoning properties of recombinant O. viverrini CALR (OvCALR) investigated. OvCALR mRNA was detected from the newly excysted juvenile to the mature parasite by RT-PCR while specific antibodies showed a wide distribution of the protein. OvCALR was localized in tegumental cell bodies, testes, ovary, eggs, Mehlis' gland, prostate gland, and vitelline cells of the mature parasite. Recombinant OvCALR showed an in vitro suppressive effect on the thermal aggregation of citrate synthase. The recombinant OvCALR C-domain showed a mobility shift in native gel electrophoresis in the presence of calcium. The results imply that OvCALR has comparable function to the mammalian homolog as a calcium-binding molecular chaperone. Inferred from the observed strong immunostaining of the reproductive tissues, OvCALR should be important for reproduction and might be an interesting target to disrupt parasite fecundity. Transacetylase activity of OvCALR as reported for calreticulin of Haemonchus contortus could not be observed.

  • Occurrence and prevalence of Marteilioides chungmueasis have been reported in several waters around Tongyeong but no report has been made for Pukman bay. Therefore, we investigated that the prevalence and infection intensities in Pukman Bay at the inside and the outside areas which are hydrographically divided by tidal current. Furthermore, various environmental parameters were investigated in order to elucidate effective parameter for parasitic infection. Infection rates of Marteilioides chungmuensis in adult oysters were ranged 3.3-20.0% at the inside area during September 2002 through January 2003, and 3.3-30.9% at the outside area during August 2002 through January 2003. External manifestation of infected oyster consisted of abnormal egg masses with nodular appearance in the soft tissue. Histopathological symptoms included massive hemocytic infiltration within or around the follicle wall and atrophic epithelium of digestive diverticula. For the environmental parameters, comparative study made differences between two side of the Bay during the infection period: inside > outside for SS while inside < outside for chlorophyll-a. A positive relationship was observed between chlorophyll-a and infection period, which might indicate the difference in food availability between two areas. The prevalence of ovarian parasite Marteilioides chungmuensis, therefore, was highly associated with food availability. Pearson's correlation analysis was made between environmental parameters and infection prevalence. Significance was observed in water temperature (p < 0.05), suspended solids (p < 0.01) and chlorophyll-a (p < 0.05). A principle component analysis showed that infection of the ovarian parasite, Marteilioides chungmuensis, exhibited effects of seasonality (component I = 55.2%) and chemical/physical environmental factors (component II = 24.4%). These results clearly indicate that the infection of ovarian parasite, M. chungmuensis in the Pacific oyster Crassostrea gigas is closely associated with seasonality and food availability.

전라남도내 미나리의 중금속 함량 및 기생충에 대한 조사 (Investigation on Contents of Heavy Metals and Parasites of the Dropwort(Oenanthe javanica) cultivated in Chollanamdo)

  • 전남 도내 5개 지역과 광주시 근교 1개 지역에서 재배되고 있는 미나리의 안전성 평가를 위해 중금속 함유량과 기생충 감염 여부를 조사하였다. 그 결과 비교적 독성을 지니고 있는 중금속인 수은과 비소는 각각 흔적 수준과 극히 미량인 $0.003{\sim}0.01\;ppm$으로 나타났다. 납은 모든 지역에서 검출되지 않았으며, 카드뮴은 $0.013{\sim}0.036\;ppm$ 범위를 보였으며, 또한 농작물 생장에 필수원소로 작용하는 구리, 망간과 아연의 함유량은 $0.285{\sim}0.627\;ppm,\;1.337{\sim}12.884\;ppm,\;2.105{\sim}4.221\;ppm$이 검출되었다. 또한 미나리에서 흡충류인 간질과 간흡충, 수인성원 충류인 람블 편모충과 크립토스포리듐속 그리고 선충류인 회충과 십이지장충 6종에 대한 기생충 검사 결과 모두 검출되지 않았다.

Congenital Malaria in Newborns Selected for Low Birth-Weight, Anemia, and Other Possible Symptoms in Maumere, Indonesia

  • Congenital malaria is assumed to be a risk factor for infant morbidity and mortality in endemic areas like Maumere, Indonesia. Infected infants are susceptible to its impact such as premature labor, low birth weight, anemia, and other unspecified symptoms. The aim of this study was to investigate the prevalence of congenital malaria and the influence of mother-infant paired parasite densities on the clinical outcome of the newborns at TC Hillers Hospital, Maumere. An analytical cross sectional study was carried out in newborns which showed criteria associated with congenital malaria. A thick and thin blood smear confirmed by nested PCR was performed in both mothers and infants. The association of congenital malaria with the newborn's health status was then assessed. From 112 mother-infant pairs included in this study, 92 were evaluated further. Thirty-nine infants (42.4%) were found to be infected and half of them were asymptomatic. Infected newborns had a 4.7 times higher risk in developing anemia compared to uninfected newborns (95% CI, 1.3-17.1). The hemoglobin level, erythrocyte amount, and hematocrit level were affected by the infants' parasite densities (P<0.05). Focusing on newborns at risk of congenital malaria, the prevalence is almost 3 times higher than in an unselected collective. Low birth weight, anemia, and pre-term birth were the most common features. Anemia seems to be significantly influenced by infant parasite densities but not by maternal parasitemia.

Molecular Genetic Findings of Spirometra decipiens and S. ranarum in Korea

  • The taxonomy of Spirometra species has been controversial despite the medical and veterinary importance. Currently, only a few Spirometra species are considered valid species in the genus Spirometra. In the present study, the distribution of Spirometra species obtained from animals in Korea were identified by molecular analysis of the mitochondrial cytochrome c oxidase I (cox1) gene. A total of 28 Spirometra species specimens were analyzed. These were all collected between 1973 and 2008 in the Republic of Korea. Mitochondrial cox1 sequences were examined for a total of 28 specimens comprising 14 S. decipiens and 14 S. ranarum. The difference in partial cox1 sequences (316 bp) between S. erinaceieuropaei (KJ599680) and S. ranarum (this study) was 9.3%, while that between S. decipiens (KJ599679) and S. ranarum (this study) was 2.2%. Genetic analyses identified 2 Spirometra species in animals such as cat, leopard cat, dog, duck and snake in Korea as S. decipiens and S. ranarum. S. decipiens and S. ranarum were present in Gyeongnam Province (P), Jeonnam P, Gangwon P, Chungbuk P, and Seoul. S. decipiens was found in tadpoles, snakes, ducks, cats, leopard cats and dogs, while S. ranarum was found in cats and dogs. The ratio of S. decipiens:S. ranarum calculated from the molecular data was 14:14 (or 1:1). These results indicate that S. decipiens and S. ranarum are sympatrically distributed in Korea.

Intestinal Parasitosis in Relation to Anti-Retroviral Therapy, CD4+ T-cell Count and Diarrhea in HIV Patients

  • Intestinal parasitic infections are one of the major causes of diarrhea in human immunodeficiency virus (HIV) seropositive individuals. Antiretroviral therapy has markedly reduced the incidence of many opportunistic infections, but parasite-related diarrhea still remains frequent and often underestimated especially in developing countries. The present hospital-based study was conducted to determine the spectrum of intestinal parasitosis in adult HIV/AIDS (acquired immunodeficiency syndrome) patients with or without diarrhea with the levels of $CD4^+$ T-cell counts. A total of 400 individuals were enrolled and were screened for intestinal parasitosis. Of these study population, 200 were HIV seropositives, and the remaining 200 were HIV uninfected individuals with or without diarrhea. Intestinal parasites were identified by using microscopy as well as PCR assay. A total of 130 (32.5%) out of 400 patients were positive for any kinds of intestinal parasites. The cumulative number of parasite positive patients was 152 due to multiple infections. A significant association of Cryptosporidium (P<0.001) was detected among individuals with $CD4^+$ T-cell counts less than $200cells/{\mu}l$.
