Stability of Anthocyanin Pigment from Juice of Raspberries (나무딸기(복분자(覆盆子)) 과즙색소(果汁色素) Anthocyanin의 안정성(安定性)에 관한 연구(硏究))

  • The effect of various pH levels, temperatures. organic acids, inorganic salts, metal ions on the stability of the anthocyanins pigment (pH 3.7) from the juice of raspberries were investigated. Initial absorption of total anthocyanin was decreased as pH increased from 1.0 to 7.0. Total amount of anthocyanin reached the highest at pH 3.7 and least at pH 7.0. The total anthocyanin content decreased rapidly with the increasing temperature. Many organic acids were found to enrich and stabilize the color density at 520nm in anthocyanin solution (pH 3.7). The hyperchromic effect of saturated n-carboxylic acid increased in the following order; formic acid> acetic acid>n-butyric acid>propionic acid. On the polycarboxylic acid, especially, malic acid showed 550$\sim$930% higher than control group. On the inorganic salts (0.5M), sodium perchlorate had the most hyperchromic effect and followed by sodium sulfate>sodium chloride>sodium phosphate, monobasic. Among the metal ions, both aluminium ion and cupric ion much more accelerated the anthocyanins degradation as compared with other metal ions.

Evaluation of Shear Strength by Experiment and Finite Element Analysis of SFRC Hollow Members (SFRC 중공 부재의 실험 및 유한요소 해석에 의한 전단강도 평가)

  • This study targets SFRC hollow members with small depth under shear force and bending. To evaluate the effect of web width on shear strength of SFRC members, experiment and finite element analysis were conducted and compared with existing equations. The web width was planned to be 1/2 times and 2/3 times, and the shear span ratio was planned to be 1.5 times. In the shear test results, the maximum shear strength increased by 10.3 to 28.0% with the web width increased by 33%. When the overall depth of specimens was increased by 1.5 times, the shear strength of the specimen with a web width of 100mm was increased by 29.2%. On the other hand, specimen with the 150mm only increased by 11.3%. These results indicate that the smaller the web width, the greater the shear strength increase with the increase of depth. Also, the smaller the web width, the greater the contribution of steel fiber. It has been shown that the KCI code evaluates the shear strength of experiments as very safe side, and that the proposed formula of Shin et al. predicts the experimental strength relatively well. As the web width increases by 2, 3, and 6 times, the mean shear strength by FEA appears to be 1.18, 1.80, and 2.19 times respectively. This indicates that the shear strength does not increase in proportion to the increase in web width.

Improvement of Yields and Organoleptic Quality of Anchovy Extract by Alkali-Protease Hydrolysis (알칼리와 효소처리에 의한 멸치 추출액의 수율 및 관능적 성질의 향상)

  • Dried anchovy (Engraulis japonica) was ground and treated with 0.3N NaOH solution and then hydrolyzed with proteolytic enzymes. Extracts obtained by centrifugation of alkali-enzyme treated anchovy slurry was compared with water extract for the yields of soluble solid, protein and ashes and organoleptic characteristics. The data for the yields of the soluble solids, protein and ash showed that a 2-3 folds increase in those yields was resulted by combined alkali-enzyme treatments when it was compared to water only extract. The organoleptic evaluation on the alkali-enzyme treated anchovy extracts also showed a 2-3 folds in flavor strength of all descriptions in odor and taste and a significant improvement in total odor or taste acceptability.

Genetic relationships of pear germplasms using simple sequence repeat marker (SSR 마커를 이용한 배 유전자원의 유연관계)

  • This study analyzed the genetic diversity of 115 pear germplasms using 15 SSR markers. Three to forty-one SSR alleles were detected for each locus with an average of 16 alleles per locus. The average availability of markers was 0.966. The average observed heterozygosity ($H_{obs}$) was 0.603 (range: 0.140 to 0.929). The average expected heterozygosity ($H_{exp}$) was 0.718 (range: 0.463 to 0.904). The average polymorphism information content (PIC) was 0.692 (range: 0.403 to 0.897). The genetic relationships of pear germplasms were classified into two major groups by geographic origins and genetic characteristics according to genetic distance. The first group was composed of European pear belonging to Pyrus communis. The second group consisted of P. pyrifolia, P. ussuriensis, P. bretschneideri, P. betulaefolia, P. calleryana, interspecific hybrids, and unclear germplasms. The results of this study suggest that genotype analysis of pear germplasms using SSR markers can identify the genetic diversity of germplasms, and can be used to provide basic information for pear breeding.

Relationship between HSP70 Gene Polymorphisms and IVF Embryo Development in Pigs (돼지에서 HSP70 유전자형과 IVF 수정란 배 발달과의 관련성)

  • This study was performed to investigate the relationship between Heat shock protein 70 (HSP70) gene polymorphism and in vitro fertilization(IVF) embryo development in the pigs. The single strand conformation polymorphism(SSCP) genotypes from HSP70 K1, K3 and K4 PCR products were detected different patterns. In cleavage rate of oocyte fertilized in vitro, HSP70 K1-AA genotype($73.1\%$) and K1-AB genotype($62.3\%$) showed significantly higher oocyte cleavage rate than HSP70 K1-BB genotype($49.3\%$)(p<0.05). And HSP70 K3-AA genotype ($72.4\%$) and K3-AB($62.2\%$) also showed significantly higher oocyte cleavage rate than HSP70 K3-BB genotype($49.1\%$)(p<0.05). The IVF embryo development of 2-cell stage according to HSP70 genotypes of sperm and pig breeds also showed a significant difference. The number of embryos developed to 2-cell stage in Landrace(28.8) and Duroc(29.8) were significantly higher than in Yorkshire(10.9)(p<0.05). And also HSP70 K4-AB genotype group(29.6) higher than HSP70 K4-AA genotype group(10.6)(p<0.05). However, the number of embryos developed to blastocyst stage did not showed significant differences among breeds as well as HSP70 genotypes. These resrults suggest that in vitro development in porcine early embryos may be affected by HSP70 genotypes and breeds.

Inheritance and Cross Resistance of Acequinocyl Resistance in Twospotted Spider Mite, Tetranychus urticae (Acequinocyl 저항성 점박이응애의 유전과 교차저항성)

  • The development of resistance to acequinocyl was found in the population of the twospotted spider mite, Tetranychus urticae, collected from rose greenhouses in Gimhae, Gyeongnam province in January 2001. This pest is reared on 5 years treated with acequinocyl (over 200 times), and increased 87.8 folds in resistance as compared to susceptible strain (S). Inheritance of acequinocyl resistant strain (R) and cross resistance of this strain to 8 acaricides against T. urticae adults and eggs was investigated. There were differences of susceptibility in the acequinocyl concentration-mortality relationships in $F_1$ progenies obtained from reciprocal cross with the S and R strain ($S(female){\times}R(male)$, $R(female){\times}S(male)$). Degrees of dominance were -0.75, -0.57 in $F_1$ progenies of adult and egg of $S(female){\times}R(male)$. Inheritance in $F_1$ progenies of $S(female){\times}R(male)$ was incomplete recessive. Degree of dominance were 0.81, 0.45 in $F_1$ progenies of adult and egg of $R(female){\times}S(male)$, respectively. These results suggest that inheritance of acequinocyl resistance is controlled by a complete dominance. The R strain exhibited cross resistance of 1.1 and 0.9 fold to amitraz, bifenazate, and negatively correlated cross resistance of 0.08 fold to emamectin benzoate in adult females. The R strain showed cross resistance of 37.7, 14.0, and 26.2 fold to amitraz, milbemectin and spriodiclofen in eggs, respectively. Particularly it showed high levels of cross-resistance to pyridaben with 6538.3 fold. These chemicals showed negatively correlated cross-resistance exhibited 0.4, 0.3, and 0.2 fold to ahamectin, bifenazate, and emamectin benzoate in eggs.

Effect of Oxygen Concentrations with Catalase on In Vitro Maturation of Porcine Follicular Oocytes and In Vitro Development of Porcine IVM/IVF Embryos (산소조건 및 Catalase가 돼지난포란의 체외성숙과 배발달에 미치는 영향)

  • The present study was carried out to examine the effect of catalase (CAT) on in vitro maturation (IVM) of porcine follicular oocytes and oxygen concentration with CAT on in vitro development (IVD) of porcine IVM/IVF embryos. The results were summarized as follows - 1 . The rates of nuclear maturation, penetrated oocytes, pronucleus formation rates, polyspermic oocytes and mean numbers of the penetrated sperm were significantly lower in oocytes matured with 100, 500 and 1,000 units/ml CAT than those of central groups (P>0.05). 2. The rates of blastocyst formation and total cell numbers of blastocyst at day 7 after in vitro fertilization were significantly lower in CAT treatment groups than those of the central groups (P>0.05). 3. There were not significant difference in the blastocyst development and total cell numbers of blastocyst on in vitro culture of NCSU-23 media with 0, 100, 500 and 1000 units/ml CAT under the 5% and 20% $O_2$ concentrations. These results suggested that the addition of CAT was not helpful for porcine oocyte maturation and further development, also the rates of blastocyst formation and total cell numbers of blastocyst at day 7 of porcine IVM/IVF embryos were not significantly different in the NCSU-23 culture medium under the 5% and 20% $O_2$ concentrations.

Changes of Saponin during the Cultivation of Soybean Sprout (콩나물 생장 중 사포닌의 변화)

  • We investigated the changes in saponins during the cultivation of soybean sprout. Crude saponin content was 4.15mg/g in germinated soybean and reached its peark (5.33mg/g) in soybean sprout cultivated for six days. Saponin content in the cotyledon, stem, and root of the soybean sprout cultivated for six days were 4.17, 7.46, and 7.45mg/g, respectively. Soyasaponins extracted from the soybean sprout were analyzed with LC-electrospray ionization (ESI)-mass spectrometry, in which a reverse phase $C_18$ column was used for separation of saponins. In the soybeen sprout, group B saponin, I, II, III, IV, and V increased 7, 2, 1.4, 8.7, and 3.3 fold, respectively, compared to those in the soybean seed. Group B saponin I, II, III, IV, and V in the stem of the soybean sprout were 10.53, 1.45, 10.49, 5.72 and 8.14 fold the level of those in the cotyldon, respectively. In the root, the contents of group B saponin I, III, IV, and V were 5.54, 2.77, 4.86 and 9.73 fold, respectively, higher than those in cotyledon, but the content of group B saponin 2 was 2.96 fold less than that in cotyledon. These results indicate that the biosyntheses of group B saponins are differentially regulated in growing soybean sprout.

A Study on the CO2 Removal Efficiency with Aqueous MEA and Blended Solutions in a Vortex Tube Type Absorber (Vortex Tube 형 흡수장치에서 MEA와 혼합흡수용액을 이용한 CO2 제거 효율 고찰)

  • In this study, the $CO_2$ removal characteristics of the Vortex tube type absorbtion apparatus were investigated to enhance the compactness of $CO_2$ absorption process and to reduce the amount of absorbing solution of the $CO_2$ separation process. The Vortex tube with the diameter of 17 mm and the length of 250mm was introduced in the experimental apparatus to treat $20Nm^3/hr$ of $CO_2$ containing flue gas. The flue gases for experiments containing 11~13 vol% of $CO_2$ were supplied from the coal-firing CFBC power plant with 12 ton/hr of steam producing capacity. The mixed solutions of 20 wt% of MEA as base solution with the adding solutions like HMDA, AMP and KOH were used as absorbents. The experiments were executed under the various conditions like the absorbing solution concentrations in the range of 20 to 50 wt%, the flow rate of $CO_2$ containing flue gases in the range of 6 to $15Nm^3/hr$ and the flow rate of absorbing solution in the range of 1.0 to 3.0 l/min. As a results, the $CO_2$ removal efficiency of mixed absorbent of 20 wt% of MEA with HMDA was remarkable. From this study, we concluded that the efficient separation of $CO_2$ from flue gases using the features of the Vortex tube type absorbing unit for gas/liquid contact and the separation of gas/liquid be possible. But more works are needed to increase the $CO_2$ removal efficiency of Vortex tube process.

A Study on the Leaching and Recovery of Lithium by Reaction between Ferric Chloride Etching Solution and Waste Lithium Iron Phosphate Cathode Powder (폐리튬인산철 양극재 분말과 염화철 에칭액과의 반응에 의한 리튬의 침출 및 회수에 대한 연구)

  • Efforts are currently underway to develop a method for efficiently recovering lithium from the cathode material of waste lithium iron phosphate batteries (LFP). The successful application of lithium battery recycling can address the regional ubiquity and price volatility of lithium resources, while also mitigating the environmental impact associated with both waste battery material and lithium production processes. The isomorphic substitution leaching process was used to recover lithium from spent lithium iron phosphate batteries. Lithium was leached by the isomorphic substitution of Fe2+ in LFP using a relatively inexpensive ferric chloride etching solution as a leaching agent. In the study, the leaching rate of lithium was compared using the ferric chloride etching solution at various multiples of the LFP molar ratio: 0.7, 1.0, 1.3, and 1.6 times. The highest lithium leaching rate was shown at about 98% when using 1.3 times the LFP molar ratio. Subsequently, to eliminate Fe, the leachate was treated with NaOH. The Fe-free solution was then used to synthesize lithium carbonate, and the harvested powder was characterized and validated. The surface shape and crystal phase were analyzed using SEM and XRD analysis, and impurities and purity were confirmed using ICP analysis.
