O/W/O(oil in water in oil)형 유화액막에 의한 2성분 탄화수소 혼합물인 toluene-cyclo hexane의 분리에 있어서 투과율, 막의 안정성과 분리계수에 미치는 영향을 계면활성제 농도, emulsion과 용매와의 교반속도 등을 변수로 하여 실험하였다. 계면활성제의 농도가 증가함에 따라 투과율은 증가하는데 0.5wt% 이상에서는 증가율이 둔화 되었다. 막 파괴율은 0.1wt%에서 가장 낮게 나타났으며, 그 이상에서는 농도가 증가하면서 파괴율도 증가하였다. 또한 분리계수는 계면활성제의 농도가 0.5wt%에서 가장 높게 나타났다. 에멀젼 제조 시 교반속도 변화에서는 투과율이나 막 파괴율에 큰 영향을 미치지 않는 것으로 나타났다. 막 강화제의 농도가 증가함에 따라 투과율은 감소하였고, 에멀젼과 용매의 접촉 시 교반속도가 증가 할수록 막 파괴율도 증가하였다.
초산은 묽은 산에 의해 처리되어 얻어진 헤미셀룰로오스 가수분해액 중 가장 양이 많을 뿐만 아니라 중대한 에탄올 발효 저해물질이다. 그 가수분해액으로부터 초산을 선택적으로 제거하는데 최적인 분리시스템을 찾기 위하여 모사 헤미셀룰로오스 가수분해액으로 자일로스, 초산과 황산의 혼합물이 선택되었다. 이러한 목적을 이루기 위해 에멀젼형 액막법이 모사 헤미셀룰로오스 가수분해액으로부터 초산을 제거하는데 적용되었다. W/O 에멀젼의 주요 구성 성분들, 즉 아민계 추출제 종류, 계면활성제 조성, 첨가제 종류와 회수제의 종류 및 농도가 모사 헤미셀룰로오스 가수분해액에 있는 초산, 자일로스와 황산의 추출에 미치는 영향이 조사되었다. 특별한 실험 조건에서, 초산의 추출율은 95% 이상이었고, 자일로스의 손실은 미미하였는데, 이것은 현 에멀젼형 액막법이 경제성이 높은 공정이 될 수 있다는 것을 의미한다.
비수유화법을 이용한 미세 O/W에멀젼의 제조 기술에 관한 연구로 표면장력, 계면장력, 탁도 및 전이온도를 통하여 비수용매/기름에서의 POE(25)octyldodecyl ether의 거동을 연구하였다. POE(25)octyldodecyl ether는 물 용매에서의 미셀 형성과는 달리 비수용매에서는 용해되었다. 글리세린과 같은 용매에서는 POE(25)octyldodecyl ether의 용해력은 약했으며 표면으로 이동되었다. 표면에서 포화된 후 POE(25)octyldodecyl ether는 석출 현상을 보였다. 최종 에멀젼의 평균입자 크기는 230nm 이었으며 한달 동안의 0, 25, 40, $50^{\circ}C$ 및 순환시험에 안정하였으나 일반적인 에멀젼은 불안정하였다. 따라서 적절한 용매의 조합에 의해 계면활성제의 흡착 효율을 증가시킬 수 있었다.
본 실험에서는 화장품에 널리 사용되는 지방산과 지방알코올을 Tween 80과 Span 80을 함께 사용하여 조성 상전이 유화법으로 O/W (oil-in-water) 저점도 나노에멀젼을 제조하였다. 지방알코올의 오일상에서의 농도가 증가할수록 나노에멀젼의 입자 크기가 증가하였다. 혼합계면활성제의 HLB를 조절하여 입도 분포가 좁고 안정한 나노에멀젼의 제조가 가능하였다. 전상점 부근에서 지방산 및 지방알코올을 함유한 계 모두에서 유사한 점도와 전기전도도의 변화를 보였으나 오일과 계면활성제만으로 구성된 계와는 다르게 넓은 수용액상 분율범위에서 높은 점도를 나타냈다. 입자 크기가 100 nm 미만의 저점도 나노에멀젼은 실온에서 한 달 이상 안정하였다. 지방산 또는 지방알코올을 함유하고, 저에너지 유화법으로 제조된 저점도 O/W 나노에멀젼 제형은 화장품의 기재로서 널리 사용될 수 있다.
Glycerin monostearate, polyoxyethylene(100) monostearate, polyoxyethylene(20) sorbitan monostearate를 혼합한 유화제를 분산상인 liquid paraffin, beeswax 혼합물에 용해한 후 연속상인 propylene glycol 수용액을 첨가하는 방법인 agent-in-oil법을 적용한 반전유화법에 의해 O/W 에멀젼을 제조하였고, 제조과정에서 나타나는 상거동 및 점성거동을 관찰하였다. 연속상을 주가함에 따라 액정 영역과 백타겔 영역이 존재하는 유화제 조성(HLB 10.1~12.3)에서만 미세하고 균일한 에멀젼이 형성되었으며, 유상${\rightarrow}$액정상${\rightarrow}$겔상${\rightarrow}$O/W 에멀젼으로 상변화가 일어남을 알 수 있었다. 제조된 에멀젼의 전단속도에 따른 전단응력곡선으로부터 항복치가 존재함에 따라 에멀젼 입자간의 내부구조가 형성됨을 알 수 있었고, 전단력에 의한 내부구조의 파괴가 겉보기상으로는 완전히 비가역적으로 진행되며, 구조 파괴의 척도인 hysteresis loop의 면적은 유화제의 HLB값이 작을수록 큰 것을 알 수 있었다. 시간에 따른 전단응력 변화곡선에서소성점성도의 시간의존성이 큰 것을 확인하였고, 시간유동화현상에 따른 완화시간(${\lambda}$)으로부터 HLB값이 작을수록 상대적 안정성이 큼을 알 수 있었으며, homomixer로 제조한 에멀젼에 비해 반전유화법에 의해 제조한 에멀젼이 더 안정함을 알 수 있었다.
본 연구에서는 coconut oil과 Tween-Span계 비이온성 혼합계면활성제를 사용하여 제조한 O/W (oil in water) 유화액의 안정성에 영향을 끼치는 인자를 평가하였다. 이를 위해 비이온성 혼합계면활성제의 HLB value와 첨가량, 균질화 속도를 변수로 설정하여 제조한 O/W 유화액의 평균액적크기, 제타포텐셜, 유화안정도지수(ESI), 열적 불안정도지수(TII) 등으로부터 유화액의 안정성을 평가하였다. 제조한 O/W 유화액의 평균액적크기는 100~200 nm의 나노에멀젼으로서 비이온성 혼합계면활성제의 첨가량 및 균질화 속도가 증가함에 따라 평균액적크기는 감소하였으며, 제타포텐셜은 증가하였다. 비이온성 혼합계면활성제의 HLB value가 6.0, 10.0, 8.0 순으로 우수하게 나타났으며, HLB value가 8.0에서 제조한 유화액의 평균액적크기는 120 nm으로 가장 작고, 제타포텐셜은 40~60 mV로 가장 크게 나타났다. ESI 및 TII를 통한 안정성 평가는 HLB value가 6.0, 10.0, 8.0 순으로 안정성이 증가하였으며, HLB value가 8.0에서의 ESI 및 TII는 각각 80% 이상과 20% 이하로 가장 우수하게 나타났다.
We have studied a relationship between the pattern of complex modulus change versus internal phase volume ratio and the rheological consistency of concentrated W/O emulsions with Magnesium Sulfate in the range 0.0 to 0.5 wt% and with different oil polarities, respectively. The rheological consistency with time of concentrated W/O emulsion was checked using Fudoh Rheometer and the coalescence of deformed water droplets was examined using polarized light microscope(LEICA DMRP). To find the pattern of complex modulus change of the concentrated emulsions versus internal phase volume ratio, the effect of varying water phase volume fraction from 0.78 up to 0.85 on viscoelastic measurements was investigated using rotational rheometer (HAAKE Rheostress RS 50). The rheological consistency was mainly destroyed by the coalescence of the deformed water droplets. The greater the increase of complex modulus was, the less coalescence occurred and the more consistent the concentrated emulsions were. And the pattern of complex modulus increase versus volume ratio has been explained with the resistance to coalescence of the deformed interfacial film of water droplets in concentrated W/O emulsion.
Porous SiC beads were prepared by freeze-drying a polycarbosilane (PCS) emulsion. The water-in-oil (w/o) emulsion, which was composed of water, PCS dissolved p-xylene, and sodium xylenesulfonate (SXS) as an emulsifier, was frozen by dropping it onto a liquid $N_2$ bath; this process resulted in 1~2 mm sized beads. Beads were cured at $200^{\circ}C$ for 1 h in air and heat-treated at $800^{\circ}C$ and $1400^{\circ}C$ for 1 h in an Ar gas flow. Two types of pores, lamella-shaped and spherical pores, were observed. Lamellar-shaped pores were found to develop during the freezing of the xylene solvent. Water droplets in the w/o emulsion were changed into spherical pores under freeze-drying. At $1400^{\circ}C$ of heat-treatment, porous SiC was synthesized with a low level of impurities.
Alginate microspheres, containing fluorescein isothiocyanate-bovine serum albumin (FITC-BSA) or green fluorescent protein (GFP) were prepared and used as a model drug to develop the oral vaccine delivery system. The alginate microspheres were coated with poly-L-lysine or chitosan. Two methods, w/o-emulsion and spray, were used to prepare alginate microspheres. To optimize preparation conditions, effects of several factors on the particle size and particle morphology of microsphere, and loading efficiency of model antigen were investigated. In both preparation methods, the particle size and the loading efficiency were enhanced when the concentration of sodium alginate increased. In the w/o-emulsion preparation method, as the concentration of Span 80 was increased from 0.5% to 2%, the particle size was decreased, but the loading efficiency was increased. The higher the emulsification speed was, the smaller the particle size and loading efficiency were. The concentration of calcium chloride did not show any effect on the particle size and loading efficiency. In the spray preparation method, the particle size was increased as the nozzle pressure $(from\;1\;kgf/m^2\;to\;3\;kgf/m^2)$ and spray rate was raised. Increasing calcium chloride concentration (<7%) decreased the particle size, in contrast to no effect of calcium chloride concentration on the w/o-emulsion preparation method. Alginate microspheres prepared by two methods were different in the particle size and loading efficiency, the particle size of microspheres prepared by the spray method was about $2-6\;{\mu}m$, larger than that prepared by the w/o emulsion method $(about\;2{\mu}m)$, and the loading efficiency was also higher with spray method. Furthermore, drying process for the microspheres prepared by the spray was simpler and easier, compared with the w/o emulsion preparation. Therefore, the spray method was chosen to prepare alginate microspheres for further experiments. Release pattern of FITC-BSA in alginate microspheres was evaluated in simulated intestinal fluid and PBS (phosphate buffered saline). Dissolution rate of FITC-BSA from alginate/chitosan microsphere was lower than that from alginate microsphere and alginate/poly-L-lysine microsphere. By confocal laser scanning microscope, it was revealed that alginate/FITC-poly-L-lysine microspheres were present in close apposition epithelium of the Peyer's patches of rabbits following inoculation into lumen of intestine, which proved that microspheres could be taken up by Peyer's patch. In conclusion, it is suggested that alginate microsphere prepared by spray method, showing a particle size of & $10\;{\mu}m$ and a high loading efficiency, can be used as a model drug for the development of oral vaccine delivery system.
Provitamin $B_5$ liquid crystal ($PVB_5$-LC) was the new emulsion system to enhance moisturizing activity on the skin. In this study, it should be mentioned that PVB5-LC could be prepared with the main compound of hydrogenated lecithin (HL) in oil-in water (O/W) emulsion. The key ingredient of humectants was contained 2% of provitamin $B_5$ ($PVB_5$) into the $PVB_5$-LC. The best suitable compositions of $PVB_5$-LC were made from 4.0 wt% of HL, 4.0 wt% of cetostearyl alcohol (CSA) as emulsifier and gelling agent, 3.0 wt% of 1,3-butylene glycol (1,3-BG) and 2.0 wt% of glycerin as moisturizers, 3.0 wt% of cyclomethicone (CMC), 3.0 wt% of isononanoate (ININ), 3.0 wt% of capric/caprylic triglyceride (CCTG), 3.0 wt% of macadamia nut oil (MNO) as emollients. As the analytical result of $PVB_5$-LC, it could know that the distribution range of particle size was 0.14 to 12.37 m level (mean size 3.24 m). It was certified the multi lamellar phase around the droplet of liquid crystal when observed the droplet particles through a polarization microscope. And it clinically was tested the effectiveness of moisturizing activity (in-vivo) compared with control sample (O/W emulsion). The effectiveness of moisturizing activity of $PVB_5$-LC with Skincon-200EX after 6 hours went up 49.0% (p<0.05, n=20) whereas the effectiveness of moisturizing activity of generally O/W emulsion cream was 25.7% (p<005, n=20). Also, in case of Comeometer CM-825, the moisturizing activity of $PVB_5$-LC after 6 hours rose 36.6% (p<0.05, n=20) whereas the effectiveness of moisturizing activity of generally O/W emulsion cream was 10.8%. Therefore, it was known that the effectiveness moisturizing activity of $PVB_5$-LC with HL was remarkably superior compared with O/W emulsion cream.
