Effects of Type of Oilseed and Level of Concentrate on Fermentation, Biohydrogenation of Fatty Acids and Conjugated Linoleic Acid Production in a Rumen-Simulated Continuous Culture System (지방급원 형태와 수준에 따른 연속배양장치 내 반추위 발효성상, 지방산의 수소첨가 현상 및 Conjugated Linoleic Acid 생산에 미치는 영향 연구)

  • This experiment employed a rumen simulated continuous culture system to examine the possibility of improving the rumen bypass of polyunsaturated fatty acids (PUFA) by using a high proportion of concentrate in the feed, and compared soya and linseed in terms of conjugated linoleic acid (CLA) production. No effect of type of fat source was observed on ruminal fermentation. A high proportion of concentrate (80%) in the feed decreased (P<0.001) vessel pH but increased (P<0.01) ammonia nitrogen, total VFA, acetate, butyrate and valerate concentrations compared with a low proportion (40%). Fat sources (soya vs. linseed) and concentrate ratio in the feed did not affect digestibilities of organic matter (OM), total nitrogen, neutral detergent fiber (NDF) and acid detergent fiber (ADF). Soya increased the flows of trans C18:1, C18:2 n-6 and C18:3 n-3 compared with linseed. The difference in fat source alone did not affect the flow of CLA but this was increased when high levels of soya and linseed were associated with a high proportion of concentrate in the feed. There was no effect of fat source on biohydrogenation of C18:1 n-9 and C18:2 n-6, but biohydrogenation of C18:3 n-3 and total C18 PUFA was higher with the linseed than with the soya treatment. A high proportion of concentrate decreased biohydrogenation of C18:2 n-6, C18:3 n-3 and total C18 PUFA compared with a low proportion.

Characteristics of Food Nutrition Components of Common Stalked Barnacle Pollicipes mitella (거북손(Pollicipes mitella)의 식품 영양성분 특성)

  • Common stalked barnacle Pollicipes mitella is a filter-feeding crustacean that attaches itself to the surfaces of rocks in the ocean intertidal zone. Pollicipes mitella is one of about a dozen species of goose barnacle that are gathered commercially for human consumption. In Portugal and Spain, they are a widely consumed and expensive delicacy known as "Percebes (Pollicipes pollicipes )". However, common stalked barnacle is less popular in Korea, and information on its nutritional component is not yet available. In the present study, we analyzed the proximate and fatty acid compositions and mineral content of common stalked barnacle (muscle and cirrus). The muscle and cirrus of common stalked barnacle were 15.1% and 14.3% protein, and 3.31% and 1.81% lipid, respectively. The prominent fatty acids in the muscle and cirrus were 22:6n-3 (docosahexaenoic acid, DHA), 16:0, 20:5n-3 (eicosapentaenoic acid, EPA), 18:0, 18:1n-9, 16:1n-7 and 14:0. In particular, common stalked barnacle contained high levels of n-3 polyunsaturated fatty acids, such as DHA (25.4-25.6%) and EPA (14.7-15.1%), and low levels of the n-6 fatty acid, 20:4n-6 (1.2-1.3%). Common stalked barnacle contained a relatively large amount of Zn (10-42 mg/100 g) in its muscle and cirrus.

Production of Gamma-Linolenic Acid in Pichia pastoris by Expression of a Delta-6 Desaturase Gene from Cunninghamella echinulata

  • Gamma-linolenic acid (GLA, C18:3 ${\Delta}^{6,9,12}$) is synthesized by a delta-6 fatty acid desaturase using linoleic acid (LA, C18:2 ${\Delta}^{9,12}$) as a substrate. To enable the production of GLA in the conventional yeast Pichia pastoris, we have isolated a cDNA encoding the delta-6 fatty acid desaturase from Cunninghamella echinulata MIAN6 and confirmed its function by heterogeneous expression in P. pastoris. Sequence analysis indicated that this cDNA sequence has an open reading frame of 1,404 bp, which encodes a 52 kDa peptide of 468 amino acids. This sequence has 64% identity to the previously reported delta-6 fatty acid desaturase from Rhizopus oryzae. The polypeptide has a cytochrome b5 domain at the N-terminus including the HPGG motif in the heme-binding region, as reported for other delta-6 fatty acid desaturases. In addition, this enzyme differs from other desaturases by the presence of three possible N-linked glycosylation sites. Analysis of the fatty acid composition demonstrated the accumulation of GLA to the level of 3.1% of the total fatty acids. Notably, the amounts of ginkgolic acid (C17:1) and palmitic acid (C16:0) were increased from 1.3% to 29.6% and from 15% to 33%, respectively. These results reveal that the modification of the fatty acid biosynthetic pathway by genetic manipulation in order to produce specific polyunsaturated fatty acids in P. pastoris is a promising technique.

Backfat Characteristics of Barrows and Gilts Fed on Tuna Oil Supplemented Diets during the Growing-finishing Periods

  • This study was conducted to evaluate the effect of supplementing tuna oil to diets of growing-finishing pigs (barrows and gilts) on backfat characteristics when slaughtered at different weights. Four hundred and eighty crossbred (Large White$\times$Landrace$\times$Duroc) pigs averaging 30 kg were allotted to 12 treatment combinations (40 pigs/treatment combination) in a completely randomized design with a $2{\times}2{\times}3$ factorial arrangement of treatments. The treatments were: dietary tuna oil supplementation (0 and 2%); sex (barrows and gilts); and slaughter weight (90, 100 and 110 kg). As pigs reached their slaughter weight, they were randomly selected (8 pigs/treatment combination; 96 pigs in total) and slaughtered. Backfat colour, hardness and fatty acid profile were assessed. There were significant (p<0.05) differences in colour (L* and a* values) among treatments. Backfat of the control group was harder than on the tuna oil (p<0.001) and that of barrows was harder than of gilts (p<0.05). In addition, the thiobarbituric acid reactive substances (TBARS) values of fat from the tuna oil group stored for 3 days were higher (p<0.001) than the control group. The TBARS values of gilts tended to be higher than those of barrows and increased with increasing slaughter weight in the tuna oil group. The cholesterol and triglyceride levels were not affected by diet and sex but the triglyceride level increased with increasing slaughter weight (p<0.01). The tuna oil group had higher polyunsaturated fatty acid (PUFA) content, ratio of PUFA: saturated fatty acid (SFA) and total n-3 fatty acids but lower monounsaturated fatty acids content and n-6:n-3 fatty acids than the control group (p<0.01). Gilts had higher PUFA and n-6 fatty acids in backfat than barrows (p<0.05). The backfat from both 90 and 100 kg slaughter-weight groups had a lower ratio of n6:n3 fatty acid than the 110 kg slaughter-weight group (p<0.05). However, this was more pronounced in the tuna oil group. The PUFA: SFA was also increased while the n-6:n-3 ratio tended to reach the recommended levels for healthy eating in human beings of <5. However, due to oxidative susceptibility, barrows should not be slaughtered at more than 100 kg for the meat to be acceptable to consumers.

Effects of Regular Dietary Fatty Acids on Body Fat Content and Blood Lipid Concentration in Middle-aged Korean Women (중년여성의 평상 식이 지방산이 체지방 함량 및 혈중 지질 함량에 미치는 영향)

  • The present study undertaken to investigate the interrelationship among their regular dietary fatty acid intake, body fat content(BFC), and plasma lipid fractions(PLF) of middle-aged Korean women. Forty-nine women (age 30 to 49) participated in this study in which the aim was to determine their total nutrient intake per day, BFC and PLF, and to analyse the intercorrelations of these. Strong correlation was seen among their body mass index, mid-upper-arm-circumference, and BFC which were positively correlated with plasma total lipids(TL) and triglycerides(TG). Dietary myristic acid and lauric acid were positively correlated with both total cholesterol(TC) and LDL-cholesterol (LDL-C) in plasma. Palmitic acid, stearic acid, oleic acid, and total monounsaturated fatty acid(MUFA) were negatively correlated with HDL-cholesterol(HDL-C). Although dietary total saturated fatty acid (SFA) was not correlated with TC and LDL-C, it was positively correlated with TL and TG in plasma. However, dieary n-3 polyunsaturated fatty acid(PUFA) in regular Korean diet was not correlated with any of lipid fractions in plasma which might be due to the high ratio of n-6/n-3 in their diet. From the above results, it was concluded that plasma lipid fractions were modulated by their dietary nutrients, especially the types of the fatty acids. Dietary SFA and MUFA intake not PUFA are strongly correlated with LDL-C and HDL-C in the Korean diet This may relate to the frequency of coronary heart disease in the age group.

Enhanced Uptake of Modified Low-Density Lipoprotein by Eicosapentaenoic Acid-Treated THP-1 Macrophages

  • Animal and clinical studies as well as epidemiological data have provided convincing evidence that n-3 polyunsaturated fatty acids can protect against atherosclerosis. However, the effects of the fatty acids on atherogenesis are contradictory. This discrepancy could derive from great susceptibility of the fatty acids to oxidation. We investigated the effect of eicosapentaenoic aced(EPA) on cellular atherogenesis via the scavenger receptor of THP-1 derived macrophages. THP-1 cells were fully differentiated into macrophages by incubating with phorbol 12-myristate 13-acetate for seven days. Atherogenic features of EPA were compared by subsitituting for linoleic acid (LA). Macrophages were also incubated without treatment of the fatty acids as controls. EPA (5-50 nmol/mL) was not cytotoxic and did not measurably induce cellular oxidation compared to bovine serum albumin (BSA) vehicle or identical doses of LA. EPA increased macrophage uptake and degradation of acetylated LDL(AcLDL) up to 14% and 88%, respectively. EPA increased markedly total cellular sterol synthesis and heparin-releasable lipoprotein lipase activity of macrophages, indicating that EPA may enhance accumulation of cellular cholesteryl ester and possibly facilitate formation of foam cells. These results demonstrate that EPA promotes the modified LDL-triggered atherosclerotic process by the modulation of the scavenger receptor and the activation of LPL in macrophages.

Association between Omega Fatty Acid Intake and Suicidality : Sex Differences in the General Korean Population

  • Objectives : Epidemiological studies in other countries show that a low intake of omega-3 fatty acids (FAs) increases the risk of depression or suicidality. However, no studies have investigated the associations of suicidality with omega-3 FAs in Korea. Hence, this cross-sectional study aimed to investigate the effects of omega FAs on suicidality in the general South Korean population. Methods : The data in this study were sourced from adults (n=215,860) who completed the Sixth Korea National Health and Nutrition Examination Survey (KNHANES VI), and the associations between omega FAs and suicidality were analyzed using multivariate logistic regressions. Results : Our results demonstrated that high omega-3 FA intake was associated with a decreased risk of suicide (OR=0.83, 95% CI : 0.71-0.98) and the high omega-6 to omega-3 FA ratio was associated with an increased risk of suicide (OR=1.25, 95% CI : 1.02-1.54). Additionally, a high intake of omega-3 FAs was associated with a decreased risk of suicide in men, but not in women (OR=0.72, 95% CI : 0.59-0.88). Conclusions : Overall, our findings suggest that a lower intake of omega-3 FA is associated with the increased risk of suicidality in the general Korean population, especially in men.

Chemical Characteristics of Commercial Korean Soy Sauce Produced by Alkali Treatment Following Acidic Hydrolysis

  • Alkali treatment following acidic hydrolysis (ATAH) is a commonly used practice for reducing the levels of monochloropropanediol in commercial Korean soy sauce. This study investigated the chemical compounds produced in commercial Korean soy sauce made by ATAB. The levels of amino-N, total acidity, pH, salinity and Brix of the soy sauce were 6.66%, 2.52%, 19.81%, 4.57 and 35.01, respectively. The major fatty acids were C18:2n-6, C16:0, Cl8:ln-9 and C18:3n-6. The concentration of polyunsaturated fatty acids were especially high, with of C18:2n-6 (49.8%) being the highest followed by C18:3n-6 (3.8%) and C18:3n-3 (2.4%) in that order. Among the non-volatile organic acids, the concentration of levulinic acid (1,206.28 mg/100 g) was the highest, while the taste value of citric acid was the highest. Among the ATP related compounds, IMP concentration (31.19 mg/100 g) was highest followed by AMP, hypoxanthine and GMP in that order. The concentrations of free and total amino acids in soy sauce were 6,136.94 mg/100 g and 8,702.76 mg/100 g, respectively. On the other hand, the taste value of glutamic acid, a major amino acid flavor determinant in soy sauce, was highest of all the amino acids, which is desirable since most free amino acids such as methionine, histidine and phenylalanine have a bitter taste that detracts from the flavor of soy sauce.

Neurocognitive Functions in Infants with Malnutrition; Relation with Long-chain Polyunsaturated Fatty Acids, Micronutrients Levels and Magnetic Resonance Spectroscopy

  • Purpose: Malnutrition may influence neurocognitive development in children by directly affecting the brain structural development, or indirectly by affecting the children's cognition experience. Malnutrition alters the cell numbers, cell migration, synaptogenesis, and neurotransmission due to inadequate availability of necessary micronutrients to support cell growth. We aimed to analyze neurocognitive development in infants with malnutrition and its association with long chain polyunsaturated fatty acids (LC-PUFA), micronutrients levels and magnetic resonance spectroscopy (MRS) findings. Methods: The study included two groups; group 1, infants with malnutrition (n=24), group 2; healthy infants (n=21). Peripheral blood was obtained from the participants for studying micronutrients and LC-PUFA levels. The neurocognitive development was analyzed by the use of an Ankara Developmental Screening Inventory test. MRS were performed on all infants. Results: All parameters of neurocognitive development and serum calcium ($9.6{\pm}0.9mg/dL$ vs. $10.4{\pm}0.3mg/dL$, p<0.05) and magnesium ($2.02{\pm}0.27mg/dL$ vs. $2.2{\pm}0.14mg/dL$, p<0.05) levels were noted as being low in infants with marked malnutrition. No difference was found in LC-PUFA levels between healthy and malnourished infants. Thalamic choline/creatine levels were significantly high in infants with malnutrition ($1.33{\pm}0.22$ vs. $1.18{\pm}0.22$, p<0.05). Total neurocognitive development in infants was positively correlated with serum calcium levels (p<0.05, r=0.381). Conclusion: Calcium supplementation may improve neurocognitive development in malnourished infants.

Dose-Response Effects of n-3 Polyunsaturated Fatty Acids Supplement on the Metabolism of Streptozotocin-Induced Diabetic Rats (n-3 고도불포화 지방상 수준을 달리한 식이가 Streptozotocin으로 유발된 당뇨쥐의 대사에 미치는 영향)

  • This study was aimed to observe dose-response effects of squid liver oil enriched in n3 polyunsaturated fatty acids(n-3 PUFA) on the metabolism of streptozotocint(STZ)-induced diabetic rats. In this experiment, 24 STZ-induced diabetic male rats of Sprague Dawley strain were divided into 4 groups and fed for 4 weeks with basal diet(0%). 33%,67% and 100% squid liver oil(SLO) of total fat content, and 6 normal rats were fed with 0% SLO diet at the same time. The results obtained were as follows: 1. Rat group fed with 33% SLO diet showed the least body weight loss and changes in blood glucose, while ones of 100% SLO diet showed the highest level. 2. Serum total protein and ratio of albumin to globulin of all the groups were below the standard level, but did not show significantly different among diet groups. 3. Serum creatinine concentration of all the groups were stayed whthin normal range, but BUN were 3 to 4 times higher than normal rats. BUN concentration of rats fed with 0% and 33% SLO diet was significantly lower than those of others. 4. Total-cholesterol level of serum increased in all the groups except 33% SLO diet, but since HDL-chol, levels and TG concentration went up with an incerase of SLO in the diets, the ratio of HDL-chol. to total-chol, of rats fed with 67% and 100% SLO diet showed higher than those with 0%, and 33% SLO, and TG concentration of rats fed with 67% and 100% SLO diet decreased significantly.
