Effect of Garlic Oil on Fatty Acid Accumulation and Glycerol-3-Phosphate Dehydrogenase Activity in Differentiating Adipocytes

  • Garlic oil (GAR, Allium sativum L.) has been studied as a feed additive to improve animal production performance and decrease methane emission in ruminants. The present study was designed to determine the possible effect of GAR on fatty acid composition and accumulation in animal fat tissue using a cell model. 3T3-L1 preadipocytes at $2{\times}10^{4}\;mL^{-1}$ were seeded to 24-well plates and allowed to proliferate to reach confluence. The cells were then treated with media containing 0, 2.5, 5, 10, 20 and 40 $\mu{g}$ $mL^{-1}$ of GAR during the differentiation period for 8 days. Media containing dexamethasone, methyl-isobutylxanthine and insulin was applied during the first 2 days of the early differentiation period. On day 8 sub-sets of the wells were stained with oil red-O and the remaining cells were harvested for determination of glycerol-3-phosphate dehydrogenase [EC 1.1.1.8] (GPDH) activity (n = 6) and cellular fatty acid concentration (n = 6). It was found that supplementation of GAR increased (p<0.05) the ratio of monounsaturated fatty acids/saturated fatty acids in the adipocytes and showed inhibitory effect (p<0.05) on the post-confluent proliferation. With relative low dosage, GAR (5-20 $\mu{g}$ $mL^{-1}$) increased (p<0.05) the GPDH activity without affecting the cellular fatty acid concentration, while a high dosage (40 $\mu{g}$ $mL^{-1}$) inhibited (p<0.05) fatty acid accumulation and decreased GPDH activity. Supplementation of GAR had an effect on cell post-confluent proliferation, differentiation and fatty acid accumulation. However, the effect may be diverse and depends on the dose applied.

Effect of Snake Venom Toxin on Inhibition of Colorectal Cancer HT29 Cells Growth via Death Receptors Mediated Apoptosis

  • Objectives : We investigated whether snake venom toxin(SVT) from Vipera lebetina turanica sensitizes HT29 human epithelial colorectal cancer cells to tumor necrosis factor(TNF)-related apoptosis-inducing ligand(TRAIL) induced apoptosis in cancer cells. Methods : Cell viability assay was used to assess the inhibitory effect of TRAIL on cell growth of HT29 human colorectal cancer cells. And 6-diamidino-2-phenylindole(DAPI), terminal deoxynucleotidyl transferase mediated dUTP nick end labeling assay(TUNEL) staining assay were used to evaluate cell-apoptosis. Western blot analysis were conducted to observe apoptosis related proteins and death receptor. To assess whether the synergized inhibitory effect of SVT and TRAIL on reactive oxygen species(ROS) generation was reversed by strong anti-oxidative agent. Results : SVT with TRAIL inhibited HT29 cell growth different from TRAIL alone. Consistent with cell growth inhibition, the expression of TRAIL receptors; Expression of death receptor(DR)4 and DR5 was significantly increased and intrinsic pro-apoptotic cleaved caspase-3, -9 was subsequently increased together with increase of Bax/Bcl-2 ratio and extrinsic pro-apototic caspase-8 was also activated. In addition, the expression of anti-apoptotic survival proteins, a marker of TRAIL resistance(eg, cFLIP, survivin, X-linked inhibitor of apoptosis protein(XIAP) and Bcl-2) was suppressed by the combination treatment of SVT and TRAIL. Pretreatment with the ROS scavenger N-acetylcysteine abolished the SVT and TRAIL-induced upregulation of DR4 and DR5 expression and expression of the intrinsic pro-apoptotic caspase-3 and-9. Conclusion : The collective results suggest that SVT facilitates TRAIL-induced apoptosis in $HT_{29}$ human epithelial colorectal cancer cells through up-regulation of the TRAIL receptors; DR4 and DR5 and consecutive induction of bilateral apoptosis via regulating apoptosis related proteins.

Manufacturing Technology for Tape Casting and Soft Magnetic Powder Using by Recycling Scrap of Fe-Si Electrical Sheet (Fe-Si 전기강판 폐스크랩을 이용한 연자성 분말 및 테이프 제조기술)

  • This study focused on examining the possibility for recycling of Fe-Si electric sheet. We manufactured Fe-6.5Si mother alloy using by Fe-Si electric sheet scrap for transformer core materials. And then, soft magnetic alloy powder which diameter and shape were $45{\sim}150{\mu}m$ and sphere type was prepared by gas atomization process. As we compared to commercial Fe-6.5Si powder, its diameter distribution and microstructure of recycled powder was a similar. To investigate the possibility of reusing the soft magnetic composite sheet for electronics, recycled powder was treated to have a high aspect ratio (AR), and we finally obtained the 65~66 AR and $2.3{\mu}m$ thickness powder. To release the residual stress of powder, heat treatment was conducted under $300{\sim}400^{\circ}C$, $N_2$ gas. And then, soft magnetic sheet was made by tape casting process using by those powders. After the density and permeability of tape was measured, and we confirmed that the recycled Fe-Si electric sheet scrap was possible to reuse the soft magnetic materials of electronics.

Effect of Feeding Different Ratios of Green Fodder and Straw Supplemented with Wheat Bran on the Performance of Male Crossbred Calves

  • Twenty male crossbred calves of about one year of age (average body weight, 196 kg) were distributed in four equal groups following complete randomized design. Wheat bran was supplemented to four different combinations of wheat straw and green fodder (Sorghum vulgare) at 40:60, 30:70, 20:80 and 10:90 ratios (on as fed basis) for the feeding of animals in Group 1, 2, 3 and 4, respectively. The feeding trial was continued for a period of 70 days including one metabolism trial of 6 days collection of feed, faeces and urine sample to determine the intake and utilization of nutrients. The intakes (g/kg $W^{0.75}$) of DM, TDN and CP were $93.0{\pm}1.8$, $55.5{\pm}1.1$ and $9.51{\pm}0.18$ in Group 1; $98.0{\pm}1.8$, $59.6{\pm}1.1$ and $10.33{\pm}0.19$ in Group 2; $98.1{\pm}2.4$, $60.5{\pm}1.5$ and $10.79{\pm}0.26$ in Group 3; and $97.7{\pm}1.7$, $59.1{\pm}1.0$ and $10.78{\pm}0.19$ in Group 4, respectively. The digestibility of nutrients did not differ significantly among the groups. Relatively higher nutrient intake and balances of nitrogen reflected non-significantly high her live weight gain in the later three groups (436, 439 and 464 g, respectively) as compared to Group 1 (400 g). The DM intake remained unchanged by increasing the proportion of green fodder beyond 20:80 ratio and thus was assessed to be satisfactory for optimum productivity in animals.

Effect of Synchronizing Starch Sources and Protein (NPN) in the Rumen on Feed Intake, Rumen Microbial Fermentation, Nutrient Utilization and Performance of Lactating Dairy Cows

  • Eight crossbred (75% Holstein Friesian) cows in mid-lactation were randomly assigned to a switchback design with a 2x2 factorial arrangement to evaluate two nonstructural carbohydrate (NSC) sources (corn meal and cassava chips) with different rumen degradability and used at two levels of NSC (55 vs. 75%) with protein source (supplied by urea in the concentrate mix). The treatments were 1) Low degradable low level of corn (55%) 2) Low degradable high level of corn (75%) 3) High degradable low level of cassava (55%) and 4) High degradable high level of cassava (75%). The cows were offered the treatment concentrate at a ratio to milk yield at 1:2. Urea-treated rice straw was offered ad libitum as the roughage and supplement with 1 kg/hd/d cassava hay. The results revealed that total DM intake, BW and digestion coefficients of DM were not affected by either level or source of energy. Rumen fermentation parameters; NH3-N, blood urea nitrogen and milk urea nitrogen were unaffected by source of energy, but were dramatically increased by level of NSC. Rumen microorganism populations were not affected (p>0.05) by source of energy, but fungal zoospores were greater for cassava-based concentrate than corn-based concentrate. Milk production and milk composition were not affected significantly by diets containing either source or level of NSC, however concentrate than corn-based concentrate averaging (4.4 and 4.2, respectively). Likewise, income over feed, as estimated from 3.5% FCM, was higher on cassava-based concentrate than corn-based concentrate averaging (54.0 and 51.4 US$/mo, respectively). These results indicate that feeding diets containing either cassava-based diets and/or a higher of oncentrates up to 75% of DM with NPN (supplied by urea up to 4.5% of DM) can be used in dairy rations without altering rumen ecology or animal performance compared with corn-based concentrate.

Amino Acid-Based Material for the Complementary Therapy of Decubitus Ulcers

  • Chronic wounds, pressure sores, lesions, and infections of microbial origin in bedridden, paralyzed, or malnutrition patients remain the object of study of many researchers. A variety of factors behind the development of these disorders are related to the patient's immune system, making it unable to respond effectively to the treatment of the wound. These factors can be properly controlled, giving particular importance to the ethiology and stage of the wound, as well as the time periods corresponding to the replacement of the dressings. The present research reports a novel foam/soft material, ${{\small}L}$-Cys-g-PCL, with an application for decubitus/pressure ulcers, especially for wounds with a difficult healing process due to infections and constant oxidation of the soft tissues. During this work, the interactions between S. aureus and ${{\small}L}$-Cys-g-PCL foam were studied under conditions that simulate decubitus ulcers; namely, pH and exudate. The effects of duration of grafting (1 or 8 h) and pH (7.0 and 8.9) on wettability, surface energy, swelling, and porosity were also evaluated. Results showed an effective microbicidal activity exhibiting an inhibition ratio of 99.73% against S. aureus. This new ${{\small}L}$-Cys-g-PCL soft material showed saftey to contact skin, ability to be shaped to fill in sunken holes (craters) - pressure ulcers stage III - and to act as a smart material responsive to pH, which can be tailored to develop better swelling properties at alkaline pH where exudates are normally higher, so as to address exudate self-cleaning and prevention of desiccation.

Bioavailability of Phosphorus in Two Cultivars of Pea for Broiler Chicks

  • The aim was to determine the relative bioavailability of phosphorus (P) in peas for 21-day old broiler chickens using slope-ratio assay. One hundred and sixty eight male Ross 308 broiler chicks were divided into 42 groups 4 balanced for body weight and fed 7 diets in a completely randomized design (6 groups/diet) from day 1 to 21 of age. The diets were a corn-soybean meal basal diet, and the corn-soybean meal basal diet to which monosodium phosphate, brown- or yellow-seeded pea was added at the expense of cornstarch to supply 0.5% or 1% total phosphorus. Monosodium phosphate was included as a reference, and hence the estimated bioavailability of P in pea cultivars was relative to that in the monosodium phosphate. Birds and feed were weighed weekly and on d 21 they were killed to obtain tibia. The brown-seeded pea contained 23.4% crude protein, 0.47% P, whereas the yellow-seeded pea contained 24.3% crude protein and 0.38% P. Increasing dietary P supply improved (p<0.05) chick body weight gain and tibia ash and bone density. The estimated relative bioavailability of p values for brown- and yellow-seeded peas obtained using final body weight, average daily gain, tibia ash, and bone mineral density were 31.5% and 36.2%, 35.6% and 37.3%, 23.0% and 5.60%, and 40.3% and 30.3%, respectively. The estimated relative bioavailability of p values for brown- and yellow-seeded peas did not differ within each of the response criteria measured in this study. In conclusion, the relative bioavailability of P in pea did not differ depending on the cultivar (brown- vs yellow-seed). However, the relative bioavailability of P in pea may vary depending on the response criterion used to measure the bioavailability.

A Comparison of the Plant Community Structures in the Burned and Unburned Areas of Mt Kumo-san (금오산에서 산회지와 비산화지의 식물군집구조 비교)

  • This is a report on the recovery of vegetation and secondary succession in the burned area studied from April, 1990 to April. 1991. The forest fire occurred in a part of Mt. $K\v{u}mo-san$ on April, 1986 and the pine forest and its understory vegetation were burned out completely. The floristic compositions of burned (B) and unburned (U) areas were composed of sixty eight and thirty one species (vascular plants), respectively. These species were divided into invaders (47 species), increasers (15 species), deceasers (3 species), neutrals (3 species), and retreaters (10 species) on the basis of summed dominance ratio ($SDR_3$). Biological spectra showed the $H-D_1-R_5-e$ type in both the burned and unburned areas. The species of Lespedeza ($SDR_3$=94.7), Miscanthus (91.95), Festuca (68.33), and Spodiopogon (52.06) were dominant in the burned areas, while the species of Pinus (76.67), Robinia (56.25), Quercus (52.08), and Carex (40.25)were dominant in the unburned area. Dominance index (C) in burned and unburned areas was 0.15 and 0.25, respectively. the index of similarity (CCs) was 0.42. The degree of succession (DS) and species diversity (H) in burned and unburned areas were 675.8, 884.2 and 4.07, 2.05, respectively. The degree of succession in the burned area graduall increased and the burned area was recovered to be simmilar to the unburned area. Evenness index in burned and unburned areas was 0.965 and 0.595, respectively.

A Study on Satisfaction and Formative Appropriateness of Leisure Sports Jumpers For Elderly Women (노년 여성 레포츠 점퍼에 대한 만족도 및 형태적합성 연구)

  • We need to develop a target market for elderly women by increasing the ratio of functional apparel. The purpose of this study was to provide basic data for developing a Leisure Sports Jumper and improving the quality of life of clothing for female elderly consumers. This study reviewed the current situation on the production of the functional Leisure Sports Jumper for the elderly women in literature. For the experimental study, we selected Leisure Sports Jumpers from five apparel brands, and evaluated a sensory test on 20 subjects representing the average body shape of the elderly women aged from 65 to 75. After the first sensory test, we manipulated the patterns for samples of the Leisure Sports Jumper. In the second sensory test, visual and cognitive appropriateness of the Leisure sports jumpers were assessed by the elderly women. The results of the study were as follows: the Leisure Sports Jumper of brand 'C' showed the highest appropriateness in terms of neck, armholes, and sleeves among the 5 brands. The Leisure Sports Jumper of brands 'B' and 'D' showed the lowest appropriateness in the width of the armpits among the 5 brands. Except for the length of the Leisure Sports Jumper, the margin, location of the pocket, and armholes had the lowest score in the first test showing over 3.5 points.

Estimation and Characteristics of Atmospheric Deposition Flux of Polycyclic Aromatic Hydrocarbons (PAHs) into the Masan and Haengam Areas of Korea (마산과 행암 지역으로 유입되는 다환방향족탄화수소(PAHs)의 대기 침적 플럭스 산정과 특성)

  • Atmospheric bulk (wet and dry) samples were monthly collected in Masan and Heangam areas of Korea, to assess the deposition flux and seasonal variation of polycyclic aromatic hydrocarbons (PAHs). Deposition fluxes of PAHs in bulk samples were determined using gas chromatography coupled to mass spectrometer detector (GC/MSD). Particle deposition fluxes from Masan and Haengam areas varied from 13 to $87\;g/m^2/year$ and from 5 to $52\;g/m^2/year$, respectively. PAHs deposition fluxes in atmospheric bulk samples in Masan and Haengam areas ranged from 135 to $464\;{\mu}g/m^2/year$ and from 62.2 to $194\;{\mu}g/m^2/year$, respectively. Atmospheric deposition fluxes of particles and PAHs in this study were comparable to or slightly lower values than those from different locations in Korea and other countries. PAHs profiles of atmospheric deposition bulk samples showed slightly different from two sampling areas, however the predominant species of PAHs were similar. Indeno (1,2,3-c,d)pyrene, benzo(g,h,i)perylene, phenanthrene compounds were the most detected PAHs in deposition bulk samples. Carcinogenic PAHs occupied the contribution of approximately $30-40\%$ of the total PAHs deposition fluxes. The non-metric multi-dimensional scaling (MDS) was used, to assess the differentiation of PAHs source between two sampling areas. The result suggests that PAHs contamination sources were different according to the location and season surveyed. There was no an apparent relationship between the PAHs deposition flux against temperature and rainfall amount, even though summer season with the highest temperature and the largest amount of precipitation showed the lowest PAHs deposition flux. Benzo(e)pyrene/benzo(a)pyrene ratio indicated that the photo-degradation process was one of important factors to the seasonal variation of PAHs with the lower deposition fluxes.
