Optimization of Conditions for the Microencapsulation of ${\alpha}-Tocopherol$ and Its Storage Stability (${\alpha}-Tocopherol$ 미세캡슐화의 최적화 및 저장안정성 규명)

  • We have produced the microcapsule composed of ${\alpha}-tocopherol$ as a core material (Cm) and the gelatinized polysaccharide as a wall material (Wm). Firstly, we have developed a simple, sensitive, and quantitative analysis method of the microencapsulation product using 5% cupric acetate pyridine solution. We could then optimize all the conditions for the microencapsulation process such as the ratio of [Cm] to [Wm], the temperature of dispersion fluid, and the emulsifier concentration using response surface methodology (RSM). As for the microencapsulation of ${\alpha}-tocopherol$, the regression model equation for the yield of microencapsulation (YM, %) to the change of an independent variable could be predicted as follows : YM=99.77-1.76([Cm]:[Wm])-1.72$([Cm]\;:\;[Wm])^2$. From the ridge of maximum response, the optimum conditions for the microencapsulation of ${\alpha}-tocopherol$ were able to be determined as the ratio of [Cm] to [Wm] of 4.6:5.4(w/w), the emulsifier concentration of 0.49%, and dispersion fluid temperature of $25.5^{\circ}C$, respectively. Finally, the microcapsules produced under the optimal conditions were applied for the analysis of storage stability. The optimal conditions for the storage were found to be the values of pH 9.0 and $25{\sim}35^{\circ}C$. And the storage stability of the microcapsules containing ${\alpha}-tocopherol$ were higher than 99% for a week at pH 9.0 and $25^{\circ}C$.

Effect of Restriction of Vitamin A and D on Carcass Characteristics in Hanwoo Steers (비타민 A와 D의 공급제한이 거세 한우의 육질등급에 미치는 영향)

  • Sixty Hanwoo steers(15 months of age; 409±29.2 kg of BW) were used to evaluate the effects of dietary vitamins A and D restriction on carcass characteristics. Steers were allotted randomly to 1 of 4 treatments: Control(diet supplemented with vitamins A, D and E), -A (diet supplemented with vitamins D and E), -D(diet supplemented with vitamins A and E) and -AD(diet supplemented with vitamin E only). Steers were fed the experimental diet for a period of 8 months(until 23 months of age), and then supplemented with vitamins A and D at 0.05% of the diet(as fed-basis) from 24 to 26 months of age, and at 0.1% of the diet from 27 to 31 months of age(harvesting time). Dietary restriction of vitamins A and D did not affect DM intake, daily gain and feed conversion ratio. But the concentration of serum retinol was significantly(P<0.05) decreased by vitamin A restriction with the lowest concentration being seen at 23 months of age(345.0 ㎍/L and 326.7㎍/L for control and -D treatment versus 169.3 ㎍/L and 175.4 ㎍/L for -A and -AD treatments). The serum concentration of 25(OH)D3 was also decreased significantly(P<0.05) by vitamin D restriction and the lowest concentration was seen at 18 months of age(53.7ng/ml and 61.8ng/ml for control and - A treatment versus 24.0 ng/ml and 24.5 ng/ml for -D and -AD treatments). After the restriction period of vitamins A and D, the concentrations of retinol and 25(OH)D3 for - A, -D and -AD treatments were recovered at those of control. Dietary restriction of vitamins A and D did not affect carcass weight, backfat thickness, ribeye area, quality grade and yield grade. But marbling score was significantly increased by vitamin A restriction compared with control(6.73, 6.87 and 5.73 for -A, -AD and control, respectively). The results of the present study suggested that dietary vitamin A restriction could improve marbling score in Hanwoo steers.

Characterization of Dissolved Organic Matter in Stream and Industrial Waste Waters of Lake Sihwa Watershed by Fluorescence 3D-EEMs Analysis (형광 3D-EEMs를 이용한 시화호유역 하천 및 공단폐수의 유기물 특성 분석)

  • This study is conducted to examine spatial variations of Dissolved Organic Matter (DOM) in stream and waste waters of the different watershed areas (agricultural, residential, and industrial complex area) by using fluorescence 3D-EEMs (3 Dimensional Excitation Emission Matrix Spectroscopy). Furthermore, the research investigates the changes of DOM characterization by synchronous and 3D-EEMs during a rainfall event. The characterizations of DOM obtained by 3D-EEMs show two noticeable peaks at humic and protein-like regions. Humic-like substances (HLS) are found in rural and urban areas, and humic and protein-like substances (PLS) are shown in industrial area. According to the fluorescence peak $T_1:C_1$ ratios, it is observed that high amount of HLS was discharged from Banweol Industrial Complex (3TG). Additionally, linear relationships (Regression rate, $r^2$=0.65, $r^2$=0.66) have been shown between PLS (peak $T_1,\;B_1$) and biochemical oxygen demand (BOD), which indicates the impact of sewage. For the rainfall event (30 mm), no remarkable difference of DOM was found at rural area except increment of fluorescence intensity comparing dry period. In contrast, HLS at urban area is highly discharged within 30 minutes from the beginning of rainfall. Also, there are high influences of HLS and PLS within 20 minutes at industrial complex (4TG). Fluorescence 3D-EEMs has not only verifies a watershed of DOM origination but also monitors diffuse and point source impacts.

Determination of Adequate Method for Protein Extraction from Rice Bran and the Substitution of Dried Skim Milk with Protein Concentrate from Rice Bran in Early Weaned Pigs

  • The present study was conducted to determine a feasible method of protein concentrate extraction from rice bran (RBPC) and its effect as a substitution for skim milk in early weaning pig diets. An investigation to extract protein concentrate from full fat rice bran was undertaken to determine the best ratio of water and rice bran, the amount of NaOH and a HCl solvent to use in a simple paddle-type mixer with modified spinning to produce RBPC. The results stated that the best ratio for water mixing in the RBPC extraction process was 1:5 with 20 g NaOH and 30 min in a paddle-type mixer at 300 rpm. A mix of 250 ml 0.2 N HCl was optimum for neutralization and protein precipitation. After the fluid was spun out with a washing machine, the sediment was left for 12-14 hours to complete the filtration. One kilogram of rice bran could produce an average of 324.5 gram RBPC and it contained 3.40% ash, 496.48 kcal of GE/100 gram, 1.94% crude fiber, 28.20% ether extract, 7.64% moisture and 16.66% crude protein, respectively. A total of 45 crossbred piglets, weaned at 3 weeks of age were allotted into control diet (A) and dietary treatments formulated with a four different rates of RBPC substitution for skim milk at a percentage of 25 (B), 50 (C), 77 (D) and 100 (E) respectively, in a randomized complete block (RCB) design. All piglets had free access to feed and water until 8 week of age when the experiment ended. Feed intake, average daily gain, growth rate and feed efficiency were not affected by dietary treatments. Blood test parameters after completion of the growth trial indicated normal health. Even though the mean of cell hemoglobin concentration was significantly different between treatments (p<0.05) it was still within the normal range. The cost difference for BW gain of 100% RBPC substituted for skim milk in the weaning diet was approximately 35% lower than that of the control and the relative cost of production was 96.67, 92.85, 70.75 and 64.48% lower for the replacement of 25, 50, 75 and 100% of skim milk respectively. These results implied that this technology is feasible for use by small scale farmers to improve their self-reliance.

ISOLATION, IDENTIFICATION AND CHARACTERIZATION OF AN IMMOBILIZED BACTERIUM PRODUCING N2 FROM NH4+ UNDER AN AEROBIC CONDITION

  • To treat wastewater efficiently by a one-step process of nitrogen removal, a new bacterial strain producing $N_2$ gas from ${NH_4}^+$ under an aerobic condition was isolated and identified. The cell was motile and a Gram-negative rod, and usually occurred in pairs. By 16S-rDNA analysis, the isolated strain was identified as Enterobacter asburiae with 96% similarity. The isolate showed that the capacity of $N_2$ production under an oxic condition was approximately three times higher than that under an anoxic condition. Thus, the consumption of ${NH_4}^+$ by the isolate was significantly different in the metabolism of $N_2$ production under the two different environmental conditions. The optimal conditions of the immobilized isolate for $N_2$ production were found to be pH 7.0, $30^{\circ}C$ and C/N ratio 5, respectively. Under all the optimum reaction conditions, $N_2$ production by the immobilized isolate resulted in reduction of ORP with both the consumption of DO and the drop of pH. The removal efficiencies of $COD_{Cr}$, and TN were 56.1 and 60.9%, respectively. The removal rates of $COD_{Cr}$, and TN were the highest for the first 2.5 hrs with the removal $COD_{Cr}/TN$ ratios of 32.1, and afterwards the rates decreased as reaction proceeded. For application of the immobilized isolate to a practical process of ammonium removal, a continuous operation was executed with a synthetic medium of a low C/N ratio. The continuous bioreactor system exhibited a satisfactory performance at 12.1 hrs of HRT, in which the effluent concentrations of ${NH_4}^+$-N was measured to be 15.4 mg/L with its removal efficiency of 56.0%. The maximum removal rate of ${NH_4}^+$-N reached 1.6 mg ${NH_4}^+$-N/L/hr at 12.1 hrs of HRT(with N loading rate of $0.08\;Kg-N/m^3$-carrier/d). As a result, the application of the immobilized isolate appears a viable alternative to the nitrification-denitrification processes.

Gasification Study of Datong Coal in a Bench Scale Unit of Entrained Flow Gasifier (Datong탄에 대한 Bench Scale Unit급 분류층 석탄가스화 연구)

  • Coal gasification experiments were performed to characterize the bench scaled unit of 0.5∼1.0 T/D entrained coal gasifier developed by KIER. Datong coal from China was selected for this study. The system was operated at the temperature range of 1300∼1550$^{\circ}C$, with 62.5% of coal water mixture on the basis of dry coal. Oxygen and slurry mixture were preheated prior to feeding into burner and the ratio of oxygen/coal was in the range of 0.8∼1.2. In the preparation of coal water mixture, 0.3 wt% of CWM1002 and 0.05 wt% of NaOH wire added to reduce viscosity as well as to enhance theological properties of slurry. The resultant gaseous products consist primarily of hydrogen, carbon monoxide, carbon dioxide, and minor amounts of methane. Formation of H$_2$and CO was increased, while CO$_2$was decreased as the reacting temperature being increased due to the char-CO$_2$reaction. Maximum production of H$_2$and CO occurred in the O$_2$/coal ratio of 0.9 at 1530$^{\circ}C$. Heating values of product gases were in the range of 1700∼2400 kcal/N㎥.

Effect of Benzyladenopurine Concentration on Growth and Morphology of Soybean Sprouts and Comparison with Selling Products (BA 처리농도에 따른 콩나물의 생장과 시판 콩나물과의 비교)

  • Commercial value of soybean sprouts should be affected by their morphology including the lateral roots. This study was carried out to determine the effect of benzyladenopurine (BA) concentration on growth and morphology of soybean sprouts in order to compare them with the selling products collected from the markets. Four cultivars (cv. Eunhakong, Pungsannamulkong, Sowonkong, and Junjery) were cultured for 6 days after imbibed for 5 hours into different BA solutions (0, 1,2,4, and 8 ppm). On the 6th day, hypocotyl length was measured to calculate the composition rate of > 7 cm (A), 4 to 7 cm (B), < 4 cm (C) and non-germination (D), and the ratio of hypocotyl to root length (H/R ratio) on the base of hypocotyl length as well as lateral root, hypocotyl diameter, and fraction fresh and dry weights. The composition rates classified with hypocotyl length showed big differences between 4 cultivars in class A and C; Class A, in which cv. Sowonkong and Junjery had higher rates than cv. Eunhakong and Pungsannamulkong, was decreased with BA concentration of higher than 4 ppm, but class C showed the reverse result to class A. Any lateral root was not formed in higher than 4 ppm BA solutions although fewer in cv. Pungsannamulkong and Junjery than in the two other cultivars. Hypocotyl and root lengths were decreased with increased BA concentrations, and in all BA concentration, hypocotyl length of cv. Junjery was the longest. Hypocotyl diameter, hypocotyl and total fresh weights were thicker and heavier in 2 to 4 ppm BA concentrations than in the other ones, although in case of BA application, cv, Eunhakong and Sowonkong were heavier. The H/R ratios were increased with increased BA concentration. The ratios of nearly all the selling products ranged from 1 to 2 ppm when compared to them from our experiment.

Characteristics of Coal Methanation in a Hydrogasifier (수소가스화기에서 석탄의 메탄화 반응 특성)

  • To investigate the characteristics of substitute natural gas (SNG) production from direct coal methanation, the continuous lab-scale entrained flow hydrogasifier (I.D. : 0.025 m, Height : 1.0 m) was used in this experiment. The hydrogasification system consisted of high pressure gas handling system, high pressure coal feeder, entrained flow hydrogasifier, and unreacted char separator. The experiment was performed at the various conditions of reaction temperature ($600{\sim}800^{\circ}C$), $H_2$/coal ratio (0.2~0.4), and coal feed rate (0.8~2.5 g/min). Although it was shown that carbon conversion was different trends with coals from the methanation results for 6 sample coals, the carbon conversion increased with increasing reaction temperature. And it increased with increasing H2/coal ratio, whereas the concentration of CH4 decreased. Also. the carbon conversion increased with the carbon content of coal sample and had a maximum value at volatile matter content of 35 wt%.

Effects of Dietary Supplementation of Vitamin C and Sea Buckthorn on the Performance and Meat Quality in Old Laying Hens (비타민 C와 비타민나무 부산물 첨가가 산란 성계의 생산성 및 계육 품질에 미치는 영향)

  • The objective of this study was to investigate the effect of dietary supplementation of vitamin C and Sea buckthorn on the performance, blood biochemistry and meat quality in old laying hens. A total 200 Hy-Line Brown laying hens (101 weeks old) were randomly allotted to 1 of 5 dietary treatments : (1) Basal : basal diet, (2) Vit. C : basal diet + 0.1% vitamin C, (3) SB 0.1 : basal diet + 0.1% Sea buckthorn, (4) SB 0.5 : basal diet + 0.5% Sea buckthorn, and (5) SB 1.0 : basal diet + 1.0% Sea buckthorn. Each treatment was replicated 4 times with 10 birds units were arranged according to randomized block design. Feeding trial lasted 4 weeks under 16L:8D lighting regimen. The diets were fed to hens on an ad libitum basis for 4 weeks. Result indicated that during feeding trial of the experiment, hen-day egg production and feed conversion ratio were not significantly influenced by treatments. However, feed intake was significantly (P<0.05) higher in Vit. C and SB treated groups than the basal during 1 wks and 3 wks. Egg weight was significantly (P<0.05) higher in basal and Vit. C than the SB 1.0 treatment. There were no differences in carcass yield during feeding trials. However, partial ratio (breast and neck) was significantly (P<0.05) higher in SB 0.5 than other treatment. There were no differences in the level of leukocytes and erythrocytes. There were no significant differences on proximate analysis (DM, crude protein, crude fat, and crude ash), meat color, water holding capacity, cooking loss, and fatty acids concentrations. In conclusion, dietary supplementation of vitamin C and Sea buckthorn to the diet of old laying hens might be a potential ingredient for increasing partial weight (breast) in old laying hens.

Purification of Oat ${\beta}-Glucan$ by ${\alpha}-Amlyase$ Treatment and Characterization of Its Physicochemical Properties (귀리 ${\beta}-glucan$${\alpha}-amlyase$를 이용한 정제와 이화학적 특성)

  • The effects of purification using ${\alpha}-amlyase$ (Termamyl 120L) on physicochemical properties of ${\beta}-glucan$ from oat bran were studied. Four fractions were selected as fraction A ($55^{\circ}C$, 15%, pH 6), fraction B ($45^{\circ}C$, 15%, pH 6), fraction C ($50^{\circ}C$, 0%, pH 7), and fraction D ($50^{\circ}C$, 10%, pH 5) from the result of physiological test, and three consecutive subfractions were obtained by repeated ${\alpha}-amlyase$ treatments on the each fractions. The contents of ${\beta}-glucan$, protein, and ash after purification were in 81.4-88.2%, 4.1-6.3% and 2.6-6.2%, respectively. The apparent viscosities of purified ${\beta}-glucan$ aqueous solutions were similar to those of hydroxy methyl cellulose. Glucose was a major monosaccharide of ${\beta}-glucan$ extracts, and xylose and arabinose were also detected as minor constituents on TLC. The average molecular weight ranged $2.0{\times}10^6-5.1{\times}10^6$ and was decreased after purification. From the result of the differential scanning calorimetry, the melting point ranged $130-140^{\circ}C$ with purification step and thermal transition enthalpy was increased. The ratio of ${\beta}-(1{\rightarrow}3)\;to\;{\beta}-(1{\rightarrow}4) $ linkages were 1:2.22-1:2.52, and increased up to 1:5.50 after purification.
