TNF$\beta$ Induces Cytotoxicity of Antibody-Activated CD$4^+$T-lymphocytes Against Herpes Virus-Infected Target Cells

  • We have extended our previous work that cross-linking CD4 molecules using specific MAb induced antigen nonspecific, MHC unrestricted killing of virally infected target cells by CD$4^+$We have extended our previous work that cross-linking CD$4^+$ molecules using specific MAb induced antigen nonspecific, MHC unrestricted killing of virally infected target cells by CD$4^+$ T cells. The killing activity of antibody activated CD$4^+$T cells was completely blocked by herbimycin A, a protein tyrosine kinase (PTK) inhibitor, but not by bisindolylamaleimide, a protein kinase C (PKC) inhibitor. Herbimycin A treated human or bovine peripheral blood CD$4^+$T cells lacked PTK activity and failed to kill virally infected target cells even after cross-linking of CD4 molecules. The CD$4^+$cross-linking failed to induce effector cell proliferation or the transcription of TNF${\beta}$ Upregulation of TNF${\beta}$ was induced by incubating the antibody activated effector cells with BHV-1 infected D17 target cells for 10 h. Anti-TNF${\beta}$ antibody partially abolished (13-44%) the direct effector cell-mediated antiviral cytotoxicity. However, this antibody neutralized 70 to 100% of antiviral activity of effector and target cell culture supernatants against BHV-1 infected D17 cells. The inhibition level of the antiviral activity by the antibody was dependent on the effector and target cell ratio. These results support the hypothesis that increased p$56^ICK enzyme activity in effector cells transduces a signal critical for effector cell recognition of viral glycoproteins expressed on the target cells. Following target cell recognition, lytic cytokines known to participate in target cell killing were produced. A better understanding of the killing activity displayed by CD$4^+$T lymphocytes following surface receptor cross-linking will provide insight into the mechanisms of cytotoxic activity directed toward virally-infected cells.T cells. Plasma Surface Modification of Graphene and Combination with Bacteria Cellulose (Graphene의 플라즈마 표면 개질과 박테리아 셀룰로오스와의 결합성 검토)

  • The study was focused to evaluate the possibility for combination membrane of bacterial cellulose (BC) and graphene with high electrical properties. BC with natural polymer matrix was known to have strong physical strength. For the combination of graphene with BC, the surface of graphene was modified with oxygen plasma by changing strength and time of radio waves in room temperature. Water contact angle of modified graphene grew smaller from $130^{\circ}$ to $12^{\circ}$. XPS analysis showed that oxygen content after treatment increased from 2.99 to 10.98%. Damage degree of graphene was examined from $I_D/I_G$ ratio of Raman analysis. $I_D/I_G$ ratio of non-treated graphene (NTG) was 0.11, and 0.36 to 0.43 in plasma treated graphene (PTG), increasing structural defects of PTG. XRD analysis of PTG membrane with BC was $2{\theta}$ same to BC only, indicating chemically combined membrane. In FT-IR analysis, 1,000 to 1,300 $cm^{-1}$ (C=O) peak indicating oxygen radicals in PTG membrane had formed was larger than NTG membrane. The results suggest that BC as an alternation of plastic material for graphene combination has a possibility in some degree on the part like transparent conductive films.

Effect of Forage Feeding Level on the Milk Production Characteristics of Holstein Lactating Cows (조사료 급여 수준이 Holstein 착유우의 산유 특성에 미치는 영향)

  • This study was performed on two groups (10 cows) for primiparous Holstein lactating cows (av. 98 days in milk : DIM) which were divided into low forage diet (LF) and high forage diet (HF) groups based on forage : concentrate ratio (F : C ratio). The F : C ratios of LF and HF groups were 37:63 and 62:38, respectively. Concentrate intake was significantly higher in the LF group than the HF group whereas the HF group showed higher forage intake (12.9 kg) compared to the LF group (7.4 kg) (p<0.05). No significant difference was observed in total feed intake between the HF (20.9 kg) and LF (19.4 kg) group (p>0.05), but the HF group tended to be higher. CP, TDN and NEL intake showed no significant difference between the two groups (p>0.05). Though, there was no significant difference on actual milk between the two groups (26 vs. 23.9 kg/d, p>0.05), the LF group tended to be higher. 4% FCMs of LF and HF groups were 22.8 and 22.3 kg/d, respectively, and showed no significant difference (p>0.05). The HF group was higher in fat content and lower in MUN. C14:0, C16:0 and C16:1n7 of milk fatty acid were significantly higher in the HF group (p<0.05), but there were no differences in other milk fatty acids between the two groups. As a result, increasing high quality forage such as BIRG silage and hay in the diet will not only fulfill nutrient requirements but also reduce milk production.

Pharmacological Activity of Chaga Mushroom on Extraction Conditions and Immunostimulating Polysaccharide (추출조건에 따른 차가버섯 생리활성 및 면역활성 다당)

  • To investigate the pharmacological activity of chaga mushroom (Inonotus obliquus) on extraction conditions, chaga was extracted using water (reflux at $50^{\circ}C$, decoction over $90^{\circ}C$, pressure at $121^{\circ}C$) or ethanol (reflux at 50, 70, or $90^{\circ}C$). When water extract was further fractionated into crude polysaccharide (IO-CP), yields of IO-CP (4.8~16.8%) were higher than those of ethanolic extracts (IO-E, 1.9~2.7%) at increased temperature. For antioxidant activity, crude polysaccharide (IO-CP-121) obtained by pressurized extraction showed the highest polyphenolic and flavonoid contents (35.10 mg TAE/g and 18.48 mg QE/g, respectively) as well as DPPH and ABTS free radical scavenging activities (26.08 and 27.99 mg AEAC/100 mg, respectively). Meanwhile, IO-CP-D (decoction) and IO-CP-50 (reflux) had more potent mitogenic effects (2.10- and 1.95-fold of saline control at 100 ${\mu}g/mL$) as well as intestinal immune system modulating activities (6.30- and 5.74-fold) compared to IO-CP-121, whereas ethanolic extracts showed no activity. Although no IO-CP showed cytotoxicity against RAW 264.7 cells at 0.1 mg/mL, IO-CP-121 significantly inhibited TNF-${\alpha}$ and NO production as pro-inflammatory factors in LPS-stimulated RAW 264.7 cells (29.2 and 63.5%, respectively). Ethanolic extracts also showed no cytotoxicity at 0.1 mg/mL, whereas inhibition of TNF-${\alpha}$ and NO production was significantly low compared to that of IO-CP-121. In addition, active IO-CP-D was further fractionated into an unadsorbed (IO-CP-I) and seven adsorbed fractions (IO-CP-II~VIII) by DEAE-Sepharose CL-6B column chromatography in order to isolate immunostimulating polysaccharide. IO-CP-II showed the most potent mitogenic effect and macrophage stimulating activity (4.51- and 1.64-fold, respectively). IO-CP-II mainly contained neutral sugars (61.86%) in addition to a small amount of uronic acid (2.96%), and component sugar analysis showed that IO-CP-II consisted mainly of Glc, Gal, and Man (molar ratio of 1.00:0.55:0.31). Therefore, extraction conditions affect the physiological activity of chaga, and immunostimulating polysaccharide fractionated from chaga by decoction is composed mainly of neutral sugars.

An Unequal Divider with Enhanced Physical Isolation Between Output Ports (출력포트 사이의 물리적 격리도를 향상시킨 비대칭분배기)

  • This paper presents the design and performance of an unequal divider with physical separation and electrical isolation. This divider has a series $18{\Omega}$ resistor and 0.7 pF capacitor circuit between two quarter-wave transmission lines at half phase angle from input terminal. This design method was improved a physical isolation between output ports and easy connected other circuit because of unnecessary of extra transmission line. To show the validity of the unequal divider with complex isolation components, a 4:1 ratio unequal divider was designed and measured at center frequency of 2 GHz. The measured divider performances have the return loss of 17 dB, insertion loss of 1.5 dB and 7.7 dB, and isolation of 18 dB. Its performance is in good agreement with the simulated results.

Numerical Study on Soot Formation in Opposed-flow Nonpremixed Flame by Mixing Toluene (톨루엔 혼합에 따른 대향류 확산화염 내 매연 생성에 대한 수치적 연구)

  • A numerical simulation has been performed to investigate effects of toluene mixing on soot formation in pure ethylene opposed-flow nonpremixed flame. Mixture ratios of toluene were 3%, 5%, 10%, and 20%. Senkin code for 0-D simulation and oppdif code for 1-D simulation based on CHEMKIN III were utilized. 0-D results by senkin showed that concentrations of methyl radicals and benzene were increased with increasing toluene mixture ratio. This implied that the mixing of toluene in pure ethylene diffusion flame produces more PAHs and soot than those of pure ethylene flame. 1-D result of 10 % toluene reaction by oppdif code showed that production rate for H radical was a crucial factor for benzene formation. These results imply that methyl radical, benzene and H radical play a important role on soot formation in diffusion flames.

Studies on the Extraction Rate of Oil from Sardine, Sardinops melanosticta (정어리 지질의 추출속도에 관한 연구)

  • The extraction ratio of oil using solvent from the muscle of sardine, Sardinops melanosticta, was studied. The results were critically evaluated in the light of the theory of oil diffusion in a porous solid model. In addition the effect of temperature and moisture on the extraction rate was examined. Sardine muscle was prepared in a manner to meet the conditions required by the diffusion theory from Fick's law. The results of the model were well coincidenced to the theory derived from Fick's law. Diffusion constants at the direction to muscle fiber($D_1$) and at direction perpendicular to fiber($D_2=D_3$) when extracted at $45^{\circ}C$ were $8.16{\times}10^{-8}cm^2/sec\;and\;4.12{\times}10^{-8}cm^2/sec$, respectivly. The extraction rate was linearly propotional to absolute temperature(T) by eleventh power under the constant condition of moisture contents and muscle size. A comparison of the experiments with the highest($74.22\%$) and the lowest ($32.48\%$) moisture indicated that difference of $1\%$ in moisture contents caused to change the slope(K) of the extraction curve $0.53{\times}10^{-6}sec^{-1}$ approximately.

Yearly Trend of Sugar-Sweetened Beverage(SSB) Intake and Nutritional Status by SSB Intake Level in Korean Middle School Students Using the 2007~2015 Korea National Health and Nutrition Examination Survey (중학생의 가당음료 섭취량 변화 추이와 가당음료 섭취 수준에 따른 영양상태 평가: 2007~2015 국민건강영양조사를 이용하여)

  • This study examined yearly trend of sugar-sweetened beverage(SSB) intake and compared nutritional status by SSB intake level in middle school students aged 12~14 years(n=2,543) using the data from 2007~2015 Korea National Health and Nutrition Examination Survey. SSB included carbonated drinks, sports drinks, and caffeinated drinks contained added sugar. Subjects were classified into three groups by SSB intake level obtained from 24-hour recall method: SSB 1(SSB intake 0 g/d), SSB 2(0 g/d < SSB intake < 50th percentile) and SSB 3(SSB intake ≥ 50th percentile). Result of daily intake of SSB was 76.1±6.2 g/d for boys and 59.5±4.7 g/d for girls and it was increased significantly for boys(p-trend 0.0004) and girls(p-trend 0.0038) by year. The most intakes were carbonated drinks followed by fruit juices and sports drinks for boys and girls. Percentage of daily intake compared to the dietary reference was increased for energy and iron while was decreased for calcium and vitamin C toward SSB 3 group. Ratio of excess intake of energy/fat was increased significantly for boys(p=0.0091) and girls(p<0.0001) toward SSB 3 group. Ratio of calcium deficiency was 86.8~94.9% for boys and girls and it was very high. Therefore, it should be emphasized to reduce SSB intake and drink plain water without added sugar, etc. and milk as a source of calcium for improving nutritional status of middle school students through dietary education and social support.

Quality characteristics and sensory evaluation of Fuji apple based on commodity price (상품 가격에 따른 사과의 품질 특성 및 관능 평가)

  • This study investigated the sensory attributes and quality characteristics of Fuji apples based on market commodity price to provide data for quality index of Fuji apples. Samples were purchased from the Garak market (Seoul Agro-Fisheries & Food Corporation) and divided into four groups depending on the price such as group A, B, C, D. There were no significant differences in their volume and weight among groups. In the soluble solid content and total free sugar, A and B group (high price) showed higher content than those of C and D (low price) group. And also, the A group and B, C, D group showed 386.29 mg% and 320.09~359.28 mg% in the total organic acid content, respectively. As an sensory evaluation results, A group and B group were evaluated higher score than those of C and D group in the uniformity of red color and glossiness of skin and unique apple sensory attributes using quantitative descriptive analysis. Consumer test showed similar to quantitative descriptive analysis results in the various sensory attributes. In the analysis results between quality characteristics and sensory attributes of Fuji apples, total acceptability was correlated positively with titratable acidity (r=0.58), soluble solid (r=0.89), soluble solid content/titratable acidity ratio (r=0.42), total free sugar (r=0.36) and total organic acid (r=0.38). Based on principal component analysis of apple's quality characteristics, apples were primary separated along the first principal component (pH, acidity, soluble solid content, total free sugar, organic acid), which accounted for 66.01% of total variance. In addition, principal component analysis of sensory evaluation revealed a total variance for the quantitative descriptive of 55. 65% and a total variance for the consumer test of 55.84%.

Effects on the Thermal Changes of Hap-Kok(LI4) and Chun-Choo(ST25) Following Acupuncture on Hap-Kok(LI4) in Man (합곡(合谷)(LI4) 자침(刺鍼)이 합곡(合谷)(LI4)과 천추(天樞)(ST25) 영역(領域) 온도변화(溫度變化)에 미치는 영향(影響))

  • This study was performed from March to September 1997 on 95 healthy students to observe the effects of acupuncture at Hap-kok (LI4) according to the meridian and qi-xue(氣血) phenomenon of oriental medicine's theory. Skin temperature on the Hap-kok (LI4) and Chun-Choo (ST25) were measured by D. 1. T. I. (Digital Infrared Thermograph Imaging) before acupuncture stimulation and 1min, 10 min after acupuncture stimulation. 1. In healthy students, the left and right mean temperature of LI4 and ST25 was $29.04\;^{\circ}C,\;29.12\;^{\cir}C\;and\;30.29\;^{\circ}C,\;30.22\;^{\circ}C$ respectively. 2. In control group, the time dependent changes for 10 min of LI4 and ST25 were $-0.32\;^{\circ}C,\;-0.36\;^{\circ}C\;and\;-0.5\;^{\circ}C,\;-0.46\;^{\circ}C$ respectively, however, the thermal differences of both sides LI4 and ST25 were not changed for 10 min. 3. In acupuncture stimulation of both sides LI4, the time dependent changes of ST25 were $-0.13\;^{\circ}C,\;-0.06\;^{\circ}C$, and the thermal differences of both sides ST25 were reduced, but not changed significantly. In acupuncture stimulation of right side LI4, the time dependent changes of LI4 were $-0.1\;^{\circ}C,\;-0.32\;^{\circ}C$ respectively, and the thermal differences of both sides LI4 were increased more than control, but not significantly. Also, that of ST25 were changed by $-0.69\;^{\circ}C,\;-0.63\;^{\circ}C$ respectively, but not significantly. 4. In acupuncture stimulation group, it was classified following the thermal differences of both sides LI4 and ST25, and the effects of acupuncture were observed by changes of classification. The acupuncture of both sides LI4 results in temperature of the left side ST25 to be high after acupuncture. The acupuncture of right side LI4 results in increased ratio of the left side higher than right LI4. The above results indicates that D. I. T. I. was a useful method to observe follow-up the effects and changes by acupuncture stimulation on objective evaluation of phenomenon for the meridian system. Thus, acupuncture on LI4 affects to thermal changes of ST25 and LI4, but exact examination of thermal changes on ST25 will have to be.
